Linker for activation of T cells, zeta-associated protein-70, and Src homology 2 domain-containing leukocyte protein-76 are required for TCR-induced microtubule-organizing center polarization.

Engagement of the T cell with Ag on an APC results in a series of immediate signaling events emanating from the stimulation of the TCR. These events include the induced phosphorylation of a number of cellular proteins with a subsequent increase in intracellular calcium and the restructuring of the microtubule and actin cytoskeleton within the T cell. This restructuring of the cytoskeleton culminates in the polarization of the T cell's secretory apparatus toward the engaging APC, allowing the T cell to direct secretion of cytokines toward the appropriate APC. This polarization can be monitored

Guardians of T-Cell Ca2+ Stores: SERCA Pumps Integrated Within Complex Functional and Disease-State Signaling Dynamics.

T cells are the central regulators of the adaptive immune system, guiding both the cell-mediated and antibody-based elements of the immune response. Crucial to T-cell activation and differentiation, the T-cell receptor must transduce antigen exposure using a sustained elevated Ca2+ signal. A substantial body of research has identified and characterized multiple players in the Ca2+ signaling pathway, yet the sarcoplasmic/endoplasmic reticulum Ca2+-ATPase (SERCA) transporters, which intervene actively to regulate Ca2+ signal patterning and duration, remain relatively poorly characterized in the

PTH-Independent CaSR Inhibition with the Oral Calcilytic Encaleret Improves Calcium Homeostasis in Post-Surgical Hypoparathyroidism: A Phase 2 Open-Label Proof-of-Concept Trial.

The calcium-sensing receptor (CaSR) maintains calcium homeostasis by regulating parathyroid hormone (PTH) secretion and directly modulating renal calcium reabsorption. In the kidney, PTH and renal CaSR signaling exert opposing effects on tubular calcium reabsorption and urinary calcium excretion, complicating efforts to define their independent contributions to overall calcium regulation. To examine the PTH-independent effects of CaSR inhibition on renal calcium handling, we administered a calcilytic (CaSR antagonist) to individuals with post-surgical hypoparathyroidism (PSH). In this open-lab

Regulatory effects of exogenous calcium on sweet cherry fruit cracking and functional analysis of the PavCML42 gene.

Rain‑induced fruit cracking during sweet cherry ripening severely restricts industry development, but whether calcium signaling regulates cracking through CML genes remains unclear. Using the crack‑prone cultivar 'Brooks', we investigated the physiological and molecular mechanisms of calcium‑mediated regulation through exogenous CaCl2 spraying, physiological index measurements, transcriptome analysis, and gene function validation. Treatment with 0.5% CaCl2 was most effective, reducing the cracking rate by 53.3%, decreasing MDA content, modulating antioxidant enzyme activities, and inhibiting c…

Microglia stabilize sleep homeostasis via adenosine A3 receptor signaling.

Sleep homeostasis maintains the sleep-wake balance through sleep pressure, a process partly orchestrated by the accumulation of extracellular adenosine (eADO). Microglial Ca2+ activity has been implicated in sleep regulation, but the mechanism whereby microglia sense sleep pressure remains unclear. Here, we show that microglia regulate sleep homeostasis through brain state-dependent calcium ion activity driven by adenosine A3 receptor (A3R) signaling. Using miniaturized two-photon microscopy in freely behaving mice, we demonstrate that microglial calcium ion activity is rapidly altered by brai

Nox4 regulates InsP3 receptor-dependent Ca2+ release into mitochondria to promote cell survival.

Cells subjected to environmental stresses undergo regulated cell death (RCD) when homeostatic programs fail to maintain viability. A major mechanism of RCD is the excessive calcium loading of mitochondria and consequent triggering of the mitochondrial permeability transition (mPT), which is especially important in post-mitotic cells such as cardiomyocytes and neurons. Here, we show that stress-induced upregulation of the ROS-generating protein Nox4 at the ER-mitochondria contact sites (MAMs) is a pro-survival mechanism that inhibits calcium transfer through InsP3 receptors (InsP3 R). Nox4 medi

Atypical signaling and functional desensitization response of MAS receptor to peptide ligands.

MAS is a G protein-coupled receptor (GPCR) implicated in multiple physiological processes. Several physiological peptide ligands such as angiotensin-(1-7), angiotensin fragments and neuropeptide FF (NPFF) are reported to act on MAS. Studies of conventional G protein signaling and receptor desensitization upon stimulation of MAS with the peptide ligands are limited so far. Therefore, we systematically analyzed G protein signals activated by the peptide ligands. MAS-selective non-peptide ligands that were previously shown to activate G proteins were used as controls for comparison on a common ce

Effects of high-fat and low-calcium diets on the structure of filiform and fungiform tongue papillae in rats.

This study investigated how high-fat and calcium-deficient diets affect the tongue's structure and function in Wistar albino rats. These diets can lead to obesity, oxidative stress, and inflammation, which alter salivary gland function, impair taste bud regeneration, and disrupt epithelial integrity partly through changes in β-catenin signaling. The research examined histological changes in e filiform and fungiform papillae and evaluated β-catenin expression in the tongue epithelium. Forty-five male Wistar rats albino rats were divided into three groups: control, high-fat diet (HFD), and low c

The pathogenesis of Alzheimers disease is it a lifelong "calciumopathy"?

Alzheimer's disease (AD) is a fatal neurodegenerative disorder that has no known cure, nor is there a clear mechanistic understanding of the disease process itself. Although amyloid plaques, neurofibrillary tangles, and cognitive decline are late-stage markers of the disease, it is unclear how they are initially generated, and if they represent a cause, effect, or end phase in the pathology process. Recent studies in AD models have identified marked dysregulations in calcium signaling and related downstream pathways, which occur long before the diagnostic histopathological or cognitive changes…

CCL2 and CXCL1 trigger calcitonin gene-related peptide release by exciting primary nociceptive neurons.

Chemokines are important mediators in immune responses and inflammatory processes. Calcitonin gene-related peptide (CGRP) is produced in dorsal root ganglion (DRG) neurons. In this study, CGRP radioimmunoassay was used to investigate whether the chemokines CCL2 and CXCL1 could trigger CGRP release from cultured DRG neurons of neonatal rats and, if so, which cellular signaling pathway was involved. The results showed that CCL2 and CXCL1 ( approximately 5-100 ng/ml) evoked CGRP release and intracellular calcium elevation in a pertussis toxin (PTX)-sensitive manner. The CGRP release by CCL2 and C

Phospholipases: Paving the Way for a New Life.

Phospholipases are a large group of enzymes with ramifications on diverse physiological processes. These enzymes are involved in releasing phospholipids, which are subsequently utilized in the production of prostaglandin and calcium signaling. Calcium signaling is central to the function of phospholipases and calcium oscillations have an impact on the variety of events of embryo development. Additionally, the diverse isoforms of the enzymes can activate gene expression and regulatory factors in a stage-specific manner. Any alterations in the level of these enzymes or the pathways regulating th…

Organelle-orchestrated cGAS-STING signaling and its role in neurodegeneration.

The cGAS-STING signaling pathway serves as a central signalling axis of the innate immune system, and its aberrant activation plays a pivotal role in inflammatory responses. Recent studies have demonstrated that its regulation depends not only on individual organelles but also on a coordinated interorganelle network. This review systematically analyze how mitochondria, centrosomes, the endoplasmic reticulum (ER), membrane contact sites (MCSs), the Golgi apparatus, endosomes, and lysosomes collectively orchestrate cGAS-STING signaling. Mitochondria initiate signaling by releasing mitochondrial

Skeletal muscle Ca(2+) mishandling: Another effect of bone-to-muscle signaling.

Our appreciation of crosstalk between muscle and bone has recently expanded beyond mechanical force-driven events to encompass a variety of signaling factors originating in one tissue and communicating to the other. While the recent identification of new 'myokines' has shifted some focus to the role of muscle in this partnership, bone-derived factors and their effects on skeletal muscle should not be overlooked. This review summarizes some previously known mediators of bone-to-muscle signaling and also recent work identifying a new role for bone-derived TGF-β as a cause of skeletal muscle weak
