Epidemiology and antifungal susceptibilities of rare yeast infections in a tertiary care center.

BACKGROUND: With advancements in healthcare services, there has been an increase in the population of immunosuppressed patients and the frequency of opportunistic pathogens including rare yeasts (RY). We aimed to provide data on the distribution of RY causing invasive infections and their antifungal susceptibility profiles. In this retrospective study, the distribution of RYs obtained between January 2015 and January 2023 examined. Antifungal susceptibility analyses of the isolates conducted using the broth microdilution method following European Committee on Antimicrobial Susceptibility Testi…

Manganese-modulated proteome and phosphoproteome dataset in the opportunistic yeast Candida albicans.

The ability of the opportunistic yeast Candida albicans to acquire and maintain homeostatic levels of manganese (Mn), particularly in the metal-limited host environment, is an important determinant of its fitness. Although significant focus has been given to mechanisms of iron and copper acquisition and their roles in C. albicans fitness, little is known about how this yeast maintains and controls Mn homeostasis. Here, we present a comparative proteomic and phosphoproteomic analysis in C. albicans cells experiencing Mn starvation. Both proteome and phosphoproteome of the C. albicans reference

Ultrastructure Expansion Microscopy (U-ExM) of the Fission Yeast Schizosaccharomyces pombe.

Among widely used super-resolution microscopy techniques, including stimulated emission depletion (STED), photoactivated localization microscopy (PALM), and stochastic optical reconstruction microscopy (STORM), expansion microscopy (ExM) is unique in achieving increased resolution through a physical manipulation of the actual sample rather than optics or postprocessing. Originally developed for applications in neuroscience, ExM now has a solid foothold across many fields and model systems, and has been adapted to work for organisms with cell walls, including budding and fission yeasts, through…

Masquerading Yeast: A Case Report of Lomentospora prolificans Fungemia With a Diagnostic Twist.

Lomentospora prolificans is an environmental mold that is increasingly recognized as an opportunistic pathogen, particularly in immunocompromised hosts. Here we present a case of disseminated Lomentospora in a patient with refractory acute myeloid leukemia. In general, disseminated mold infections can be challenging to diagnose. The diagnosis in this case was delayed due to a false positive direct from a blood culture multiplex polymerase chain reaction result for Candida tropicalis due to amplification of nonviable DNA and misidentification of conidia as yeast forms on gram stain. The Lomento

Environmental Yeast Abundance and Diversity Assessment in Recreation Areas of Bangkok, Thailand.

The diversity of environmental yeast communities is underestimated in tropical and sub-tropical regions. Numerous studies demonstrated that human activity can alter the yeast diversity and increase pathogenic yeast proportions, indicating that people who frequently visit those areas are at risk of being infected. The main purpose of this study was to investigate the abundance and diversity of yeasts obtained from recreation areas in Bangkok. In this study, 158 soil and water samples were collected from 12 public parks. The analysis of the yeast communities revealed different patterns among rec…

Oxidation Performance of the Aga2-Laccase Fusion Produced by Yeast Surface Display.

Laccases are multicopper oxidases with numerous applications because of their wide substrate variety. The yeast surface display (YSD) technology is a powerful tool for the expression of recombinant laccases that can be used to design robust oxidative biocatalysts and combine stability and flexibility in a whole-cell system. In order to gain insights on improving the laccase performance, we investigated the release, from the yeast's surface, of a recombinant laccase (OB1) from Saccharomyces cerevisiae EBY100's cell surface. The laccase, or derivatives thereof, was released as a fusion protein c

Environmental characteristics and weather impact yeast populations and their dynamics in spontaneous fermentations.

Winemaking is influenced by many factors, from the vineyard to the fermentation process. While traditional approaches have focused on vineyard treatments and controlled fermentations to ensure a stable product, global warming has made it necessary to reassess the effects of these factors on winemaking. This study aimed to evaluate the impact of vintage, management, and environmental settings on the composition of yeast populations driving grape spontaneous fermentations and defining the characteristics of the final product. Our results emphasize how climate change and the vineyard's yeast popu

Screening a 681-membered yeast collection for the secretion of proteins with antifungal activity.

Fungal pathogens pose a threat to human health and food security. Few antifungals are available and resistance to all has been reported. Novel strategies to control plant and human pathogens as well as food spoilers are urgently required. Environmental yeasts provide a functionally diverse, yet underexploited potential for fungal control based on their natural competition via the secretion of proteins and other small molecules such as iron chelators, volatile organic compounds or biosurfactants. However, there is a lack of standardized workflows to systematically access application-relevant ye…

The glutathione system maintains the thiol redox balance in the mitochondria of fission yeast.

The thioredoxin and glutathione (GSH)-glutaredoxin electron donor pathways provide a reducing environment to the cell and maintain homeostasis of numerous redox reactions. The abundant tripeptide GSH has multiple roles, including redox buffering, detoxification, peroxide scavenging and iron-sulfur cluster assembly. Glutathione reductase, Pgr1 in fission yeast, maintains glutathione reduced, and it is essential in most organisms. Cells lacking Pgr1 exhibit severe pleiotropic defects. We used multiple approaches to unravel the compartment-specific roles of Pgr1. Our findings confirmed that Pgr1

Methanol biotransformation for the production of biodegradable plastic monomer L-lactate in yeast.

Methanol is an ideal feedstock for biomanufacturing, and production of the biodegradable plastic monomer lactate from methanol is a promising approach for mitigating white pollution. However, it is challenging to engineer microbes for lactate production from methanol because of strong competition between product synthesis and cell growth. Here, Ogataea polymorpha is rewired to synthesize L-lactate from methanol, where the cofactor ratio of NADPH/NADP+ is higher than that of NADH/NAD+. By engineering gene expression, enhancing cell viability, modifying cofactor homeostasis, and performing mitoc

Magnetic Nanoworm-Based Screening of Yeast Library for Anti-PEG Nanobodies.

PEGylation is commonly utilized to modify nanoparticles, vaccines, therapeutic proteins, and biomaterials. The increased use of PEG-containing cosmetics and medicines results in anti-PEG immunoglobulins in humans, including IgG, IgM, and IgE types. Recent studies have shown that high-affinity antibodies against PEG can be isolated through immunization. These antibodies have been used in immunoassays and for decorating PEGylated nanoparticles with targeting ligands. However, conventional antibodies have limitations, such as their large size and potential stability issues. Here, we developed a p

Evaluation of flavor properties in rice bran by solid-state fermentation with yeast.

Rice bran (RB) as an available agricultural by-product is currently underutilized due to its susceptibility to oxidation and rancidity. This study investigated the evaluation of flavor properties in RB by solid-state fermentation with yeast (Saccharomyces cerevisiae). The volatile compounds (VOCs) in fermented RB were detected by chromatography-ion mobility spectrometry (GC-IMS), and headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS). A total of 76 volatile compounds (VOCs) including 10 aldehydes, 6 ketones, 9 alcohols, 20 esters, 10 hydrocarbons, 10 aci

Structural basis for hygromycin B inhibition of yeast pseudouridine-deficient ribosomes.

Eukaryotic ribosomes are enriched with pseudouridine, particularly at the functional centers targeted by antibiotics. Here, we investigated the roles of pseudouridine in aminoglycoside-mediated translation inhibition by comparing the structural and functional properties of the yeast wild-type and the pseudouridine-free ribosomes. We showed that the pseudouridine-free ribosomes have decreased thermostability and high sensitivity to aminoglycosides. When presented with a model internal ribosomal entry site RNA, elongation factor eEF2, GTP (guanosine triphosphate), and sordarin, hygromycin B pref

Structural Basis for Oxidized Glutathione Recognition by Yeast Cadmium Factor 1.

Transporters from the ABCC family have an essential role in detoxifying electrophilic compounds, including heavy metals and drugs, often after conjugation with glutathione. The Yeast Cadmium Factor 1 (Ycf1) transports glutathione conjugated to toxic heavy metals including Cd2+, Hg2+, and the metalloid As3+ into the vacuole to safely sequester them in the cell. It also has a major role in transporting oxidized glutathione to help maintain redox balance and recycle glutathione. To understand the complicated selectivity and promiscuity of binding, we determined the cryo-EM structure of Ycf1 bound

Breeding bafilomycin A1-resistant sake yeast to improve fermentative capacity.

Sake yeast exhibits remarkable fermentative capacity in mash environments, even under various stress conditions. Although breeding techniques aimed at improving the flavor of sake by modifying aroma compounds and organic acid composition have been employed, these approaches often result in reduced fermentative capacity. Furthermore, existing breeding methods aimed at enhancing fermentative capacity often result in increased acidity in the final product. In this study, we aimed to improve sake yeast fermentative capacity while limiting sake acidity. Depletion of intracellular ATP may enhance fe

Red yeast rice with monacolin K for the improvement of hyperlipidemia: A narrative review.

Low-density lipoprotein cholesterol (LDL-C) is the most causal risk factor for atherosclerotic cardiovascular disease (ASCVD). Red yeast rice (RYR) is a nutraceutical widely used as a lipid-lowering dietary supplement. The main cholesterol-lower agents in RYR are monacolins, particularly monacolin K, a weak reversible inhibitor of 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase, whose daily consumption (up to 10 mg/day) reduces LDL-C plasma levels up to 34% within 6-8 weeks when compared to placebo. The reduction in LDL-C is often accompanied by lower levels of plasma apolipoprotein B, total

Machine learning enables identification of an alternative yeast galactose utilization pathway.

How genomic differences contribute to phenotypic differences is a major question in biology. The recently characterized genomes, isolation environments, and qualitative patterns of growth on 122 sources and conditions of 1,154 strains from 1,049 fungal species (nearly all known) in the yeast subphylum Saccharomycotina provide a powerful, yet complex, dataset for addressing this question. We used a random forest algorithm trained on these genomic, metabolic, and environmental data to predict growth on several carbon sources with high accuracy. Known structural genes involved in assimilation of
