Wine by-products, in particular grape pomace, can be an important source of polyphenols and dietary fibers and are increasingly being used as a starting material in the industrial production of plant food supplements, such as other matrices containing biomolecules, with antioxidant properties. The risk associated with the consumption of these products was recently analyzed through a study of potential genotoxic and carcinogenic compounds that can be found in the marketed products. In particular, occurrence data about contamination with the mycotoxin ochratoxin A were also reported. This short

Ochratoxin A (OTA) is a mycotoxin naturally occurring in different foods. OTA is arguably a risk factor for Balkan endemic nephropathy (BEN). The aims of this study are to (1) test the OTA-BEN association in BEN-groups and controls and (2) determine whether urine β2-microglobulin, a marker of impaired ability of the kidneys to re-absorb, is related to OTA. BEN patients had significantly higher OTA serum levels. Within the offspring, OTA was significantly related to higher β2-microglobulin excretion. OTA (2005/2006) was related to a higher incidence of BEN after 2008, providing further evidence

Zearalenone (ZEN) is a common foodborne mycotoxin with intestinal toxicity, but its role in colitis-associated colorectal cancer (CAC) remains unclear. This study evaluated the association between ZEN exposure and CAC progression and explored the related molecular, microbial, and metabolic alterations. Network toxicology was used to predict potential ZEN-related targets and pathways associated with colorectal cancer, suggesting possible involvement of PI3K-Akt, MAPK, and apoptosis-related pathways. Subsequently, an azoxymethane/dextran sulfate sodium (AOM/DSS)-induced CAC model was established

INTRODUCTION: Zearalenone (ZEN), a mycotoxin from Fusarium species, is widely present in contaminated grains and animal products. It exerts estrogen-like effects, disrupting hormonal balance and reproductive function, partly through oxidative stress-induced DNA damage. The ATM-Chk2-p53 pathway is a key mediator of the DNA damage response. Melatonin (MT), a natural antioxidant, supports ovarian function by regulating hormone secretion and reducing oxidative stress. This study explores whether MT alleviates ZEN-induced ovarian and granulosa cell damage via the ATM-Chk2-p53 pathway.

The metabolic capacity of swine caecum-derived Lactobacillus spp. to biotransform mycotoxins presents promising potential as a host-probiotic strategy to improve pig health and support host-targeted probiotic research. In the present study, Lactobacillus spp. isolated from the pig caecum were examined for their ability to detoxify fumonisin B1 (FB1) in vitro. Three experimental groups were established (i) Control 1 (C1: buffer + caecal chyme), (ii) Control 2 (C2: buffer + FB1), (iii) Experimental group (E: buffer + caecal chyme + FB1), each with 12 replicates per group (4 replicates per time p…

Mycotoxin contamination in foods is a major risk factor for human and animal health due to its prevalence in cereals and their by-products. Deoxynivalenol (DON), mainly produced by Fusarium genera, is the most common mycotoxin detected in cereal products. Deoxynivalenol disrupts intestinal barrier function and decreases protein levels of tight junction proteins (TJP). However, the overall mechanism by which DON regulates specific TJP turnover and epithelial cell integrity remains unclear. Herein, we show that DON (2 μM) decreases the protein stability and accelerates the degradation of TJP in

Deoxynivalenol (DON) is a prevalent Fusarium mycotoxin, occurs predominantly in the global environment, especially in cereals, animal feed and food commodities. The widespread contamination causes a serious risk to human and animal health. DON usually impairs weight gain, which is presumably from its capacity to reduce feed intake by interfering with intestinal motility. To clarify the role of smooth muscle cells (SMCs) contractility in intestinal motility and growth inhibition caused by DON, twelve weaned piglets were firstly divided into two groups to feed control or Fusarium mycotoxin-conta

Contamination of the world's food supply and animal feed with mycotoxins is a growing concern as global temperatures rise and promote the growth of fungus. Zearalenone (ZEN), an estrogenic mycotoxin produced by Fusarium fungi, is a common contaminant of cereal grains and has also been detected at lower levels in meat, milk, and spices. ZEN's synthetic derivative, zeranol, is used as a growth promoter in United States (US) and Canadian beef production. Experimental research suggests that ZEN and zeranol disrupt the endocrine and reproductive systems, leading to infertility, polycystic ovarian s…

The immunotoxicity of zearalenone (ZEA) and deoxynivalenol (DON), two of the most common environmental mycotoxins, has been well investigated. However, due to the complexity of the immune system, especially during bacterial infection, many types of immune cells are involved in invasion resistance and bacterial clearance. Of these, T helper 2 (Th2) cells, which are members of the helper T cell family, assist B cells to activate and differentiate into antibody-secreting cells, participate in humoral immune response, and, ultimately, eliminate pathogens. Thus, it is important to identify the stag…

The nearly-ubiquitous food and feed-borne mycotoxin aflatoxin B₁ (AFB₁) is carcinogenic and mutagenic, posing a food safety threat to humans and animals. One of the most susceptible animal species known and thus a good model for characterizing toxicological pathways, is the domesticated turkey (DT), a condition likely due, at least in part, to deficient hepatic AFB₁-detoxifying alpha-class glutathione S-transferases (GSTAs). Conversely, wild turkeys (Eastern wild, EW) are relatively resistant to the hepatotoxic, hepatocarcinogenic and immunosuppressive effects of AFB₁ owing to functional gene

BACKGROUND: The western corn rootworm (WCR) is one of the economically most important pests of maize. A better understanding of microbial communities associated with guts and eggs of the WCR is required in order to develop new pest control strategies, and to assess the potential role of the WCR in the dissemination of microorganisms, e.g., mycotoxin-producing fungi.
METHODOLOGY/PRINCIPAL FINDINGS: Total community (TC) DNA was extracted from maize rhizosphere, WCR eggs, and guts of larvae feeding on maize roots grown in three different soil types. Denaturing gradient gel electrophoresis (DGGE)

Fusarium moniliforme is a widespread fungal pathogen which primarily infects corn, but can also infect rice or wheat. Fusarium moniliforme produce several mycotoxins, the most prominent of which is called fumonisin B1 (FB1). Epidemiological studies have indicated that ingestion of fumonisins correlates with a higher incidence of oesophageal cancer in Africa and China. Fumonisins also cause a neurodegenerative disease in horses, induce hepatic cancer in rats, are nephrotoxic in rats, or cause pulmonary oedema in swine. Structurally, fumonisins resemble sphingolipids and can alter sphingolipid b…

Deoxynivalenol (DON) is an important mycotoxin that contaminates cereal grains and poses serious threats to livestock and poultry production and human health. This review systematically summarizes the cytotoxicity of DON in the intestinal, immune, nervous, and reproductive systems. Particular emphasis is placed on the mechanism in which the direct ribotoxic stress response (direct RSR) acts as the initial trigger, followed by an imbalance in signaling networks and secondary amplification of ROS and endoplasmic reticulum (ER) stress. Through crosstalk among the MAPK, PI3K/AKT/mTOR, NF-κB, JAK2/

Mycotoxigenic fungi, principally Aspergillus, Fusarium, and Penicillium spp. contaminate 60-80% of global feed commodities and cause multi-billion-dollar losses through animal health impairment, reduced productivity, and food chain carry-over. Despite decades of reliance on synthetic antifungals (azoles, polyenes, echinocandins), the emergence of resistant strains and growing regulatory pressure demand fundamentally different solutions. Existing reviews have catalogued plant-derived phytochemicals as promising alternatives yet have failed to provide an integrated, mechanistically grounded, and

Ustilaginoidin D is a type of bis-naphtho-γ-pyrone mycotoxin produced by Ustilaginoidea virens, the causal agent of rice false smut. Although previous studies have demonstrated the inhibitory effect of ustilaginoidin D on ATP synthesis and cancer cell growth in mice, its specific health risks remain unclear. Here, we reveal that ustilaginoidin D is highly toxic to mice with an LD50 value of 213 mg /kg·bw. Dose-dependent weight loss and liver damage were observed, accompanied by altered markers of liver cell damage, including the enzyme activities of alanine aminotransferase and aspartate amino
