Highly specific and sensitive anti-Strongyloides venezuelensis IgY antibodies applied to the human strongyloidiasis immunodiagnosis.

Due to the epidemiological problem of the neglected condition of human strongyloidiasis, rapid and effective diagnosis is extremely important, with the development of new diagnostic tools being essential to reduce infections and chronic cases. Avian immunoglobulin Y (IgY) technology is an alternative for antibody production that has high specificity and profitability. This study aimed to produce and fractionate IgY antibodies from the egg yolks of hens that were immunized with the total antigenic extracts of Strongyloides venezuelensis infectious filariform larvae (iL3) and parthenogenetic fem…

Strongyloidiasis in humans: diagnostic efficacy of four conventional methods and real-time polymerase chain reaction.

INTRODUCTION: Strongyloides stercoralis is an intestinal parasitic nematode that causes hyperinfection and/or a dissemination syndrome in hosts, which is often difficult to diagnose. This study aims to compare the diagnostic efficacy of four conventional methods used to diagnose strongyloidiasis with real-time polymerase chain reaction (qPCR) to detect S. stercoralis in fecal samples. METHODS: We analyzed 143 fecal samples collected from Colombian regions with varying degrees of risk for intestinal infections caused by S. stercoralis to assess the validity, performance, overall efficiency, an…

Metaperiodate deglycosylation of Strongyloides venezuelensis larvae: Immunochemical characterization and antigen production for human strongyloidiasis diagnosis.

Strongyloidiasis is an important helminthiasis affecting million people worldwide. The aim of this study was to use sodium metaperiodate (MP) treatment to immunochemically characterize Strongyloides venezuelensis filariform larvae and use MP-treated heterologous antigen to detect IgG and subclasses in serum. Samples from individuals with definitive diagnosis of strongyloidiasis (n = 50), other parasitic diseases (n = 60) and negative endemic (n = 50) were tested. TG-ROC and two-way ANOVA were applied. MP-treatment resulted on differential localization of carbohydrates at larval structure and n…

IMMUNODIAGNOSIS OF HUMAN STRONGYLOIDIASIS: USE OF SIX DIFFERENT ANTIGENIC FRACTIONS FROM Strongyloides venezuelensis PARASITIC FEMALES.

The aim of this study was to evaluate six different antigenic fractions from Strongyloides venezuelensis parasitic females for the immunodiagnosis of human strongyloidiasis. Soluble and membrane fractions from S. venezuelensis parasitic females were prepared in phosphate-buffered saline (SSF and SMF, respectively), Tris-HCl (TSF and TMF, respectively), and an alkaline buffer (ASF and AMF, respectively). Serum samples obtained from patients with strongyloidiasis or, other parasitic diseases, and healthy individuals were analyzed by enzyme-linked immunosorbent assay (ELISA). Soluble fractions SS…

Strongyloidiasis in a young French woman raises concern about possible ongoing autochthonous transmission in Spain.

Strongyloidiasis is one of the most common geohelminth infections in tropical and subtropical areas. Accurate diagnosis remains challenging, leading to an overall underestimation of strongyloidiasis prevalence. The possibility of ongoing autochthonous transmission in some temperate areas and especially in southern Europe is still debated, and data supporting this hypothesis are scarce. The case of a young French woman, who had travelled frequently to Spain and had acquired Strongyloides stercoralis infection as revealed by gastrointestinal symptoms and hypereosinophilia, is reported here. Phys…

[Human T-lymphotropic virus 1 (HTLV-1), strongyloidiasis and scabies. Infections and associations to considerate].

Human T-lymphotropic virus 1 (HTLV-1) is classified into the family of Retroviridae and preferentially causes a chronic infection of CD4+ T cells. Most people infected with HTLV-1 remain asymptomatic. However, less than 105 of infected individuals eventually develop any of the associated severe diseases such as inflammatory syndromes, neoplastic diseases and opportunistic infections including Strongyloides stercoralis and Sarcoptes scabiei hyperinfections. Recently, it has been described that a weak Th2 cell-mediated response is associated with HTLV-1 infection and Strongyloides hyperinfection

[Retrospective study of 1,934 cases of strongyloidiasis in Paris (1970-1986). II. Diagnosis, Eosinophilia. Treatment].

The best methods of detection of strongyloidiasis were by "combined concentrations" described in 1965 and 1972: a faecal sample was concentrated with or without ether, then floating. Baermann's extraction was positive in only 80% of the cases. The average blood eosinophilia was higher in Europeans (16.4%) than in subjects from intertropical countries (13.4%). In West Indians; it was higher in cases of strongyloidiasis than in ankylostomiasis (9.4%) and intestinal bilharziasis (8.3%). Treatment with thiabendazole: overall efficacy in 621 patients was 89%. The most effective dosage (97%) with th…

Case Report: From chronic infection to disseminated strongyloidiasis: a case of corticosteroid-induced hyperinfection syndrome.

BACKGROUND: Strongyloides stercoralis (S. stercoralis) is a soil-transmitted nematode predominantly endemic to tropical and subtropical regions, capable of causing life-threatening hyperinfection syndrome and disseminated infection in immunocompromised individuals. With the improvement of sanitation conditions, strongyloidiasis has become increasingly uncommon. Early-stage infection presents with mild, nonspecific symptoms, posing diagnostic challenges. CASE DESCRIPTION: We report a case of S. stercoralis infection in a 73-year-old female patient, evolving from initial erythematous rash to di…

Improving Molecular Epidemiological Surveillance of Strongyloidiasis Upon Differentiation of Strongyloides fuelleborni fuelleborni From Strongyloides stercoralis.

Molecular epidemiological surveillance for zoonotic strongyloidiasis is confounded by a genus-specific TaqMan probe assay that conflates Strongyloides fuelleborni fuelleborni with Strongyloides stercoralis. To improve surveillance, we developed and validated a novel duplex species-specific TaqMan probe assay, screening a representative collection of available clinical samples. Our assay was highly discriminatory, evidencing no cross-reactivity, and had a lowest DNA detection limit of 1 pg/µL. However, as the genus-specific DNA assay has greater detection ability, we propose a 2-step protocol w…

Improved diagnostic sensitivity of human strongyloidiasis using point-of-care mixed recombinant antigen-based immunochromatography.

Strongyloidiasis is a neglected tropical disease that can cause fatal complications due to hyperinfection and disseminated strongyloidiasis in immunocompromised patients. We used two Strongyloides stercoralis recombinant antigenic proteins, L3NieAg.01 (NIE) and IgG-immunoreactive antigen (SsIR), to develop the recombinant antigen-based immunochromatography test (ICT) kit. We constructed and compared kits using either the NIE (NIE ICT kit) or the SsIR (SsIR ICT kit) antigens and a kit using a mixture of both (NIE-SsIR ICT kit) for detection of anti-Strongyloides IgG antibody in human serum samp…

Fatal Outcome of Disseminated Strongyloidiasis despite Detectable Plasma and Cerebrospinal Levels of Orally Administered Ivermectin.

Strongyloides stercoralis affects over 100 million people worldwide. Those people most susceptible to infection are those with an immunocompromising condition, such as cancer or human immunodeficiency virus (HIV). Local disease may spread throughout the body of the host, causing a condition termed disseminated strongyloidiasis. Standard treatment for Strongyloides stercoralis infection is oral ivermectin. We describe a patient with chronic lymphocytic leukemia diagnosed with disseminated strongyloidiasis two weeks after initial presentation. After repeated dosing of oral ivermectin with no cli…

Strongyloidiasis associated with human T-cell lymphotropic virus type I infection in a nonendemic area.

Concomitant strongyloidiasis and human T-cell lymphotropic virus type I (HTLV-I) infection has been reported from areas in Japan where both organisms are endemic. We present four cases of concomitant infection with these organisms from an area that is not endemic for Strongyloides stercoralis. Three of the four patients had adult T-cell leukemia, an aggressive neoplasm resulting from HTLV-I infection, while the other was an asymptomatic carrier of HTLV-I. Three of the patients had spent their childhoods in an endemic location for both organisms, suggesting an initial infection at that time. Th…

Is eosinophilia a reliable diagnostic clue for chronic strongyloidiasis? a case series from Khuzestan Province, Iran.

Strongyloidiasis, a disease caused by Strongyloides stercoralis (S. stercoralis) nematode, can present in acute, chronic, hyperinfection, and disseminated forms. It is of particular important due to its high mortality rate, especially among immunocompromised patients, and the absence of a gold standard diagnostic method. Eosinophilia, as a key sign, can help in the diagnosis of the infection. This report presents five cases of strongyloidiasis who referred to the 17 Shahrivar Hospital, Abadan County, Iran, for various reasons, including cardiovascular diseases, elective surgeries, and routine …

Strongyloidiasis in a heart transplant recipient: Exploring the use of veterinary-formulated subcutaneous ivermectin and a review of the literature.

Strongyloides stercoralis can complete its entire life cycle within the human host, allowing infections to persist for years. While it may cause chronic, often asymptomatic infection in immunocompetent individuals, it can lead to life-threatening hyperinfection syndrome or disseminated disease in those who are immunocompromised. We report a case of Strongyloides hyperinfection syndrome in a heart transplant recipient who had tested negative on conventional serologic screening as part of his pretransplant workup three years prior to transplantation. The patient developed severe disease characte
