Effects of physical stress on peroxide scavengers in normal and sickle cell trait erythrocytes.

Sickle cell trait subjects are potentially at risk with microvascular complications frequently associated with high altitude and extreme exertion. Studies conducted in our laboratory suggest that exposure of sickle cell trait subjects to treadmill exercise resulted in an increase not only in the susceptibility of red blood cells to oxidation but also in the density of RBC membrane. The trend of increased susceptibility of red cells to oxidation as a consequence of exercise was associated with relatively excessive production of H2O2, inefficient breakdown of H2O2 by GSH-Px- and catalase-catalyz

[The effect of the peroxidation of ribamidil-containing liposomes on the results of chemotherapy in experimental viral infections].

A relationship between ribamydil concentration and the intensity of accumulation of peroxidation products was found while storing ribamydil-containing liposomes which indicated a significant pro-oxidant activity of ribamydil. Increasing lethality of guinea pigs and white mice infected with Venezuelan equine encephalomyelitis virus was shown while using liposomal ribamydil containing from 4.78 to 6.82 nmol. ml-1 malonic dialdehyde which appeared to be associated with disordered function of the antioxidant system of the experimental animals.

[Studies on the influence of long-term doses of lipid peroxide generators on rat pancreas].

The role of oxygen derived free radicals or tissue lipid peroxides in the pathogenesis of chronic pancreatitis has not been established. To evaluate long-term effects of tissue lipid peroxides in the pathogenesis of chronic pancreatitis, we treated Wistar male rats with 2,2'-azo-bis-(2-amidino-propane) dihydrochloride (AAPH) and/or linoleic acid (LA) for 3 or 6 months. Rats were divided into eight groups. A: Saline-treated rats for 3 months as control, B: AAPH 40 mg/kgw intraperitoneally, twice a week for 3 months, C: LA 0.5 ml/kgw intraperitoneally, every other week for 3 months, D: AAPH and …

Roles of catalase and the glutathione redox cycle in the regulation of anterior-chamber hydrogen peroxide.

The effects of inhibition of both glutathione synthesis and of glutathione reductase and catalase activities have been determined in the regulation of hydrogen peroxide (H2O2) in the anterior chamber of pigmented rabbits. Glutathione reductase inhibition using intravitreal 1,3-bis-(2-chloroethyl)-1-nitrosourea (BCNU) did not significantly alter either total glutathione or the percent oxidized glutathione fraction in the iris-ciliary body. Intravitreal buthionine sulfoximine (BSO) significantly reduced the total glutathione content of iris-ciliary body and corneal endothelium, while not alterin

[Effects of different buffers, ethanol and hydrogen peroxide on the morphology of cultured brain cells].

Effects of different buffers: N-2-Hydroxyethylpiperazine-N-2-ethanesulfonic acid (HEPES), Ethanolamine (2-Aminoethanol), sodium bicarbonate (NaHCO3) in the presence of 0.003% H2O2 or 0.5% ethanol were investigated on cell cultures from brain (chick embryo 5 1/2 or 9 day in ovo). Morphological parameters: density of cells, reaggregation, size of cell aggregates were used. H2O2 in the presence of NaHCO3 led to a decrease of isolated cells and an increase of reaggregation with degeneration in the great clusters. When H2O2 and 5 mM HEPES were present together in the absence of NaHCO3, the aggregat

Antioxidant enzyme status and lipid peroxidation in various tissues of diabetic and starved rats.

The effect of short term (2-wk) diabetes induced by streptozotocin and starvation (1-wk) on antioxidant enzymes and lipid peroxidation in the liver, kidney and heart of rats was investigated. The activity of mitochondrial oxidative markers was increased in diabetic liver and kidney, while the activity in tissues of starved rats tended to be decreased. Immunoreactive manganese superoxide dismutase was increased only in diabetic liver and was unchanged or decreased in the rest of the tissues. Glutathione peroxidase activity was increased in tissues of diabetic but not starved rats. The changes i

Increased hydrogen peroxide in the expired breath of patients with acute hypoxemic respiratory failure.

Acute hypoxemic respiratory failure (AHRF) can result from diverse lung insults. Toxic oxygen metabolites have been implicated in this clinical condition and in animal models of pulmonary edema. Hydrogen peroxide (H2O2), an oxygen metabolite, mediates tissue injury. We measured H2O2 levels by a spectrophotometric technique in the breath condensate of 68 mechanically ventilated patients; 13 patients with normal lungs undergoing elective surgery had no such detectable levels of H2O2. Fifty-five patients in the ICU meeting criteria for the adult respiratory distress syndrome (ARDS) had a higher c

Catalase and glutathione peroxidase are equally active in detoxification of hydrogen peroxide in human erythrocytes.

Genetic deficiencies of glucose-6-phosphate dehydrogenase (G6PD) and NADPH predispose affected erythrocytes to destruction from peroxides. Conversely, genetic deficiencies of catalase do not predispose affected erythrocytes to peroxide-induced destruction. These observations have served to strengthen the assumption that the NADPH/glutathione/glutathione peroxidase pathway is the principal means for disposal of H2O2 in human erythrocytes. Recently, however, mammalian catalase was found to have tightly bound NADPH and to require NADPH for the prevention and reversal of inactivation by its toxic …

Hydrogen peroxide and methyl mercury are primary stimuli of eicosanoid release in human platelets.

Hydrogen peroxide (H2O2) and methyl mercury induced the liberation of arachidonate and its metabolites from human washed platelets. [14C]Eicosanoids were extracted from the supernatants of [14C] arachidonate-prelabelled platelets and analysed by thin layer chromatography and radioscanning. Thromboxane B2 (TXB2), 12(S)-hydroxy-5,8,10-heptadecatrienoic acid (HHT) and 12(S)-hydroxy-5,8,10,14-eicosatetraenoic acid (12-HETE) were found as stable metabolites, together with unreacted arachidonate. In the presence of dazoxiben, a shift in eicosanoid metabolism was observed towards prostaglandin E2 (PG

[Induction of lipid peroxidation and change of fatty acid composition in rats fed methylmercury].

Four groups of rats, with 5 animals in each, were fed a diet to which 1, 2, 4 or 8 ppm of methylmercury (MeHg) were added while another group of 5 rats was fed a diet containing no MeHg as a control. Moreover, each rat was daily supplied the same volume of these diets. During the experimental period of 40 days, body weights of the rats were daily checked individually. On day 40, expired air of the rats was collected to analyse for ethane and pentane production, and then the rats were sacrificed to obtain the liver, kidneys, brain and plasma. Comparison of a salicylic acid cleanser and a benzoyl peroxide wash in the treatment of acne vulgaris.

A four-week crossover study to compare the efficacy of an acne cleanser containing 2% salicylic acid with that of a 10% benzoyl peroxide wash was conducted in 30 patients with acne vulgaris. The results demonstrated that only patients treated with the salicylic acid cleanser had a significant reduction in comedones. Patients treated with the salicylic acid cleanser for the first two weeks showed a significant improvement in acne, but worsened during benzoyl peroxide therapy over the following two weeks. In contrast, patients initially treated with the benzoyl peroxide wash for the first two we

The role of phospholipase A2 in microsomal lipid peroxidation induced with t-butyl hydroperoxide.

The role of phospholipase A2 (PlA2) in lipid peroxidation induced with t-butyl hydroperoxide was examined in rat liver microsomes. Exposure of microsomes to t-butyl hydroperoxide was associated with activation of endogenous PlA2. When PlA2 was inhibited with chlorpromazine, mepacrine, or p-bromphenacyl bromide, the accumulation of thiobarbituric acid reactive substances (TBARS) was reduced in a dose dependent manner. In contrast, the accumulation of conjugated dienes was not affected by chlorpromazine, and was slightly increased by mepacrine. When endogenous PlA2 was activated with mellitin pr

Oxidative inactivation of rhodanese by hydrogen peroxide produces states that show differential reactivation.

Controlled conditions have been found that give complete reactivation and long term stabilization of rhodanese (EC 2.8.1.1) after oxidative inactivation by hydrogen peroxide. Inactivated rhodanese was completely reactivated by reductants such as thioglycolic acid (TGA) (100 mM) and dithiothreitol (DTT) (100 mM) or the substrate thiosulfate (100 mM) if these reagents were added soon after inactivation. Reactivability fell in a biphasic first order process. At pH 7.5, in the presence of DTT inactive rhodanese lost 40% of its reactivability in less than 5 min, and the remaining 60% was lost more

[Lipid peroxidation and endogenous phospholipase hydrolysis during trauma and graded hypothermic load].

Rates of lipid peroxidation and endogenous phospholipase hydrolysis were studied under conditions of simultaneous moderate hypothermia and traumatic ischemia of muscles. Activation of the reactions studied, which was expressed as an increase in content of malonic dialdehyde and free fatty acids, was of importance for pathogenesis of muscles impairments occurring in trauma. Local cooling affected favourably these patterns of lipid metabolism.

Free radicals and lipid peroxidation in ethanol- or aspirin-induced gastric mucosal injury.

In this study the role of free radicals and lipid peroxidation as mediators of chemically induced mucosal damage was investigated. Two enzymatic antioxidants, superoxide dismutase or catalase injected intravenously, reduced mucosal damage either by ethanol or aspirin. Of six nonenzymatic antioxidants, given in a wide dose range subcutaneously 30 min before intragastric administration of absolute ethanol, only propyl gallate decreased mucosal damage, while four of the antioxidants tested against aspirin were protective. These nonenzymatic antioxidants were antisecretory in the pylorus-ligated r

Role of glutathione in protecting endothelial cells against hydrogen peroxide oxidant injury.

To determine the mechanism responsible for the enhanced susceptibility of endothelial cells to oxidant injury in the absence of glucose, we induced endothelial cell injury with oxygen radicals in the presence of various oxygen radical scavengers and measured endothelial cell levels of glutathione after oxidant injury in the presence and absence of glucose. Endothelial cells were damaged with toxic oxygen radicals generated by phorbol myristate acetate (PMA)-activated polymorphonuclear leukocytes (PMNs) or xanthine-xanthine oxidase in the presence and absence of glucose and catalase (scavenger
