The hemolymph of Biomphalaria snail vectors of schistosomiasis supports a diverse microbiome.

The microbiome - the microorganism community that is found on or within an organism's body - is increasingly recognized to shape many aspects of its host biology and is a key determinant of health and disease. Microbiomes modulate the capacity of insect disease vectors (mosquitoes, tsetse flies, sandflies) to transmit parasites and disease. We investigate the diversity and abundance of microorganisms within the hemolymph (i.e. blood) of Biomphalaria snails, the intermediate host for Schistosoma mansoni, using Illumina MiSeq sequencing of the bacterial 16S V4 rDNA. We sampled hemolymph from fiv…

C6-O-alkylated 7-deazainosine nucleoside analogues: Discovery of potent and selective anti-sleeping sickness agents.

African trypanosomiasis, a deadly infectious disease caused by the protozoan Trypanosoma brucei spp., is spread to new hosts by bites of infected tsetse flies. Currently approved therapies all have their specific drawbacks, prompting a search for novel therapeutic agents. T. brucei lacks the enzymes necessary to forge the purine ring from amino acid precursors, rendering them dependent on the uptake and interconversion of host purines. This dependency renders analogues of purines and corresponding nucleosides an interesting source of potential anti-T. brucei agents. In this study, we synthesiz…

Interpreting Morphological Adaptations Associated with Viviparity in the Tsetse Fly Glossina morsitans (Westwood) by Three-Dimensional Analysis.

Tsetse flies (genus Glossina), the sole vectors of African trypanosomiasis, are distinct from most other insects, due to dramatic morphological and physiological adaptations required to support their unique biology. These adaptations are driven by demands associated with obligate hematophagy and viviparous reproduction. Obligate viviparity entails intrauterine larval development and the provision of maternal nutrients for the developing larvae. The reduced reproductive capacity/rate associated with this biology results in increased inter- and intra-sexual competition. Here, we use phase contra…

Recent Updates on Development of Drug Molecules for Human African Trypanosomiasis.

Human African trypanosomiasis (HAT, better called as sleeping sickness), caused by two morphologically identicalprotozoan parasite Trypanosoma bruceiis transmitted by the bite of tsetse flies of Glossinagenus, mainly in the rural areas of the sub-Saharan Africa. HAT is one of the neglected tropical diseases and is characterized by sleep disturbance as the main symptom, hence is called as sleeping sickness. As it is epidemic in the poorest population of Africa, there is limited availability of safe and cost-effective tools for controlling the disease. Trypanosoma bruceigambiense causes sleeping…

The cost of tsetse control using 'Tiny Targets' in the sleeping sickness endemic forest area of Bonon in Côte d'Ivoire: Implications for comparing costs across different settings.

BACKGROUND: Work to control the gambiense form of human African trypanosomiasis (gHAT), or sleeping sickness, is now directed towards ending transmission of the parasite by 2030. In order to supplement gHAT case-finding and treatment, since 2011 tsetse control has been implemented using Tiny Targets in a number of gHAT foci. As this intervention is extended to new foci, it is vital to understand the costs involved. Costs have already been analysed for the foci of Arua in Uganda and Mandoul in Chad. This paper examines the costs of controlling Glossina palpalis palpalis in the focus of Bonon in

Expansions of chemosensory gene orthologs among selected tsetse fly species and their expressions in Glossina morsitans morsitans tsetse fly.

Tsetse fly exhibit species-specific olfactory uniqueness potentially underpinned by differences in their chemosensory protein repertoire. We assessed 1) expansions of chemosensory protein orthologs in Glossina morsitans morsitans, Glossina pallidipes, Glossina austeni, Glossina palpalis gambiensis, Glossina fuscipes fuscipes and Glossina brevipalpis tsetse fly species using Café analysis (to identify species-specific expansions) and 2) differential expressions of the orthologs and associated proteins in male G. m. morsitans antennae and head tissues using RNA-Seq approaches (to establish assoc

The proteome and transcriptome of the infectious metacyclic form of Trypanosoma brucei define quiescent cells primed for mammalian invasion.

The infectious metacyclic forms of Trypanosoma brucei result from a complex development in the tsetse fly vector. When they infect mammals, they cause African sleeping sickness in humans. Due to scarcity of biological material and difficulties of the tsetse fly as an experimental system, very limited information is available concerning the gene expression profile of metacyclic forms. We used an in vitro system based on expressing the RNA binding protein 6 to obtain infectious metacyclics and determined their protein and mRNA repertoires by mass-spectrometry (MS) based proteomics and mRNA seque

[One century of "sleeping sickness" in West Africa].

This paper summarizes the geography of sleeping sickness disease (or Human African Trypanosomiasis, HAT) over the last 100 years in West Africa, with the objective of identifying today's priority areas for the sleeping sickness surveillance. The history and geography of the disease are based on a bibliographic review of old reports and recent publications on recent results obtained from medical surveys conducted in West Africa up to 2007. This allowed us to situate the historical geography of HAT from the beginning of the 20th century to nowadays. For instance, active HAT foci seem to have mov

[Trypanosomiasis--a real risk for tourists visiting national parks in Tanzania].

African sleeping sickness is no longer a rare disease among tourists visiting national parks in Tanzania. The disease is caused by a parasite, Trypanosoma brucei, which is transmitted by the tsetse fly. Two species infect humans: Trypanosoma brucci gambiense and Trypanosoma brucei rhodesiense; the last form is re-emerging in parts of Africa. Untreated this disease carries a mortality of nearly 100%. This article describes a case of African sleeping sickness in a tourist visiting Tanzania, which was diagnosed at the Nordic Clinic, Dar es Salaam. The most important symptoms, diagnostic investiga

African trypanosomiasis research: 100 years of progress, but questions and problems still remain.

Past and present progress in our understanding of African trypanosomiasis is briefly reviewed. Although tremendous scientific strides have been achieved, an epidemic of the disease is currently underway. Three areas of research which are believed necessary for the control of African trypanosomiasis are discussed. It is suggested that a better understanding of the host-parasite relationship is essential; more emphasis and a broader approach to drug development is required; and finally, further research into the socio-economic aspects of African trypanosomiasis is urgently needed before the huma

[Technical reasons for the re-emergence of sleeping sickness].

In the 1920s the epidemic outbreak of human African trypanosomiasis was so deadly that government authorities decided to take large-scale action. It was by giving Jamot absolute administrative and financial autonomy to apply his ideas that the disease was successfully controlled. After Jamot determined efforts against the disease continued so that, by the dawn of decolonization, many considered the problem of sleeping sickness as resolved. Control programs progressively slowed and virtually ceased. Neglected and all but forgotten, the disease has able to make a natural and forceful comeback. O

Tsetse-trypanosomiasis challenge to village N'Dama cattle in The Gambia: field assessments of spatial and temporal patterns of tsetse-cattle contact and the risk of trypanosomiasis infection.

The severity of the trypanosomiasis problem in a particular location is traditionally assessed in terms of a challenge index-the product of some measure of tsetse abundance and infection-rate-which is assumed to be proportional to the force of infection. However, this index masks variation in the force of infection between herds and among individuals within herds. It is also not comparable between sites since the relative abundance of tsetse to hosts may vary. We have studied spatial distribution of herds of cattle in relation to tsetse in The Gambia and calculated an index of challenge based

Quantitative phenotyping of N'Dama cattle for aspects of trypanotolerance under field tsetse challenge.

Matching animal health and performance data were recorded over the 2 year period from weaning at 10 months of age on 255 N'Dama cattle in a high natural tsetse challenge situation in Zaire. Four parameters that are regarded as possible indicators of trypanotolerance, species of trypanosomes detected, length of time parasitaemic, intensity of parasitaemia (parasitaemia score), and anaemic condition as estimated by packed cell volume (PCV) values, were measured and the relative effects of changes in these parameters on trypanocidal drug requirements and growth were assessed. The effects of speci

A comparison of susceptibility of two allopatric populations of Glossina pallidipes for stocks of Trypanosoma congolense.

A colony of Glossina pallidipes Austen which originated from Nguruman, Rift Valley Province, Kenya, was significantly more susceptible to infection (19.3%) with a stock of Trypanosoma congolense Broden isolated from G. pallidipes in Nguruman than a colony of the same species which originated from Shimba Hills, Coast Province, Kenya (5.6%). Male G.pallidipes from Nguruman were significantly more susceptible than females to this T.congolense stock whilst the susceptibility of both sexes of G.pallidipes from Shimba Hills did not differ significantly. Vector population responses to control interventions.

In 1981, Kenyan authorities attempted eradication of tsetse (Glossina pallidipes) from the Lambwe valley, by sequential aerial spraying of endosulfan. Fly populations were reduced by over 99.9% in the main habitats, but recovered to their original levels within one year. In 1986, an Argentine research team attempted local elimination of domestic Triatominae (Triatoma infestans) in an area of Santiago del Estero. House infestation rates were reduced to an apparent zero, but recovered to pre-control levels in two years. In Sri Lanka, a combination of mosquito control with active case detection a

Variation in resistance to isometamidium chloride and diminazene aceturate by clones derived from a stock of Trypanosoma congolense.

Nine clones were derived from a drug-resistant Trypanosoma congolense stock (IL 2856) and characterized in mice for their sensitivity to isometamidium chloride and diminazene aceturate. All clones were derived from the stock without drug selection and expressed high levels of resistance to isometamidium chloride (50% curative dose [CD50] values ranging from 1.5 to 5.1 mg/kg) and intermediate to high levels of resistance to diminazene aceturate (CD50 values ranging from 5.1 to 21.0 mg/kg). By contrast, the isometamidium chloride and diminazene aceturate CD50 values for a drug-sensitive clone, T

Comparative analysis of the costs and benefits of alternative disease control strategies: vector control versus human case finding and treatment.

Recent estimates of the costs of detection and treatment and vector control for human trypanosomiasis are summarized and the main factors influencing their levels discussed. In order to proceed to an economic analysis a benefit unit is defined as equivalent to one year's infection avoided due to the control strategy for one person. Different monetary values could be assigned to this unit and it has the advantage of being easily integrated into epidemiological models, being represented by the difference in prevalence with and without control. A simple spreadsheet-based economic model was develo
