A general polyploid model for analyzing gene segregation in outcrossing tetraploid species.

Polyploidy has played an important role in higher plant evolution and applied plant breeding. Polyploids are commonly categorized as allopolyploids resulting from the increase of chromosome number through hybridization and subsequent chromosome doubling or autopolyploids due to chromosome doubling of the same genome. Allopolyploids undergo bivalent pairing at meiosis because only homologous chromosomes pair. For autopolyploids, however, all homologous chromosomes can pair at the same time so that multivalents and, therefore, double reductions are formed. In this article, we use a maximum-likel…

Insights into the genetic and biochemical basis of Gibberella ear rot resistance in maize.

Fusarium graminearum colonizes the maize ear, causing Gibberella ear rot (GER) and producing harmful mycotoxins, including deoxynivalenol (DON) and zearalenone (ZEA). The disease can be managed in part by breeding and planting resistant maize cultivars. Resistance to GER is a quantitative and complex trait. Evaluation of diverse germplasm to identify regions and candidate genes associated with resistance may be useful for crop improvement efforts. Screening for GER is time-consuming and costly. Thus, identifying other traits that may serve as a proxy for GER resistance may accelerate resistanc

The Membrane Continuum Governing Cadmium Uptake, Partitioning, and Detoxification in Plants.

Cadmium (Cd) contamination threatens agro-ecological safety and food security; thus, elucidating mechanisms of Cd uptake and accumulation in plants is key to breeding low-Cd crops. Within the defensive network plants deploy against Cd stress, the plasma membrane and endomembrane system act as physical barriers to metal influx and as the core regulatory hub governing signal perception, transmembrane transport, and vacuolar compartmentalization for detoxification. This review examines the physiological and molecular mechanisms by which the cell membrane system influences Cd transport and accumul

Automated Video Tracking to Phenotype Plant Resistance to Aphid-Transmitted Yellow Dwarf Viruses in Grass Seed Crops.

Grass seed crops are susceptible to yellow dwarf viruses transmitted by aphids. The Willamette Valley in Oregon, United States, is the leading producer of cool-season grass seed crops globally, and industry reports have attributed seed yield loss and shortened stand longevity to aphid-transmitted yellow dwarf viruses. Genetic resources are needed for effective and sustainable management of this pest, specifically the Rhopalosiphum padi-PAV pathosystem, in grass seed production to reduce foliar insecticide applications and maintain optimum seed yield potential. High-throughput phenotyping metho

SlMYB30 relieves high vapor pressure deficit stress by modulating salicylic acid biosynthesis and stomatal morphology in tomato.

High vapor pressure deficit (HVPD) stress presents challenges to global agriculture, impacting crop productivity, water resource management, ecosystem stability, and food security. Tomato (Solanum lycopersicum L.) is an economically important crop grown extensively around the world; however, the molecular mechanisms that enable its resistance to HVPD stress are not yet fully understood. In this study, V-MYB AVIAN MYELOBLASTOSIS VIRAL ONCOGENE HOMOLOG 30 (SlMYB30), an R2R3-type MYB transcription factor responsive to HVPD stress in tomato, was identified through transcriptomic and metabolomic an

Genome-Wide Screening and Functional Verification of the Tobacco SGR Family Identify Transcription-Factor-Like NtSGRL as a Negative Regulator of Tomato Yellow Leaf Curl Virus Resistance.

Viral infection often induces chlorosis, a symptom closely associated with chlorophyll (Chl) degradation. STAY-GREEN (SGR) proteins are key regulators of Chl breakdown, but their roles in plant virus resistance remain poorly understood. In this study, we identified six NtSGR family genes in Nicotiana tabacum and functionally characterized NtSGRL1 during tomato yellow leaf curl virus (TYLCV) infection. TYLCV drastically upregulates NtSGRL1 (149-fold at 4 days post-inoculation [dpi] vs. control), suggesting its involvement in the tobacco-TYLCV interaction. Subcellular localization revealed that

CRISPR/Cas9-mediated knockout of MsMLO1 confers enhanced powdery mildew resistance in alfalfa.

Powdery mildew, a disease primarily caused by Erysiphe pisi, is one of the most devastating foliar diseases of alfalfa (Medicago sativa L.), a globally important forage crop. The MLO gene is a well-known susceptibility factor for powdery mildew in many plant species, but its role in alfalfa remains unclear. In this study, we used the CRISPR/Cas9 system to edit MsMLO1 in alfalfa cultivar 'Aohan'. We designed an sgRNA based on a conserved target site in the exon region of this gene, and introduced the genome-editing vector into alfalfa via Agrobacterium tumefaciens-mediated transformation. Mulberry ERF Transcription Factor MnERF23 Is a Positive Regulator of Plant Drought Tolerance.

Mulberry (Morus alba L.) is an important economic tree species, and its leaves are increasingly being used to prepare healthy dishes in addition to serving as food for silkworms. Drought is one of the main stress factors affecting the quality of mulberry leaves; however, the molecular mechanisms through which mulberry plants respond to drought stress are currently unclear. The AP2/ERF gene family is involved in plant growth, development and stress response processes. In response to drought stress, the expression of mulberry MnERF23 is induced. The overexpression of MnERF23 in Arabidopsis thali

Bioactive Molecules Derived from Plants in Managing Dengue Vector Aedes aegypti (Linn.).

Mosquitoes are the potential vectors of several viral diseases such as filariasis, malaria, dengue, yellow fever, Zika fever and encephalitis in humans as well as other species. Dengue, the most common mosquito-borne disease in humans caused by the dengue virus is transmitted by the vector Ae. aegypti. Fever, chills, nausea and neurological disorders are the frequent symptoms of Zika and dengue. Thanks to various anthropogenic activities such as deforestation, industrialized farming and poor drainage facilities there has been a significant rise in mosquitoes and vector-borne diseases. Control

Novel PCR markers for discriminating RMia resistance alleles in Prunus rootstock breeding.

Root-knot nematodes are pest of many plant species, particularly in southern Europe, where Meloidogyne incognita, M. arenaria and M. javanica are the most common species. In Prunus rootstock breeding, restrictions on nematicide use has increased importance of incorporating genetic resistance. Three resistance genes have been identified in Prunus: Ma, RMja and RMia. The latter is a TNL gene located on the linkage group 2 that confers resistance to M. arenaria, M. incognita and M. ethiopica. Resistance is expressed in both homozygous and heterozygous dominant genotypes, but not in homozygous rec

Combining pathway engineering and precursor enrichment to build a multi-trait soybean platform for aquaculture feed applications.

Oilseed crops offer a scalable aquaculture feed production platform for generating oils with long-chain ω-3 polyunsaturated fatty acids and high-value antioxidants, but metabolic flux through heterologous pathways is often constrained by precursor availability and endogenous regulatory limits. Here we engineered soybean seeds to co-produce eicosapentaenoic acid (EPA), astaxanthin, and tocotrienols by introducing nine microbial and plant enzymes spanning fatty acid desaturation/elongation, ketocarotenoid synthesis, and vitamin E tocochromanol biosynthesis. Transgenic events accumulated EPA up t

Mechanisms and Advances in Plant Lipid Regulatory Responses Under Biotic and Abiotic Stress.

Biotic stresses (pest feeding, pathogenic fungal/bacterial/viral infection) and diverse abiotic stresses (extreme temperature, drought, waterlogging, saline-alkali soil, heavy metal pollution, nutrient deficiency, UV-B, ozone) severely restrict crop growth and global agricultural yield. Lipids act as core membrane structural constituents and vital secondary signaling messengers, executing multi-layered adaptive balancing functions during cell-type interactive stress acclimation, rather than uniform whole-plant lipid responses. They sustain membrane structural integrity across distinct cell pop

AutoSiQ: a curated haploid Arabidopsis thaliana inflorescence dataset with a fine-grained silique ontology and a deep learning application for haploid fertility quantification.

Doubled haploid (DH) technology can fast-track crop breeding. Haploid induction yields haploids with only one set of genomes, which are usually sterile. Haploid fertility (HF) is the ability of haploid plants to set seed, and it is a critical bottleneck in DH pipelines. Genetic mechanisms to restore HF hold immense potential in DH crop breeding, yet its phenotyping remains manual, destructive, and inconsistent. While recent advances in imaging and machine learning have improved throughput for general plant traits, no curated image dataset exists for Arabidopsis thaliana that explicitly represe

Flooding stress resilience and crop improvement.

Promoting the long-term viability of agriculture in the face of climate change has spurred a rise in alternative approaches to enhance crop resilience. Universally, the significant risk of crop yield reduction in cereal crops due to flooding has become a prominent concern, and this threat is anticipated to intensify in various regions worldwide due to future climatic anomalies. Flooding stress induces oxygen (O2) deficiency within the plant system, serving as a major restriction for plant growth during submerged and waterlogged environments. Plants experience fluctuating levels of O2 contents,
