Reprogramming bone homeostasis: The recoupling-oriented framework of bidirectional regulation strategies for osteoporosis.

Current osteoporosis treatments reduce fracture risk by suppressing bone resorption, stimulating bone formation, or combining both effects. Yet these clinical benefits do not show whether formation has resumed at sites where bone was previously resorbed. This Review uses a remodeling recoupling framework to examine this resorption-to-formation transition. We define recoupling as restoration of the sequence from resorption through reversal to osteoblast recruitment and bone formation. This differs from bidirectional regulation, in which osteoblast activity increases and osteoclast activity decr

MMBs_TransNeXt: a hierarchical adaptive feature learning model for pig behavior recognition in precision livestock farming.

Accurate recognition of pig behavior plays a critical role in enhancing animal welfare and management efficiency for precision livestock farming. Nevertheless, existing methods are commonly limited by rigid feature fusion strategies, insufficient preservation of spatial details, and weak interactions across hierarchical features, which severely compromise recognition performance in complex farm scenarios. To address these challenges, we present a novel deep learning architecture termed MMBs_TransNeXt for pig behavior recognition. This model establishes a unified, hierarchical and adaptive feat

In vitro space of Disse model for exploration of drug induced hepatotoxicity.

The limited predictive power of animal models remains a major bottleneck in drug development, particularly in assessing drug-induced liver injury (DILI). To address this, we developed a novel in vitro liver-on-a-chip platform focused on modelling the space of Disse (sD) - a key microenvironment, yet insufficiently studied, which mediates cross-talk between endothelial and hepatic systems. The system integrates a biocompatible sodium alginate hydrogel whose mechanical properties were optimised to mimic physiological liver stiffness, enabling molecular cross-talk between human liver sinusoidal e

Remodeling the Tumor Microenvironment: Mechanistic Insights and Translational Frontiers of TCM-Derived Bioactive Monomers.

Recent studies have shown that tumor development and therapeutic resistance are closely linked to the tumor microenvironment (TME), a dynamic ecosystem composed of vascular networks, immune and stromal cells, extracellular matrix components, cytokines, and extracellular vesicles. This review summarizes current mechanistic evidence on how bioactive monomers and purified active fractions derived from traditional Chinese medicine (TCM), including polysaccharides, saponins, alkaloids, terpenoids, flavonoids, and polyphenols, regulate TME-associated processes. These compounds have been reported to

Microglia, cerebellar inflammation, and autism spectrum disorders: Developmental mechanisms and therapeutic perspectives.

Autism spectrum disorders (ASD) are neurodevelopmental conditions characterized by deficits in social interaction and communication, as well as restricted and repetitive behaviors. These conditions often co-occur with medical issues linked to synaptic alterations, which compromise synaptic integrity and are associated with brain circuit dysfunction. Both human and animal studies have identified cerebellar structural and functional alterations in subsets of ASD cases, with evidence of abnormal morphology and disrupted connectivity correlating with symptom severity. Numerous studies further sugg

Lunar cycle and moonlight intensity influence nocturnal migration patterns in a small songbird.

Lunar cycle and moonlight exposure have significant impacts on animal behaviour and physiology. The presence or absence of moonlight, along with predictable changes in brightness throughout the lunar cycle, can shape reproduction, foraging, communication, and other aspects of an animal's world. While it has been shown that invertebrates use the moonlight for orientation, little is known on the effect of the lunar cycle on migratory birds. We found that the lunar cycle affected the nocturnal migration activity of a diurnal songbird species, the Eurasian Skylark Alauda arvensis. The occurrence o

A Bio-Inspired Artificial Nerve Simulator for Ex Vivo Validation of Implantable Neural Interfaces Equipped with Plug Electrodes.

The development of implantable neural interfaces is essential for enabling bidirectional communication between the nervous system and prosthetic devices, yet their evaluation still relies primarily on in vivo models which are costly, variable, and ethically constrained. Here, we report a bio-inspired artificial nerve simulator engineered as a reproducible ex vivo platform for pre-implantation testing of plug-type electrodes. The simulator is fabricated from a conductive hydrogel composite based on reduced graphene oxide (rGO), polyaniline (PANI), agarose, sucrose, and sodium chloride, with emb

The Unexplored Role of Connexin Hemichannels in Promoting Facioscapulohumeral Muscular Dystrophy Progression.

DUX4 is typically a repressed transcription factor, but its aberrant activation in Facioscapulohumeral Muscular Dystrophy (FSHD) leads to cell death by disrupting muscle homeostasis. This disruption affects crucial processes such as myogenesis, sarcolemma integrity, gene regulation, oxidative stress, immune response, and many other biological pathways. Notably, these disrupted processes have been associated, in other pathological contexts, with the presence of connexin (Cx) hemichannels-transmembrane structures that mediate communication between the intracellular and extracellular environments

Farnesoid X receptor: From Structure to Function and Its Pharmacology in Liver Fibrosis.

The farnesoid X receptor (FXR), a ligand-activated transcription factor, plays a crucial role in regulating bile acid metabolism within the enterohepatic circulation. Beyond its involvement in metabolic disorders and immune imbalances affecting various tissues, FXR is implicated in microbiota modulation, gut-to-brain communication, and liver disease. The liver, as a pivotal metabolic and detoxification organ, is susceptible to damage from factors such as alcohol, viruses, drugs, and high-fat diets. Chronic or recurrent liver injury can culminate in liver fibrosis, which, if left untreated, may

What is the role of the hippocampus and parahippocampal gyrus in the persistence of tinnitus?

The hippocampus and parahippocampal gyrus have been implicated as part of a tinnitus network by a number of studies. These structures are usually considered in the context of a "limbic system," a concept typically invoked to explain the emotional response to tinnitus. Despite this common framing, it is not apparent from current literature that this is necessarily the main functional role of these structures in persistent tinnitus. Here, we highlight a different role that encompasses their most commonly implicated functional position within the brain-that is, as a memory system. We consider tin

Hypothalamic circuits and aging: keeping the circadian clock updated.

Over the past century, age-related diseases, such as cancer, type-2 diabetes, obesity, and mental illness, have shown a significant increase, negatively impacting overall quality of life. Studies on aged animal models have unveiled a progressive discoordination at multiple regulatory levels, including transcriptional, translational, and post-translational processes, resulting from cellular stress and circadian derangements. The circadian clock emerges as a key regulator, sustaining physiological homeostasis and promoting healthy aging through timely molecular coordination of pivotal cellular p

Cross-frequency coupling between slow harmonics via the real brainstem oscillators: An in vivo animal study.

Brain waves of discrete rhythms (gamma to delta frequency ranges) are ubiquitously recorded and interpreted with respect to probable corresponding specific functions. The most challenging idea of interpreting varied frequencies of brain waves has been postulated as a communication mechanism in which different neuronal assemblies use specific ranges of frequencies cooperatively. One promising candidate is cross-frequency coupling (CFC), in which some neuronal assemblies efficiently utilize the fastest gamma range brain waves as an information carrier (phase-amplitude CFC); however, phase-phase

Single-cell transcriptome analysis reveals liver injury induced by glyphosate in mice.

BACKGROUND: Glyphosate (GLY), as the active ingredient of the most widely used herbicide worldwide, is commonly detected in the environment and living organisms, including humans. Its toxicity and carcinogenicity in mammals remain controversial. Several studies have demonstrated the hepatotoxicity of GLY; however, the underlying cellular and molecular mechanisms are still largely unknown. METHODS: Using single-cell RNA sequencing (scRNA-seq), immunofluorescent staining, and in vivo animal studies, we analyzed the liver tissues from untreated and GLY-treated mice. RESULTS: We generated the fi

Muscle Involvement in Amyotrophic Lateral Sclerosis: Understanding the Pathogenesis and Advancing Therapeutics.

Amyotrophic lateral sclerosis (ALS) is a fatal condition characterized by the selective loss of motor neurons in the motor cortex, brainstem, and spinal cord. Muscle involvement, muscle atrophy, and subsequent paralysis are among the main features of this disease, which is defined as a neuromuscular disorder. ALS is a persistently progressive disease, and as motor neurons continue to degenerate, individuals with ALS experience a gradual decline in their ability to perform daily activities. Ultimately, muscle function loss may result in paralysis, presenting significant challenges in mobility,

In praise of adventitious sounds.

Study of the characteristics, neural processing, and behavioral effects of communication sounds are an important focus of hearing research. The goal of this paper is to bring attention to another, rather neglected class of sounds that are omnipresent and that are arguably at least equally important for the daily survival of many species and have been in the evolution of the mammalian auditory system. All terrestrial mammals produce adventitious sounds when they locomote. The handful of studies that examined the characteristics of such sounds suggests that high-frequency transients are an impor

Mechanistic roles for altered O-GlcNAcylation in neurodegenerative disorders.

Neurodegenerative diseases such as Alzheimer's and Parkinson's remain highly prevalent and incurable disorders. A major challenge in fully understanding and combating the progression of these diseases is the complexity of the network of processes that lead to progressive neuronal dysfunction and death. An ideal therapeutic avenue is conceivably one that could address many if not all of these multiple misregulated mechanisms. Over the years, chemical intervention for the up-regulation of the endogenous posttranslational modification (PTM) O-GlcNAc has been proposed as a potential strategy to sl

Astroglia in Autism Spectrum Disorder.

Autism spectrum disorder (ASD) is an umbrella term encompassing several neurodevelopmental disorders such as Asperger syndrome or autism. It is characterised by the occurrence of distinct deficits in social behaviour and communication and repetitive patterns of behaviour. The symptoms may be of different intensity and may vary in types. Risk factors for ASD include disturbed brain homeostasis, genetic predispositions, or inflammation during the prenatal period caused by viruses or bacteria. The number of diagnosed cases is growing, but the main cause and mechanism leading to ASD is still uncer

Neuromodulation by mGluRs in Sound Localization Circuits in the Auditory Brainstem.

The ability of humans and animals to localize the source of a sound in a complex acoustic environment facilitates communication and survival. Two cues are used for sound localization at horizontal planes, interaural time and level differences (ITD and ILD), which are analyzed by distinct neural circuits in the brainstem. Here, we review the studies on metabotropic glutamate receptor (mGluR)-mediated neuromodulation of both intrinsic and synaptic properties of brainstem neurons in these circuits. Both mammalian and avian animal models have been used, with each having their advantages that are n

Metabolism-Disrupting Chemicals and the Constitutive Androstane Receptor CAR.

During the last two decades, the constitutive androstane receptor (CAR; NR1I3) has emerged as a master activator of drug- and xenobiotic-metabolizing enzymes and transporters that govern the clearance of both exogenous and endogenous small molecules. Recent studies indicate that CAR participates, together with other nuclear receptors (NRs) and transcription factors, in regulation of hepatic glucose and lipid metabolism, hepatocyte communication, proliferation and toxicity, and liver tumor development in rodents. Endocrine-disrupting chemicals (EDCs) constitute a wide range of persistent organi

Spinal cord injury and the human microbiome: beyond the brain-gut axis.

In addition to standard management for the treatment of the acute phase of spinal cord injury (SCI), implementation of novel neuroprotective interventions offers the potential for significant reductions in morbidity and long-term health costs. A better understanding of the systemic changes after SCI could provide insight into mechanisms that lead to secondary injury. An emerging area of research involves the complex interplay of the gut microbiome and the CNS, i.e., a brain-gut axis, or perhaps more appropriately, a CNS-gut axis. This review summarizes the relevant literature relating to the g
