Short Interspersed Nuclear Element (SINE) Sequences in the Genome of the Human Pathogenic Fungus Aspergillus fumigatus Af293.

Novel families of short interspersed nuclear element (SINE) sequences in the human pathogenic fungus Aspergillus fumigatus, clinical isolate Af293, were identified and categorised into tRNA-related and 5S rRNA-related SINEs. Eight predicted tRNA-related SINE families originating from different tRNAs, and nominated as AfuSINE2 sequences, contained target site duplications of short direct repeat sequences (4-14 bp) flanking the elements, an extended tRNA-unrelated region and typical features of RNA polymerase III promoter sequences. The elements ranged in size from 140-493 bp and were present in

The Potential of Noncoding RNAs-siRNAs in Cancer Research and Therapy: Challenges and Solutions.

Noncoding RNAs (ncRNAs) are RNA molecules, which play critical roles in regulating gene expression and cellular activities, influencing processes like differentiation, proliferation, and cell survival, unlike messenger RNAs (mRNAs) that act as templates for protein production. Examples of ncRNAs include small interfering RNAs (siRNAs), microRNAs (miRNAs), ribosomal RNAs, transfer RNAs, small nucleolar RNAs (sno RNAs), and small nuclear RNAs (snRNAs). Small interfering RNAs (siRNAs) are a type of noncoding RNA that function primarily by silencing specific genes through a process called RNA inte

Roles of exosomal non-coding RNAs in osteoarthritis.

Osteoarthritis (OA) is increasingly recognized as a chronic low-grade inflammatory joint disorder characterized by progressive cartilage degeneration, synovial inflammation, subchondral bone remodeling, and disrupted intercellular communication. Growing evidence indicates that exosomal non-coding RNAs (ncRNAs), including microRNAs, long non-coding RNAs, and circular RNAs, are key regulators of the OA microenvironment. By mediating intercellular signal transfer among chondrocytes, synovial fibroblasts, macrophages, osteoblasts, and osteoclasts, exosomal ncRNAs influence inflammatory mediator pr

Multiple Targeting of HSP Isoforms to Challenge Isoform Specificity and Compensatory Expression.

Heat shock proteins (HSPs) are molecular chaperones that assist in protein folding, trafficking, and metabolism. Intracellular chaperone functions of HSPs had been well-investigated, but extracellular and exosomal HSPs have been recently found. Exosomal HSPs are intercellularly transferred, while extracellular HSPs play cytokine-like roles called chaperokines. We have shown that exosomal HSPs play key roles in intercellular communication between tongue carcinoma and tumor-associated macrophages in the tumor microenvironment. Notably, HSP90 isoforms consist of HSP90alpha, HSP90beta, mitochondri

Pancreatic β-cell tRNA hypomethylation and fragmentation link TRMT10A deficiency with diabetes.

Transfer RNAs (tRNAs) are non-coding RNA molecules essential for protein synthesis. Post-transcriptionally they are heavily modified to improve their function, folding and stability. Intronic polymorphisms in CDKAL1, a tRNA methylthiotransferase, are associated with increased type 2 diabetes risk. Loss-of-function mutations in TRMT10A, a tRNA methyltransferase, are a monogenic cause of early onset diabetes and microcephaly. Here we confirm the role of TRMT10A as a guanosine 9 tRNA methyltransferase, and identify tRNAGln and tRNAiMeth as two of its targets. Using RNA interference and induced pl

Temperature-sensitive steps in the biosynthesis of Venezuelan equine encephalitis virus.

In contrast to Eastern equine encephalitis virus, the replication of Venezuelan equine encephalitis (VEE) virus was strongly inhibited at 44 C in chick embryo cells. The inhibited steps were analyzed by shifting the incubating temperatures up or down, and by determining during the shifts the rate and extent of infectious ribonucleic acid (RNA) synthesis, intact virus synthesis, and formation of complement-fixing antigen or of antigen detectable by a direct fluorescent-antibody technique. The inhibition appeared to be due to two temperature-sensitive steps involved in the synthesis of VEE virus

[Studies on the biological action of uterine RNA in rat implantation (author's transl)].

With a view to investigating the mechanism of the action of estrogen involved in nidation, the biologically active rat uterine RNA thereby derived was extracted and then administered in the uterine cavity of castrated rats to observe the ensuing morphological changes in the endometrium and compare them with those resulting from the administration of decidual RNA which is non-estrogenised. There was a tendency toward increasing proliferation noted in the endometrial epithelium of the castrated rats 48 hours after the administration of the biologically active uterine RNA, as in the endometrial e

Electrospun PEO/gelatin nanofibers for pDNA and mRNA delivery.

Electrospun nanofiber mats are a versatile delivery system that can be applied locally at a target site, making them attractive for localized gene delivery. Although some studies reported electrospinning of nucleic acids, polyethylene oxide (PEO) was primarily used as a co-spinnable additive rather than as a standalone carrier in previous studies. In this study, PEO and gelatin nanofiber mats were prepared via blend and coaxial electrospinning to encapsulate pDNA and mRNA polyplexes prepared with a commercial transfection reagent. Notably, mRNA complexes were directly encapsulated within elect

Bacterial Schlafen proteins mediate phage defence.

Human Schlafen proteins restrict viral replication by cleaving tRNA, thereby suppressing protein synthesis. Although the ribonuclease domain of Schlafen proteins is conserved across all domains of life, its function in prokaryotes has remained unclear. Here we demonstrate that prokaryotic Schlafen nucleases are widespread antiviral effectors that protect bacteria from bacteriophages and are fused to a diverse array of phage-sensing domains. We expressed seven Enterobacterales Schlafen systems in Escherichia coli, identifying two that confer defence against coliphages. We focused on a system wh

Complexation of Polypeptides and Polypeptoids with Nucleic Acids: Does Chirality Matter for Salt Stability and Morphology?

Liquid-liquid phase separation (LLPS), especially via coacervation, offers a novel drug delivery strategy by encapsulating therapeutic agents within phase-separated droplets, thereby improving stability, solubility, and controlled release. Polypeptides and polypeptoids are ideal biomaterials for these systems due to their versatility and tunable properties. Polypeptoids are particularly advantageous, offering enhanced enzymatic resistance and greater control over molecular interactions, making them suitable for complexation studies. This research explores the binary complexation of L-, D,L-, a

Nontoxic mRNA vehicles based on electrically neutral self-assembling peptides.

The cationic molecules and low-molecular-weight amphiphiles commonly used in nonviral gene delivery vehicles often induce adverse nonspecific effects, notably cytotoxicity and detergent-like unfavourable membrane activity. To address these limitations, here we report the development of a nontoxic mRNA-condensing agent based on electrically neutral self-assembling peptides (ENSPs). Driven by cooperative hydrogen bonding and hydrophobic interactions, ENSPs coassemble with mRNA into discrete nanotube-shaped complexes. ENSP-based mRNA vehicles are almost nontoxic to mammalian cells and exhibit neg

Self-Amplifying mRNA Enhances Transamniotic Fetal mRNA Delivery.

Intraamniotic injection of encapsulated linear mRNA routes to the fetus after placental uptake but has a limited half-life and translation. Self-amplifying mRNA (saRNA) is a synthetic mRNA containing a replicase sequence and gene of interest (GOI) with replicase-targeting motifs that allow for the amplification of both the full saRNA and GOI. We compared the fetal distribution of saRNA and linear mRNA with the same GOI after intraamniotic injections in a rat model. Fetuses (n = 99) from seven Sprague-Dawley dams were divided into three groups based on volume-matched intraamniotic injections pe

Continuous in situ synthesis of a complete set of tRNAs sustains steady-state translation in a recombinant cell-free system.

Construction of a self-regenerating biochemical system is critical for building a synthetic cell. An essential step in building a self-regenerative system is producing a complete set of tRNAs for translation, which remains a significant challenge. Here, we reconstitute a complete set of 21 in vitro transcribed tRNAs and optimize their abundance to improve protein yield. Next, we show that protein expression in the PURE transcription-translation system can be achieved by in situ transcribing tRNAs from 21 linear tRNA templates or a single plasmid template. To enable synthesis of mature tRNAs fr

Serum tRF-18-HR05X6D2 may serve as a promising potential diagnostic biomarker for gastric cancer.

Gastric cancer (GC) presents with insidious early symptoms that lack distinctiveness. Coupled with insufficient screening awareness and limited diagnostic methods, these symptoms are difficult to distinguish from manifestations of benign conditions including gastritis and gastric ulcers, leading patients to seek medical attention only after advanced-stage symptoms develop. Traditional serum markers exhibit low positivity and specificity in early-stage GC; therefore, actively identifying novel molecules for combined diagnosis in conjunction with traditional biomarkers proves essential for the e

tiRNA-Gly-GCC-001 in major depressive disorder: Promising diagnostic and therapeutic biomarker.

BACKGROUND AND PURPOSE: In major depressive disorder (MDD), exploration of biomarkers will be helpful in diagnosing the disorder as well as in choosing a treatment and predicting the treatment response. Currently, tRNA-derived small ribonucleic acids (tsRNAs) have been established as promising non-invasive biomarker candidates that may enable a more reliable diagnosis or monitoring of various diseases. Herein, we aimed to explore tsRNA expression together with functional activities in MDD development. EXPERIMENTAL APPROACH: Serum samples were obtained from patients with MDD and healthy contro

Mitochondrial tRNA variants in 811 Chinese probands with Leber's hereditary optic neuropathy.

Leber's hereditary optic neuropathy (LHON) is the maternal inheritance of eye disorder. LHON-linked mitochondrial DNA (mtDNA) mutations affect the ND1, ND4 or ND6 genes encoding essential subunits of complex I. However, the role of mitochondrial tRNA defects in the pathogenesis of LHON is poorly understood. In this report, Sanger sequence analysis of 22 mitochondrial tRNA genes identified 139 variants in a cohort of 811 Han Chinese probands and 485 control Chinese subjects. Among these, 32 (4 known and 28 novel/putative) tRNA variants in 71 probands may contribute to pathogenesis of LHON, as t

Historical Roles of Selenium and Selenoproteins in Health and Development: The Good, the Bad and the Ugly.

Selenium is a fascinating element that has a long history, most of which documents it as a deleterious element to health. In more recent years, selenium has been found to be an essential element in the diet of humans, all other mammals, and many other life forms. It has many health benefits that include, for example, roles in preventing heart disease and certain forms of cancer, slowing AIDS progression in HIV patients, supporting male reproduction, inhibiting viral expression, and boosting the immune system, and it also plays essential roles in mammalian development.
