Dysregulation of lipid homeostasis is common in cancers, and biophysical studies have suggested that increased incorporation of saturated lipids and cholesterol may reduce cell membrane permeability to oxygen (O2). Low intracellular oxygen, in turn, may promote tumor aggressiveness, as well as resistance to radiotherapy. A prior lipidomics study found membrane phospholipids associated with de novo fatty acid synthesis to be unusually abundant in breast tumors with poor prognosis, including the saturated lipids 1-myristoyl-2-palmitoyl-sn-glycero-3-phosphocholine (MPPC14,16, or PC14:0/16:0) and …

BACKGROUND: Triglyceride-type n-3 polyunsaturated fatty acids (n-3 PUFAs) are often used to improve the quality of meat products, while the potential of phospholipid-type n-3 PUFAs remains underexplored. This study evaluated the gel and flavor quality of minced pork gel (MPG) with marine-sourced large yellow croaker roe phospholipids (LYPLs) as phospholipid-type n-3 PUFA supplements.
RESULTS: The addition of LYPLs increased the n-3 PUFA content in MPG and caused no visually significant color changes. Textural properties, water-holding capacity and binding water percentage increased with the i…

This study aimed to evaluate the effects of phytosterols (PSs), alone or in combination with phospholipids (PLs), on dyslipidemia and oxidative stress, using both a high-fat diet-induced rat model and a randomized controlled trial in individuals with borderline hypercholesterolemia. In the animal study, Sprague-Dawley rats were divided into five groups (control, model, PS, PL, and PS+PL) and treated for 8 weeks. The PS+PL combination significantly reduced body weight, liver index, and serum glutathione peroxidase activity, while improving hepatic and adipose morphology. In the human trial, 82 …

Moreover, mitochondrial membrane potential was decreased and peroxidation of mitochondrial phospholipids was increased by iPLA2γ knockdown. It was also shown that co-treatment of bromoenol lactone, an iPLA2 inhibitor, increased CDDP-induced apoptosis. These results indicated that iPLA2γ plays an important role in protecting bladder cancer cells from CD …

X-linked adrenoleukodystrophy (X-ALD) is the most common leukodystrophy. Despite intensive research in recent years, it remains unclear, what drives the different clinical disease courses. Due to this missing pathophysiological link, therapy for the childhood cerebral disease course of X-ALD (CCALD) remains symptomatic; the allogenic hematopoietic stem cell transplantation or hematopoietic stem-cell gene therapy is an option for early disease stages. The inclusion of dried blood spot (DBS) C26:0-lysophosphatidylcholine to newborn screening in an increasing number of countries is leading to an

OBJECTIVES: Phospholipids (PLs) are increasingly recognized as key molecules with potential diagnostic value in acute inflammation, CVD and atherosclerosis. We introduce a pioneer mass spectrometry (MS)-based approach aiming to investigate the relationship of specific plasma PL-subsets with atherogenic blood parameters in young patients with familial hyperlipidemia representing high-CVD-risk groups.
METHODS: Plasma of carefully phenotyped FH and FCH patients as well as normolipidemic subjects (age 13 ± 5 years, n = 20) was used. Clinical parameters were assessed using standard laboratory tech…

Net degradation of cellular components occurs in plant cells cultured under starvation conditions, and autophagy contributes to the degradation of intracellular proteins. In this study, we investigated the degradation of membrane phospholipids by autophagy in cultured tobacco (Nicotiana tabacum) cells. The amounts of total phospholipids and a major phospholipid, phosphatidylcholine (PC), decreased, whereas phosphorylcholine, a degradation product of PC, increased in response to deprivation of sucrose. The addition of glycerol to the culture medium inhibited both the degradation of phospholipid…

Chronic hypoxia due to chronic obstructive pulmonary disease (COPD) constitutes a stress to cerebral metabolic homeostasis. Previous studies using phosphorus-31 magnetic resonance spectroscopy (31P MRS) have suggested that the brains of such patients utilize anaerobic glycolysis, which in neonatal, animal, and in vitro studies is associated with a protective intracellular alkalosis. To identify such a compensatory intracellular alkalosis in hypoxic COPD patients, in vivo cerebral 31P MRS was performed in eight patients and eight controls. The mean intracellular pH (pHi) in patients with COPD w

Since iodide (I-) inhibits TSH stimulation of cAMP formation, which mediates most of the effects of the hormone, it has been assumed that this accounts for the inhibitory action of iodide on the thyroid. However, TSH stimulation of 32P incorporation into phospholipids and stimulation of thyroid metabolism by other agonists, such as carbachol, phorbol esters, and ionophore A23187, is not cAMP mediated. The present studies examined the effect of iodide on stimulation of glucose oxidation and 32P incorporation into phospholipids by TSH and other agonists to determine if the inhibition of cAMP for…

Qualitative and quantitative assays for human antibodies to cardiolipin and other phospholipids were used in tests for these reactions in sera from patients with systemic lupus erythematosus (SLE) and syphilis. Of 22 SLE serum samples tested by the qualitative assay, 8 showed positive staining to cardiolipin, phosphatidic acid, and/or phosphatidylserine. All 47 syphilitic sera reacted with these three phospholipids. The apparent affinity of anticardiolipin binding was estimated by normalizing absolute binding levels as a function of serum concentration to the maximum percent bound. It was evid…

Fatty acids of the phospholipid fraction of bronchoalveolar lavage fluid from patients with bacterial pneumonia or with the adult respiratory distress syndrome were chromatographed and the patterns compared with those for a control group. In the control group, palmitic acid (16:0) was the predominant fatty acid, accounting for 58.0% (SD 8.25%) of the total fatty acid, a proportion significantly higher (p less than 0.001) than in the distress-syndrome group (42.1%, SD 4.88%) or the acute pneumonia group (32.1%, SD 1.73%). There was a greater proportion of oleic acid (18:1) in the disease groups

Plasma levels of cholesterol (C), triglycerides (TG), phospholipids (PL) (in total plasma, very low density [VLDL], low density (LDL), and high density [HDL] lipoproteins) and of two apolipoproteins (apo-B and apo-A) were studied in 13 hyperlipidemic patients suffering from hypertension and/or stable angina and treated by metoprolol or propranolol. Propranolol reduced the low density and high density lipoprotein phospholipids by 26% and 11%, respectively, and increased the very low density phospholipids by 24%. Metoprolol had only a transient effect on high density lipoprotein phospholipids. V…

A purified preparation of the oligomycin-sensitive ATPase from yeast mitochondria has been shown to elicit an oligomycin- and uncoupler-senstitive ATP-32Pi exchange in the presence of phospholipids. Reconstitution was normally achieved by dialysis of an ATPase-phospholipid-cholate mixture. Following this procedure, vesicles with diameters between 200 and 1,500 A were seen by electron microscopy. As in mitochondria, ATPase activity in the reconstituted system was stimulated by a range of uncouplers which inhibited ATP-32Pi exchange. These and other findings suggest that the coupling mechanism m…

Adipose-derived mesenchymal stem cells (ADMSCs) are a crucial source of new adipocytes and play a significant role in regulating adipose tissue physiology. Interestingly, mature adipocytes differentiated from the ADMSCs maintain some characteristic features of their respective depots as well as the metabolic status of the donor patients. The above is true with respect to the cells' secretory profile and their glycerolipid and sphingolipid composition. In the current paper, we focused our attention on the phospholipid profile of the adipocytes derived from ADMSCs, with a particular emphasis pla
