Nanoantibiotics: A Novel Rational Approach to Antibiotic Resistant Infections.

BACKGROUND: The main drawbacks for using conventional antimicrobial agents are the development of multiple drug resistance due to the use of high concentrations of antibiotics for extended periods. This vicious cycle often generates complications of persistent infections, and intolerable antibiotic toxicity. The problem is that while all new discovered antimicrobials are effective and promising, they remain as only short-term solutions to the overall challenge of drug-resistant bacteria. OBJECTIVE: Recently, nanoantibiotics (nAbts) have been of tremendous interest in overcoming the drug resis

Self-reported pathways through which illicitly manufactured fentanyl enters the stimulant supply: Novel insights from people incarcerated for drug manufacturing and distribution.

BACKGROUND: The unintentional consumption of stimulants containing fentanyl among people who intend to only use stimulants contributes to overdose mortality in North America. Research exploring how fentanyl appears in the stimulant supply among people who manufacture and/or distribute drugs (PWDD) is critical to understanding supply-side factors that shape stimulant and opioid-involved overdose risk. METHODS: From April to July 2023, thirty PWDD incarcerated at the Rhode Island Department of Corrections completed an in-depth interview about stimulant and opioid-involved overdose. Data were th

Prevalence of QT-Prolonging Drug-Drug Interactions in Psychiatry: A Systematic Review and Meta Analysis.

Background: Drug-drug interactions (DDIs) are considered an emerging threat to the patients if undetected. DDIs can prolong QT interval, leading to fatal ventricular arrhythmia. Antipsychotics and antidepressants prescribed commonly to psychiatric patients have the propensity to prolong QT interval and can precipitate Torsades de pointes (TdP). This review aimed to summarize the prevalence of QT interval prolonging DDIs in psychiatric patients. Methods: This meta-analysis was carried out following the MOOSE (Meta-analysis of Observational Studies in Epidemiology) statement. Databases like Pubm

Targeting SARS-CoV-2 nucleocapsid oligomerization: Insights from molecular docking and molecular dynamics simulations.

The outbreak of COVID-19 caused by SARS-CoV-2 virus continually led to infect a large population worldwide. Currently, there is no specific viral protein-targeted therapeutics. The Nucleocapsid (N) protein of the SARS-CoV-2 virus is necessary for viral RNA replication and transcription. The C-terminal domain of N protein (CTD) involves in the self-assembly of N protein into a filament that is packaged into new virions. In this study, the CTD (PDB ID: 6WJI) was targeted for the identification of possible inhibitors of oligomerization of N protein. Herein, multiple computational approaches were

The FICI paradigm: Correcting flaws in antimicrobial in vitro synergy screens at their inception.

Antibiotics have become the corner stone of modern medicine. However, our society is currently facing one of the greatest challenges of its time: the emergence of antimicrobial resistance. It is estimated that if no new therapies are implemented by 2050, 10 million people will die worldwide every year as a result of infections caused by bacteria resistant to current antibiotics; new antimicrobials are thus urgently needed. However, drug development is a tedious and very costly endeavor of hundreds of millions that can take up to 15-20 years from the bench discovery to the bedside. Under this s

Iatrogenic Microplastic Exposure: A Possible and Underrecognized Healthcare-Associated Exposure Framework in Human Medicotoxicological Risk.

Microplastics (MPs) are emerging environmental contaminants detected not only in water, soil, and air but also in human biological samples. To date, three main exposure routes have been identified. Currently, the principal exposure routes examined in scholarly works are oral, inhalational, and dermal. This paper explores iatrogenic microplastic exposure (IME) as an underrecognized healthcare-associated source of exposure and suggests that, in certain clinical contexts involving invasive, device-mediated, or direct systemic contact, IME may be considered a possible fourth route of exposure. IME

Silica-containing products at autopsy: An uncommon incidental finding.

Silica-containing products are widely encountered in consumer and pharmaceutical packaging as moisture-absorbing agents. Although generally considered non-toxic, accidental ingestion of silica desiccants has been documented in pediatric and elderly populations and may result in mechanical complications such as choking or gastrointestinal obstruction, particularly with cylindrical canister formulations. The authors present two rare forensic cases in which silica desiccant material was found within the gastrointestinal tract during postmortem examination. Case 1 involved a 39-year-old woman with

"Our interventions are still here to support communities during the pandemic": Resuming mass drug administration for neglected tropical diseases after COVID-19 implementation delays.

The COVID-19 pandemic disrupted essential health services, including those provided by national neglected tropical disease (NTD) programs. Most mass drug administration (MDA) programs were postponed for 6-12 months following World Health Organization guidance released in April 2020 to temporarily halt NTD programs and launch necessary COVID-19 precautions. While NTD-endemic countries have since resumed MDA activities, it is critical to understand implementers' perspectives on the key challenges and opportunities for program relaunch, as these insights are critical for maximizing gains towards

VMAT2 Inhibitors and the Path to Ingrezza (Valbenazine).

The dopaminergic system plays a key role in the central nervous system, regulating executive function, arousal, reward, and motor control. Dysregulation of this critical monoaminergic system has been associated with diseases of the central nervous system including schizophrenia, Parkinson's disease, and disorders such as attention deficit hyperactivity disorders and addiction. Drugs that modify the dopaminergic system by modulating the activity of dopamine have been successful in demonstrating clinical efficacy by providing treatments for these diseases. Specifically, antipsychotics, both typi

In-silico investigation integrated with machine learning to identify potential inhibitors targeting AKT2: Key driver of cancer cell progression and metastasis.

BACKGROUND AND OBJECTIVE: In search of a key driver for the invasive growth of cancer metastasis, AKT2 is found to be exceptionally expressed in colorectal cancer and its metastasis. Again, exceeding genomic arrangements of AKT2 can be held responsible for HGSC (High-grade serous ovarian cancer) and breast cancer cell metastasis. FDA-approved capivasertib, a potential drug targeting the AKT signaling pathway, has a few side effects such as plausible alterations of liver function and gastrointestinal issues. Hence, this research aims to detect compounds with higher drug potency for selective AK

Therapeutic indications and other use-case-driven updates in the drug ontology: anti-malarials, anti-hypertensives, opioid analgesics, and a large term request.

BACKGROUND: The Drug Ontology (DrOn) is an OWL2-based representation of drug products and their ingredients, mechanisms of action, strengths, and dose forms. We originally created DrOn for use cases in comparative effectiveness research, primarily to identify historically complete sets of United States National Drug Codes (NDCs) that represent packaged drug products, by the ingredient(s), mechanism(s) of action, and so on contained in those products. Although we had designed DrOn from the outset to carefully distinguish those entities that have a therapeutic indication from those entities that

cellMCD Effectively Discovers Drug Resistance and Sensitivity Genes for Acute Myeloid Leukemia.

BACKGROUND: Rapid advances in biotechnology provide researchers with the opportunity to integrate omics profiles (genomics, epigenomics, transcriptomics, proteomics, etc.) with multiple phenotypes or experimental conditions. In cancers such as acute myeloid leukemia (AML), where combination therapies are standard of care, identifying genetic drivers of drug resistance requires evaluating how genes are associated with multiple drug response phenotypes. Statistical analyses associating omics profiles with multiple phenotypes yield multiple significance values and rankings for each of many genes.

Combined Quantitative and Qualitative Statistical Analyses Improve Benzodiazepine Target Discovery in Label-free Affinity-Based Protein Profiling Data.

Affinity-based protein profiling (AfBPP) allows us to identify target proteins that bind drugs or other small molecules of interest in complex samples. As an enrichment technique, label-free AfBPP often generates data with high missingness, particularly in negative control samples. We developed an R package, chemoprotR, which enables both quantitative and qualitative statistical analyses of chemoproteomic data, and applied it to the identification of specific benzodiazepine drug targets in the brain. Benzodiazepines comprise a class of drugs that affect GABAA receptors through positive alloste

Self-disintegrating protein-leaking microparticles for topical protein delivery.

Advanced drug delivery systems, if engineered to enhance drug stability and extend its kinetic bioavailability, are expected to expand the spectrum of local therapeutic approaches for dermatological conditions. In this context, the objective of this study was to explore whether artificial secretory granules, namely nontoxic, microscale granular Zn-assisted protein depots that leak the forming protein in a time-prolonged way, could be suited as functional agents in topical delivery. A model, recombinant form of the human fibroblast growth factor 2 (FGF-2), formulated as secretory granules, prom

Clinically Approved Antiviral Drug in an Orally Administrable Nanoparticle for COVID-19.

There is urgent therapeutic need for COVID-19, a disease for which there are currently no widely effective approved treatments and the emergency use authorized drugs do not result in significant and widespread patient improvement. The food and drug administration-approved drug ivermectin has long been shown to be both antihelmintic agent and a potent inhibitor of viruses such as Yellow Fever Virus. In this study, we highlight the potential of ivermectin packaged in an orally administrable nanoparticle that could serve as a vehicle to deliver a more potent therapeutic antiviral dose and demonst
