Plant-based vaccine candidate against Infectious bursal disease: An alternative to inactivated vaccines for breeder hens.

Infectious bursal disease (IBD) is an acute, highly contagious immunosuppressive disease that affects young birds causing important economic losses in the poultry industry worldwide. Strict hygiene management together with effective vaccination programs are the most important strategies to prevent Infectious bursal disease virus entry in poultry production facilities. Hyperimmunisation of dams with inactivated vaccines just before the laying period provides passive immunity to the progeny that protects them during the critical first few weeks after hatching before vaccination with live attenua

A novel approach to enhance resistance to vascular disease by expressing cell-death-inducing fungal elicitors in the xylem tissue.

Constitutive expression of hypersensitive cell-death-inducing effectors secreted by pathogens has proved to be difficult in plants. Herein, xylem-specific promoters (PVND7 or PVND2) were utilized to express three cell-death-inducing proteins (FocSIX1A, FocSIX6 or TvSm1) of fungal origin in banana plants. The regenerated plants were not only healthy but also showed improved resistance to fungal (Foc vcg0124 and Foc vcg0125) infection. These banana plants developed systemic resistance-like symptoms, including elevated jasmonic acid content, callose depositions, increased xylem wall thickness and

Plant-derived bioactive compounds modulate the gut microbiota in Alzheimer's disease: Metabolite signaling, neuroimmune circuits, and systems-level regulation.

BACKGROUND: Alzheimer's disease (AD) is increasingly recognized as a multisystem disorder shaped not only by central neurodegeneration but also by peripheral metabolic and immune dysregulation. Growing evidence highlights the gut microbiota and its metabolites as key modulators of amyloid accumulation, tau phosphorylation, neuroinflammation, and microglial dysfunction. PURPOSE: This review aims to synthesize current advances on how plant-derived bioactive compounds modulate AD pathophysiology through microbiota-dependent metabolic and neuroimmune mechanisms, and to establish a systems-level f

OsTGAL1 suppresses the resistance of rice to bacterial blight disease by regulating the expression of salicylic acid glucosyltransferase OsSGT1.

Many TGA transcription factors participate in immune responses in the SA-mediated signaling pathway in Arabidopsis. This study identified a transcription factor OsTGAL1, which is induced upon infection by Xoo. Overexpression of OsTGAL1 increased the susceptibility of rice to Xoo. Plants overexpressing OsTGAL1 could affect the expression of many SA signaling-related genes. OsTGAL1 was able to interact with the promoter of OsSGT1, which encodes a key enzyme for SA metabolism. The transcript of OsSGT1 was induced by Xoo and this responsive expression was further increased in plants overexpressing

JMJ14 encoded H3K4 demethylase modulates immune responses by regulating defence gene expression and pipecolic acid levels.

Epigenetic modifications have emerged as an important mechanism underlying plant defence against pathogens. We examined the role of JMJ14, a Jumonji (JMJ) domain-containing H3K4 demethylase, in local and systemic plant immune responses in Arabidopsis. The function of JMJ14 in local or systemic defence response was investigated by pathogen growth assays and by analysing expression and H3K4me3 enrichments of key defence genes using qPCR and ChIP-qPCR. Salicylic acid (SA) and pipecolic acid (Pip) levels were quantified and function of JMJ14 in SA- and Pip-mediated defences was analysed in Col-0 a

Purification of monoclonal antibody against Ebola GP1 protein expressed in Nicotiana benthamiana.

Monoclonal antibodies (mAbs) are one of the fastest growing drug molecules targeting the treatment of diseases ranging from arthritis, immune disorders, and infectious diseases to cancer. Due to its unique application principle, antibodies are commonly produced in large quantities. Plants, such as Nicotiana benthamiana, offer a unique production platform for bio-therapeutics due to their ability to produce large amounts of biomolecules in a relatively quick manner. However, purification of a target protein from plant is an arduous task due to the presence of toxic compounds in ground plant tis

Manipulation of Microbial Symbionts in Bemisia tabaci and Trialeurodes vaporariorum (Hemiptera: Aleyrodidae) Reveals Divergent Impacts on Insect Host Fitness and Plant Defense Modulation.

Insect-microbe symbioses play pivotal roles in host ecology and plant-insect interactions, yet their species-specific functions in agricultural pests remain less understood. This study elucidates the functional role of symbiotic microbial communities in mediating insect host fitness and plant defense responses in two economically important whitefly species, Bemisia tabaci Gennadius and Trialeurodes vaporariorum Westwood (Hemiptera: Aleyrodidae). Using integrated molecular and physiological approaches, we characterized species-specific responses to antibiotic treatments (rifampicin and tetracyc

Mitigating effects of Bean yellow mosaic virus infection in faba bean using new carboxymethyl chitosan-titania nanobiocomposites.

Bean yellow mosaic virus (BYMV) is the main cause of the mosaic and malformation of many plants, worldwide. Thus, the triggering of plant systemic resistance against BYMV is of great interest. In this endeavor, we aimed to explore the capacity of new carboxymethyl chitosan-titania nanobiocomposites (NBCs, NBC1,2) to trigger faba bean plants resistance against BYMV. Effects of NBCs on faba bean (Vicia faba L.) disease severity (DS), growth parameters, and antioxidant defense system activity were investigated under BYMV stress. Noticeably that the DS in NBCs-treated faba bean was significantly r

Arabidopsis triphosphate tunnel metalloenzyme2 is a negative regulator of the salicylic acid-mediated feedback amplification loop for defense responses.

The triphosphate tunnel metalloenzyme (TTM) superfamily represents a group of enzymes that is characterized by their ability to hydrolyze a range of tripolyphosphate substrates. Arabidopsis (Arabidopsis thaliana) encodes three TTM genes, AtTTM1, AtTTM2, and AtTTM3. Although AtTTM3 has previously been reported to have tripolyphosphatase activity, recombinantly expressed AtTTM2 unexpectedly exhibited pyrophosphatase activity. AtTTM2 knockout mutant plants exhibit an enhanced hypersensitive response, elevated pathogen resistance against both virulent and avirulent pathogens, and elevated accumula

Canonical EDS1/PAD4 small-molecule binding sites are required for LRR-RP-mediated pattern-triggered immunity.

Pattern-triggered immunity (PTI) is a key plant defense mechanism initiated by the recognition of pathogen-associated molecular patterns (PAMPs) by cell-surface receptors, including leucine-rich repeat receptor proteins (LRR-RPs). Upon PAMP perception, LRR-RPs associate with SOBIR1 and recruit SERK coreceptors, such as BAK1, into the receptor complex to initiate PTI. The lipase-like protein EDS1 and PAD4 are central components of effector-triggered immunity (ETI). Small molecule (SM) phosphoribosyl-AMP/ADP, generated by TIR-domain NADase activity, promotes the recruitment of helper NLRs of the

H3K27me3-Mediated Epigenetic Silencing of FgHMG1 Enables Fungal Host Immune Evasion.

Histone H3 lysine 27 trimethylation (H3K27me3) is essential for fungal pathogenicity, yet its contribution to pathogen-host interactions remains incompletely understood. Here, we profiled H3K27me3 dynamics in Fusarium graminearum during infection and identified 132 H3K27me3-marked genes (FgHMGs). Among these, FgHMG1 encodes a secreted glycoside hydrolase family 11 (GH11) protein that functions as a pathogen-associated molecular pattern (PAMP), triggering PAMP-triggered immunity (PTI) in Nicotiana benthamiana through the receptor kinases BAK1 and SOBIR1, independently of its enzymatic activity.

In silico designing and characterization of outer membrane protein (OmpC) gene from Salmonella enterica and its expression in Nicotiana tabacum for developing a plant-based vaccine against salmonellosis.

Salmonella, a gram-negative bacteria, is the leading cause of foodborne illness globally. Two serovars of Salmonella, S. enteritidis and S. typhimurium are responsible for the majority of human salmonellosis. Prolonged salmonellosis caused by Salmonella species leads to the development of colon cancer, which is 3rd most common cancer in the world. Porins in the outer membrane of Salmonella can be used to elicit immune response. The production of plant-based vaccine against salmonellosis and the subsequent colon cancer using outer membrane proteins can be helpful for the people of developing co

Intranasal vaccination with a plant-derived H5 HA vaccine protects mice and ferrets against highly pathogenic avian influenza virus challenge.

Highly pathogenic avian influenza H5N1 infection remains a public health threat and vaccination is the best measure of limiting the impact of a potential pandemic. Mucosal vaccines have the advantage of eliciting immune responses at the site of viral entry, thereby preventing infection as well as further viral transmission. In this study, we assessed the protective efficacy of hemagglutinin (HA) from the A/Indonesia/05/05 (H5N1) strain of influenza virus that was produced by transient expression in plants. The plant-derived vaccine, in combination with the mucosal adjuvant (3',5')-cyclic dimer

Coevolution of plant-microbe interactions, friend-foe continuum, and microbiome engineering for a sustainable future.

The coevolution of plant-microbe (PM) associations over approximately 450 million years has been a fundamental driver of terrestrial life, giving rise to mutualistic, commensal, and pathogenic relationships along a dynamic friend-foe continuum. The need to adapt to the host environment has driven the convergent evolution of common strategies among mutualists and pathogens, enabling them to evade or modulate the plant immune system. This review synthesizes PM coevolution within a deep-time, three-pillar framework: organellogenesis, root evolution, and immune gatekeeping, linking ancient endosym

Cyclic lipopeptides from Bacillus subtilis activate distinct patterns of defence responses in grapevine.

Non-self-recognition of microorganisms partly relies on the perception of microbe-associated molecular patterns (MAMPs) and leads to the activation of an innate immune response. Bacillus subtilis produces three main families of cyclic lipopeptides (LPs), namely surfactins, iturins and fengycins. Although LPs are involved in induced systemic resistance (ISR) activation, little is known about defence responses induced by these molecules and their involvement in local resistance to fungi. Here, we showed that purified surfactin, mycosubtilin (iturin family) and plipastatin (fengycin family) are p

The polymerase-associated factor 1 complex modulates the growth-defense tradeoff in Arabidopsis.

Plants have evolved sophisticated mechanisms to balance growth and defense. The evolutionarily conserved polymerase-associated factor 1 complex (PAF1C) plays multiple roles in transcription. Here, we show that PAF1C regulates the growth-defense tradeoff by repressing defense genes expression in Arabidopsis. Loss of PAF1C leads to increased expression of defense genes, enhanced disease resistance, but compromised growth. Mechanistically, PAF1C binds to defense genes, where it interacts with histone deacetylases, such as HDA6, to promote histone deacetylation, thereby repressing defense genes ex
