The essential role of heparan sulfate in the entry of PDCoV and other porcine coronaviruses.

Porcine enteric coronaviruses, including porcine deltacoronavirus (PDCoV), porcine epidemic diarrhea virus (PEDV), swine acute diarrhea syndrome coronavirus (SADS-CoV), and transmissible gastroenteritis coronavirus (TGEV), can cause acute diarrhea, vomiting, dehydration, and high mortality in suckling piglets. Recent studies revealing human PDCoV infections and the potential of SADS-CoV to penetrate human cell lines have heightened apprehensions about the zoonotic transmission risks of these viruses. While heparan sulfate (HS) serves as a receptor in PDCoV binding, the key host genes involved

Sulfation Attenuates Mitigating Effects of 25-Hydroxycholesterol on Postmenopausal Bone Loss.

Postmenopausal osteoporosis is a prevalent skeletal disorder with an unmet need for safer therapeutic strategies. While certain oxysterols show osteoinductive potential, the role of 25-hydroxycholesterol (25HC) in bone homeostasis remains unclear. Furthermore, 25HC can undergo sulfation, a largely underexplored chemical modification, to form 25HC3S. This study aimed to investigate the effects of 25HC and 25HC3S on bone loss and the underlying mechanisms. We found that treatment with 25HC, but not 25HC3S, ameliorated bone loss in an ovariectomized (OVX) mouse model of osteoporosis, as evidenced

Sulfated glycosaminoglycans inhibit LCMV entry and modulate antiviral immunity and pathology.

Viral infections remain a major challenge due to the limited availability and efficacy of current treatments. Existing antivirals primarily target viral replication but are often virus-specific and can lead to drug resistance. Sulfated glycosaminoglycans (GAGs) have emerged as promising broad-spectrum agents that block viral binding and entry into host cells. Here, we show that highly sulfated GAGs restrict the infectivity of both pathogenic and non-pathogenic Arenaviruses. Using the lymphocytic choriomeningitis virus (LCMV) model, we demonstrate that GAG exposure reduces viral entry and infec

Influence of alkali treatment and ultrasonication on the bioactivities and rheological properties of sulfated polysaccharides.

The rheological, antioxidant and anticoagulant properties of 17 different structurally uniform and hybrid sulfated polysaccharides are presented in this study. Alkali treatment was used to modify carrageenan precursors, while ultrasonication was employed to partially depolymerize polysaccharide chains. Native and modified sulfated polysaccharides with well-resolved molecular structures were characterized by size exclusion chromatography and dynamic rheometry methods. Extensive ultrasonication (1-64 min) reduced the molecular weights of the samples from approximately 384-1156 kDa to 47-95 kDa.

Acute Hypercalcemia Resulting From Antibiotic-Impregnated Calcium Sulfate Beads.

An 86-year-old man with a history of chronic kidney disease sustained a hip fracture after a mechanical fall, which eventually led to a total hip arthroplasty (THA). During this THA, he was found to have a prosthetic joint infection and underwent debridement, antibiotics, and implant retention, where calcium sulfate beads (CSBs) were placed. He subsequently developed hypercalcemia, requiring intravenous fluids, diuretics, and calcitonin for treatment. This case highlights the development of hypercalcemia after the placement of CSBs, which is a known, although uncommon, side effect.

Influence of magnesium sulfate on the pharmacodynamic characteristics of rocuronium. A randomized clinical trial.

BACKGROUND: As a multimodal anesthesia adjuvant, magnesium sulfate (MgSO4) plays an important role in the anesthetic arsenal, due to its properties and substantial synergistic effects with other drugs such as opioids, hypnotics and neuromuscular blocking drugs (NMBD). Rocuronium is a non-depolarizing NMBD used widely in general anesthesia, and its association with MgSO4 is still a concern. This study aimed to evaluate the influence of MgSO4 at a dose of 30 mg kg-1 on the pharmacodynamic characteristics of rocuronium. METHODS: It was a double-blinded, randomized controlled trial in adult femal

Aromatic Pentaamide Macrocycles as High-Affinity Sulfate Binders and Efficient Transporters.

The binding and transport of the highly hydrophilic sulfate ion pose a significant challenge. Receptors capable of both strong sulfate binding and efficient transport are rare. Macrocycles c5, featuring a constrained aromatic amide backbone that convergently locks multiple amide NH and phenyl NH donors, along with five large amide dipoles, create a preorganized, electropositive cavity that binds sulfate in a 1:1 stoichiometry, with affinities (Ka's) from 108 to 1013 M-1 in solvents of low to high polarity. The water-soluble c5c binds sulfate, forming a 4:1 complex in aqueous solution. Macrocyc

Elevated Dehydroepiandrosterone Sulfate Levels in 2 Patients With Prolactinomas: An Underrecognized Association.

While adrenal androgen production is primarily regulated by ACTH, some data suggest a less common association between hyperprolactinemia and elevated dehydroepiandrosterone sulfate (DHEA-S). We describe 2 patients with this underrecognized connection. Patient 1, a 60-year-old male, was incidentally found to have bilateral adrenal masses on computed tomography during evaluation of gastrointestinal complaints. Further workup revealed elevated levels of DHEA-S and prolactin, with pituitary macroadenoma identified as the cause, given normalization of both prolactin and DHEA-S on cabergoline therap

Indoxyl sulfate: clinical implications for anemia management in chronic kidney disease.

PURPOSE OF REVIEW: This review examines the role of indoxyl sulfate, a gut-derived uremic toxin, in the development of anemia in chronic kidney disease. It dissects the cellular and biochemical mechanisms through which indoxyl sulfate suppresses erythropoietin production, disrupts iron metabolism, and promotes oxidative stress and inflammation. RECENT FINDINGS: Indoxyl sulfate interferes directly with the hypoxia-inducible factor pathway, thereby reducing the transcriptional activation of erythropoietin. In parallel, indoxyl sulfate-induced oxidative stress damages red blood cells and acceler

Cyanobacterial Ampholyte Hydrogels Developed by the Cationization of Sulfated Polysaccharides and Their Cell-Compatibility.

Sacran is a cyanobacterial supergiant polysaccharide with carboxylate and sulfate groups that exhibits antiallergic and antiinflammatory properties. However, its high anionic functions restrict cell compatibility. Quaternary ammonium groups were substituted to form sacran ampholytes, and the cell compatibility of the cationized sacran hydrogels was evaluated. The cationization process involved the reaction of N-(3-chloro-2-hydroxypropyl)trimethylammonium chloride with the primary amine or hydroxyl groups of sacran. The degree of cationization ranged from 32 to 87% for sugar residues. Hydrogels

Incorporation of Silver into Sulfate Groups Enhances Antimicrobial and Antiviral Effects of Fucoidan.

The COVID-19 pandemic has significantly impacted our daily lives. Routine infection-control measures present an effective preventive strategy for a new infectious disease outbreak. Fucoidan, a fucose-rich sulfated polysaccharide found in brown algae, exhibits antiviral activity. Moreover, fucoidan exerts an antimicrobial effect; however, it requires considerably higher concentrations than those needed for its antiviral effect. In this study, we aimed to enhance the antimicrobial activity of fucoidan and prepared a fucoidan silver salt (Ag-Fuc) by incorporating silver ions into the sulfate grou

A comprehensive review of sulfated fucan from sea cucumber: Antecedent and prospect.

Sulfated fucan from sea cucumber is mainly consists of L-fucose and sulfate groups. Recent studies have confirmed that the structure of sulfated fucan mainly consists of repeating units, typically tetrasaccharides. However, there is growing evidence indicating the presence of irregular domains with heterogeneous units that have not been extensively explored. Moreover, as a key contributor to the nutritional benefits of sea cucumbers, sulfated fucan demonstrates a range of biological activities, such as anti-inflammatory, anticancer, hypolipidemic, anti-hyperglycemic, antioxidant, and anticoagu

Preparation and structural characterization of a sulfated octasaccharide with heparin-like anticoagulant activity.

Heparins are sulfated polysaccharides with a heterogeneous mixture derived from animal tissues, subject to supply limitations and the risk of animal virus residues. Patients using heparin also face the risks of spontaneous bleeding and thrombocytopenia. Here we reported an efficient riclinoctaose-based approach for rapid chemical synthesis of a structurally defined heparin-like anticoagulant sulfated octasaccharide (SRO). We used sulfur trioxide-pyridine, sulfur trioxide-trimethylamine, and sulfur trioxide-triethylamine complexes as solvents for one-pot O-sulfation and determined the optimal c

A Comprehensive Review on Iron-Based Sulfate Cathodes for Sodium-Ion Batteries.

Sodium-ion batteries (SIBs) are advantageous for large-scale energy storage due to the plentiful and ubiquitous nature of sodium resources, coupled with their lower cost relative to alternative technologies. To expedite the market adoption of SIBs, enhancing the energy density of SIBs is essential. Raising the operational voltage of the SIBs cathode is regarded as an effective strategy for achieving this goal, but it requires stable high-voltage cathode materials. Sodium iron sulfate (NFSO) is considered to be a promising cathode material due to its stable framework, adjustable structure, oper

Investigation of a chondroitin sulfate-based bioactive coating for neural interface applications.

Invasive neural implants allow for high-resolution bidirectional communication with the nervous tissue and have demonstrated the ability to record neural activity, stimulate neurons, and sense neurochemical species with high spatial selectivity and resolution. However, upon implantation, they are exposed to a foreign body response which can disrupt the seamless integration of the device with the native tissue and lead to deterioration in device functionality for chronic implantation. Modifying the device surface by incorporating bioactive coatings has been a promising approach to camouflage th
