Effects of Athamanta turbith fruit essential oils on CCl4-induced hepatic failure in mice and their antioxidant properties.

The effects of essential oils isolated from mature fruits of Athamanta turbith ssp. hungarica (Borbás) Tutin and A. turbith ssp. haynaldii (Borbás & Uechtr.) Tutin (Umbelliferae) on some liver biochemical parameters in mice intoxicated with carbon tetrachloride were investigated. Pretreatment with both essential oils extenuated the effects caused by carbon tetrachloride. In order to investigate in vitro antioxidant properties of the oils, three methods were applied: scavenging of both 2,2-diphenyl-1-picrylhydrazyl (DPPH) and OH radicals, as well as a test of inhibition of Fe(2+)/ascorbic-induc

The effect of quinine on outer hair cell shape, compliance and force.

Quinine intoxication causes a well-described syndrome that includes tinnitus, sensorineural hearing loss and vertigo. The pathophysiology of quinine's effects on hearing is unknown, but may include a peripheral component. The cochlear outer hair cell is known to be motile and to contribute force to amplify the vibration pattern of the organ of Corti. The outer hair cell is also a target of diseases involving tinnitus and sensorineural hearing loss, including salicylate intoxication. These effects may be mediated through changes either in motile force or in mechanical properties. Quinine's effe

Aminophylline: could it act as an antioxidant in vivo?

Potential antioxidant properties of aminophylline and theophylline were investigated. We have found that these drugs, though ineffective against superoxide anion and hydrogen peroxide, are scavengers of hydroxyl radical (OH.). Indeed, second-order rate constants (k) of aminophylline and theophylline with OH. are about 1.9 x 10(10) mol-1 s-1 and 4.5 x 10(9) mol-1 s-1, respectively. Ethylenediamine, which is present in the aminophylline molecule, significantly contributes to this marked OH. scavenging activity, since it is characterized by a high k value, i.e. about 8 x 10(9) mol-1 s-1. However,…

High field proton NMR investigations of the metabolic profiles of multidrug-sensitive and -resistant leukaemic cell lines: evidence for diminished taurine levels in multidrug-resistant cells.

High field proton (1H) nuclear magnetic resonance (NMR) spectroscopy has for the first time been employed to investigate and compare the metabolic profiles of vinblastine-sensitive and -resistant T-lymphoid leukaemic cell lines (CCRF-CEM and CEM/VLB100 respectively) and evidence is presented for a significantly lower taurine content in the CEM/VLB100 resistant subline when expressed relative to that of its drug-sensitive parental counterpart. These data suggest differences in the nature and relative involvements of taurine biosynthetic pathways between the two cell lines, a phenomenon that may

Sulfate-mediated electrooxidation of X-ray contrast media on boron-doped diamond anode.

Recently, electrochemical activation of sulfate ions to sulfate radical species and nonradically activated persulfate has been demonstrated … diatrizoate were observed in sulfate anolyte. These products have not been reported previously for hydroxyl radical-mediated oxidation of ICM. Given that electron-transfer mechanism is more typical for sulfate than for hydroxyl radicals, formation of hydroxylamine and nitro-derivatives of ICM was assigned to one-electron charge transfer to sulfate radical species …

Heterogeneous activation of peroxymonosulfate by hierarchical CuBi2O4 to generate reactive oxygen species for refractory organic compounds degradation: morphology and surface chemistry derived reaction and its mechanism.

Activation of peroxymonosulfate (PMS) by a novel hierarchical CuBi2O4 generated reactive oxygen radical for degradation refractory organic compounds in aqueous solution, which would be controlled by the morphology and surface chemistry of solid catalyst. It's found that the activation ability of CuBi2O4 toward PMS was highly dependent on the morphology and surface hydroxyl group, as using rhodamine B (RhB) as the model compound. The spherical CuBi2O4, which possessed higher density of surface hydroxyl group, exhibited better catalytic activity in RhB degradation than scattered cluster CuBi2O4,

Role of the surface characteristics of hyper-crosslinked polymers on the transformation of adsorbed trichlorophenol: Implications for understanding the surface reactivity of biochar derived from waste biomass.

The surface reactivity of biochar derived from waste biomass has not been well understood due to its complex composition and heterogeneity. … f aromatization and graphitization were negatively correlated. It was found that greater amounts of hydroxyl radicals were produced with increasing amounts of hydroxyl groups on the synthesized HCPs. Batch degradation experiments with trichlorophenols (TCPs) suggested that all HCPs could decompose TCP molecules upon contact. The degree of TCP degradation (~45 %) w …

Fluoroquinolone antimicrobials: singlet oxygen, superoxide and phototoxicity.

The fluoroquinolone antibacterial agents possess photosensitizing properties that lead to phototoxic responses in both human and animal subjects. The phototoxicity order reported in humans is: fleroxacin > lomefloxacin, pefloxacin >> ciprofloxacin > enoxacin, norfloxacin and ofloxacin. Studies both in vivo and in vitro have related this phototoxicity to the generation of reactive oxygen species including hydrogen peroxide and the hydroxyl radical. We determined the quantum yields of singlet oxygen generation (phi delta) by detection of the singlet oxygen (1O2) luminescence at 1270 nm for sever…

Use of salicylate as a probe for .OH formation in isolated ischemic rat hearts.

Salicylic acid was used as a probe for .OH formed during reperfusion of the ischemic myocardium. .OH adds to the phenolic ring of salicylate to yield dihydroxybenzoic acid species. The two principal dihydroxybenzoic acids formed are the 2,3- and 2,5-derivatives and can be isolated and quantitated using HPLC combined with electrochemical detection. In these experiments, dihydroxybenzoic acids were detectable in the f molar range. Rat hearts were perfused in the Langendorff mode with Krebs-Henseleit buffer containing 100 microM salicylate. Following 20 min of global ischemia a 173% increase in t

Free radicals and activated oxygen.

Superoxide anion (0(-2)), hydrogen peroxide (H2O2) and hydroxyl radical (OH.) are products of the biological reduction of 0(2). They are very reactive and poorly tolerated within living systems and enzymes that catalytically scavenge these products have been evolved as defense mechanisms. These include superoxide dismutases (SOD), catalase and peroxidases. Large amounts of O-2 are produced by different enzymatic and non enzymatic biological processes. Large amounts of activated oxygens are produced by phagocytosing cells such as macrophages and polymorphonuclear cells. This production is assoc…

Cysteine and resistance to oxidative stress: implications for virulence and antibiotic resistance.

Reactive oxygen species (ROS), including the superoxide radical anion (O2•-), hydrogen peroxide (H2O2), and the hydroxyl radical (•HO), are inherent components of bacterial metabolism in an aerobic environment. Bacteria also encounter exogenous ROS, such as those produced by the host cells during the respiratory burst. As ROS have the capacity to damage bacterial DNA, proteins, and lipids, detoxification of ROS is critical for bacterial survival. It has been recently recognised that low-molecular-weight (LMW) thiols play a central role in this process. Here, we review the emerging role of cyst…

Interaction of rhodanese with intermediates of oxygen reduction.

Cyanide-promoted inactivation of the enzyme rhodanese [thiosulfate sulfurtransferase (EC 2.8.1.1)] in the presence of ketoaldehydes is caused by reduced forms of molecular oxygen generated during autoxidation of the reaction products. The requirement of both catalase and superoxide dismutase to prevent rhodanese inactivation indicates that hydroxyl radical could be the most efficient inactivating agent. Rhodanese, also in the less stable sulfur-free form, shows a different sensitivity towards oxygen activated species. While the enzyme is unaffected by superoxide radical, it is rapidly inactiva…

EPR studies of in vivo radical production by lipopolysaccharide: potential role of iron mobilized from iron-nitrosyl complexes.

Although oxidative stress has been implicated in the pathogenesis of sepsis, there is little evidence for the formation of radicals other th … oxide synthesis and is iron-dependent. One of the secondary produced radicals was identified as the hydroxyl radical. Its formation is proposed to occur because of the mobilization of redox-active iron required to repair the nitrosyl complexes produced by LPS. The results suggest that iron chelation may be a useful adjuvant therapy for treating sepsis.

Study of free iron and pyridinoline in hypertrophic scars and normal skin.

In order to investigate the possible involvement of free radicals in the formation of hypertrophic scar, normal skin sections were subjected in vitro to a free radical generating system and the pyridinoline levels determined by enzyme-linked immunoassay. Pyridinoline concentration increased from 5.123 pmol/mg wet weight to 8.760 pmol/mg wet weight after treatment with a free radical generating system. Free iron, a catalyst for the generation of hydroxyl radicals from hydrogen peroxide, was found to be significantly higher in hypertrophic scar tissue (n = 6, mean 50.70 ng/mg wet weight) compare…
