Viral and host factors that contribute to pathogenicity of enterovirus 71.

The single-stranded RNA virus enterovirus 71 (EV71), which belongs to the Picornaviridae family, has caused epidemics worldwide, particularly in the Asia-Pacific region. Most EV71 infections result in mild clinical symptoms, including herpangina and hand, foot and mouth disease. However, serious pathological complications have also been reported, especially for young children. The mechanisms of EV71 disease progression remain unclear. The pathogenesis of adverse clinical outcomes may relate to many factors, including cell tropism, cell death and host immune responses. This article reviews the

Exploring gaps in our knowledge on Lyme borreliosis spirochaetes--updates on complex heterogeneity, ecology, and pathogenicity.

The Lyme borreliosis complex is a heterogeneous group of tick-borne spirochaetes of the genus Borrelia (Spirochaetales: Spirochaetaceae) that are distributed all over the temperate zone of the northern hemisphere. Due to the usage of new methods for phylogenetic analysis, this group has expanded rapidly during the past 5 years. Along with this development, the number of Borrelia spp. regarded as pathogenic to humans also increased. Distribution areas as well as host and vector ranges of Lyme borreliosis agents turned out to be much wider than previously thought. Furthermore, there is evidence

Mechanisms of meningococcal colonisation.

Despite advances against infectious diseases over the past century, Neisseria meningitidis remains a major causative agent of meningitis and septicaemia worldwide. Its adaptation for survival in the human nasopharynx makes the meningococcus a highly successful commensal bacterium. Recent progress has been made in understanding the mechanisms that enable neisserial colonisation, in terms of the role of type IV pili, the impact of other adhesins, biofilm formation, nutrient acquisition and resistance to host immune defences. Refinements in cell-based and in vivo models will lead to improved unde

Bacillus anthracis: molecular taxonomy, population genetics, phylogeny and patho-evolution.

Bacillus anthracis, the etiological agent of anthrax, manifests a particular bimodal lifestyle. This bacterial species alternates between short replication phases of 20-40 generations that strictly require infection of the host, normally causing death, interrupted by relatively long, mostly dormant phases as spores in the environment. Hence, the B. anthracis genome is highly homogeneous. This feature and the fact that strains from nearly all parts of the world have been analysed for canonical single nucleotide polymorphisms (canSNPs) and variable number tandem repeats (VNTRs) has allowed the d

Effect of host change on demographic fitness of the parasitoid, Trichogramma brassicae.

Trichogramma brassicae (Bezdenko) is the most important species of Trichogramma parasitoids in Iran. The cotton bollworm, Helicoverpa armigera (Hubner) is a polyphagous insect pest that attacks many crops including cotton, maize, soybean, tomato, etc. The bollworm egg is a suitable target for many Trichogramma species. Factitious hosts such as eggs of the flour moth, Anagasta kuehniella (Zeller) and cereal moth, Sitotroga cerealella (Hubner) are used for mass rearing purposes. But a problem that arises sometimes in laboratory cultures is the development of a tendency toward laboratory hosts fo

Modeling the viral dynamics of influenza A virus infection.

Influenza virus causes an acute, mostly self-limited, infection of the upper respiratory tract. Yearly epidemics of influenza infect up to 20% of the population, and in the US cause an average of 36,000 deaths every year. Because influenza is a short-term infection lasting 4 to 7 d in most cases, studying the dynamics of the virus and the immune response in vivo is difficult. Here we review the most recent attempts at mathematical modeling of influenza dynamics within the host to better understand the kinetics of the virus and associated immune responses. These models have been developed based

Hepatitis C virus infection: a "liaison a trois" amongst the virus, the host, and chronic low-level inflammation for human survival.

This review covers the various aspects of the immune system that allows the relationship between the hepatitis-C virus, the host and chronic low-level inflammation, to be highly flexible and able to defend the host from persistent infections. This ambiguity mainly stems from the property of the immune system that can be both protective and harmful. Immunity cannot be fully protective without producing a certain degree of damage (acute hepatitis resulting in resolving HCV infection). In addition, the balance between protection and tissue damage is critical for the development of chronic HCV inf

Variability in inhibition of host RNA synthesis by entero- and cardioviruses.

Both entero- and cardioviruses have been shown to suppress host mRNA synthesis. Enteroviruses are also known to inhibit the activity of rRNA genes, whereas this ability of cardioviruses is under debate. This study reported that mengovirus (a cardiovirus) suppressed rRNA synthesis but less efficiently than poliovirus (an enterovirus). In contrast to poliovirus infection, the incorporation of BrUTP, fluorouridine and [14C]uridine in rRNA precursors was observed even during the late stages of mengovirus infection, although at a significantly reduced level. The cleavage of TATA-binding protein, co

Histoplasma capsulatum at the host-pathogen interface.

Histoplasma capsulatum is the most common cause of invasive fungal pulmonary disease worldwide. The interaction of H. capsulatum with a host is a complex, dynamic process. Severe disease most commonly occurs in individuals with compromised immunity, and the increasing utilization of immunomodulators in medicine has revealed significant risks for reactivation disease in patients with latent histoplasmosis. Fortunately, there are well developed molecular tools and excellent animal models for studying H. capsulatum virulence and numerous recent advances have been made regarding the pathogenesis o

Molecular mechanisms underlying group A streptococcal pathogenesis.

Group A Streptococcus (GAS) is a versatile human pathogen causing diseases ranging from uncomplicated mucosal infections to life-threatening invasive disease. The development of human-relevant animal models of GAS infection and introduction of new technologies have markedly accelerated the pace of discoveries related to GAS host-pathogen interactions. For example, recently investigators have identified pili on the GAS cell surface and learned that they are key components for adherence to eukaryotic cell surfaces. Similarly, the recent development of a transgenic mouse expressing human plasmino…

Paracoccidioides brasiliensis: phylogenetic and ecological aspects.

The knowledge on the biological aspects of Paracoccidioides brasiliensis has evolved greatly since the first description of the disease in 1908. From the pioneers, who were able to clearly demonstrate the fungal nature of the agent, to the recent genomic era, important advances have been achieved. P. brasiliensis is a true fungus, belonging to the Ascomycetous Division, although its sexual phase has not been demonstrated morphologically. A better understanding of the fundamental aspects of the agent, especially its phylogeny and evolutionary history, will provide us with valuable insights allo

Vascular endothelium: the battlefield of dengue viruses.

Increased vascular permeability without morphological damage to the capillary endothelium is the cardinal feature of dengue haemorrhagic fever (DHF)/dengue shock syndrome (DSS). Extensive plasma leakage in various tissue spaces and serous cavities of the body, including the pleural, pericardial and peritoneal cavities in patients with DHF, may result in profound shock. Among various mechanisms that have been considered include immune complex disease, T-cell-mediated, antibodies cross-reacting with vascular endothelium, enhancing antibodies, complement and its products, various soluble mediator

Pathogen Effector-Mediated Reprogramming of Plant Alternative Splicing: From Immune Regulation to Crop Disease Resistance.

Plants have evolved sophisticated immune systems to defend against diverse pathogens, whereas pathogens deploy effectors to manipulate host cellular processes for successful infection. Recent studies have revealed that pathogen effectors can target host RNA processing, particularly pre-mRNA alternative splicing (AS), to reshape transcript isoform profiles and modulate plant immunity. However, the common principles and specific differences among pathogen effector-mediated regulation of host AS and their impacts on plant immunity have not been systematically summarized. In this review, we summar

Investigation of Whole Cell Meningococcal Glycan Interactions Using High Throughput Glycobiology Techniques: Glycan Array and Surface Plasmon Resonance.

A growing body of evidence suggests that glycans are important for meningococcal host-pathogen interactions and virulence. The development of glycobiology techniques such as glycan array analysis and surface plasmon resonance (SPR) has increased awareness of the importance of glycans in biological processes and has increased the interest of their study. While these techniques are more routinely used with purified proteins, there is growing interest in their applicability to cell-based studies, to better emulate host-pathogen interactions in vivo. Here we describe the use of glycan array analys…

Reframing clathrin-mediated viral entry: evidence standards, context dependence and host-directed vulnerabilities.

Viral entry is the first obligatory step of productive infection and a rational point for host-directed antiviral intervention. This structured narrative review synthesizes evidence from database searches and citation tracking performed up to 18 June 2026. Clathrin-mediated endocytosis (CME) can internalize virus-receptor complexes and deliver them to endosomal compartments that support penetration, fusion, capsid remodeling, uncoating or genome release. However, CME should be interpreted as a series of experimentally separable checkpoints rather than as a single pathway label. Viral use of CM

Carbon and nitrogen sources can fuel fungal virulence.

For all pathogenic microbes, metabolic processes are essential for growth, development, and survival during infection. However, host niches are dynamic environments where nutrient availability can vary, and host defense mechanisms often restrict access to host-derived nutrients. Therefore, microbes must adapt to these microenvironments by regulating nutrient acquisition and metabolism, and by employing mechanisms such as virulence factors that release host-derived nutrients. This metabolic plasticity is a key feature that enables certain microbes to thrive during host-microbe interactions. Hen

Microgravity reshapes bacteriophage-host coevolution aboard the International Space Station.

Bacteriophage-host interactions play a fundamental role in shaping microbial ecosystems. While extensively studied on Earth, their behavior in microgravity remains largely unexplored. Here, we report the dynamics between T7 bacteriophage and Escherichia coli in microgravity aboard the International Space Station (ISS). Phage activity was initially delayed in microgravity but ultimately successful. We identified de novo mutations in both phage and bacteria that enhanced fitness in microgravity. Deep mutational scanning of the phage receptor binding domain revealed striking differences in the nu

Topological dependence of viral mutation spread in complex host-interaction networks.

The increase in the connectivity between hosts in recent times has facilitated the emergence of more aggressive mutant viral strains, making their containment and eradication significantly more challenging compared to the original variants. We focus on the evolution of a new more aggressive mutant strain that appears at specific nodes in the system and competes with a previous version until reaching fixation. The role of host-interaction network topology in the emergence and spatial diffusion of these highly contagious strains is analyzed, as well as how network-based interventions can help mi
