Noise rich in low frequency components, a new comorbidity for periodontal disease? An experimental study.

INTRODUCTION: Exposure to noise rich in low frequency components induces abnormal proliferation of extracellular matrix and collagens. The p … comparing control and exposed groups (P ≤ 0.004). The t-test for independent samples was applied to collagen type IV where no significant differences were found (P = 0.410), when comparing to the control group. DISCUSSION: As in other organs, we can observe fibrosis and the newly formed collagen is likely to be "nonfunctional", which could have clinical impact. …

The implantation of sepharose beads in mouse livers as an aid in the study of hepatic schistosomal fibrosis.

An experimental model of schistosomal portal fibrosis is described. Sepharose beads the size of schistosome eggs, loaded or not with soluble egg antigen (SEA) from Schistosoma mansoni, are injected into the coecal vein of C3H/Sn mice and become embolized in the liver. Only SEA-coated beads evoke a granulomatous reaction; this is enhanced by simultaneous priming of the mice with spleen cells from Schistosoma mansoni-infected syngeneic animals. The fibrosis, which ensues around the beads, is stable and is much more evident after priming. Preliminary collagen tissue immunotyping reveals the prese

In vitro model suggests oxidative stress involved in keratoconus disease.

Keratoconus (KC) affects 1:2000 people and is a disorder where cornea thins and assumes a conical shape. Advanced KC requires surgery to maintain vision. The role of oxidative stress in KC remains unclear. We aimed to identify oxidative stress levels between human corneal keratocytes (HCKs), fibroblasts (HCFs) and keratoconus cells (HKCs). Cells were cultured in 2D and 3D systems. Vitamin C (VitC) and TGF-β3 (T3) were used for 4 weeks to stimulate self-assembled extracellular matrix (ECM). No T3 used as controls. Samples were analyzed using qRT-PCR and metabolomics. qRT-PCR data showed low lev

Glomerular pathology in Alport syndrome: a molecular perspective.

We have known for some time that mutations in the genes encoding 3 of the 6 type IV collagen chains are the underlying defect responsible for both X-linked (where the COL4A5 gene is involved) and autosomal (where either COL4A3 or COL4A4 genes are involved) Alport syndrome. The result of these mutations is the absence of the sub-epithelial network of all three chains in the glomerular basement membrane (GBM), resulting, at maturity, in a type IV collagen GBM network comprising only α1(IV) and α2(IV) chains. The altered GBM functions adequately in early life. Eventually, there is onset of protei

Ruptured thoracoabdominal aortic aneurysm in a renal transplant patient with Alport's syndrome.

Alport's syndrome is a rare genetic disorder of type IV basement membrane collagen synthesis that typically presents with nephropathy, deafness, and ocular abnormalities. To the best of our knowledge, this is the first report in the world's literature of ruptured thoracoabdominal aortic aneurysm in a young patient with Alport's syndrome and a renal transplant. Hypotheses on an association between collagen disease in Alport's syndrome and aortic aneurysms are discussed.

[Familial hematuric nephropathies].

Alport syndrome is a hereditary hematuric nephritis progressing to end-stage renal failure, often associated with hearing loss and specific ocular changes. It is characterized by the presence of thickening and splitting of the glomerular basement membrane. Defects in type IV collagen, the main component of basement membranes, are responsible for the disease. The prevalence rate of Alport syndrome is 1/5,000 and it is the cause of about 2% of end-stage renal failure. The disease is heterogenous both at the clinical and genetic level. In most kindreds, it is transmitted as an X-linked dominant t

Loss of bullous pemphigoid antigen in peritumoral lacunas of basal cell carcinomas.

Peritumoral lacunas of basal cell carcinoma (BCC) were for a long time misinterpreted as fixation artifacts. However, recent studies showed that they might be considered as a dynamic process related to degenerescence for the palisade cells. We studied three antigens of the epidermal basement membrane zone--type IV collagen, laminin, and bullous pemphigoid antigen--by indirect immunofluorescence in six cases of basal cell carcinoma. We could observe that type IV collagen as well as laminin were expressed as a linear continuous staining pattern surrounding each of the BCC buds. When there was a

Morphological, immunohistochemical and ultrastructural changes in dimethylnitrosamine [correction of dimenthylnitrosamine] induced liver injury. Effect of malotilate.

Dimethylnitrosamine (DMN) induced liver injury in rats with cell necrosis, inflammation, hemorrhages, increased collagen type III synthesis and basement membrane component laminin and collagen IV localization in perisinusoidal sites. Malotilate ingestion during DMN treatment abolished inflammation and decreased interstitial collagen deposits and vascularization. It affected clearly less DMN-caused hemorrhage. When malotilate treatment was started subsequently to development of DMN-injury, it also caused decrease in inflammation, though less, as well as in collagen III, BM and fibronectin depos

Recessive osteogenesis imperfecta caused by missense mutations in SPARC.

Secreted protein, acidic, cysteine-rich (SPARC) is a glycoprotein that binds to collagen type I and other proteins in the extracellular matrix. Using whole-exome sequencing to identify the molecular defect in two unrelated girls with severe bone fragility and a clinical diagnosis of osteogenesis imperfecta type IV, we identified two homozygous variants in SPARC (GenBank: NM_003118.3; c.497G>A [p.Arg166His] in individual 1; c.787G>A [p.Glu263Lys] in individual 2). Published modeling and site-directed mutagenesis studies had previously shown that the residues substituted by these mutations form

Twenty-one novel mutations identified in the COL4A5 gene in Chinese patients with X-linked Alport's syndrome confirmed by skin biopsy.

BACKGROUND: The clinical and pathological features of Alport syndrome are characterized by abnormalities in the basement membrane collagen network which are composed of the α3, α4 and α5 chains of type IV collagen and usually associated with hearing loss and ocular lesions. The predominant form (85% of AS) is inherited as X-linked mode (XLAS) caused by mutations encoding the α5 chain of type IV collagen gene, COL4A5. Different mutations in the COL4A5 gene have been reported widely, but only a few mutations were identified in Chinese patients. METHODS: We studied 71 Chinese patients from 35 un

Allograft rejection and glomerular basement membrane antibodies in Alport's syndrome.

BACKGROUND: Anti-glomerular basement membrane (GBM) antibodies occasionally occur in Alport patients after renal allograft transplantation. METHODS: We report a patient with Alport's syndrome who lost four transplants each within the first year post transplantation. We searched for the presence of anti-GBM antibodies using recombinant NC1 domains of type IV collagen. Immunoblotting, enzyme linked immunosorbent assay (ELISA), and immunofluorescence were used to detect the presence of antibodies against the glomerular basement membrane. RESULTS: High antibody titers to the alpha3 chain (the Go

Gingival basement membrane antigens in desquamative lesions of the gingiva.

Desquamative gingival lesions are a clinical manifestation of several entities where histologic alterations in the gingival basement membrane are frequent. Normal and lesional gingival specimens from six patients with desquamative lesions of the gingiva associated with cicatricial pemphigoid or lichen planus and from one normal subject were examined by indirect immunofluorescence for immunologic alterations in basement membrane antigens. In four out of five desquamative gingival lesions from patients with lichen planus, type IV collagen and EBA basement membrane antigens were interrupted, thic

Ultrastructural study of the dermis in a case of type IV Ehlers-Danlos syndrome.

Light microscopic and ultrastructural examination of the skin in a case of type IV Ehlers-Danlos syndrome (EDS) showed the presence of morphological changes involving all the dermis components. Collagen fibres were smaller, more irregular and more spaced apart than in normal dermis. Conversely, elastic, microfibrillar and amorphous components were considerably increased. Collagen fibrils presented poorly defined outlines and a loose texture, besides being markedly reduced in size. Fibroblasts were characterized by variable degrees of dilatation of RER cisternae containing flocculent material.

The effects of age, vessel size, and Ehlers-Danlos type IV syndrome on the waviness index of arteries.

Ehlers-Danlos Syndrome is a rare hereditary connective tissue disorder produced by collagen abnormalities. The Type IV variant causes vascular rupture in the second and third decades. Definitive diagnosis at present is based on demonstrating a deficiency of Type III collagen or procollagen chemically or in fibroblast cultures. Both require fresh tissue. We have shown in fixed autopsy tissue that the elastin in two EDS IV patients appears accordion-pleated with a high waviness index (WI) of 1.58 +/- 0.028 (n = 90) compared to 1.35 +/- 0.08 (n = 446) for 21 controls aged 9 to 71. The WI is the r

Effects of methylprednisolone on glomerular and medullary mRNA levels for extracellular matrices in puromycin aminonucleoside nephrosis.

We examined the effects of methylprednisolone (MPSL) on type IV collagen, laminin and heparan sulfate proteoglycan (HSPG) mRNA levels in the renal glomeruli and medulla of puromycin aminonucleoside (PAN) nephrosis. mRNA levels encoding for type IV collagen and laminin increased markedly, whereas those for HSPG decreased significantly in glomeruli of PAN nephrosis. Administration of MPSL partially ameliorated the abnormal gene expression for basement membrane components. Furthermore, we showed that medullary mRNA levels for all these basement membrane components decreased with age in PAN nephro

Presence of type I and VI collagen mRNAs in endothelial cells in cutaneous neurofibromas.

In situ hybridization and peroxidase anti-peroxidase immunodetection were used in the same tissue sections to elucidate the spatial distribution of collagen gene expression in cutaneous neurofibromas, particularly in relation to blood vessels; the latter structures were identified by the presence of factor VIII-related antigen. The data indicate a clear relationship between the vascular structures and sites of locally elevated expression of type I and VI collagen genes. Specifically, some, but not all, blood vessels were surrounded by stromal cells highly active in expressing pro alpha 1(I) an

Perisigmoid Abscess Leading to a Diagnosis of Ehlers-Danlos Syndrome Type IV.

The Ehlers-Danlos syndromes (EDS) are a group of connective tissue disorders characterized by triad of joint hypermobility, skin extensibility, and tissue fragility. Ehlers-Danlos syndrome type IV places patients at risk for life-threatening, spontaneous, vascular or visceral rupture due to reduced or abnormal secretion of type III collagen. We present an adolescent male who was found to have a perisigmoid abscess with a fistula connecting to adjacent sigmoid colon secondary to undiagnosed EDS type IV. Conservative management with antibiotics and bowel rest was pursued to allow for elective re

Fatal hemoptysis in Ehlers-Danlos syndrome. Old malady with a new curse.

We describe the case of a 27-year-old man with Ehlers-Danlos syndrome, type IV. The patient had recurrent and eventually fatal pulmonary hemorrhage. Type IV Ehlers-Danlos syndrome is a rare disorder of type III collagen synthesis. It is characterized by an unusual facies, thin, translucent skin with venous vascular pattern, and hypermobility of the small joints. The cause of death is usually due to rupture of a viscus or a major arterial hemorrhage and, in women, rupture of the gravid uterus. Fatal lung hemorrhage in Ehlers-Danlos syndrome, to our knowledge, has not been previously described.

Loss of function of Colgalt1 disrupts collagen post-translational modification and causes musculoskeletal defects.

In a screen for organogenesis defects in N-ethyl-N-nitrosourea (ENU)-induced mutant mice, we discovered a line carrying a mutation in Colgalt1 [collagen beta(1-O)galactosyltransferase type 1], which is required for proper galactosylation of hydroxylysine residues in a number of collagens. Colgalt1 mutant embryos have not been previously characterized; here, we show that they exhibit skeletal and muscular defects. Analysis of mutant-derived embryonic fibroblasts reveals that COLGALT1 acts on collagen IV and VI, and, while collagen VI appears stable and its secretion is not affected, collagen IV
