Evidence that naloxone attenuates the consumption of food by domestic pigeons through a central influence.

Three experiments were performed on domestic pigeons deprived of food for 24 hr to determine whether the anorexic influence of naloxone, which is observed in this species, results from a central effect of this drug. Injections were given 15 min before the delivery of food, and repeated measures of the consumption of food were obtained during the next 20 min. The systemic administration of either 2 or 10 mumol of naloxone, but not of equimolar amounts of quaternary naloxone, that does not traverse the blood-brain barrier, reduced the consumption of food of the pigeons. At a dose of 20 mumol, qu

Effects of 4-hydroxyamphetamine on in vivo brown adipose tissue thermogenesis and feeding behavior in the rat.

The influence of 4-hydroxyamphetamine (4-OHAM) on food and water intake and in vivo brown adipose thermogenesis was examined in two experiments. In Experiment 1, female rats were treated with 0.00, 0.25, 0.50, 1.00, or 2.00 mg/kg 4-OHAM (ip) prior to assessment of interscapular brown adipose tissue (IBAT) thermogenesis. The 4-OHAM treatment induced dose-dependent activation of IBAT thermogenesis consistent with the enhanced serum levels of norepinephrine and epinephrine observed in 4-OHAM-treated rats immediately after temperature measurement. In Experiment 2, the influence of 4-OHAM on food a

Induction of hyperphagia in rats by intracerebroventricular infusion of sodium pentobarbital. A method for testing anorexigenic compounds.

In rats hyperphagia could be reversibly induced by intracerebroventricular infusion of sodium pentobarbital. Hyperphagia was elicited in a dose dependent manner, the optimal infusion rate being 1000 micrograms/h sodium pentobarbital. At this rate, feeding behaviour was induced selectively, with only minor side effects being apparent. Systemic application of d-methamphetamine or propylhexedrine given prior to infusion resulted in a dose dependent reduction of hyperphagia. The method, therefore, could be useful for testing putative anorexigenic compounds in hyperphagic rats.

Effects of behavioral and pharmacological weight loss programs on nutrient intake.

Behavior therapy is a widely used method for short-term weight control. Formal nutrition counseling is not included in many behavioral programs. To determine what types of diets were self-selected by patients and to see how they compared with individuals participating in a relaxation group with anorectic medication, the eating diaries compiled by participants in such a program were analyzed for 20 different nutrients. In general, pretreatment intakes were nutritious and typical for Americans. Both the behavioral and a relaxation-medication treatment produced weight loss and reduction in calori

A spatial and projection-based transcriptomic atlas of paraventricular hypothalamic cell types.

The paraventricular hypothalamus (PVH) controls behavioral and physiologic processes, including appetite, social behavior, autonomic outflow, and pituitary hormone secretion. However, molecular markers for centrally projecting PVH neuron populations remain largely undefined, and a complete census of PVH cell types has not been established. Therefore, we performed extensive single-cell/nucleus RNA sequencing to catalog PVH neuron subtypes and multiplexed error-robust fluorescence in situ hybridization (MERFISH) to map them spatially. Our spatial transcriptomic atlas resolves 26 Sim1+ and 29 GAB

Anorexic behavior and elevation of hypothalamic malonyl-CoA in socially defeated rats.

Suppression of body weight and eating disorders, such as anorexia, are one of the major symptoms of psychiatric disorders such as depression. However, the mechanisms of weight loss and reduced appetite in depressive patients and in animal models of depression are largely unknown. In this study, we characterized the mechanism of anorexia resulting from depression using socially defeated rats as an animal model of depression. Socially defeated rats showed suppressed body weight gain, enlarged adrenal glands, decreased home cage activity, decreased food intake, and increased immobility in the for

Amylin causes anorexigenic effects via the hypothalamus and brain stem in chicks.

This study was conducted to determine the effects of amylin on appetite-related processes in chicks. Broiler chicks were centrally and peripherally injected with amylin, and feed and water intake were quantified. Feed intake was reduced after both central and peripheral amylin, but water intake was not affected. To determine if the hypothalamus and brainstem were involved in the anorexigenic effect, chicks were centrally and peripherally injected with amylin, and c-Fos immunoreactivity was quantified in the lateral hypothalamus (LH), ventromedial hypothalamus (VMH), area postrema (AP) and the

Grief in adults with mental retardation: preliminary findings.

Structured direct interviews of 43 adults with mental retardation were conducted to evaluate how they express grief and deal with loss. In addition, 100 providers, social workers, and clergy were surveyed to assess their views of how adults with mental retardation cope and respond during a grief situation. Adults with moderate to severe mental retardation display grief responses similar to all adults: a mixture of sadness, anger, anxiety, confusion, and pain. Individuals continue to experience emotional turmoil 1 year beyond the initial death and personal loss. Caregivers and professionals wer

Acute stress suppresses hunger-driven food seeking through PVN activation: Reversal by anxiolytic drug and ghrelin receptor agonist, with anxiolytic-like effects of refeeding.

Feeding behavior is influenced by both metabolic drive and emotional context, yet how acute stress interferes with hunger-driven motivation remains poorly understood. Using a conflict-based open-field feeding paradigm, we examined how 30-min restraint stress alters food-seeking behavior in fasted mice and how pharmacological or physiological interventions modulate this effect. Acute restraint abolished fasting-induced increases in food seeking and intake, an effect that was reversed by intraperitoneal diazepam, an anxiolytic, and by MK-677, a ghrelin receptor agonist that enhances appetite. Vagal sensory neurons encode internal protein status to guide eating.

Animals adaptively adjust nutrient intake based on internal physiological need. Although protein deficiency elicits robust behavioral and endocrine responses, the sensory mechanisms that detect dietary protein and guide selective feeding remain incompletely understood. Here, we identify a population of vagal sensory neurons that respond selectively to intragastric protein and are required for adaptive regulation of protein intake. Using activity-dependent genetic labeling and in vivo calcium imaging, we show that these neurons are activated by dietary protein, exhibit enhanced responses in pro

Effectiveness of Tri-Kaysorn-Mas Extract in Ameliorating Cognitive-like Behavior Deficits in Ovariectomized Mice via Activation of Multiple Mechanisms.

Postmenopausal women have a higher probability of experiencing cognitive alterations compared to men, suggesting that the decline in female hormones may contribute to cognitive deterioration. Thailand traditionally uses Tri-Kaysorn-Mas (TKM), a blend of three medicinal herbs, as a tonic to stimulate appetite and relieve dyspepsia. Due to its antioxidant and anti-acetylcholinesterase activities, we investigated the effects of TKM (50 and 100 mg/kg/day, p.o., for 8 weeks) on cognitive deficits and their underlying causes in an ovariectomized (OVX) mouse model of menopause. OVX mice showed cognit

Safety and mechanism of appetite suppression by a novel hydroxycitric acid extract (HCA-SX).

A growing body of evidence demonstrates the efficacy of Garcinia cambogia-derived natural (-)-hydroxycitric acid (HCA) in weight management by curbing appetite and inhibiting body fat biosynthesis. However, the exact mechanism of action of this novel phytopharmaceutical has yet to be fully understood. In a previous study, we showed that in the rat brain cortex a novel HCA extract (HCA-SX, Super CitriMax) increases the release/availability of radiolabeled 5-hydroxytryptamine or serotonin ([3H]-5-HT), a neurotransmitter implicated in the regulation of eating behavior and appetite control. The ai

Genetic parameters of feeding behavior traits and multi-tissue transcriptomic comparison of divergent feeding patterns in ducks.

Feeding behavior is a key factor influencing growth performance and feed efficiency in poultry, yet its complex phenotypes remain difficult to quantify accurately, limiting the application of large-scale behavioral data in genetic and molecular analyses. In this study, feeding behavior and feed efficiency-related phenotypes were recorded in 829 Tianfu Nonghua ducks using an automated feeding monitoring system integrated with radio frequency identification (RFID) technology. Genetic parameter estimation indicated that the heritability of feeding behavior traits ranged from 0.25 to 0.39. Individ

Not only gut feelings: pancreatic hormone, amylin, controls emotionality and sociability, in a sex divergent manner.

Amylin, a pancreatic peptide hormone, has gained attention for its role in appetite regulation. Its analogues are approved for use in diabetic patients, and it is now under investigation as a potential anti-obesity therapeutic. However, despite its clinical use, its effects on emotionality remain poorly understood. Given that many other food intake-modulating substances influence psychiatric functions, here we determined the role of systemic amylin in modulating key aspects of emotionality and sociability in male and female rats. We further evaluated whether the central amygdala (CeA) is suffi

Food responsiveness, addiction, and hyperphagia in Prader-Willi syndrome: a cross-sectional study of 210 Chinese patients.

OBJECTIVE: Prader-Willi syndrome (PWS) is the most common genetic syndromic obesity, characterized primarily by hyperphagia. It was described as excessive appetite, defective satiety, and obsession with food. However, the underlying mechanisms of hyperphagia in PWS remain obscure. This study aimed to determine the eating behavior patterns and food addiction tendencies in patients with PWS. METHODS: 210 patients with PWS from 26 provinces in China were enrolled in this study. The translated Children's Eating Behavior Questionnaire and modified Yale Food Addiction Scale for Children 2.0 were ad

Effect of ninjin'yoeito on age-related decline in responsiveness of the brain reward system in mice.

The reward system involved in the regulation of appetite and motivated behaviors involves mesolimbic dopamine signaling from the ventral tegmental area (VTA) to the nucleus accumbens (Nac). Age-related loss of dopaminergic neurons and weakening of dopamine signals reportedly cause reward system dysfunction, and similarly, age-related changes in microglia in the VTA have been suggested to be involved in this dysfunction. Ninjin'yoeito (NYT), a traditional Japanese herbal medicine, has been reported to improve anorexia, apathy, and the decline in motivation-related behavior in frail Alzheimer's

Acute nicotine activates orectic and inhibits anorectic brain regions in rats exposed to chronic nicotine.

Nicotine use produces psychoactive effects, and chronic use is associated with physiological and psychological symptoms of addiction. However, chronic nicotine use is known to decrease food intake and body weight gain, suggesting that nicotine also affects central metabolic and appetite regulation. We recently showed that acute nicotine self-administration in nicotine-dependent animals produces a short-term increase in food intake, contrary to its long-term decrease of feeding behavior. As feeding behavior is regulated by complex neural signaling mechanisms, this study aimed to test the hypoth

Dopamine D2 receptors in the bed nucleus of the stria terminalis modulate alcohol-related behaviors.

Dysregulation of the dopamine (DA) system is a hallmark of substance use disorders, including alcohol use disorder (AUD). Of the DA receptor subtypes, the DA D2 receptors (D2Rs) play a key role in the reinforcing effects of alcohol. D2Rs are expressed in numerous brain regions associated with the regulation of appetitive behaviors. One such region is the bed nucleus of the stria terminalis (BNST), which has been linked to the development and maintenance of AUD. Recently, we identified alcohol withdrawal-related neuroadaptations in the periaqueductal gray/dorsal raphe to BNST DA circuit in male…
