This viewpoint presents a unifying concept for the treatment of atopic dermatitis (AD) that is based on the improvement of deficient Notch signalling, which appears to represent the fundamental epithelial defect of AD resulting in epidermal and immunological barrier dysfunction. One study of AD patients demonstrated a marked epidermal deficiency of Notch receptors and several mouse models with genetically suppressed Notch signalling exhibit dry skin, signs of scratching, skin barrier abnormalities, increased transepidermal water loss and Th2 cell-mediated immunological changes closely resembli

BACKGROUND & OBJECTIVES: Atopic diseases, including atopic dermatitis (AD), allergy and asthma, are complex diseases resulting from the effect of multiple genetic and interacting environmental factors on their pathophysiology. The genetic basis is incompletely understood; however, recent studies have shown an association between loss-of-function variants of the filaggrin gene (FLG) and atopic dermatitis. The aim of this study was to determine whether FLG variants can serve as a predictor for atopic diseases in Korean individuals.
METHODS: A total of 648 subjects were genotyped for the FLG P47

Atopic dermatitis (AD) is the most common childhood inflammatory skin disorder. The purpose of this study was to elucidate the knowledge level of daycare center teachers about AD and related factors. Study subjects were 297 teachers (287 female, 10 male) from 20 randomly selected daycare centers in Istanbul. The knowledge level of teachers was assessed using a questionnaire with 21 questions about AD. The teachers were asked nine additional questions about demographic and other characteristics of the child care centers and about themselves. The mean age of the teachers was 26.4 ± 8.1 years (ra

Prior studies have revealed the key roles played by Th1/Th2 cell dysregulation, IgE production, mast cell hyperactivity, and dendritic cell signaling in the evolution of the chronic, pruritic, inflammatory dermatosis that characterizes atopic dermatitis (AD). We review here increasing evidence that the inflammation in AD results primarily from inherited abnormalities in epidermal structural and enzymatic proteins that impact permeability barrier function. We also will show that the barrier defect can be attributed to a paracellular abnormality due to a variety of abnormalities in lipid composi

Bullous pemphigoid (BP) is an autoimmune blistering disorder that typically affects elderly patients. Rarely, it can occur in childhood. Vesicular pemphigoid is an atypical variant of BP. We report a case of childhood vesicular pemphigoid in an infant aged 6 months that was initially misdiagnosed as severe atopic dermatitis (AD). To the best of our knowledge, only one other case of childhood vesicular pemphigoid has been reported in the literature.

PURPOSE: The authors describe a patient with keratoglobus and a history of vernal keratoconjunctivitis and atopic dermatitis who acutely developed corneal hydrops and severe microbial keratitis. The infectious keratitis responded poorly to medical management and resulted in enucleation of the eye.
METHODS: A 25-year-old man presented with an acutely painful, red left eye. He had an ocular history of keratoglobus in association with vernal keratoconjunctivitis and atopic dermatitis. His visual acuity was light perception in the left eye and 20/40 in the right eye. Ocular examination showed a g

Diseases of the skin and visible mucous membranes are important signs of HIV infection. Dermatitis and eczema present mostly as seborrheic dermatitis. Thirty-one % of asymptomatic patients in seropositive latency period and up to 83% of AIDS patients present with this common symptom of HIV infection. Up to now there is no clear answer about the importance of atopic manifestations in AIDS. In children with pediatric AIDS, atopic eczema has been described in up to 50% of cases. In adult patients however there are contradictory results about the frequency of atopic diseases.

The spontaneous binding of sheep erythrocytes to human lymphocytes with the formation of rosettes was used as a measure of thymus-derived lymphocytes and the rosette formation with complement-coated sheep erythrocytes as a measure of bursa-dependent lymphocytes in the peripheral blood of patients with atopic dermatitis. Both percentage proportion and the absolute number of thymus-derived lymphocytes were slightly but significantly reduced. In a few patients with atopic dermatitis the percentage and absolute number of bursa-dependent lymphocytes were increased. However, when all values were con

PURPOSE OF REVIEW: To review the current literature for updates on pediatric nonsteroidal topical therapies (NSTT) for the treatment of atopic dermatitis (AD), psoriasis, alopecia areata (AA), vitiligo, and seborrheic dermatitis (SD).
RECENT FINDINGS: The FDA approved difamilast and expanded the approved age ranges for roflumilast, tapinarof, and ruxolitinib to treat AD. Recent studies also show positive results with delgocitinib. In the field of psoriasis, current research shows promise for tapinarof and roflumilast in an expanded age range. Among new AA treatments, relevant literature suppo

BACKGROUND: Myopathy in its severe forms including rhabdomyolysis is a very rare adverse effect occurring during monotherapy with the HMG-CoA reductase inhibitors ('statins') and is associated with pronounced signs of oxidation injury. This has been found at a local (muscle) as well as at a systemic level (blood). Several lines of evidence indicate that even mild forms of myopathy during statin treatment may be associated with in vivo oxidation injury. In contrast, statin therapy has been shown to be associated with a decrease in oxidation injury.
OBJECTIVE: The aim of this study was to inves

The retinoic acid receptor-related orphan receptors RORα and RORγ are critical for the functions of specific subsets of T cells and innate lymphoid cells, which are key drivers of inflammatory disease in barrier tissues. Here, we investigate the anti-inflammatory potential of SR1001, a synthetic RORα/γ inverse agonist, in mouse models of atopic dermatitis and acute irritant dermatitis. Topical treatment with SR1001 reduces epidermal and dermal features of MC903-induced atopic dermatitis-like disease and suppresses the production of type 2 cytokines and other inflammatory mediators in lesional

Atopic dermatitis (AD) is a chronic relapsing eczematous skin disease. Certain populations of patients are resistant to standard therapies with topical steroids and/or calcineurin inhibitors, and require systemic medication, such as immunosuppressants. Recently, several reports have shed light on the anti-allergic effects of carotenoids. Therefore, we investigated the effect of p.o. administration of β-carotene or lycopene on AD-like symptoms of HR-1 hairless mice fed with a low zinc/magnesium diet. Mice were divided into four groups: (i) fed with a standard diet (Co group); (ii) low zinc/magn

The development of atopic dermatitis in infancy, and subsequent allergies, such as asthma in later childhood, is known as the atopic march. The mechanism is largely unknown, however the course of disease indicates an inter-epithelial crosstalk, through the onset of inflammation in the skin and progression to other mucosal epithelia. In this study, we investigated if and how skin-lung epithelial crosstalk contributes to the development of the atopic march. First, we emulated inter-epithelial crosstalk through indirect coculture of bioengineered atopic-like skin disease models and three-dimensio

Abrocitinib, an oral selective Janus kinase 1 inhibitor, is approved for the treatment of moderate-tosevere atopic dermatitis (AD) in adults, but realworld evidence remains limited. We retrospectively collected data from 187 adult patients (mean age:37.9 years; 60.4% male), with AD and up to oneyear exposure to abrocitinib either 100 or 200 mg, between 2023 and 2025 across 22 Italian dermatology centres. Nearly half of patients were bio-experienced and 80 % had difficult-to-treat areas. Beginning at week 8, with either 100 (59%) or 200 mg, 52 % achieved Investigator Global Assessment 0/1, sign

The NLRP3 inflammasome is upregulated by various agents, such as nuclear factor-kappa B (NF-κB), lipopolysaccharide (LPS), and adenosine triphosphate (ATP). The NLRP3 inflammasome facilitations the maturation of interleukin (IL)-1β, a proinflammatory cytokine that is critically involved in the pathogenesis of atopic dermatitis (AD). Although the NLRP3 inflammasome clearly exacerbates AD symptoms such as erythema and pruritus, drugs for AD patients targeting the NLRP3 inflammasome are still lacking. Based on the previous findings that Mentha arvensis essential oil (MAEO) possesses strong anti-i
