Unravelling the consortium of the cercarial dermatitis in lake from a basin of Argentinian Central Andes: histological and phylogenetic insights of Chilina snail and their fluke partner.

In the context of a research program dealing with the symbiotic associations between aquatic snails and trematodes, we studied a host-parasite consortium in Lake El Nihuil (Mendoza, Argentina), a water body from Atuel river basin (Central Andes) in which recent outbreaks of cercarial dermatitis ('swimmer's itch') occurred. According to the morphological traits and molecular phylogenetic analyses (mitochondrial COI and 16S rRNA genes), the gastropod host was assigned to the endemic genus Chilina. The snails hosted and released a schistosomatid brevifurcate apharingeate cercaria, whose phylogene

Absence of Giardia spp. in fecal samples from capybaras (Hydrochoerus hydrochaeris) inhabiting urban areas in the state of Mato Grosso do Sul, Brazil.

Giardiasis is a major cause of diarrhea in humans and animals worldwide. Currently, there are nine species of Giardia, including Giardia duodenalis, which infects most vertebrates. The capybara (Hydrochoerus hydrochaeris) is the largest herbivorous rodent in the world. Although capybaras are hosts of several parasites of public health importance, including helminths and protozoa, there is a paucity of research on their zoonotic potential. We investigated the prevalence of Giardia spp. in populations of capybaras living in urban areas. Fecal samples from 247 capybaras were collected in Lagoa Ma

Schistosoma species detection by environmental DNA assays in African freshwaters.

BACKGROUND: Schistosomiasis is a neglected tropical parasitic disease associated with severe pathology, mortality and economic loss worldwide. Programs for disease control may benefit from specific and sensitive diagnostic methods to detect Schistosoma trematodes in aquatic environments. Here we report the development of novel environmental DNA (eDNA) qPCR assays for the presence of the human-infecting species Schistosoma mansoni, S. haematobium and S. japonicum. METHODOLOGY/PRINCIPAL FINDINGS: We first tested the specificity of the assays across the three species using genomic DNA preparatio

Identification and characterization of the Onchocerca volvulus Excretory Secretory Product Ov28CRP, a putative GM2 activator protein.

Onchocerca volvulus is the nematode pathogen responsible for human onchocerciasis also known as "River blindness", a neglected tropical disease that affects up to 18 million people worldwide. Helminths Excretory Secretory Products (ESPs) constitute a rich repertoire of molecules that can be exploited for host-parasite relationship, diagnosis and vaccine studies. Here, we report, using a range of molecular techniques including PCR, western blot, recombinant DNA technology, ELISA, high performance thin-layer chromatography and mass spectrometry that the 28 KDa cysteine-rich protein (Ov28CRP) is

Schistosoma mansoni Stomatin like protein-2 is located in the tegument and induces partial protection against challenge infection.

BACKGROUND: Schistosomiasis affects more than 200 million individuals worldwide, with a further 650 million living at risk of infection, constituting a severe health problem in developing countries. Even though an effective treatment exists, it does not prevent re-infection, and the development of an effective vaccine still remains the most desirable means of control for this disease. METHODOLOGY/PRINCIPAL FINDINGS: Herein, we report the cloning and characterization of a S. mansoni Stomatin-like protein 2 (SmStoLP-2). In silico analysis predicts three putative sites for palmitoylation (Cys11,

The Pristionchus HOX gene Ppa-lin-39 inhibits programmed cell death to specify the vulva equivalence group and is not required during vulval induction.

In the two nematode species Caenorhabditis elegans and Pristionchus pacificus the vulva equivalence group in the central body region is specified by the Hox gene lin-39. C. elegans lin-39 mutants are vulvaless and the vulval precursor cells fuse with the surrounding hypodermis, whereas in P. pacificus lin-39 mutants the vulval precursor cells die by apoptosis. Mechanistically, LIN-39 might inhibit non-vulval fate (cell fusion in C. elegans, apoptosis in P. pacificus), promote vulval fate or do both. To study the mechanism of lin-39 function, we isolated P. pacificus cell death mutants and iden

Screening diagnostic candidates for schistosomiasis from tegument proteins of adult Schistosoma japonicum using an immunoproteomic approach.

BACKGROUND: Schistosomiasis is one of the world's most prevalent zoonotic diseases and a serious worldwide public health problem. Since the tegument (TG) of Schistosoma japonicum is in direct contact with the host and induces a host immune response against infection, the identification of immune response target molecules in the schistosome TG is crucial for screening diagnostic antigens for this disease. METHODOLOGY/PRINCIPAL FINDINGS: In this study, an immunoproteomics approach used TG proteins as screening antigens to identify potential diagnostic molecules of S. japonicum. Ten spots corres

[Cloning, expression and protective efficacy evaluation of radiation sensitive protein 23 (RAD23) from Schistosoma japonicum].

Radiation sensitive protein 23 (RAD23) is a nucleotide excision repair (NER) protein that plays an important role in Ubiquitin-proteasome pathway (UPP). Schistosoma japonicum radiation sensitive protein23 (SjRAD23) cDNA sequences were amplified by PCR and cloned into pET28a (+) vector to construct recombinant expression plasmid pET28a(+)-SjRAD23. The recombinant protein was expressed as both inclusion bodies and the supernatant in Escherichia coli BL21 (DE3) cell. Immunofluorescence observation shows that SjRAD23 was mainly distributed on the tegument surface of the worms. ELISA assay reveals

Immunolocalization of anti-hsf1 to the acetabular glands of infectious schistosomes suggests a non-transcriptional function for this transcriptional activator.

Schistosomiasis is a chronically debilitating disease caused by parasitic worms of the genus Schistosoma, and it is a global problem affecting over 240 million people. Little is known about the regulatory proteins and mechanisms that control schistosome host invasion, gene expression, and development. Schistosome larvae, cercariae, are transiently free-swimming organisms and infectious to man. Cercariae penetrate human host skin directly using proteases that degrade skin connective tissue. These proteases are secreted from anucleate acetabular glands that contain many proteins, including heat

First identification of eggs of the Asian fish tapeworm Bothriocephalus acheilognathi (Cestoda: Bothriocephalidea) in human stool.

We report the first case of egg isolation of the Asian fish tapeworm Bothriocephalus acheilognathi (Bothriocephalidea) from human stool. A male patient from Saint Laurent du Maroni (French Guiana) presenting abdominal pain was examined in France for the diagnosis of intestinal parasites. Diphyllobothrium-like eggs were observed in his stool. However, molecular phylogenetic analyses based on sequences of rDNA and COI genes showed that the eggs observed belong to a bothriocephalidean cestode B. acheilognathi. The adult life stages of B. acheilognathi cestodes are known as invasive parasites of a

Sequence variation in the cytochrome oxidase I, internal transcribed spacer 1, and Ts14 diagnostic antigen sequences of Taenia solium isolates from South and Central America, India, and Asia.

We examined the genetic variability in the pig-human tapeworm, Taenia solium, by sequencing the genes for cytochrome oxidase I, internal transcribed spacer 1, and a diagnostic antigen, Ts14, from individual cysts isolated from Peru, Colombia, Mexico, India, China, and the Philippines. For these genes, the rate of nucleotide variation was minimal. Isolates from these countries can be distinguished based on one to eight nucleotide differences in the 396 nucleotide cytochrome oxidase I (COI) sequence. However, all of the 15 isolates from within Peru had identical COI sequences. The Ts14 sequences

Characterization of the heat-shock protein 60 chaperonin from Onchocerca volvulus.

Chaperonin 60 (cpn60) belongs to the group of ubiquitous molecular chaperones that comprise the heat shock proteins, nucleoplasmins and chaperonins. Antibodies to recombinant CPN60 from humans was used to screen a cDNA library of Onchocerca volvulus and antigen-positive clones were selected. Sequencing of the DNA inserts confirmed their identity as cpn60 transcripts. These are distinct from a cpn60 sequence recorded previously from O. volvulus (GenBank accession number Y09416) that appears to be of endobacterial origin, rather than derived from the parasite itself. The full-length sequence of

Hitching a ride across the continental divide of Mexico: the twospot livebearer Pseudoxiphophorus bimaculatus carried its Gyrodactylus spp. when translocated from the Atlantic to the Pacific Ocean versant.

We studied flatworm parasites of the genus Gyrodactylus associated with twospot livebearer Pseudoxiphophorus bimaculatus both in this host's native distribution in the Gulf of Mexico (Atlantic Ocean) versant, as well as in its invasive range across the continental divide, in the Pacific Ocean watershed. We surveyed 12 river basins, six on each slope of the continent, and recorded -apparently- overdispersed parasite distributions, both in terms of localities within basins and on fish hosts. Six gyrodactylid species were previously known to infect this poeciliid host, and we add five taxa to the

First molecular identification and characterization of Taenia lynciscapreoli in the Eurasian lynx (Lynx lynx) and Hydatigera taeniaeformis sensu stricto in domestic cats (Felis catus) from Central Asia.

Investigations of taeniid fauna in felids, including the Eurasian lynx, in Central Asia have so far relied exclusively on traditional morphological methods. However, species identification based solely on morphological features can be unreliable. Therefore, the aim of the present study was to identify and characterize taeniid species from the Eurasian lynx and domestic cats in Kazakhstan using molecular techniques. Taeniid cestodes were collected from the intestines of 18 Eurasian lynx from northern and central Kazakhstan. In addition, 856 fecal samples from domestic cats in Astana, Almaty, an

Phylogeny of two accacoeliid species (Digenea: Hemiuroidea) ex Mola mola (Linnaeus, 1758) (Tetraodontiformes: Molidae) from Northwest Pacific, with first molecular data on Odhnerium Yamaguti, 1934.

Accacoeliid digeneans associated with fish of the family Molidae exhibit enigmatically high taxonomic diversity. However, the phylogenetic relationships between species within this digenean taxon are poorly understood. In the present study, the first nuclear 28S rRNA gene, ITS2 region of nuclear DNA, and mitochondrial cox1 gene sequence datasets were obtained for two members of the Accacoeliidae, a type and only species of the genus Odhnerium Yamaguti 1934 and an unidentified Accacladocoelium sp. collected from Mola mola (Linnaeus, 1758) off Iturup Island. Analyses of molecular differentiation

New multiplex PCR method for the simultaneous diagnosis of the three known species of equine tapeworm.

Although several techniques exist for the detection of equine tapeworms in serum and feces, the differential diagnosis of tapeworm infection is usually based on postmortem findings and the morphological identification of eggs in feces. In this study, a multiplex polymerase chain reaction (PCR)-based method for the simultaneuos detection of Anoplocephala magna, Anoplocephala perfoliata and Anoplocephaloides mamillana has been developed and validated. The method simultaneously amplifies hypervariable SSUrRNA gene regions in the three tapeworm species in a single reaction using three pairs of pri
