Ultrasound-responsive hydrogel with ROS scavenging and NO controllable release capabilities for accelerated healing of diabetic wounds.

Diabetic wounds represent a common and challenging complication of diabetes mellitus, characterized by a complex pathological microenvironment that includes excessive reactive oxygen species (ROS), chronic inflammation, hypoxia, and impaired nitric oxide (NO) synthesis. These factors collectively contribute to delayed wound healing, increased infection risk, and potential progression to chronic non-healing ulcers. Herein, we constructed a multifunctional injectable hydrogel (Prussian blue nanozyme and S-nitrosothiol-loaded hydrogel, PBE&SNO@HG) co-loaded with multi-enzyme-mimicking Prussian bl

Integrated cascade catalysis of AuPtCu nanozymes and glycolysis inhibition for synergistic breast cancer therapy via metabolism regulation.

Nanozymes have emerged as powerful therapeutic agents due to their robust catalytic performance, but their efficacy is often constrained by the complex tumor microenvironment (TME) and the unique metabolic pathway of cancer cells. To overcome this circumstance, a multifunctional nanoplatform (G5.NHAc-PG@APC) was developed to integrate targeted triple-enzyme cascade catalysis with responsive glycolytic inhibition. By using phenylboronic acid (PBA)-modified G5 PAMAM dendrimers as targeted nanocarriers, trimetallic AuPtCu nanozymes were encapsulated, followed by conjugating the glycolytic inhibit

pH-responsive folate-targeted catalase conjugate for enhanced cellular uptake and oxidative stress protection in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) remains one of the most aggressive breast cancer subtypes, with limited targeted treatment options. This study investigates the efficacy of folic acid (FOL) as a targeting molecule for TNBC. It also examines whether the cellular uptake of FOL is influenced by acidic pH, a major feature of tumor tissue. A multifunctional FITC-Catalase-Folate (FITC-CAT-FOL) conjugate was developed and optimized to combine fluorescence tracking, enzymatic activity, and receptor targeting. Two conjugation strategies were compared to evaluate how reaction order affects labeling

Cyclodextrins: Synthesis, Pharmaceutical Applications, Drug Delivery Systems and Safety Perspectives.

Cyclodextrins (CDs) are cyclic oligosaccharides composed of α-(1,4)-linked glucopyranose units that have emerged as multifunctional and versatile pharmaceutical excipients. One of the major challenges in modern drug development is that nearly half of newly discovered drug molecules exhibit poor aqueous solubility, which adversely affects formulation development, bioavailability, and therapeutic efficacy. CDs address this limitation by forming non-covalent inclusion and non-inclusion complexes, thereby enhancing drug solubility, stability, dissolution rate, and overall biopharmaceutical perform

Preparation and Evaluation of the Synergistic Benefits of a Glycoside-Pyrone-Based Multifunctional System as a Possible Regulator for Melanogenesis.

The skin is constantly exposed to external factors throughout a person's life, ultraviolet (UV) radiation being one of the most harmful. The primary defence against UV-induced damage is skin pigmentation, which is achieved through the synthesis of melanin. However, overproduction of melanin can lead to skin disorders such as pigment spots, melasma, and even melanoma. Therefore, the present study aimed to obtain a new multifunctional bioactive system (MBS) starting from a supramolecular co-assembled gel (SG) based on amino acids and short peptides enhanced with a glycoside-pyrone-based complex

Molecular Pathogenesis of and Regenerative Strategies for Osteonecrosis of the Femoral Head.

Osteonecrosis of the femoral head (ONFH) is a progressive, multifactorial bone disease characterized by ischemia-induced osteocyte death, microenvironmental imbalance, and failed tissue regeneration. According to recent advances in pathophysiological understanding, vascular injury, oxidative stress, and inflammatory storms form a pathogenic cascade leading to osteogenic dysfunction, adipogenic lineage drift of mesenchymal stem cells (MSCs), and epigenetic alterations that exacerbate bone degeneration. Despite improvements in early detection, traditional interventions-including bisphosphonates,

Sequential clearance of neutrophil extracellular traps for precision therapy of sepsis.

Sepsis, a life-threatening syndrome driven by dysregulated host response to infection, is critically exacerbated by the uncontrolled formation of neutrophil extracellular traps (NETs), which amplify inflammation, promote microthrombosis, and precipitate multiple organ dysfunction. However, current therapeutic approaches remain inadequate in effectively targeting and modulating NETs, leaving a significant clinical gap in managing sepsis progression. Herein, we propose a "sequential NETs-clearance" strategy to precisely mitigate NET-associated pathologies by simultaneously inhibiting NET overpro

Reprogramming the Diabetic Wound Microenvironment by Enzyme Cascade-Driven, ROS-Responsive Microspheres.

Diabetes represents a major global health burden, and diabetic wounds remain particularly challenging due to delayed healing and high infection risk. A dysregulated wound microenvironment, characterized by hyperglycemia, excessive reactive oxygen species (ROS), and persistent hypoxia, critically impairs angiogenesis and tissue regeneration. Herein, we report a multifunctional microsphere system integrating ROS-responsive curcumin-loaded nanoparticles, glucose oxidase (GOx), and catalase (CAT) to simultaneously regulate glucose, oxygen, and oxidative stress in diabetic wounds. HERC5-mediated ISGylation of SARS-CoV-2 nsp8 facilitates its degradation and inhibits viral replication.

Severe acute respiratory syndrome coronavirus 2 non-structural protein 8 (SARS-CoV-2 nsp8) is a multifunctional protein essential for viral replication and immune evasion. However, the host factors that regulate nsp8 stability and function remain unclear. In this study, we identify HECT and RCC-like domain-containing protein 5 (HERC5) as an essential E3 ligase that regulates nsp8 stability through ISGylation, a ubiquitin-like post-translational modification that facilitates proteasome-dependent degradation. HERC5 overexpression significantly enhances nsp8 degradation in an enzymatic activity-d

Ultrasound-driven manganese oxide nanocatalyts augment multipronged catalytic tumor therapies by unleashing mutually enhanced and sustained oxidative strom and ferroptosis.

The catalytic strategies selectively eradicate tumors, but their efficacy is hindered by antioxidant defense mechanisms of tumor microenvironment (TME) and insufficient catalytic efficiency. Herein, we present a multifunctional manganese oxide nanosphere (MnOx NSs)-based antitumor platform that synergizes ultrasound (US)-triggered reactive oxygen species (ROS) generation with oxygen-independent sulfate radical (SO4•-) production to amplify oxidative damage. Under US irradiation, MnOx NSs generate hydroxyl radicals (•OH) and singlet oxygen (1O2) via sonodynamic effects, and Mn2+/Mn3+ species ac

Platinum nanozymes system with tunable enzyme activity for on-demand supply therapy of infected wounds.

The rapid healing of bacterially infected wounds remains a major clinical challenge. The pathological microenvironment-characterized by bacterial infection, reactive oxygen species (ROS) accumulation, persistent inflammation, and impaired tissue repair-severely impedes this process. In this work, we constructed a photothermally triggered, microenvironmental pH-regulating platinum nanozyme system, ACC@LPDAPt nanoparticles (NPs), leveraging the pH-dependent peroxidase-like and catalase-like activities of platinum nanozymes. This system achieves enzyme-like activity conversion by modulating the w

Sustainable membranes from industrial waste valorization for microbial electrochemical desalination systems.

Freshwater scarcity and the environmental burden of reverse osmosis (RO) reject streams demand sustainable desalination alternatives. Microbial desalination cells (MDCs), integrated with bioelectrochemical processes, have emerged as eco-friendly platforms for simultaneous wastewater treatment, ion removal, and energy recovery. However, their reliance on costly, ion-exchange membranes limits scalability. This review critically examines low-cost waste-valorized membranes fabricated from agricultural residues, polymeric wastes, ceramics, and industrial byproducts as green substitutes for conventi

N-acetyltransferases required for iron uptake and aminoglycoside resistance promote virulence lipid production in Mycobacterium marinum.

Phagosomal lysis is essential for mycobacterial infection of macrophages. Acetylation is a protein modification mediated enzymatically by N-acetyltransferases (NATs) that impacts bacterial pathogenesis and physiology. To identify NATs required for lytic activity, we leveraged Mycobacterium marinum, a nontubercular pathogen and a model for Mycobacterium tuberculosis. M. marinum hemolysis is a proxy for phagolytic activity. We generated M. marinum strains with deletions in conserved NAT genes and screened for hemolytic activity. Several conserved lysine acetyltransferases (KATs) contributed to h
