[Studies on the iodide metabolism and the expression of thyroglobulin and thyroid peroxidase mRNA in the thyroid of BB/W rats].

BioBreeding/Worcester (BB/W) rats develop insulin dependent diabetes mellitus (IDDM) and lymphocytic thyroiditis (LT) spontaneously. Our previous studies have shown that BB/W (Saitama-Tokyo colony) rats develop LT at about 10 weeks of age. Their serum TSH values increase as LT extends, although their serum thyroid hormone levels remain normal. This indicates that BB/W rats suffer from subclinical hypothyroidism. To investigate whether BB/W rats have a defect in iodide metabolism, the thyroidal radioactive iodine uptake (RAIU) in BB/W rats was examined. Thyroidal RAIU at 3hr in both 8 and 16 we

Formation of molecular iodine during oxidation of iodide by the peroxidase/H2O2 system. Implications for antithyroid therapy.

The first step in the biogenesis of thyroid hormones is the oxidation of iodides taken up by the thyroid gland. Oxidation of I- by the H2O2/peroxidase system leads to the formation of iodinium ions I+ which bond to thyroglobulin by electrophilic substitution. However, it is not clear whether I- is transformed directly to I+ or whether it passes through a molecular iodine intermediate. This latter possibility is indicated by the oxidation potentials of the reactions. I2 can be detected in vitro from the formation of I3- ions, although this has yet to be confirmed in vivo. The present study was

Age-related changes of glutathione content, glutathione reductase and glutathione peroxidase activity of human erythrocytes.

1. In order to investigate the effect of aging on the erythrocyte glutathione system, total glutathione (GSH), glutathione reductase (GSH-red) and glutathione peroxidase (GSH-px) levels were measured in erythrocytes from 33 young (mean age = 30.5 +/- 9.7 years) and 28 aged (mean age = 68.9 +/- 11.4 years) healthy individuals. 2. GSH was 3.5 +/- 1.8 microM/g Hb for the young group, a value significantly greater (P less than 0.01) than 2.3 +/- 0.9 microM/g Hb found for the aged group. Similarly, GSH-red activity, 5.5 +/- 1.8 IU/g Hb, was higher (P less than 0.05) for the young group than 3.4 +/-

Differential effects of methylmercuric chloride and mercuric chloride on oxidation and iodination reactions catalyzed by thyroid peroxidase.

Thyroid peroxidase (TPO), the major enzyme in the thyroid hormone synthesis, multifunctionally catalyzes (1) iodide oxidation, (2) iodination of the precursor protein, and (3) a coupling reaction of iodotyrosyl residues. The present study was carried out to examine the mercurial effects on the iodination, the second step of TPO. Purified porcine thyroglobulin or bovine serum albumin as acceptor protein was iodinated with [125I]NaI and H2O2 by purified porcine TPO. Iodinated protein was separated by acid precipitation on membrane filter or paper chromatography. Both CH3HgCl and HgCl2 dose-depen

[The dynamics of the T-lymphocyte subpopulations and the peroxidase activity of the neutrophils in children immunized with a live mumps vaccine].

The amount of T-helpers and T-suppressors and the T-helper/T-suppressor ratio were determined in 30 practically healthy children aged 1.5-2 years, immunized with live parotitis vaccine prepared from strain -3. One immunization dose of the vaccine contained 7,950 HADU50. In all children peroxidase activity in the cytoplasm of neutrophils was studied. This investigation revealed that in all examined children the formation of immune response to immunization with live parotitis vaccine was accompanied by the development of an imbalance of immunoregulatory T-cell subpopulations and by functional ch

A horseradish peroxidase study of the central projections of primary trigeminal neurons innervating the hard palate in the cat.

The central projections of primary sensory neurons innervating the hard palate in the cat were studied by transganglionic transport of horseradish peroxidase conjugated to wheat germ agglutinin (WGA-HRP). Following injection of WGA-HRP in the incisive papilla terminal labeling was observed in all subdivisions of the sensory trigeminal nuclear complex. In the main sensory nucleus labeling was located in the dorsal part, especially in its most rostral portion. At the levels of nuclei oralis and interpolaris labeling was observed along the medial borders of the nuclei. In addition, at these level

Thyroglobulin and thyroid peroxidase share common epitopes recognized by autoantibodies in patients with chronic autoimmune thyroiditis.

Monoclonal antibodies specific for human thyroid peroxidase (TPO) were prepared by the hybridoma technique using hyperimmune spleen cells from mice immunized with TPO purified from thyroid glands from patients with Graves' disease. Use of the microenzyme-linked immunosorbent assay method revealed that some of the monoclonal antibodies cross-reacted strongly with human thyroglobulin (Tg). Conversely, monoclonal anti-Tg antibodies cross-reacted with TPO, albeit to a lesser degree. Some anti-Tg autoantibodies in serum from patients with chronic autoimmune thyroiditis purified by Tg affinity chrom

Cisplatin-induced changes of selenium levels and glutathione peroxidase activities in blood of testis tumor patients.

Haematocrit and glutathione peroxidase activity in blood, as well as selenium levels in blood, erythrocytes and plasma, were determined in 15 patients during four courses of cisplatin combination treatment for testicular teratoma. The haematocrit steadily declined, necessitating frequent blood transfusions during or after treatment. For patients without blood transfusions during treatment the reduction of the haematocrit averaged 40%. Glutathione peroxidase activity in blood declined also; for patients without blood transfusion the reduction was 30%, which is fully explained by the decrease of

Plasma-selenium, glutathione peroxidase in erythrocytes and mercury in plasma in patients allegedly subject to oral galvanism.

Twelve patients with subjective symptoms, ascribed by the patients themselves to mercury released from dental restorations, were investigated. In addition to a general dental examination the following parameters were registered: the total number of amalgam surfaces in the mouth; potential and polarization of existing and accessible dental metallic restorations for calculation of intraoral currents. As regards the highest calculated intraoral current for each individual there was a statistically significant difference between the patient group and a control group consisting of 12 persons. An an

Thalamic projections to the posterior suprasylvian gyrus and the ventrally adjacent cortex in the cat traced with horseradish peroxidase.

The present study has been undertaken to elucidate the thalamic projections to the posterior suprasylvian gyrus (PSG) and the ventrally adjacent areas in the cat using retrograde axonal transport of horseradish peroxidase. The posterior part of the PSG, area 19 b, receives fibers mainly from the dorsolateral part of the medial pulvinar nucleus, and a lesser number from the lateral pulvinar nucleus. It receives a few fibers from the inferior pulvinar nucleus and the medial interlaminar nucleus (NIM) of the lateral geniculate. The anterior part of the PSG, area 21 b, receives a large number of f

Sensory nerve endings of the incisive papilla of rat hard palate studied by peroxidase cytochemical methods.

This study utilized peroxidase cytochemical methods to survey and analyze the distribution, morphology, and functional relationship of the various components of sensory nerve endings in a focal region of the anterior hard palate (incisive papilla) of the rat. The studies included an anatomical survey of this relatively unknown oral structure, confirmation of the trigeminal origin (labeled by retrograde axonal transport of WGA-HRP injected into the incisive papilla) of the sensory nerve endings, and a complete analysis of the distribution of these nerve endings (labeled by anterograde axonal tr

Comparative study of conjugation between horseradish peroxidase and D-cytochrome b5. Incorporation into subcellular membranes.

Horseradish peroxidase was conjugated to D-cytochrome b5 by three different two-step methods. The yield of conjugates based on the peroxidase enzymatic activity recovered after gel filtration was very low in the glutaraldehyde method, but higher in the N-succinimidyl 3-(2-pyridyldithio)propionate (SPDP) and periodate methods. The molecular size of the conjugates was analyzed by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. Monomeric conjugates were mostly formed via the glutaraldehyde and SPDP methods in the presence of appropriate molar ratios of proteins. Most of the conju

Iron Deficiency Decreases Suberization in Bean Roots through a Decrease in Suberin-Specific Peroxidase Activity.

The suberin content of young root parts of iron-deficient and iron-sufficient Phaseolus vulgaris L. cv Prélude was determined. The aliphatic components that could be released from suberin-enriched fractions by LiAID(4) depolymerization were identified by gas chromatography-mass spectrometry. In the normal roots, the major aliphatic components were omega-hydroxy acids and dicarboxylic acids in which saturated C(16) and monounsaturated C(18) were the dominant homologues. Iron-deficient bean roots contained only 11% of the aliphatic components of suberin found in control roots although the relati

Analysis of N-linked oligosaccharide chains of glycoproteins on nitrocellulose sheets using lectin-peroxidase reagents.

A rapid and convenient method was established for analysis of the N-linked carbohydrate chains of glycoproteins on nitrocellulose sheets. Proteins were separated by polyacrylamide gel electrophoresis, transferred to nitrocellulose sheets, reacted with peroxidase-coupled lectins, and detected by color development of the enzyme reaction. Four glycoproteins having N-linked oligosaccharide chains were used as test materials: Taka-amylase A (which has a high-mannose-type chain), ovalbumin (high-mannose-type chains and hybrid-type chains), transferrin (biantennary chains of complex type), and fetuin

Organization within the cranial IX-X complex in ranid frogs: a horseradish peroxidase transport study.

Cranial nerves IX and X in frogs have been described as originating from a nuclear group referred to as the IX-X complex. We studied the central nervous system components of this complex in Rana pipiens and R. catesbiana by labeling peripheral branches of cranial nerves IX and X and identifying the central nervous system contributions of these branches. Various peripheral nerves (IX and the cardiac, gastric, pulmonary, and laryngeal branches of X) were identified and soaked in horseradish peroxidase (HRP). One to 2 weeks later, the frogs were killed and processed for HRP by the tetramethylbenz

Peroxidase activity in relation to iodide, 17 beta-oestradiol and thioureylene drug uptake in human polymorphoneutrophils.

In polymorphoneutrophils (PMNs) phagocytosis is accompanied by an increase in peroxidase activity. Accumulation of iodide, thioureylene antithyroid drugs and 17 beta-oestradiol also occurs during the process. There is no evidence of an active iodide transport system in the PMNs as pertechnetate is not concentrated and neither ouabain nor perchlorate abolishes iodide accumulation. The uptakes of 125I, [35S]PTU and [3H]-17 beta-oestradiol were compared in phagocytosing PMNs and the effects of various compounds examined. In addition, chemiluminescence generation from luminol by PMNs and by horser

Studies on glutathione S-transferase, glutathione peroxidase and glutathione reductase in human normal and cataractous lenses.

The cause(s) for decreased reduced glutathione (GSH) content of human senile cataracts were investigated by determination of three enzymes of GSH metabolism: GSH S-transferase, GSH peroxidase and GSH reductase. There are significant decreases in activities of GSH S-transferase of early cataracts (-76%) and of dark advanced cataracts (-73%) as compared to clear age-matched lenses. However, the activities of GSH peroxidase were only slightly decreased in cataracts as compared to clear age-matched lenses. The decrease in enzymes of GSH metabolism does not account for increased GSH degradation. Th

The origin of the preganglionic parasympathetic fibres to the mandibular and sublingual salivary glands in the rat: a horseradish peroxidase study.

Horseradish peroxidase was applied to a ligation of the chorda tympani nerves running alongside the submandibular duct in order to label the preganglionic parasympathetic salivatory neurons in the brain stem. Labelled neurons were found in the ipsilateral motor trigeminal nucleus, lateral reticular formation and in the facial motor nuclei of both sides. However, the labelling of neurons in the motor nuclei of the trigeminal and facial nucleus was caused by the uptake of horseradish peroxidase by axons in the musculature surrounding the injection site which were probably damaged by the dissecti
