cis-Diamminedichloroplatinum (II) (cis-DDP), the antitumor drug, is cytotoxic in vitro primarily by binding to DNA and disrupting its normal functions. We have studied cis-DDP modification of DNA in nucleated peripheral blood cells (buffy coat cells) of testicular and ovarian cancer patients receiving cis-DDP chemotherapy, and of untreated controls. Yolk sac tumors were induced in fetectomized rats by displacement of the visceral yolk sac outside the uterus with or without injection of murine sarcoma virus (MSV) into the placenta. Evidence was obtained that the response of blood lymphocytes to phytohemagglutinin was reduced in some but not all rats with massive hereditary renal tumors (less than 50 mm mean diameter). Erythrocyte-antibody sensitized, complement dependent (EAC) rosette formation by sensitized spleen cells was also depressed in these animals, but plaque forming cell activity was essentially normal. Lesser tumor growth was associated with general cell-mediated and humoral immune responses which were within the range found for non-tumor-bearing syngeneic rats.

This article describes the first successful virus induction of sarcomas in adult immunosuppressed cats. The method of tumor induction, the immunosuppression regimen, and the gross, histologic, and angiographic findings in these induced sarcomas are described. The simplicity of the model, the rapid and reliable growth of the tumors, and the size of the animals permit repeated angiography. The malignant neurogenic rat cell line BT4C developed tolerance to further heat injury after continuous heating in culture at 41.0 and 42.0 degrees C for 8 and 2 hours, respectively. Survival was evaluated by colony forming ability. The proliferative capacity of the surviving cells was reduced after heating as shown by a decrease in size of the colonies.

Sprague-Dawley rats were injected weekly with either 1,2-dimethylhydrazine (DMH) or methylazoxymethanol acetate (MAM). Addition of 4 ppm selenium (sodium selenite) in the drinking water reduced the number of rats developing DMH-induced colon tumors from 13 to 6 groups of 15 each.
