Exploration of network-based, machine learning, MD simulations, and MM/GBSA approaches revealed luteolin from Gynura procumbens as key inhibitor of MMP9 in NSCLC.

The primary leading reason for cancer death is non-small-cell lung cancer (NSCLC) still up to date globally. Even with today's advanced technology, it has very limited successful treatment options and very low survival rates. The intricate molecular pathogenesis of NSCLC, involving the dysregulation of PI3K-AKT and EGFR biological pathways, requires novel multi-targeted approaches. A medicinal plant rich in phytochemicals, Gynura procumbens, has emerged as a potential candidate to be explored for NSCLC, named Gynura procumbens. This plant has anticancer, antioxidant, and anti-inflammatory prop

Radiomics and machine learning for characterizing CKD-associated cortical bone texture patterns in HR-pQCT tibia scans: a slice- and patient-level methodological framework.

Chronic kidney disease (CKD) is associated with alterations in cortical bone structure and composition that contribute to increased fracture risk but are incompletely captured by standard clinical imaging, including DXA. This study evaluated whether radiomics combined with machine learning can enhance detection of CKD-related cortical bone characteristics using high-resolution peripheral quantitative computed tomography (HR-pQCT). HR-pQCT images (60.7 μm isotropic resolution; 168-slice stacks acquired at 7.3% and 30% proximal to the tibial endplate) were analyzed from 72 participants (38 non-C

Detection and Pattern Recognition of Chemical Warfare Agents by MOS-Based MEMS Gas Sensor Array.

Chemical warfare agents (CWAs), including hydrogen cyanide (AC), 2-[fluoro(methyl)phosphoryl]oxypropane (GB), 3-[fluoro(methyl)phosphoryl]oxy-2,2-dimethylbutane (GD), ethyl S-(2-diisopropylaminoethyl) methylphosphonothioate (VX), and di-2-chloroethyl sulfide (HD), pose a great threat to public safety; therefore, it is important to develop sensing technology for CWAs. Herein, a sensor array consisting of 24 metal oxide semiconductor (MOS)-based MEMS sensors with good gas sensing performance, a simple device structure (0.9 mm × 0.9 mm), and low power consumption (<10 mW on average) was developed

Application of multi-omics data integration and machine learning approaches to identify epigenetic and transcriptomic differences between in vitro and in vivo produced bovine embryos.

Pregnancy rates for in vitro produced (IVP) embryos are usually lower than for embryos produced in vivo after ovarian superovulation (MOET). This is potentially due to alterations in their trophectoderm (TE), the outermost layer in physical contact with the maternal endometrium. The main objective was to apply a multi-omics data integration approach to identify both temporally differentially expressed and differentially methylated genes (DEG and DMG), between IVP and MOET embryos, that could impact TE function. To start, four and five published transcriptomic and epigenomic datasets, respectiv

Identification and validation of key PANoptosis-related genes via integrative machine learning and single-cell sequencing in AILI.

Acetaminophen (APAP) overdose is a leading cause of drug-induced liver injury and acute liver failure. PANoptosis, a recently defined form of programmed cell death, is closely linked to immune regulation; however, its role in APAP-induced liver injury (AILI) remains unclear. Here, we aimed to identify PANoptosis-related biomarkers and elucidate their functions in AILI. By integrating bulk RNA-seq, weighted gene co-expression network analysis, machine learning, and single-cell RNA-seq, we identified Cdkn1a and Pdk1 as key PANoptosis-related genes with high diagnostic potential. Immune infiltrat

Exploring diagnostic biomarkers of type 2 cardio-renal syndrome based on secreted proteins and bioinformatics analysis.

Chronic heart failure (CHF) can induce chronic kidney disease (CKD), called type 2 cardio-renal syndrome (CRS2). The mechanism is not completely clear, and there is a lack of early warning biomarkers of CKD in the context of CHF. Two CKD, one CHF-PBMC and four CHF-cardiac tissue expression profile datasets were obtained from GEO database. Differential expression analysis and WGCNA were used to detect CKD key genes and CHF-related secreted proteins. Protein-protein interaction (PPI), functional enrichment, and cMAP analysis reveal potential mechanisms and drugs of CHF-related CKD. Five machine

Machine Learning Approach for Frailty Detection in Long-Term Care Using Accelerometer-Measured Gait and Daily Physical Activity: Model Development and Validation Study.

BACKGROUND: Frailty affects over 50% of older adults in long-term care (LTC), and early detection is critical due to its potential reversibility. Wearable sensors enable continuous monitoring of gait and physical activity, and machine learning has shown promise in detecting frailty among community-dwelling older adults. However, its applicability in LTC remains underexplored. Furthermore, dynamic gait outcomes (eg, gait stability and symmetry) may offer more sensitive frailty indicators than traditional measures like gait speed, yet their potential remains largely untapped. OBJECTIVE: This st

A multi-omics framework integrating gut microbiota, blood metabolites, and immune cells to elucidate the pathogenesis of Alzheimer's disease.

BACKGROUND: Alzheimer's disease (AD) develops through complex interactions between the central nervous system and peripheral systems. The microbiota-metabolite-immune axis has emerged as an important focus of AD research. However, the coordinated mechanisms that regulate this axis remain poorly understood. METHODS: We used a multi-stage, multi-omics strategy to systematically investigate peripheral-central interactions in AD. The analytical framework integrated Mendelian randomization (MR), summary-data-based Mendelian randomization (SMR), differential expression analysis, machine learning, s

Pilot Study Using Machine Learning to Identify Immune Profiles for the Prediction of Early Virological Relapse After Stopping Nucleos(t)ide Analogues in HBeAg-Negative CHB.

Treatment with nucleos(t)ide analogues (NAs) may be stopped after 1-3 years of hepatitis B virus DNA suppression in hepatitis B e antigen (HBeAg)-negative patients according to Asian Pacific Association for the Study of Liver and European Association for the Study of Liver guidelines. However, virological relapse (VR) occurs in most patients. We aimed to analyze soluble immune markers (SIMs) and use machine learning to identify SIM combinations as predictor for early VR after NA discontinuation. A validation cohort was used to verify the predictive power of the SIM combination. In a post hoc a

Development and Validation of a Machine Learning Model for Predicting 30-Day Mortality and Early Endoscopic Hemostatic Intervention in Acute Upper Gastrointestinal Bleeding.

OBJECTIVE: Accurate pre-endoscopic risk stratification is essential in acute upper gastrointestinal bleeding (UGIB). Established scores predict mortality but have limited ability to identify patients requiring early therapeutic endoscopy. We developed and temporally validated a pre-endoscopic machine learning model to predict 30-day mortality and early (≤24 h) endoscopic hemostatic intervention and assessed model interpretability. METHODS: This retrospective cohort study included consecutive adults admitted with UGIB between November 2023 and November 2025. Only pre-endoscopic variables were

Multimodal machine learning reveals strain-specific flavor and kinetic dynamics in chinese spicy cabbage fermentation.

This study presents a novel multimodal machine learning framework integrating physicochemical kinetics with volatile metabolomics to elucidate strain-specific fermentation dynamics in Chinese spicy cabbage (CSC). Over a 90-day fermentation under ten single-strain LAB inoculation conditions, the modified Weibull model accurately described distinct growth-decline patterns, categorizing strains into three physiological types: slow-growing, robust-survival, and fast-growing. By integrating multimodal data including organic acids, reducing sugars, and volatile organic compounds (VOCs), multiple mac
