A lack of evidence for down-modulation of CD3 zeta expression in colorectal carcinoma and pregnancy using multiple detection methods.

Loss of the T cell receptor-associated CD3 zeta chain has been proposed as a possible mechanism of the acquired immunosuppression in both tumour-bearing hosts, and in symptomatic patients with HIV infection. However, other reports suggest that the zeta-chain loss may in part be caused by protease activity of contaminating phagocytes ex vivo. Using flow cytometry and Western blot analysis on highly purified T cells, and ensuring adequate addition of protease inhibitors, we have studied the expression of CD3zeta on peripheral blood T cells from patients with colorectal carcinoma, and compared th

Proliferation of CD3+ B220- single-positive normal T cells was suppressed in B-cell-deficient lpr mice.

It is known that lpr mice develop systemic lymphadenopathy and lupus erythematosus-like autoimmune disease that are associated with the accumulation of CD4- CD8- (double-negative; DN) CD3+ B220+ abnormal T cells as well as normal mature CD4+ or CD8+ single-positive (SP) CD3+ T cells. In order to clarify the role of B cells in the lymphoproliferation and autoimmunity of lpr mice, we created B-cell-deficient C57BL/6 (B6) lpr mice (B6lpr/lpr microMT/microMT) by crossing B6lpr/lpr mice with B6 microMT/microMT mice in which the B-cell development was arrested at pre-B stage owing to a targeted disr

Palmoplantar pustulosis: a clinical and immunohistological study.

Pustulosis palmoplantaris (PPP) is a common chronic skin disease, which is very resistant to treatment. It is not known why the lesions are located in the palms and soles. There are few studies of the disease and in particular studies of the histology. Fifty-nine patients with PPP answered a questionnaire concerning their medical history and 39 of them were clinically examined. Biopsy specimens were taken from involved skin in 22 of the 39 patients and studied immunohistologically for tryptase+ mast cells, EG2+ eosinophils, lipocalin+ neutrophils and CD3+ T lymphocytes. The sweat gland and swe

Constant detection of CD2, CD3, CD4, and CD5 in fixed and paraffin-embedded tissue using the peroxidase-mediated deposition of biotin-tyramide.

Immunohistochemical methods are widely used for diagnostic purposes in histopathology. However, the use of most monoclonal anti-leukocyte antibodies is limited to frozen tissues. Initially, it was believed that formalin fixation in particular, which is the gold standard for morphological tissue preservation, destroys most of the antigen binding sites. In recent years, protease digestion and the introduction of microwave techniques have significantly enhanced the sensitivity of immunohistochemical techniques, and a variety of hidden antigen sites in formalin-fixed tissue have been retrieved for

Aggressive gastric peripheral T-cell lymphoma of CD8+ type associated with lymphomatous meningitis.

Most gastric lymphomas are of B-cell origin, and many are either low-grade or high-grade B-cell lymphomas of mucosa-associated lymphoid tissue type. Gastric T-cell lymphomas are very rare, usually of CD4+ type, and few have had genotypic studies. We report the case of a gastric peripheral T-cell lymphoma of CD8+ type. Genotypic studies demonstrated partial deletion with rearrangement of the T-cell receptor beta-chain gene. Lymphomatous meningitis developed, and the patient died after only 2 months despite chemotherapy. The case is compared with the other gastric T-cell lymphomas that have been

The use of therapeutic drug monitoring to optimise immunosuppressive therapy.

Most experience of the therapeutic drug monitoring of immunosuppressive agents has been acquired in the field of solid organ transplantation; however, agents such as cyclosporin (cyclosporin A) are being increasingly utilised for the management of autoimmune diseases. Cyclosporin is the most widely studied immunosuppressant, but in spite of this many controversies still exist as to the optimum strategy for monitoring this drug. Owing to its widely variable pharmacokinetics and metabolism, and the absence of a simple method to measure therapeutic effectiveness, many factors should be considered

Recruitment during infectious mononucleosis of CD3+CD4+CD8+ virus-specific cytotoxic T cells which recognise Epstein-Barr virus lytic antigen BHRF1.

Epstein-Barr virus (EBV) is the aetiological agent of infectious mononucleosis (IM) which is a common sequel to primary EBV infection. Thereafter, the virus is maintained as a lifetime latent infection. Although the proteins expressed during the latent EBV infection provide a rich source of immunogenic epitopes, very little is known about cytotoxic T lymphocyte (CTL) control of primary EBV infection. The present report is based on an analysis of CTL clones derived from a patient suffering from acute IM. An intriguing feature of six CTL clones that displayed an HLA-restricted pattern of cell ly

Distribution and phenotypes of duodenal intraepithelial gamma/delta T cells in patients with various types of primary B-cell deficiency.

Expression of the gamma/delta T-cell receptor (TCR) on CD3+ intraepithelial lymphocytes (IEL) was studied in situ by two-color immunofluorescence on duodenal tissue sections from 34 infection-prone, adult patients with various types of primary hypogammaglobulinemia, classical Bruton's, Bruton-like or congenital, and common variable immunodeficiency. TCR gamma/delta+ IEL proportions (median 4.3%, range 0.3-43.3%) were within the range (0.3-38.3%) for histologically normal controls (n = 11), and there was no significant difference between the three patient categories. The total number of CD3+ IE

Translocation of CD3D gene in an acute myeloid leukemia (M5) with t(11;17)(q23;21).

Cytogenetic studies in patients with acute leukemia showed structural abnormalities on chromosome 11 at band q23 in five cases. Four of these had acute lymphoblastic leukemia (ALL) associated with t(4;11)(q21;q23) and one case had acute nonlymphoblastic leukemia (ANLL) (M5) associated with t(11;17)(q23;q21). We examined the CD3D and c-ets-1 genes in the t(11;17)(q23;q21) patient to ascertain any association between them and the chromosome change. In situ hybridization results showed that unlike in other studied cases with rearrangements of 11q23, the CD3D gene in the t(11;17)(q23;21) is transp

Peripheral engraftment of fetal intestine into athymic mice sponsors T cell development: direct evidence for thymopoietic function of murine small intestine.

Adult athymic, lethally irradiated, F1-->parent bone marrow-reconstituted (AT x BM) mice were engrafted bilaterally with day 16-18 fetal intestine or fetal thymus into the kidney capsule and were studied for evidence of peripheral T cell repopulation of 1-12 wk postengraftment. Throughout that time period, both types of grafts were macroscopically and histologically characteristic of differentiated thymus or intestine tissues, respectively. Beginning at week 2 postengraftment, clusters of lymphocytes were present within intestine grafts, particularly in subepithelial regions and in areas below

Recombination activating genes and CD3-TCR molecules are coexpressed in a subset of human T-cell acute lymphoblastic leukemias.

The V-(D) -J recombination that occurs during lymphoid differentiation is the basic event which settles the diversity of antigen receptors. Two recently cloned genes, RAG1 and RAG2, are sufficient to induce rearrangements of synthetic substrates when transfected into fibroblasts. In order to determine at which differentiation steps where the human RAG are expressed and in order to investigate the possible impairment of their expression by the leukemic process, we have recently studied a large series of B and T lymphoid neoplasias. We show that RAG are expressed in almost all sIg- B-ALL but not

Quantitative analysis of the immunocompetent cells in periapical granuloma: correlation with the histological characteristics of the lesions.

Granuloma formation includes an immune response in oral tissues to various microorganisms and their products. The immunocompetent cells of both series (T and B) are present in the periapical lesions. In order to further analyze the relative contribution and pathophysiological significance of the T cell subsets in granuloma formation, we undertook the quantitative analysis of the CD3-positive, CD4-positive, CD8-positive and Ig-positive cells in these lesions by using indirect immunofluorescence. Evidence is provided showing predominance of T cells in diffuse and B cells in focal mononuclear inf

TCR gamma delta bearing lymphocyte clones with lymphokine-activated killer activity against autologous leukemic cells.

Activated T lymphocytes with the T-cell receptor (TCR) gamma delta (CD3+ and TCR delta 1+) exhibit strong cytotoxic activity against the standard natural killer (NK) and lymphokine-activated killer (LAK) sensitive target cells. In order to test the cytotoxic activity of gamma delta T lymphocytes against autologous leukemic cells, 84 clones of gamma delta T lymphocytes were obtained from the peripheral blood of three acute lymphoblastic leukemia (ALL) patients. Forty-four of these T-cell clones were active against an LAK-sensitive cell line and the other 40 were active against K562, an NK targe

Expression of disulfide-linked and non-disulfide-linked forms of the T cell receptor gamma/delta heterodimer in human intestinal intraepithelial lymphocytes.

Immunohistochemistry has been used to investigate disulfide- and non-disulfide-linked forms of the T cell receptor gamma/delta heterodimer (TcR gamma/delta) in blood and intestinal epithelium of normal human small intestine, intestine of patients with untreated coeliac disease (in whom T cells expressing TcR gamma/delta are disproportionately raised), intestine of patients with tropical malabsorption, and in the human fetus. In blood from adult volunteers, 90% of T cells expressing TcR gamma/delta use the disulfide-linked form. In contrast in the epithelium in normal small intestine, coeliac d

The Ca2+-dependent phosphatase calcineurin controls the formation of the Carma1-Bcl10-Malt1 complex during T cell receptor-induced NF-kappaB activation.

T cell receptor (TCR) ligation induces increased diacylglycerol and Ca(2+) levels in T cells, and both secondary messengers are crucial for TCR-induced nuclear factor of activated T cells (NF-AT) and NF-κB signaling pathways. One prominent calcium-dependent enzyme involved in the regulation of NF-AT and NF-κB signaling pathways is the protein phosphatase calcineurin. However, in contrast to NF-AT, which is directly dephosphorylated by calcineurin, the molecular basis of the calcium-calcineurin dependence of the TCR-induced NF-κB activity remains largely unknown. Here, we demonstrate that calci

High-energy irradiation of matrix isolated acetic acid yields a ·CH3⋯CO2 complex: A spectroscopic and ab initio study.

The mechanism of the chemical transformations of isolated small organic molecules induced by high-energy radiation is of basic interest for astrophysics and astrochemistry. In this work, we first applied a combination of electron paramagnetic resonance (EPR) and Fourier-transform infrared (FTIR) spectroscopy to identify the products of the radiation-induced transformations of isolated CH3COOH and CD3COOH molecules. As revealed by EPR, ·CH3 (or ·CD3) is the principal primary radical generated from acetic acid in solid argon and xenon, while the FTIR results suggest that this radical is trapped

Integrating CD45 + CD3+ and CD3 + CD8+ T-cell markers into nomograms for predicting survival in gastroesophageal junction carcinoma.

To develop and validate prognostic nomograms for overall (OS) and progression-free survival (PFS) in gastroesophageal junction (GEJ) carcinoma to enable risk-stratified clinical decision-making. In this retrospective study of 618 patients, prognostic factors were selected using LASSO regression. Nomograms were constructed and validated by assessing discrimination (C-index, time-dependent ROC curves), calibration, and clinical utility (decision curve analysis). Patients were stratified into high- and low-risk groups based on nomogram-derived scores. The OS nomogram incorporated serum CD3 + CD8+

DNA Hypomethylating Agents Preserve T-cell Stemness and Potentiate the Efficacy of CD3-Bispecific Antibodies.

Bispecific antibodies targeting tumor-associated antigens and CD3 are promising therapeutic agents for both solid and hematologic cancers. CD3-bispecifics induce T-cell activation and cytotoxicity; however, prolonged T-cell receptor (TCR) stimulation can lead to chromatin rewiring and T-cell dysfunction, thereby limiting their full therapeutic potential. In this study, we investigate the combination of CD3-bispecifics with the DNA hypomethylating agent decitabine (DAC) and observe enhanced tumor growth inhibition in various preclinical models. Utilizing a prostate-specific membrane antigen (PS

Chimeric PRR T-cell-engager targeting cell surface β-1,3-glucan for invasive candidiasis.

Invasive candidiasis, primarily caused by Candida albicans, represents the most common fungal disease among hospitalized patients and poses a significant threat to human health. Intrinsic or acquired immunosuppression serves as a critical risk factor predisposing individuals to this disease, while simultaneously reducing the efficacy of conventional antifungal therapies and worsening clinical outcomes. Given the central role of immune dysfunction in the pathogenesis of invasive candidiasis, immunotherapeutic strategies hold substantial promise. We targeted dectin-1, the primary pattern recogni

The frequency of CD3+ lymphocytes in non-myocarditis endomyocardial biopsies.

Lymphocytic myocarditis is a serious disease with significant morbidity and mortality. Cardiovascular pathology has an important role in its diagnosis, a diagnosis historically made using the presence of a lymphocytic infiltrate and myocyte injury (Dallas Criteria). The European Society of Cardiology (ESC) criteria, additionally, use a threshold of immune cells, determined by CD3 immunohistochemical stains to render the diagnosis of myocarditis on endomyocardial biopsy. However, the frequency of immune cells in non-myocarditis endomyocardial biopsy cases is unclear and dependent on different e
