Inducible anti-parasitic effector mechanisms in human uroepithelial cells: tryptophan degradation vs. NO production.

In murine cells the most important effector mechanism directed against the intracellular pathogen Toxoplasma gondii is the production of toxic nitrogen oxides. In contrast the induction of the tryptophan degrading enzyme indolamine 2,3-dioxygenase (IDO) has been described to be the most effective anti-parasitic mechanism in most human cells. In this report we analysed IDO induction and NO production in the human uroepithelial carcinoma cell line RT4. We found that after stimulation with IFN-gamma these cells were able to restrict toxoplasma growth. This was due to an activation of IDO, and the…

L-tryptophan-related eosinophilia-myalgia syndrome possibly associated with a chronic B-lymphocytic leukemia.

A female patient presenting with B-CLL and coincident eosinophilia-myalgia syndrome (EMS) after ingestion of L-tryptophan is described. The manifestations of EMS disappeared completely during treatment with cyclophosphamide/prednisone. and there was an intermittent clinical remission of CLL with absence of the monoclonal B-cell population. A few years later, the B-CLL relapsed, but without sign and symptoms of EMS. Whereas other eosinophilic syndromes such as eosinophilic fasciitis, panniculitis, or cellulitis Wells have been found to occur in relation to malignant underlying diseases, only a …

Molecular cloning of chick pineal tryptophan hydroxylase and circadian oscillation of its mRNA levels.

We have previously shown that the level of [35S]methionine incorporation into tryptophan hydroxylase (TPH) shows a circadian rhythm in cultured chick pineal cells. The TPH protein oscillation persists in constant darkness, peaks in the early night and can be phase-shifted by light, in parallel to the effect of these treatments on melatonin synthesis. We have cloned and sequenced a full-length cDNA for chick pineal TPH. Levels of TPH mRNA show a robust diurnal oscillation both in vivo and in vitro. The rhythm in TPH mRNA also persists in constant darkness, suggesting that TPH mRNA synthesis and…

Molecular cloning and characterization of cDNA encoding tryptophan hydroxylase from rat central serotonergic neurons.

Tryptophan hydroxylase (TPH) from central serotonergic neurons in the dorsal raphe nucleus (DRN) and that from the endocrine pineal gland (PG) have been shown to exhibit difference biochemical characteristics. We further report here that the isoelectric point determined by chromatofocusing differs between TPH from the rat brainstem and PG. In addition, the levels of TPH mRNA are much greater in the PG than the DRN despite a higher enzymatic activity in the DRN. These data raise the question as to whether different forms of TPH may exist in the DRN and the PG. To address this question, we ampli…

Aflatoxin B1 alters central and systemic tryptophan and tyrosine metabolism: influence of immunomodulatory drugs.

Semi-chronic exposure of ICR male Mice to Aflatoxin B1 (AFB1) in non-toxic doses results in elevated lung tryptophan (TRP) levels without change in serotonin (5-HT) or 5-hydroxyindole-3-acetic acid (5-HIAA) levels. This change is organ specific in that TRP levels are not altered in spleen, duodenum, heart or central nervous system (CNS). Acute (48 hour) flunixin treatment decreases lung TRP levels and reverses the AFB1 mediated increase in lung TRP levels. On the other hand, flunixin treatment decreases CNS TRP levels in control mice but not in AFB1 treated mice. Aflatoxin B1 treated mice have…

Cerebrospinal fluid amine metabolites, tryptophan and clinical parameters in depression. Part 1. Background variables.

5-Hydroxyindoleacetic acid, homovanillic acid, and tryptophan were measured in the lumbar cerebrospinal fluid of 33 female patients with severe hospitalized depression. Total plasma tryptophan was also determined in blood samples taken simultaneously at the time of lumbar puncture. Twelve clinical parameters were recorded using operational definitions for variables not measurable in physical units, and were compared in patient subgroups containing individuals with the highest and the lowest metabolite levels. Only age proved to be significantly higher in patients with low CSF 5-HIAA, and body …

Cerebrospinal fluid amine metabolites, tryptophan and clinical parameters in depression. Part 2. Psychopathological symptoms.

The group of 33 severely depressed female patients was divided into 3 equally large subgroups, separately according to their CSF 5-HIAA, HVA, tryptophan and total plasma tryptophan, and the severity of 17 operationally defined psychopathological symptoms was compared between the subgroups containing the highest and lowest biochemical values. Nonparametric statistical testing yielded significant results on several symptoms, when the subgroups were formed from CSF 5-HIAA and HVA. There was only one significant symptom difference between subgroups separated by CSF tryptophan and a few other diffe…

Suppression of androgen production by D-tryptophan-6-luteinizing hormone-releasing hormone in man.

Four male transsexual subjects were given a superactive luteinizing hormone-releasing hormone (LHRH) analogue, D-tryptophan-6-LHRH at daily doses of 100 micrograms for 3--6 mo. A decrease in beard growth, acne, and erectile potency was noted; the latter was documented objectively with the recordings of nocturnal penile tumescence episodes. Plasma testosterone and dihydrotestosterone levels fell to castrate values; basal prolactin and luteinizing hormone levels showed a small decline, whereas the acutely releasable luteinizing hormone was significantly suppressed. A rise of plasma testosterone …

Short-term metabolism of (14-C) tryptophan in rats infected with Trypanosoma brucei gambiense.

The pathways of short-term tryptophan metabolism in rats infected with Trypanosoma brucei gambiense were studied to determine whether this amino acid undergoes rapid conversion to tryptophol and indole lactic acid, two metabolites produced in vitro by T. brucei gambiense. Intravenously administered L-(3-14-C)-tryptophan was metabolized to at least two compounds in infected rats: tryptophol (indole ethanol) and indole acetic acid. Indole lactic acid was not detected as a metabolite. Approximately 76 percent of the labeled substrate was metabolized along these pathways in 30 min. Control rats ex…

Tryptophan Indole Metabolites Reduce Anastomotic Leakage Through Aryl Hydrocarbon Receptor-Driven Interleukin-22 Production.

BACKGROUND & AIMS: Colorectal cancer (CRC) often requires surgical resection of the tumor and an anastomosis. Anastomotic leakage (AL) occurs in 2.8% to 30% of patients, which increases postoperative morbidity and complications. The preoperative microbiome composition is implicated in AL. Recent studies have shown that microbial tryptophan (Trp) metabolism into aryl hydrocarbon receptor (AhR) indole-derivatives contributes to intestinal tissue healing. Here, we addressed the role of Trp and its metabolites in AL in a CRC patient cohort and a colon-anastomosis mouse model. METHODS: Targeted qu…

Enzymatic Prenylation of Proteins and Peptides: From Cysteine S-Prenylation to Tryptophan-Selective Biocatalysis.

Posttranslational prenylation endows polypeptides with lipophilic properties, facilitating essential biological functions such as membrane t … g the use of longer-chain variants and geometric isomers, as well as regioselective modification of tryptophan. Furthermore, we compare the biophysical properties of emerging tryptophan C-prenylation with those of conventional cysteine S-prenylation and linear fatty acylation, highlighting their distinct roles in modulating polypeptide function and behavior. …

OASA1D-Mediated Tryptophan Enrichment Improves Redox and Ionic Homeostasis Under Salt Stress in Rice.

Salinity restricts rice growth by disrupting cellular ion balance and promoting oxidative damage. Although exogenous melatonin can improve rice salt tolerance, whether expansion of the endogenous tryptophan pool enhances melatonin biosynthetic capacity and stress acclimation remains unclear. Here, we investigated a homozygous transgenic rice line constitutively expressing OASA1D, a feedback-insensitive D323N variant of the anthranilate synthase α-subunit OASA1. The OASA1D-expressing line exhibited strong resistance to 5-methyltryptophan and accumulated approximately twofold more tryptophan tha…

Tryptophan-enriched diet improves social behavior in a mouse model of tuberous sclerosis complex.

Background: Tryptophan is an essential dietary amino acid and the sole precursor of serotonin (5-HT), a neurotransmitter that plays a key role in cognition, mood regulation, and sensory processing. Because central serotonin synthesis depends on dietary tryptophan availability, nutritional modulation of tryptophan intake has emerged as a potential strategy to influence brain function and behavior. Disruptions in tryptophan metabolism and serotonergic signaling have been implicated in autism spectrum disorder (ASD), particularly in Tuberous Sclerosis Complex (TSC), a neurogenetic condition frequ…

Dietary Tryptophan Allocation in Depression: Serotonin-Kynurenine Balance, Microbial Indole Pathways, and Inflammatory Phenotypes.

Depression is defined by clinical symptoms, but its biology varies considerably among patients. In this review, dietary tryptophan allocation describes the routes taken by tryptophan after intestinal absorption. Some is incorporated into proteins. Some enters serotonin and melatonin synthesis or the kynurenine pathway, while gut bacteria convert another fraction into indole compounds. Brain availability also depends on the circulating free pool and competition with other large neutral amino acids. This term does not imply a new biochemical pathway. It allows these known processes to be conside…

The Tryptophan Biosynthesis Gene trpE Contributes to Virulence in Mesophilic Aeromonas salmonicida SRW-OG1.

The aquatic pathogen Aeromonas salmonicida SRW-OG1 is responsible for substantial mortality in Epinephelus coioides and many other cultured fish species. This study investigates the contribution of trpE to the pathogenicity of A. salmonicida SRW-OG1 in E. coioides. Specifically, trpE encodes a key enzyme in tryptophan biosynthesis; however, its function in regulating virulence in A. salmonicida remains uncertain. Phenotypic assays indicate that deletion of trpE markedly impaired bacterial growth and significantly reduced multiple virulence-associated traits, including swimming motility, biofil…

Disrupted Tryptophan Metabolism Mediates Manganese-Induced Neurogenesis and Neuroinflammatory Impairments: Rescue by Exogenous Melatonin.

Manganese (Mn) is a common environmental pollutant, and excessive exposure can lead to motor dysfunction resembling Parkinson's disease. Inc … ontributed to a vicious cycle of neural damage. Multiomics analyses identified dysregulation of the tryptophan metabolic pathway as a key mechanism, with a marked reduction in melatonin levels following Mn exposure. Notably, exogenous melatonin supplementation effectively rescued Mn-induced impairments in neurogenesis, neuronal integrity, and neuroinflammati …

The tryptophan metabolite 3-hydroxyanthranilic acid alleviates hyperoxia-induced bronchopulmonary dysplasia via inhibiting ferroptosis.

Bronchopulmonary dysplasia (BPD) is a prevalent chronic respiratory condition in preterm infants with an increasing incidence, severely affe … nificant reduction in 3-hydroxyanthranilic acid (3-HAA), which was confirmed to be partly caused by tryptophan-metabolizing enzyme disorders. In vivo and in vitro models were subsequently established to assess the efficacy and underlying mechanisms of 3-HAA in relation to BPD. Compared with the BPD group, 3-HAA nebulization improved lung development and supp …
