Impact of cancer cachexia on respiratory muscle function and the therapeutic potential of exercise.

Cancer cachexia is defined as a multi-factorial syndrome characterised by an ongoing loss of skeletal muscle mass and progressive functional impairment, estimated to affect 50-80% of patients and responsible for 20% of cancer deaths. Elevations in the morbidity and mortality rates of cachectic cancer patients has been linked to respiratory failure due to atrophy and dysfunction of the ventilatory muscles. Despite this, there is a distinct scarcity of research investigating the structural and functional condition of the respiratory musculature in cancer, with the majority of studies exclusively

Chronic progressive external ophthalmoplegia. I. A quantitative histochemical study of skeletal muscles.

This study quantitates the major morphological and cytochemical changes in limb muscle biopsies from 37 patients with the syndrome of chronic progressive external ophthalmoplegia (CPEO). The aim was to assess the value of limb muscle biopsy in the diagnosis of this syndrome; to define the myopathological changes and to determine whether there were any specific clinico-pathological correlations. Patients were divided into three clinical groups--11 patients with CPEO with facial and/or limb muscle weakness; 10 with CPEO with facial and/or limb muscle weakness and a positive family history; 16 wi

[Change in the ultrastructure of the muscles in thyrotoxic periodic paralysis].

The article presents data on the clinico-morphological study of three patients suffering from thyrotoxicosis with attacks of periodic paralysis. The muscles obtained by biopsy in two patients at the moment of attack and in one patient--in the absence of attack were investigated by the electron microscopy method. The data obtained make it possible to assume that in the thyrotoxic periodic paralysis the action of the thyroid hormones causes not only impairment of the mineral metabolism, but brings about changes in the structure of the membranes of the sarcolemma and T-system, which leads to dist

Diet-induced obesity does not exacerbate cachexia in male mice bearing Lewis-lung carcinoma tumors.

Cachexia is a muscle-wasting syndrome commonly observed in patients with cancer, which can significantly worsen clinical outcomes. Because of a global rise in obesity, the coexistence of cachexia in obese individuals poses unique challenges, with the impact of excessive adiposity on cachexia severity and underlying pathophysiology not well defined. Understanding the interplay between cachexia and obesity is crucial for improving diagnosis and treatment strategies for these patients; therefore, the present study examined differences in cachexia between lean and obese mice bearing Lewis lung car

Variation in retinal changes and muscle pathology in mitochondriopathies.

A variety of retinal changes that have so far not been classified under mitochondriopathies can now be included in this group, since muscle biopsy has identified ragged-red fibers with pathological mitochondriae. The ophthalmological findings in our relatively large group of 12 patients with mitochondrial myopathies are compared with the spectrum of myopathic findings. No obvious correlation exists between the severity of the pathological retinal changes and the characteristic of the myopathic alterations. In addition to fine pigmentation and depigmentation, severe dystrophic changes of the re

Metformin Reverses the Enhanced Myocardial SR/ER-Mitochondria Interaction and Impaired Complex I-Driven Respiration in Dystrophin-Deficient Mice.

Besides skeletal muscle dysfunction, Duchenne muscular dystrophy (DMD) exhibits a progressive cardiomyopathy characterized by an impaired calcium (Ca2+) homeostasis and a mitochondrial dysfunction. Here we aimed to determine whether sarco-endoplasmic reticulum (SR/ER)-mitochondria interactions and mitochondrial function were impaired in dystrophic heart at the early stage of the pathology. For this purpose, ventricular cardiomyocytes and mitochondria were isolated from 3-month-old dystrophin-deficient mice (mdx mice). The number of contacts points between the SR/ER Ca2+ release channels (IP3R1

Targeted inhibition of calpain in mitochondria alleviates oxidative stress-induced myocardial injury.

The protein levels and activities of calpain-1 and calpain-2 are increased in cardiac mitochondria under pathological conditions including ischemia, diabetes, and sepsis, and transgenic overexpression of mitochondrial-targeted calpain-1 induces dilated heart failure, which underscores an important role of increased calpain in mitochondria in mediating myocardial injury. However, it remains to be determined whether selective inhibition of calpain in mitochondria protects the heart under pathological conditions. In this study, we generated transgenic mice overexpressing mitochondrial-targeted ca

The Effects of Two Different Exercise Programs on the Ultrastructural Features of the Sciatic Nerve and Soleus Muscle After Sciatic Crush.

Peripheral nerve injuries constitute a significant medical problem and the recovery is critically dependent on post-injury treatment. In this study, following sciatic nerve crush, we investigated the effects of a 4-week endurance training program (ET) and balance and coordination training program (BCT) on the ultrastructural features of the sciatic nerve and soleus muscle. The animals were randomly divided into Sham, non-trained (NT), ET, and BCT groups each of which included three animals. Ultra-thin cross and longitudinal sections (70-85 nm) were digitized and analyzed comparatively. The ele

[Characteristics of energy metabolism in different regions of the heart, liver and kidneys in myocardial infarct after administration of prednisolone].

The effect of therapeutic doses of Prednisolone (1 mg/kg) on the indices of energy metabolism in different regions of the heart, liver and kidneys was studied in 70 dogs within the initial 10 days after the ligation of the left coronary artery. The administration of such doses of Prednisolone was found to increase the respiration of the mytochondria and especially the binding of the inorganic phosphate in the zone of the infarction, to switch over from the poorly effective oxidation in the nondamaged area of the left ventricle to the one firmly conjugated with phosphorylation, to level the bal

Multiple deletions in mitochondrial DNA in a patient with progressive external ophthalmoplegia, leukoencephalopathy and hypogonadism.

Progressive external ophthalmoplegia (PEO) is one of a number of major types of mitochondrial disorders. Most sporadic PEO patients have a heteroplasmic large deletion of mitochondrial DNA (mtDNA) in the mitochondria in skeletal muscles. We herein analyzed mtDNA deletions using sub-cloning and Sanger sequencing of PCR products in a 31-year-old Japanese man with multiple symptoms, including PEO, muscle weakness, hearing loss, leukoencephalopathy and hypogonadism. A large number of multiple deletions was detected, as well as four kinds of deletion breakpoints identified in different locations, i

Effects of acute swimming exercise on muscle and erythrocyte malondialdehyde, serum myoglobin, and plasma ascorbic acid concentrations.

Acute exercise may induce free-radical production in mitochondria during basal metabolism of aerobic cells. Ascorbic acid is a strong antioxidant agent, whereas myoglobin is known to act as an oxygen reservoir. The purpose of this study was to investigate the effects of acute exercise on malondialdehyde levels in gastrocnemius, vastus medialis, and triceps brachi muscles and erythrocytes. In addition, we investigated the ascorbic acid levels and serum myoglobin concentrations in rats, following acute swimming exercise. We found that the levels of muscle malondialdehyde and serum myoglobin incr

[Electron microscope study of skeletal muscle fibers during physical exercise in high-altitude hypoxia].

Experiments on rats exposed to intense exercise and high-altitude hypoxia have shown the reduction in physical efficiency and in the training effect of muscle loads as compared to the same characteristics examined under the conditions of plain. Electron microscopy of rat skeletal muscle fibers has discovered significant changes in their ultrastructure. Morphometric analysis of the same changes has shown a reduction in the relative volume of mitochondria, an increase in the relative volume of sarcoplasmic reticulum an abrupt fall of the glycogen reserves in the early stages of experiment with a

Maternal high-fat diet aggravates fructose-induced mitochondrial damage in skeletal muscles and causes differentiated adaptive responses on lipid metabolism in adult male offspring.

Maternal high-fat diet (HFD) is associated with metabolic disturbances in the offspring. Fructose is a highly consumed lipogenic sugar; however, it is unknown whether skeletal muscle of maternal HFD offspring respond differentially to a fructose overload. Female Wistar rats received standard diet (STD: 9% fat) or isocaloric high-fat diet (HFD: 29% fat) during 8 weeks before mating until weaning. After weaning, male offspring received STD and, from 120 to 150 days-old, they drank water or 15% fructose in water (STD-F and HFD-F). At 150th day, we collected the oxidative soleus and glycolytic ext

Association of skeletal muscle oxidative capacity with muscle function, sarcopenia-related exercise performance, and intramuscular adipose tissue in older adults.

Muscle function and exercise performance measures, such as muscle endurance capacity, maximal strength, chair stand score, gait speed, and Timed Up and Go score, are evaluated to diagnose sarcopenia and frailty in older individuals. Furthermore, intramuscular adipose tissue (IntraMAT) content increases with age. Skeletal muscle oxidative capacity determines muscle metabolism and maintains muscle performance. This study aimed to investigate the association of skeletal muscle oxidative capacity with muscle function, exercise performance, and IntraMAT content in older individuals. Thirteen older

Maturational arrest of fetal muscle in neonatal myotonic dystrophy. A pathologic study of four cases.

Skeletal muscles from four infants with a severe neonatal form of myotonic muscular dystrophy showed histopathologic features of immaturity. Three of the infants died in the neonatal period and were studied at autopsy; one of these and the still-living infant had a gastrocnemius muscle biopsy. The most severely involved muscles were those associated with arthrogrypotic joints regardless of function as flexors or extensors. Pharyngeal muscles and the diaphragm were also severely involved. Immature features included irregularly distributed small, round muscles fibers with large vesicular interna

[Disorder of swallowing muscles in iron deficient rabbits].

Plummer-Vinson syndrome is a clinical entity associated with dysphagia, sideropenic anemia and atrophic glossitis. Using rabbits with iron deficiency anemia, the author investigated the cause of dysphagia in this syndrome. Iron deficient animals were produced by bolus feeding without iron and intramuscular injection of an iron-chelating agent. The fibers of swallowing muscles (the thyro-pharyngeal, cricopharyngeal and cervicoesophageal muscles) were classified into three types (Type 1, 2A and 2B fibers) by actomyosin ATPase staining. No significant difference between the muscles of sideropenic

Effects of rotator cuff ruptures on the cellular and intracellular composition of the human supraspinatus muscle.

Ruptures of the rotator cuff tendons of the human shoulder are a common incidence and lead to functional impairment of the four muscles connected to the cuff, entailing profound changes of their cellular tissue composition. Most importantly, such tendon tears lead to atrophy, fatty degeneration and fibrosis of the corresponding muscles. The muscle most commonly affected with such changes is the M. supraspinatus. The present study uses biopsy samples from the supraspinatus muscle of 12 elderly patients and 6 controls to examine the rupture-induced muscle change at both the cellular and the intr

[Electron microscopic study of the myocardium in cyanide poisoning].

Effect of the KCN poisoning on the cardiac muscle was studied by the aid of electron microscopic cytochemical methods. The early lesions revealed were: alterations in the region of the Z-lines, "septum formation" in the mitochondria, crystolysis, enlargment of the sarcotubules. Later destruction of the thick and thin filaments alteration of the intercalar discs were observed. Activity of the succinate-dehydrogenase and cytochromoxydase decreased. Authors emphasize the non-specific character of the alterations described.
