The spontaneous unitary activity and the response to contralateral tone-burst were analyzed in the inferior colliculus (IC) of guinea pigs during the sleep-waking cycle and under the effects of pentobarbital anesthesia. Minor changes were observed in both spontaneous and evoked activity between wakefulness (W) and slow wave sleep (SWS). On the other hand, a consistent increase in the mean spontaneous firing rate and a significant decrement in the signal-to-noise ratio (S/N ratio) was observed during paradoxical sleep (PS). Pentobarbital anesthesia reduced the spontaneous and evoked firing rate

STUDY OBJECTIVES: We sought to elucidate hypotheses for the pathophysiology of idiopathic hypersomnia and discuss the mechanisms by which low-sodium oxybate (LXB) improves idiopathic hypersomnia symptoms.
METHODS: Published literature on idiopathic hypersomnia sleep abnormalities, symptoms, treatments, and underlying pathophysiology was reviewed.
RESULTS: Patients with idiopathic hypersomnia often show sleep architecture alterations, such as increased sleep efficiency and reduced slow-wave sleep, although the literature lacks consensus. The current understanding of pathophysiologic changes i

BACKGROUND: Sleep is a natural part of every individual's life. Delta sleep-inducing peptide (DSIP) is a nonapeptide that could promote sleep through the induction of slow wave sleep. However, little is known about the pharmacological effect of DSIP on insomnia.
OBJECTIVES: The main objective of this study was to analyze the pharmacological effect of DSIP on insomnia.
METHODS: We designed a fusion protein containing N-terminal TAT-based transduction domain followed by human serum albumin and DSIP and designated this protein as PHD fusion protein. The PHD fusion protein were expressed in Pich

OBJECTIVE: Evidence concerning whether sleep disturbances in older adults predict mortality is mixed. However, data are limited to self-reported sleep problems and may be confounded with other comorbidities. We examined whether electroencephalographic (EEG) sleep parameters predicted survival time independently of known predictors of all-cause mortality.
METHODS: A total of 185 healthy older adults, primarily in their 60s through 80s, with no history of mental illness, sleep complaints, or current cognitive impairment, were enrolled in one of eight research protocols between October 1981 and

OBJECTIVES: To describe how the neural generators of different sleep components can provoke seizure discharge propagation during NREM sleep and can suppress it during REM sleep.
METHODS: Experimental manipulations of discrete physiological components were conducted in feline epilepsy models (n=64), mostly in the systemic penicillin epilepsy model of primary generalized epilepsy and the amygdala kindling model of the localization-related seizure disorder, temporal lobe epilepsy. Procedures included seizure induction as well as quantifying norepinephrine concentrations (microdialysis) and the s

We reported a 13-year-old boy diagnosed as multiple sclerosis associated with narcolepsy. He had suffered from retrobulbar optic neuritis at the age of 11 years which was improved gradually by prednisolone. Four months later he was admitted because of excessive somnolence. The diagnosis of narcolepsy was made based on hypnagogic hallucination, sleep paralysis, changes of personality and the sleep onset sREM (SOREM). The elevation of anti-measles antibody and a positive oligoclonal band in CSF, low density areas in the bilateral internal capsule on CT scan, and high signal areas in the same reg

The electroencephalographic abnormalities seen in Landau-Kleffner syndrome (LKS) (language deterioration) are non-specific, and consist of a variety of epileptiform discharge patterns including continuous slow spike-wave discharges during sleep, focal sharp waves with spikes, and centrotemporal (rolandic) spikes. Similarly, the EEG abnormalities seen in autistic epileptiform regression (language and social/behavioral deterioration) are non-specific and overlap with those seen in LKS. By contrast, distinct epilepsy syndromes in otherwise normal children occur in the EEG-defined benign focal epi

As we age, we experience changes in our nighttime sleep and daytime wakefulness. Individuals afflicted with Alzheimer's disease (AD) can develop sleep problems even before memory and other cognitive deficits are reported. As the disease progresses and cognitive changes ensue, sleep disturbances become even more debilitating. Thus, it is imperative to gain a better understanding of the relationship between sleep and AD pathogenesis. We postulate a bidirectional relationship between sleep and the neuropathological hallmarks of AD; in particular, the accumulation of amyloid-β (Aβ) and tau. Our re

Post-traumatic sleep-wake disturbances are common after acute traumatic brain injury. Increased sleep need per 24 h and excessive daytime sleepiness are among the most prevalent post-traumatic sleep disorders and impair quality of life of trauma patients. Nevertheless, the relation between traumatic brain injury and sleep outcome, but also the link between post-traumatic sleep problems and clinical measures in the acute phase after traumatic brain injury has so far not been addressed in a controlled and prospective approach. We therefore performed a prospective controlled clinical study to exa

Disturbances of sleep are typical for most depressed patients and belong to the core symptoms of the disorder. Since the 1960s polysomnographic sleep research has demonstrated that besides disturbances of sleep continuity, depression is associated with altered sleep architecture, i.e., a decrease in slow wave sleep (SWS) production and disturbed rapid eye movement (REM) sleep regulation. Shortened REM latency (i.e., the interval between sleep onset and the occurrence of the first REM period), increased REM sleep duration and increased REM density (i.e., the frequency of rapid eye movements per

AIMS: Sex differences in the prevalence of chronic sleep disorders (CSD), including insomnia, have been documented. Questionnaire-based surveys generally report a higher prevalence in women, whereas polysomnographic assessments often indicate a higher prevalence in men. To elucidate sex differences in sleep disturbances and associated impairments in glucose metabolism, we employed a mouse model of CSD induced by psychophysiological stress.
METHODS: We evaluated the effects of one week of CSD under normal chow on activity and sleep-wake rhythms and six weeks of CSD under a high-fat high-sucros

BACKGROUND: Sleep disturbances are highly prevalent in depressive episodes and are linked to higher mood severity and suicidal behaviors. Slow wave sleep (SWS) and REM sleep are compromised in depression. Current evidence suggests that rapid antidepressant effects of intravenous (IV) ketamine in patients with treatment-resistant depression (TRD) is mediated by its effects on SWS and REM sleep. Sleep phenotypes may help predict ketamine response.
METHOD: In this observational study, we investigated differences in rates of response among sleep phenotypes defined by QIDS-SR in a cohort of patien

BACKGROUND: The administration of certain sedatives has been shown to promote sleep in humans. Related agents induce sleep-like behavior when administered to horses. Interpretation of electroencephalograms (EEGs) obtained from sedated horses should take into account background activity, presence of sleep-related EEG events, and the animal's behavior.
HYPOTHESIS: Sedatives induce states of vigilance that are indistinguishable on EEGs from those that occur naturally.
ANIMALS: Six healthy horses.
METHODS: Digital EEG with video was recorded after administration of 1 of 4 sedatives (acepromazin

AIMS: The type 2 muscarinic receptor (M2R) differs from the other G-protein-coupled muscarinic receptor (type 4, or M4R) in tissue distribution and physiologic effects. We studied the impact of these receptors on sleep and arousal by using M2R and M4R knock-out (KO) mice.
MAIN METHODS: M2R and M4R KO and genetically intact mice were compared in terms of normal patterns of sleep, responses to sleep loss, infectious challenge and acoustic startle, and acoustic prepulse inhibition of startle (PPI).
KEY FINDINGS: Under basal conditions, M2R and M4R KO mice do not differ from the background strai

We report on two girls and one boy with clinical and electroencephalographic features of late-onset childhood epilepsy with occipital paroxysms of the "Gastaut type", showing an unusual evolution. Neurological examination and brain imaging were normal in all three. At the age of 7.5 years, eight years and ten years respectively, the three children presented with episodes of visual symptoms when awake, and in one of them, the seizures were occasionally followed by oculocephalic deviation. The interictal EEG showed bilateral occipital spike-wave activated by eye closing. In two patients, the occ

RATIONALE: The nuclear receptor retinoid X receptor (RXR) belongs to a nuclear receptor superfamily that modulates diverse functions via homodimerization with itself or several other nuclear receptors, including PPARα. While the activation of PPARα by natural or synthetic agonists regulates the sleep-wake cycle, the role of RXR in the sleep modulation is unknown.
OBJECTIVES: We investigated the effects of bexarotene (Bexa, a RXR agonist) or UVI 3003 (UVI, a RXR antagonist) on sleep, sleep homeostasis, levels of neurochemical related to sleep modulation, and c-Fos and NeuN expression.

BACKGROUND: Thalamocortical interactions play a critical role in sleep oscillatory activities. However, in individuals with insomnia disorder (ID), the structural organization of these circuits, their relationship with sleep oscillatory coupling, and their potential modulation by repetitive transcranial magnetic stimulation (rTMS) remain unclear.
METHODS: In Study 1, we recruited 62 ID patients and 62 healthy controls and assessed white matter tract strength between thalamic subregions and cortical areas using probabilistic tractography. In Study 2, we administered 20 sessions of 1 Hz rTMS (a

Aging is associated with a variety of physiological changes originating peripherally and centrally, including within the autonomic nervous system. Sleep-wake disturbances constitute reliable hallmarks of aging in several animal species and humans. Recent studies have been interested in N-acetylmannosamine (ManNAc) a potential therapeutic agent for improving quality of life, as well as preventing age-related cognitive decline. In this study, ManNAc (5.0 mg/ml) was administered in the drinking water of middle-aged male C57BL/6J mice (55 weeks old) for 7 days. Mice were housed under a 12:12 h lig

Insomnia affects 10-30 % of adults, with higher prevalence among older individuals, women, and those with comorbid medical or psychiatric conditions. It disrupts cognitive function, emotional regulation, and physical health, elevating risks for depression, cardiovascular disease, and diminished quality of life. Although pharmacological treatments have offered short-term symptom relief, their use has been constrained by side effects, dependency potential, and limited long-term efficacy, underscoring the need for effective non-pharmacological alternatives. Exercise has emerged as a well-supporte
