Systemic inhibition of mammalian target of rapamycin inhibits fear memory reconsolidation.

BACKGROUND: Established traumatic memories have a selective vulnerability to pharmacologic interventions following their reactivation that c … ically with the mTOR inhibitor rapamycin (1-40mg/kg), at various time points relative to contextual fear conditioning training or fear memory retrieval, and compared to vehicle or anisomycin-treated groups (N=10-12 in each group). RESULTS: Inhibition of mTOR via systemic administration of rapamycin blocks reconsolidation of an established fear memory …

[Allergy to penicillins and to cephalosporins: should we fear cross-reactivity?].

The diagnosis of allergy to penicillins is based on a positive history and cutaneous tests. The sensitivity of these tests is around 70%, therefore it is sometimes necessary to supplement them with in vitro tests, more rarely with drug provocation challenges. Among patients allergic to penicillin, 1 to 2% will develop an allergy to cephalosporins in the event of exposure to this antibiotic class. In this paper, we describe options for investigating and treating allergy to penicillins and cephalosporins.

Systemic or intrahippocampal delivery of histone deacetylase inhibitors facilitates fear extinction.

Several recent studies have shown that chromatin, the DNA-protein complex that packages genomic DNA, has an important function in learning a … SA) intrahippocampally prior to a brief (3-min) contextual extinction session causes context-evoked fear to decrease to levels observed with a long (24-min) extinction session. Fear of severe acute respiratory syndrome (SARS) among health care workers.

In this study, the authors examined fear related to severe acute respiratory syndrome (SARS) among 2 samples of hospital staff in Hong Kong. Sample 1 included health care workers (n=82) and was assessed during the peak of the SARS epidemic. Sample 2 included hospital staff who recovered from SARS (n=97). The results show that participants in both samples had equal, if not more, concern about infecting others (especially family members) than being self-infected. Sample 1 participants had stronger fear related to infection than Sample 2 participants, who seemed to be concerned more about other h…

Conditioned fear-related ultrasonic vocalizations are emitted as an emotional response.

Ultrasonic vocalizations (USVs) are observed along with freezing behavior when rats are placed under fearful situation. Coping style with stress is categorized into two forms, namely passive and emotional avoidance and active operant avoidance. In this study, fear-induced USV was compared between two conditioning protocols. Two groups of male Wistar rats were contextually conditioned for 10 days by exposing to the shock box. One group was shock-unavoidable and the other shock-avoidable by operant control of emitting the USVs. They were then proceeded to the extinction phase. As the results, th…

Isoflurane causes anterograde but not retrograde amnesia for pavlovian fear conditioning.

BACKGROUND: Production of retrograde amnesia by anesthetics would indicate that these drugs can disrupt mechanisms that stabilize memory. Such disruption would allow suppression of memory of previous untoward events. The authors examined whether isoflurane provides retrograde amnesia for classic (Pavlovian) fear conditioning. METHODS: Rats were trained to fear tone by applying three (three-trial) or one (one-trial) tone-shock pairs while breathing various constant concentrations of isoflurane. Immediately after training, isoflurane administration was either discontinued, maintained unchanged,…

Nonconscious fear conditioning, visceral perception, and the development of gut feelings.

When people are presented with backward-masked images of fear-relevant stimuli and only some of these images are paired consistently with electric shocks, they can predict the occurrence of shocks even though they do not consciously know which images they have seen. We postulated that they may use the perception of visceral cues from the conditional fear response to facilitate the prediction of shocks. In this study, ability to detect heartbeats was used to index sensitivity to visceral cues. The results showed that subjects who could detect their heartbeats performed better than chance in pre…

Fear-potentiated startle in adolescent offspring of parents with anxiety disorders.

BACKGROUND: The startle reflex and its potentiation by aversive states was used as a possible vulnerability marker for anxiety disorders in adolescent offspring of parents with this condition. METHODS: The participants were 39 low-risk adolescents (16 male/23 female) with a parental history of no psychiatric disorder and 35 high-risk adolescents (18 male/17 female) with a parental history of anxiety disorders. The magnitude of startle was examined at baseline and during anticipation of an aversive stimulus (fear-potentiated startle). RESULTS: Startle was found to discriminate between childre…

Fear, threat, and perceptions of efficacy from frightening skin cancer messages.

Fear appeal messages were designed and tested to ascertain their effectiveness in promoting skin protective behaviors. Theoretically guided by the Extended Parallel Process Model, the fear appeal messages in this study conveyed the seriousness of exposed and unprotected skin for college students in the Southwestern United States, and recommended a series of behaviors that would protect individuals from serious sunburns. The results demonstrated that fear appeals are effective in promoting skin protective behaviors. Specifically, highly threatening fear appeal messages were most effective when …

Evidence that GABA transmission mediates context-specific extinction of learned fear.

Six experiments used rats to study the effects of the beta-carboline FG 7142 on extinction of fear responses (freezing) to an auditory cue that had signalled footshock. Subcutaneous injection of FG 7142 interfered with the development of extinction without having any detectable effect on the rats' levels of fear prior to extinction. Injection of FG 7142 also reversed extinction, partially reinstating fear responses that had been extinguished previously. A similar reinstatement of extinguished fear was seen when rats were tested for fear of the cue in a different chamber. The reinstatement prod…

Toward a standard experiment for studying post-treatment return of fear.

Fear sometimes returns after successful fear attenuation via in vivo exposure to fear signals. Post-treatment return of fear is of considerable interest both practically and theoretically, but factors associated with return of fear are poorly understood due to conflicting results from procedurally diverse experiments. This paper reports two very similar experiments in which fear of animal specimens was weakened then allowed to return so that factors associated with return of fear could be studied. In each experiment attentional focus versus distraction during exposure served as a between-subje…

Confirmatory factor analysis of the Fear Questionnaire with social phobia patients.

BACKGROUND: The Fear Questionnaire is an internationally popular measure in anxiety disorder research, but its validity with social phobia patients has not been clearly demonstrated. METHOD: A confirmatory factor analysis was conducted using the Fear Questionnaire responses of 122 social phobia patients. RESULTS: The results indicated that the proposed three-factor model of the scale (agoraphobia, social phobia, and blood/injury dimensions) provided a good fit to the data. CONCLUSIONS: These data provide strong support for the validity of the scale and for the multidimensional view of fears…

Cerebral correlates of anticipated fear: a PET study of specific phobia.

Cerebral correlates of anticipatory anxiety was studied in 14 women with specific snake or spider phobia, and in 6 nonphobic controls. Videofilms with neutral scenes were shown during positron emission tomographic measurements of regional cerebral blood flow (rCBF). The phobics, but not the controls, anticipated scenes with spiders or snakes to appear in the videofilms. This was reflected in higher anxiety ratings in the phobics. The primary visual cortex rCBF was lower in phobics than controls, while no rCBF differences were found in the other brain regions investigated. The results are sugge

Scopolamine selectively disrupts the acquisition of contextual fear conditioning in rats.

Muscarinic cholinergic antagonism produces learning and memory deficits in a variety of hippocampal-dependent tasks. Hippocampal lesions produce both acquisition deficits and retrograde amnesia for contextual fear conditioning, but do not impact fear conditioning to discrete cues. In order to examine the effects of muscarinic antagonism in this paradigm, rats were given scopolamine (1 mg/kg) either before or for 3 days after a Pavlovian fear-conditioning session in which tones were paired with aversive footshocks. Fear to the context and the tone was assessed by measuring freezing in separate …

Fear-relevant illusory correlations: what types of associations promote judgmental bias?

A. J. Tomarken, S. Mineka, and M. Cook (1989) found that high-fear individuals markedly overestimated the covariation between fear-relevant stimuli and aversive outcomes. The authors assessed what features of stimulus-outcome associations promote illusory correlations. In Experiment 1, participants with high snake fear exhibited significant covariation bias for slides of snakes and shocks, but not for slides of damaged electric outlets (DEOs) and shocks. In Experiment 2, individuals with high and low snake fear rated DEOs and shocks as belonging together better than snakes and shocks. However,…

[Minimizing pain and fear of venipunctures in 6-year-old children].

The aim is to describe the interventions used by nurses to reduce pain and fear in children during venipuncture. The aim of the various methods is not to eliminate all the pain, but rather to encourage the child to adopt a behavior which will not hamper even long-term treatment. The material was collected from Oulu University Hospital by means of open questionnaires to a total of 59 nurses and laboratory assistants regarding their work. A content analysis was performed, replies significant from the point of view of the present topic being tabulated and the results depicted in the form of frequ…

A 10-year follow-up of patients treated for dental fear.

Different types of treatment have been used in trying to reduce dental anxiety in adult patients with severe fear and avoidance behavior. However, there is an obvious shortage in long-term treatment follow-up. The present investigation reports results over a period of 10 yr of 14 patients treated for dental fear in a specialized dental fear treatment and research clinic. The aims of this project were to evaluate patients' regularity in dental behavior between 1977 and 1987, changes in and present level of dental fear and other long-standing positive/negative effects of the respective initial t…
