Evaluation of CRISPR/CasRx-Mediated VEGF mRNA Knockdown in Mouse Retina.

Neovascular eye diseases (NEDs) are a group of diseases caused by the abnormal overgrowth of blood vessels in the eye. Normal vasculature is maintained through a dynamic balance of the vascular endothelial growth factor (VEGF). In diseases such as advanced diabetic retinopathy (DR) and neovascular age-related macular degeneration (AMD), an overexpression of VEGF leads to the formation of structurally weak and leaky blood vessels, resulting in vision impairment and, without intervention, legal blindness. In the clinic, NEDs are presently managed through anti-VEGF agents that specifically bind a

Metabolic Syndrome Predisposes Ossabaw Minipig Retina to an Early Neurodegenerative Milieu.

The miniature (mini) Ossabaw pigs are proposed as a translational preclinical model for testing and developing novel therapeutics for human diseases, including cystic fibrosis, cancer, and metabolic syndrome (MetS). In recent years, pigs have gained similar attention for studying retinal abnormalities and disorders owing to their close resemblance in size, anatomy, vasculature, and pathology to the human eye compared with their rodent counterparts. In our previous study, Ossabaw minipigs fed a Western diet for 10 weeks and followed for 3.5 months exhibited early signs of retinal degeneration a…

Dissociating retina- and face-centered reference frames of human face recognition.

Face identity recognition is a core function of the human brain, predominantly driven by horizontally oriented facial cues. While face proce … ormance should stay tuned to the horizontal irrespective of rotation angle. Any contribution of the retina-centered reference frame should cause the peak of orientation tuning to shift away from the horizontal, in the same direction as the stimulus rotation. While overall identity recognition performance decreased when faces were rotated by ±90° compared …

Introducing a Porcine Inflammatory Ex Vivo Retina Model for Diabetic Retinopathy.

This study aimed to develop an ex vivo retinal model to examine inflammatory processes in diabetic retinopathy (DR) without animal testing. Porcine eyes were collected from a local abattoir, dissected, and cultivated for four days in five experimental groups: control group (Co), 25 mM and 50 mM mannitol groups (Man25, Man50) as osmotic controls, and 25 mM and 50 mM glucose groups (Glc25, Glc50) as diabetic groups. A TUNEL assay was used to determine relative cell death. Immunofluorescence and quantitative real-time polymerase chain reaction (qRT-PCR) were performed to detect inflammatory marke…

Retina-Inspired 2D Semiconductor NIR Sensor with PRO Architecture for Photodetection.

Light detection and ranging (LiDAR) technology is critical for autonomous driving, which relies on abundant near-infrared (NIR) photodetectors to accurately capture three-dimensional spatial information. CMOS-based LiDAR detection systems are less efficient with respect to computational efficiency, inherently limited by their von Neumann architecture, leading to high latency and significant computational demands. Retina-inspired neuromorphic sensors integrated from sensing and computation present a promising alternative but need additional analog-to-digital converters (ADCs). Here we present t…

Microglia activation and migration in retina during acute high-altitude exposure.

High-altitude retinopathy (HAR), caused by hypobaric hypoxia, leads to retinal dysfunction. However, its pathogenic mechanism remains elusive. Microglia, the resident immune cells of the retina, play crucial roles in various retinal disorders. Here, we investigated the functional alterations and underlying mechanisms of microglial activation in mouse models exposed to high-altitude (5000m HAE). Hematoxylin and eosin (H&E) staining showed edema in the entire, inner, and outer retinal layers after high-altitude exposure (HAE). Electroretinogram (ERG) testing revealed impaired retinal function un…

The Developing Retina Undergoes Mitochondrial Remodeling via PINK1/PRKN-dependent Mitophagy.

Mitophagy, the selective degradation of mitochondria, is essential for retinal ganglion cell (RGC) differentiation and retinal homeostasis. However, the specific mitophagy pathways involved and their temporal dynamics during retinal development and maturation remain poorly understood. Using proteomics analysis of isolated mouse retinas across developmental stages and the mitophagy reporter mouse line, mito-QC, we characterized mitophagy throughout retinogenesis. While mitolysosomes were more prevalent in the mature retina, we observed two distinct mitophagy peaks during embryonic development. …

Nanowire-based artificial photoreceptors with multi-hyperuniformity inspired by chicken retina.

Retinal cells of chicken possess a hyperuniform distribution enabling exceptional spatial sampling. Recently, interest has grown in mimicking biological imaging systems for optoelectronics technology. However, since biological retinal cells and current imaging pixels have a significant scale mismatch such as super-micron and sub-micron, the applicability of hyperuniform distribution must be investigated at sub-micron scale, considering wave-optics phenomena. In this study, we present theoretical analyses and design strategies for nanowire-based artificial photoreceptors with hyperuniformity. F…

Primate retina trades single-photon detection for high-fidelity contrast encoding.

How the spike output of the retina enables human visual perception is not fully understood. Here, we address this at the sensitivity limit of vision by correlating human visual perception with the spike outputs of primate ON and OFF parasol (magnocellular) retinal ganglion cells in tightly matching stimulus conditions. We show that human vision at its ultimate sensitivity limit depends on the spike output of the ON but not the OFF retinal pathway. Consequently, nonlinear signal processing in the retinal ON pathway precludes perceptual detection of single photons in darkness but enables quantal…

Isolation and Culture of Primary Retinal Ganglion Cells from Rodent Retina.

Primary retinal ganglion cell (RGC) cultures are widely used for evaluating the neuroprotective and neurogenic effects of candidate compounds. The axons of RGCs make up the optic nerve and are responsible for transmitting electrochemical signals to the brain. As the retina is an outgrowth of the brain, both it and the optic nerve are part of the central nervous system (CNS). In the process of culturing RGC, the eye and retina are dissected, meaning the RGC, disconnected from the brain, degenerate without intervention due to the traumatic damage they have endured. Therefore, this in vitro model…

Alpha retinal ganglion cells in pigmented mice retina: number and distribution.

Purpose: To identify and characterize numerically and topographically the population of alpha retinal ganglion cells (αRGCs) and their subtypes, the sustained-response ON-center αRGCs (ONs-αRGCs), which correspond to the type 4 intrinsically photosensitive RGCs (M4-ipRGCs), the transient-response ON-center αRGCs (ONt-αRGCs), the sustained-response OFF-center αRGCs (OFFs-αRGCs), and the transient-response OFF-center αRGCs (OFFt-αRGCs) in the adult pigmented mouse retina. Methods: The αRGC population and its subtypes were studied in flat-mounted retinas and radial sections immunodetected against…

Cellular mechanisms mediating activity-dependent extracellular space shrinkage in the retina.

Volume transmission plays an essential role in CNS function, with neurotransmitters released from synapses diffusing through the extracellular space (ECS) to distant sites. Changes in the ECS volume fraction (α) will influence the diffusion and the concentration of transmitters within the ECS. We have recently shown that neuronal activity evoked by physiological photic stimuli results in rapid decreases in ECS α as large as 10% in the retina. We now characterize the cellular mechanisms responsible for this ECS shrinkage. We find that block of inwardly rectifying K+ channels with Ba2+ , inhibit…
