The blood-brain barrier (BBB) is a unique neurovascular interface essential for central nervous system homeostasis. Beyond its classical protective role, accumulating evidence points to the BBB as a dynamic structure actively involved in brain function in health and disease. The present review synthesizes clinical, molecular, and neuroimaging evidence implicating BBB dysfunction in major depressive disorder (MDD), bipolar disorder (BD), and schizophrenia. The paper was prepared in accordance with the recommendations of the Scale for the Assessment of Narrative Review Articles and was based on

BACKGROUND: Intraventricular haemorrhage (IVH) is the primary neuropathology in infants born very preterm. IVH describes bleeding into the ventricular space of the newborn brain, originating from the germinal matrix, termed germinal matrix haemorrhage. IVH is diagnosed at a rate of 1 in 5 infants born very preterm (less than 32 weeks' gestation), but the incidence increases with earlier gestation at birth. IVH is graded in severity (I to IV), and the neurological sequelae of IVH in infants born very preterm are significant, with more than 1 in 4 infants with any grade of IVH subsequently diagn

BACKGROUND: Retinopathy of prematurity (ROP) remains the leading cause for blindness in children. Limited hyperoxia induced proliferative retinopathy (L-HIPR) was recently introduced as a potential animal model for ROP and persistent fetal vasculature; however, the detailed pathological changes remain unclear.
METHODS: To model L-HIPR, we placed C57BL/6J mice in 65% oxygen from birth to post-natal day 7 (P7). We examined eyes at intervals between P12 and P30. Retinal morphometry, thickness, and preretinal fibrosis were quantified at different time points on histological sections stained with

Exposure to hypoxic-ischemic (HI) insults in newborns can predispose them to severe neurological sequela. The mechanisms underlying HI-related brain injury have not been completely elucidated. The neurovascular unit (NVU) is a composite of structures that protect the brain from the influx of detrimental molecules. Changes in the NVU after HI are important because they could reveal endogenous neuroprotective pathways in the cerebral microvasculature. Furthermore, the time course of changes in the NVU after exposure to HI in the newborn remains to be determined. In this study, we examined the ef

Microglial activation is implicated in retinal vasoregression of the neurodegenerative ciliopathy-associated disease rat model (i.e., the polycystic kidney disease (PKD) model). microRNA can regulate microglial activation and vascular function, but the effect of microRNA-124 (miR-124) on retinal vasoregression remains unclear. Transgenic PKD and wild-type Sprague Dawley (SD) rats received miR-124 at 8 and 10 weeks of age intravitreally. Retinal glia activation was assessed by immunofluorescent staining and in situ hybridization. Vasoregression and neuroretinal function were evaluated by quanti

Pericytic tumors encompass several entities sharing morphologic and immunohistochemical features. A subset of perivascular myoid tumors associated with the SRF-RELA fusion gene was previously described. Herein, we report a series of 13 tumors belonging to this group, in which we have identified new fusion genes by RNA-sequencing, thus expanding the molecular spectrum of this entity. All patients except 1 were children and infants. The tumors, frequently located in the head (n=8), had a mean size of 38 mm (range 10 to 150 mm) and were mostly (n=9) well-circumscribed. AIM: Idiopathic normal pressure hydrocephalus (iNPH) is one subtype of dementia characterized by cerebrospinal fluid (CSF) disturbance, but with unknown cause. We recently reported that frontal cortex biopsies of iNPH patients disclosed degenerative alterations of the capillary basement membrane, including degenerated pericyte processes. Given that pericyte degeneration is associated with blood-brain barrier (BBB) dysfunction, the present study was undertaken to examine whether BBB leakage of blood proteins can be revealed by light microscopy (LM) immunohistochemistry in iNPH.

BACKGROUND: The eye is considered as a window of the disease, and a better understanding of neurodegenerative changes in the eye may help diagnose and manage neurodegenerative diseases including the diseases of brain, heart, kidney and liver. In the eye, the blood retinal barrier (BRB] is maintained by a combination of endothelial cells, pericytes, and glia. This BRB integrity is fundamental to the physiology of retinal cellular function and accurate vision. The role of endothelial dysfunction as a consequence of endothelial activation in the initiation and prolongation of neurovascular diseas…

There are three principal malignant vasoformative tumors that can be found in the head and neck--hemangiopericytoma, angiosarcoma, and Kaposi's sarcoma. All are uncommon and provide challenges for the pathologist and the therapist both. The histogenesis of each tumor is different. Kaposi's sarcoma has many features which suggest that it is an altered immune-response disease. Angiosarcoma is a malignancy of endothelium. Hemangiopericytoma is a tumor whose cell of origin is considered to be the perithelial pericyte. The general prognosis for patients with Kaposi's sarcoma is good. The biologic c

Alzheimer's disease (AD) is a progressive neurodegenerative disorder in which blood-brain barrier (BBB) dysfunction may arise early. We performed an age-stratified, cross-sectional analysis of pericyte- and endothelial-associated markers and junctional proteins (claudin-1, -3, -5, occludin, and VE-cadherin) in cerebral capillary-enriched fractions from male Non-Tg and 3xTg-AD mice at asymptomatic, early, and advanced stages (3, 6, 12, and 16 months of age). Protein abundance was assessed by Western blot, and BBB permeability to 3-5 kDa FITC-dextran was evaluated at 3 months in an independent c

Cerebral small vessel disease (CSVD) is a heterogeneous group of disorders involving the cerebral microvasculature and is a major cause of stroke and vascular cognitive impairment. The role of blood-brain barrier (BBB) dysfunction in CSVD is now recognized as a key pathological process; however, conventional vascular risk factors do not fully explain its initiation, progression, or clinical heterogeneity. Astrocyte-BBB crosstalk has been implicated in endothelial dysfunction, pericyte injury, and vascular remodeling, and the mechanisms by which early barrier instability progresses to neurovasc

Primary percutaneous coronary intervention restores epicardial flow after ST-segment elevation myocardial infarction, but tissue perfusion may remain impaired. Coronary microvascular dysfunction, angiographic no-reflow, cardiac magnetic resonance-defined microvascular obstruction, and intramyocardial hemorrhage are overlapping but non-equivalent phenotypes. This review examines reduced mitochondrial aldehyde dehydrogenase 2 (ALDH2) activity as a susceptibility amplifier rather than an established independent cause of these outcomes. Reperfusion generates reactive oxygen species that initiate l

Critical-sized bone defects demand rapid vascularization for graft success, yet conventional pre-vascularization strategies have failed to coordinate vessel formation with osteoinduction. Here, we developed a GelMA-based 3D construct encapsulating co-cultured spheroids composed of human adipose-derived stem cells and human umbilical vein endothelial cells, incorporating BMP-2-immobilized nanofibers that directly deliver BMP-2 within the spheroid microenvironment. This spatially confined delivery system allowed BMP-2 to concurrently enhance hADSC osteogenesis, HUVEC functionality, and pericyte-

Loss of retinal function due to manifestation of chronic inflammation and oxidative stress in hyperglycemia is well addressed. However, the effect of hyperlipidemia on retinal inflammation and microvascular integrity, and the modulatory effects of oxidation-stable oleic acid and long-chain n-3 fatty acids have never been addressed. The objective of this investigation was to assess the retinoprotective effect of oxidation stable oleic acid and oxidation-susceptible EPA + DHA on retinal inflammation and microvascular integrity, under hyperlipidemic conditions.
