Muscle fibres and cultured muscle cells express the B7.1/2-related inducible co-stimulatory molecule, ICOSL: implications for the pathogenesis of inflammatory myopathies.

Inducible co-stimulator ligand (ICOSL), a member of the B7 family of co-stimulatory molecules related to B7.1/2, regulates CD4 as well as CD … and pathological conditions. We also detected mRNA and cell surface protein expression of ICOSL on myoblasts cultured from control subjects and patients as well as in TE671 muscle rhabdomyosarcoma cells. ICOSL expression was upregulated by tumour necrosis factor-alpha (TNF-alpha), whereas interferon-gamma (IFN-gamma) had no such effect. Co-culture experime …

miRNAs as cell-intrinsic epigenetic regulators in muscle in type 2 diabetes.

miRNAs are important metabolic regulators and are altered at both the cellular and secreted levels in diseases, including type 2 diabetes (T2D). However, to what extent these alterations are in response to factors in the in vivo milieu or are cell-intrinsic remains unclear. Here we used a disease-in-a-dish model in which iPSCs from T2D patients and controls were differentiated into myoblasts (iMyos), and their cellular and secreted miRNAs were profiled. We found that iMyos from T2D donors exhibit cell-intrinsic alterations in miRNA expression and secretion in small extracellular vesicles (sEVs…

Assessing bacterial invasion of cardiac cells in culture and heart colonization in infected mice using Listeria monocytogenes.

Listeria monocytogenes is a Gram-positive facultative intracellular pathogen that is capable of causing serious invasive infections in immun … iotropism of L. monocytogenes isolates. Methods are presented for the infection of H9c2 rat cardiac myoblasts in tissue culture as well as for the determination of bacterial colonization of the hearts of infected mice. These methods are useful not only for identifying strains with the potential to colonize cardiac tissue in infected animals, but may also fa …

Development of a decellularized hypopharynx with vascular pedicle scaffold for use in reconstructing hypopharynx.

BACKGROUND: Hypopharynx reconstruction after hypopharyngectomy is still a great challenge. Perfusion decellularization is for extracellular matrix (ECM) scaffolding and had been used in organ reconstruction. Our study aimed to prepare an acellular, natural, three-dimensional biological hypopharynx with vascular pedicle scaffold as the substitute materials to reconstruct hypopharynx. RESULT: Scanning electron microscope and histology staining showed that the decellularized hypopharynx with vascular pedicle scaffold retained intact native anatomical ECM structure. Myoblasts were observed on the…

The Importance of CXCL1 in Physiology and Noncancerous Diseases of Bone, Bone Marrow, Muscle and the Nervous System.

This review describes the role of CXCL1, a chemokine crucial in inflammation as a chemoattractant for neutrophils, in physiology and in selected major non-cancer diseases. Due to the vast amount of available information, we focus on the role CXCL1 plays in the physiology of bones, bone marrow, muscle and the nervous system. For this reason, we describe its effects on hematopoietic stem cells, myoblasts, oligodendrocyte progenitors and osteoclast precursors. We also present the involvement of CXCL1 in diseases of selected tissues and organs including Alzheimer's disease, epilepsy, herpes simple…

Discovery of novel pyridazine derivatives as glucose transporter type 4 (GLUT4) translocation activators.

We report herein the synthesis and structure-activity relationships (SAR) of a series of pyridazine derivatives with the activation of glucose transporter type 4 (GLUT4) translocation. Through a cell-based phenotype screening in L6-GLUT4-myc myoblasts and functional glucose uptake assays, lead compound 1a was identified as a functional small molecule. After further derivatization, the thienopyridazine scaffold as the central ring (B-part) was revealed to have potent GLUT4 translocation activities. Consequently, we obtained promising compound 26b, which showed a significant blood glucose loweri…

[Surgery for anal incontinence: developments in the past two decades and future directions].

Studies showing the frequency of anal incontinence and its social and economic impact have driven progress in surgical treatment, from muscl … requency in 2000-2010, and development in 2012 of cellular therapy based on injection of autologous myoblasts. Progress in functional gut exploration (anorectal manometry, electrophysiological tests, endoanal ultrasonography, MRI, colonic transit time) and better knowledge of colonic and ano-rectal physiology will lead to further surgical advances.

Dual effect of transforming growth factor beta on rat thyroid cells: inhibition of thyrotropin-induced proliferation and reduction of thyroid-specific differentiation markers.

Transforming growth factor beta (TGF beta) is now known to have a number of effects other than inducing phenotypic transformation of fibrobl … in. Moreover, in analogy to the effect produced upon other differentiated cells in culture, such as myoblasts and adipocytes, TGF beta 1 modulates the expression of FRTL5-specific thyroid markers, reducing thyroglobulin biosynthesis and the ability to concentrate the iodide.

Hybrid Treatment Approach for a Rare Middle Cranial Fossa Intracranial Tumor in a Pediatric Patient: A Case Report.

Pediatric central nervous system (CNS) tumors are often classified by distinct histologic and molecular features; however, some tumors remain unclassified, resulting in diagnostic and therapeutic challenges. We report a case of a previously healthy 3-year-old female who presented with right eyelid ptosis and headache. Imaging revealed a right middle cranial fossa mass. Following surgery and histopathologic and molecular analyses, the diagnosis was a malignant neoplasm with mixed neural and myoblastic differentiation, not elsewhere classified based on the current World Health Organization (WHO)

RAGE in the pathophysiology of skeletal muscle.

Emerging evidence suggests that the signalling of the Receptor for Advanced Glycation End products (RAGE) is critical for skeletal muscle physiology controlling both the activity of muscle precursors during skeletal muscle development and the correct time of muscle regeneration after acute injury. On the other hand, the aberrant re-expression/activity of RAGE in adult skeletal muscle is a hallmark of muscle wasting that occurs in response to ageing, genetic disorders, inflammatory conditions, cancer, and metabolic alterations. In this review, we discuss the mechanisms of action and the ligands

The splicing factor hnRNPL demonstrates conserved myocardial regulation across species and is altered in heart failure.

Heart failure (HF) is highly prevalent. Mechanisms underlying HF remain incompletely understood. Splicing factors (SF), which control pre-mR … g sites suggested hnRNPL-mediated alternative splicing of tropomyosin, which was confirmed in C2C12 myoblasts. These findings identify hnRNPL as a sensor of cardiac dysfunction and suggest that disturbances of hnRNPL affect alternative splicing in HF.

The impact of Megf10/Drpr gain-of-function on muscle development in Drosophila.

Recessive mutations in multiple epidermal growth factor-like domains 10 (MEGF10) underlie a rare congenital muscle disease known as MEGF10 m … not in other tissues, and with overexpression during intermediate stages of myogenesis, but not in myoblasts. We find that tubular muscle subtypes are particularly sensitive to Megf10/Drpr overexpression. Complementary genetic analyses show that Megf10/Drpr and Notch may interact to regulate myogenesis. Our findings provide a basis for investigating MEGF10 …

Silica nanoparticles actively engage with mesenchymal stem cells in improving acute functional cardiac integration.

AIM: To assess functional effects of silica nanoparticles (SiO2-NPs) on human mesenchymal stem cell (hMSC) cardiac integration potential. M … ent also enhanced hMSC expression of Connexin-43, favoring hMSC interaction with cocultured cardiac myoblasts in an ischemia-like environment. CONCLUSION: These findings provide strong evidence that SiO2-NPs actively engage in mediating biological effects, ultimately resulting in augmented hMSC acute cardiac integration potential.

[Primary cilia of cells of cardiovascular apparatus].

The primary cilium is a mechanosensory, solitary, non-motile microtubule-based structure that in the quiescent phase of the cell cycle projects from the surface of the majority of human cells, including embryonal, stem and mesenchymal cells, fibroblasts, myoblasts, cardiomyocytes, vascular smooth muscle and endothelial cells. Primary cilia are in increased frequency also present on the surface of endothelial cells in atherosclerotic predilection sites, lipoid streaks and dots and atheromatous plaques. The primary cilium is formed from the mother centriole. Primary cilia are currently studied i…

Differentiation of an epiperiosteal sheath in avian hyperostosis induced by myeloblastosis associated virus MAV-2 (0).

If myeloblastosis associated virus MAV-2(0) is injected in chick embryos, it causes a severe, irreversible and generalized hyperostosis which subsequently follows a chronic course. An epiperiosteal sheath forms at the site of the lesion, whose development follows that of the hyperostosis. The sheath contains not only cell infiltrates, blood vessels and myoblasts, but also highly specialized cells such as Pacinian corpuscles. The structural lesions are absolutely different from these described in human, mammal or avian spontaneous or induced osteopetrosis. Our initial description of the "hypero…

Regenerating skeletal muscle cells express insulin-like growth factor I.

The expression of the trophic peptide insulin-like growth factor I (IGF-I; somatomedin C) was investigated in the regenerating soleus muscle of mice after injury by the snake venom taipoxin. No specific IGF-I immunoreactivity was observed in muscle cells during control conditions. Within 2 days after taipoxin injection, IGF-I immunoreactivity could be demonstrated in activated satellite cells. Myoblasts and myotubes expressed high IGF-I immunoreactivity. The IGF-I immunoreactivity was strictly cytoplasmatic and obviously associated with polyribosomes. No vesicular or membraneous IGF-I immunore…

Repression-free utrophin-A 5'UTR variants.

Mutation in the dystrophin gene results Duchenne Muscular Dystrophy (DMD), an X-linked fatal neuromuscular disorder. Dystrophin deficiency can be compensated by upregulation of utrophin, an autosomal homologue of dystrophin. But the expression of utrophin in adults is restricted to myotendinous and neuromuscular junctions. Therefore utrophin upregulation throughout the muscle fiber can only be achieved if we understand regulatory mechanisms behind its expression. Utrophin-A 5'UTR mediated repression of translation was reported earlier. In this article, we present evidences of two transcript va

Multi-omics and machine learning reveal a critical mediator of PRKCA in quercetin-mediated protection against Sarcopenia.

BACKGROUND: Sarcopenia, the age-associated loss of skeletal muscle mass and function, poses a growing public health challenge. Although dietary flavonoids have been proposed as protective agents due to their antioxidant and anti-inflammatory properties, their precise roles in sarcopenia prevention remain unclear. METHODS: We integrated data from the National Health and Nutrition Examination Survey (NHANES, 2017-2018) and the Food and Nutrient Database for Dietary Studies (FNDDS) to examine the relationship between flavonoid intake and sarcopenia risk. Using machine learning, LASSO, CatBoost,
