Conditional expression of human bone Gla protein in osteoblasts causes skeletal abnormality in mice.

Bone Gla protein (BGP), also known as osteocalcin, is one of the most abundant γ-carboxylated noncollagenous protein in bone matrix and plays important roles in mineralization and calcium ion homeostasis. BGP is synthesized specifically in osteoblasts; however, its precise function in bone metabolism has not been fully elucidated. To investigate the in vivo function of human BGP (hBGP), we generated CAG-GFP(floxed)-hBGP transgenic mice carrying a transgene cassette composed of the promoter and a floxed GFP linked to hBGP cDNA. The mice were crossed with ColI-Cre mice, which express the Cre rec…

Effects of vitamin K on bone mass and bone metabolism.

Vitamin K is involved in blood coagulation and in bone metabolism via the carboxylation of glutamate residues in (hepatic) blood coagulation factors and (osteoblastic) bone proteins. The bioavailability of nutritional vitamin K depends on the type of food, the dietary fat content, the length of the aliphatic side chain in the K-vitamer and probably also the genetically determined polymorphism of apolipoprotein E. Although undercarboxylation of blood coagulation factors is very rare, undercarboxylated osteocalcin (bone Gla-protein) is frequently found in postmenopausal women. Supplementation of…

[Osteoporosis in Turner's syndrome].

Axial and appendicular osteoporosis are common features of Turner's syndrome. The histology of this osteoporosis is similar in some respects to postmenopausal osteoporosis. Therefore, we treated a woman with Turner's syndrome who developed a vertebral wedge fracture, despite estrogen substitution since adolescence, with bone stimulating drugs. Absence of a beneficial effect on bone biological indices or bone mineral density, could be explained by other pathogenetic factors like a direct result of the chromosomal abnormality or other hormonal deficiencies.

Hyperparathyroidism in metastases of prostatic carcinoma: a biochemical, hormonal and histomorphometric study.

Secondary hyperparathyroidism can develop as a result of bone metastases from prostatic cancer, but this has not been studied from the multiple aspects of biochemistry, hormonal status and histomorphometry. In 20 patients with stage-D prostatic cancer, a transiliac bone biopsy was performed for histomorphometric study. In all of them, molecular parathormone (PTH-M) and osteocalcin were determined by radioimmunoassay together with other parameters considered to be biological markers of bone remodelling. Of these 20 patients, only 2 (10%) had elevated PTH-M (240 +/- 20.6 pmol/l), differing signi…

Role of Inflammatory Biomarkers in Mediating Causal Effect of Life Course Body Composition on Hypertension.

BACKGROUND: The mediating role of inflammatory biomarkers in the causal relationship between body composition and hypertension remains uncle … e, FinnGen, and other consortia. The inflammatory biomarkers included leptin, insulin, adiponectin, osteocalcin, FGF23 (fibroblast growth factor 23), and PTH (parathyroid hormone). RESULTS: The observational analysis revealed that increased fat mass positively influenced diastolic blood pressure through osteocalcin, while fat-free mass had an inverse effect. …

Lower bone mass in prepubertal overweight children with prediabetes.

Childhood studies of the fat-bone relationship are conflicting, possibly reflecting the influence of metabolic abnormalities in some but not all obese children. Bone mass was compared between prepubertal overweight children with (n = 41) and without (n = 99) prediabetes. Associations of bone mass with measures of total and central adiposity, glucose intolerance, insulin sensitivity, lipid profile, systemic inflammation, and osteocalcin also were determined. In 140 overweight children aged 7 to 11 years, an oral glucose tolerance test was used to identify those with prediabetes and for determin…

High blood pressure, bone-mineral loss and insulin resistance in women.

Increasing evidence indicates that high blood pressure is associated with abnormalities in calcium metabolism. Sustained calcium loss may lead to increased bone-mineral loss in subjects with elevated blood pressure. Furthermore, recent findings indicate a possible linkage between abnormal calcium metabolism and insulin resistance. In the present study, we investigated the relationship(s) among bone-mineral density (BMD), blood pressure, calcium-related and bone metabolic parameters (plasma intact parathyroid hormone (I-PTH), 1,25-dihydroxyvitamin D [1,25(OH)2D], osteocalcin, and urinary deoxyp…
