Intracerebroventricular injection of anti-prolactin serum suppresses excessive grooming of pituitary homografted rats.

Rats with endogenous hyperprolactinaemia, as induced by pituitary homografts under the kidney capsule, displayed increased grooming behavior as compared to that of sham-operated animals. Twelve days after surgery, intracerebroventricular injection of anti-prolactin serum (dilution 1:100, 1 microliter) suppressed the excessive grooming of homografted rats. These observations suggest that prolactin from a peripheral source may reach the central nervous system to affect brain mechanisms involved in grooming behavior.

Effect of calmodulin antagonists on calcium and ethanol-induced sleeping time in mice.

This investigation was carried out to determine if calcium prolongation of ethanol-induced sleep is mediated by calmodulin and a calmodulin-dependent protein kinase. The duration of ethanol-induced sleeping time in ddY male mice was measured following the administration of CaCl2 (20, 40, 80 and 200 mumol/kg, intraperitoneally (IP) both with and without the calmodulin antagonists, W-7: [N-(6-Aminohexyl)-5-chloro-1-naphthalenesulfonamide] (4.2 micrograms/mouse, intraventricular (IVT) or trifluoperazine (TFP; 1.8 micrograms/mouse, IVT). When CaCl2 was administered in a dose dependent manner the d

Influence of H2 receptors blockade upon some central effects of prostacyclin in rats.

Prostacyclin (PGI2) administered icv into the lateral rat brain ventricle in a dose of 1 and 10 micrograms caused hypothermia and catalepsy. Joint administration of PGI2 and chlorpromazine produced a greater cataleptic effect than that observed after the neuroleptic alone. Cimetidine (CMT) 2 g/kg po administered 60 min before PGI2 icv injection inhibited hypothermic and cataleptogenic action of PGI2. CMT blocked the cataleptogenic effect of chlorpromazine as well as combination of it with PGI2. CMT inhibited cataleptogenic effect of haloperidol, but it did not block the catalepsy induced by jo

Intrahypothalamic 5,7-dihydroxytryptamine: temporal analysis of effects on 5-hydroxytryptamine content in brain nuclei and on facilitated lordosis behavior.

The long-term relationship between serotonin (5-HT) levels in discrete hypothalamic nuclei and female rat sexual behavior, the lordosis response, was examined following intrahypothalamic injection of 5,7-dihydroxytryptamine (5,7-DHT). One week following 5,7-DHT injection, 5-HT levels in the ventromedial hypothalamic nucleus, dorsomedial nucleus, anterior hypothalamic nucleus and the medial preoptic nucleus were approximately 90% depleted as compared to sham animals. Other hypothalamic and preoptic areas including the arcuate-median eminence, vertical nucleus of diagonal band and lateral septal

Role for the subfornical organ in vasopressin release.

The effect of subfornical organ (SFO) lesions on plasma vasopressin and drinking was tested in rats given either of two stimuli: subcutaneous injection of hypertonic saline or injection of a single dose of angiotensin II (AII) into the dorsal third cerebral ventricle. Drinking in response to hypertonicity was significantly attenuated, whereas AII-induced drinking was unaffected by the lesions. In contrast, SFO lesions were associated with significantly reduced vasopressin responses following either of these potent stimuli to vasopressin secretion. Partial lesions that damaged structures adjoin

Physiological role of alpha-melanocyte-stimulating hormone in modulating the secretion of prolactin and luteinizing hormone in the female rat.

Long-term ovariectomized (OVX) rats were injected in the third cerebral ventricle with 5 microliter of the globulin fraction of an antiserum raised against alpha-melanocyte-stimulating hormone (alpha-MSH) or an equal volume of the globulin fraction of normal rabbit serum (NRS). Immunoneutralization of brain alpha-MSH produced an increase in the area under the secretion curve of prolactin (Prl), the amplitude of Prl pulses, and mean plasma Prl (P less than 0.01). In animals that had received two injections of NRS or anti-MSH and were subjected to a 2-min ether stress, Prl levels significantly i

Cerebroventricular sites for enkephalin inhibition of the central actions of angiotensin.

The effect of leucine5-enkephalin (Leu-enkephalin) administered into the lateral (LV) or fourth (4V) cerebroventricle on angiotensin II (ANG II)-stimulated increase in blood pressure, plasma vasopressin concentration ([AVP]), and drinking behavior was determined in conscious rats. Leu-enkephalin (20, 50, or 100 micrograms) was injected into the LV (5 microliters) or 4V (2 microliters) less than or equal to 15 s before ANG II (50 or 500 ng, LV). Naloxone effects on blood pressure, analgesia and diuresis in spontaneous hypertensive and normotensive rats.

Studies were performed in unanesthetized normotensive and spontaneously hypertensive rats (SHR) to compare the effects of naloxone. In normotensive Wistar rats, naloxone did not change blood pressure (BP) and nociceptive threshold, but it induced a dose-related diuretic response. Whereas in SHR naloxone decreased nociceptive threshold and lowered BP when given intracerebroventricularly, it failed to significantly modify diuresis. These differences between hypertensive and normotensive rats in their responses to naloxone may be explained by the fact that vasopressin (VP) levels and opioid activ

Naloxone: analeptic action unrelated to opiate receptor antagonism?

The effects on the duration of sleeping time (ST) of the opiate receptor antagonists, naloxone and naltrexone, were determined in rats anesthetized by intraperitoneal injection of ketamine, halothane, or pentobarbital. Intracerebroventricular administration of naloxone shortened the duration of sleeping time induced by all three anesthetic agents in a dose-related manner. Centrally administered naltrexone (240 microgram) and systemically administered naloxone (50 mg/kg) prolonged the duration of pentobarbital sleeping time without altering duration of ketamine or halothane sleeping time. Naltr

Influence of analgesic antipyretics on the central cardiovascular effects of clonidine in rats.

The centrally acting antihypertensive drug clonidine has been found to stimulate the synthesis of PGF2 alpha in the brain. Centrally administered PGF2 alpha, in turn, induces rises of blood pressure and heart rate. We therefore studied the influence of inhibitors of prostaglandin (PG) synthesis on the cardiovascular effects of clonidine in urethane-anaesthetised rats. Pretreatment with indomethacin or paracetamol (100 micrograms/rat into the fourth cerebral ventricle) antagonised the central hypotensive effect of clonidine (0.125-16.0 micrograms/rat into the fourth cerebral ventricle). The bra

Miconazole for treatment of disseminated coccidioidomycosis. Unfavorable experience.

Miconazole at dosages up to 30 mg/kg/day was given intravenously to seven patients with complicated courses of disseminated coccidioidomycosis. Six had received treatment with amphotericin B previously and five of these patients could be evaluated for the efficacy of the treatment. In three patients the condition failed to respond to therapy, another patient required intratracheal administration of amphotericin B later, and the fifth patient had an equivocal response to treatment. Severe phlebitis, pruritus, nausea, vomiting, hyperlipidemia, and thrombocytosis were frequent side effects. These

Alcohol drinking: abnormal intake caused by tetrahydropapaveroline in brain.

Tetrahydropapaveroline (THP), a dopamine-dopaldehyde condensation product, was delivered directly into the cerebral ventricle of rats automatically every 15 minutes for 12 days. The animals were given access to both water and ethylalcohol, the latter being presented in 12 concentrations from 3 to 30 percent. Within 3 to 6 days of the start of the infusion of THP, the rats, which normally rejected alcohol, drank alcohol solutions in increasingly excessive amounts; this was accompanied by symptoms that were similar to those of withdrawal and intoxication. These results provide evidence that an a

Role of central and peripheral mechanisms in the action of alpha-methyldopa on blood pressure and renin secretion.

The mechanism by which alpha-methyldopa lowers arterial pressure and suppresses renin secretion was investigated in pentobarbital-anesthetized dogs in which changes in renal perfusion pressure were prevented by adjusting a suprarenal aortic clamp. After intravenous alpha-methyldopa (100 mg/kg) mean arterial pressure (MAP) decreased form 127+/-3 to a mean minimum of 107+/-4 mm Hg (P less than .01) and plasma renin activity (PRA) decreased from 20.6+/-4.8 to 10.9+/-1.7 ng/ml/3 hr (P less than .05). Blockade of peripheral dopa decarboxylase with intravenous carbidopa (20 mg/kg) significantly atte

Convulsive seizure induced by intracerebral injection of semicarbazide (an anti-vitamin B6) in the mouse.

The direct injection of semicarbazide (SC), an antivitamin B6 (anti-B6), into the lateral ventricle of the mouse brain induced convulsion and tremors at a smaller dose after a shorter latent period than that in systemic administration. The symptoms were prevented by pyridoxine, aminooxyacetic acid or acetone, while they enhanced by pyridoxal, pyridoxal phosphate, or some other anti-B6. In mice fed a vitamin B6 (B6)-deficient diet, convulsion and tremors occurred at smaller doses of SC than those in mice given control food, and were counteracted by pyridoxine. On the other hand, mice into which

Experimental measles encephalitis: a genetic analysis.

The encephalitogenic potential of nine temperature-sensitive mutants of measles virus was determined in newborn golden Syrian hamsters. The parental virus produced acute encephalitis without any prior adaptation. Six of the mutants were attenuated, two were virulent, and one was associated with hydrocephalus with acute onset. The attenuated mutants, blocked before measles virus antigen and ribonucleic acid synthesis at 39 C, were all members of one complementation group. The virulent temperature-sensitive mutants, defective in hemolysin antigen synthesis at 39 C, were members of a second compl

Role of substance P in suppressing growth hormone release in the rat.

To evaluate a possible physiological role of endogenous substance P (SP) in the control of growth hormone (GH; somatotropin) secretion, a specific antiserum against SP (anti-SP) was injected intraventricularly (3 microliters into the third cerebral ventricle) in unanesthetized unrestrained normal male rats. Control rats received an equivalent volume of normal rabbit serum (NRS). Intraventricular injection of the NRS lowered plasma GH concentrations significantly. The lowering was detected on first measurement at 10 min after injection and was maximal at 30 min. This was followed by a return to

Effects of catecholamines and adenosine derivatives given into the brain of fowls.

1. Adult fowls (Gallus domesticus) with cannulae chronically implanted into the IIIrd cerebral ventricle and various other sites of the brain received microinfusions or injections of catecholamines, adenosine, 3',5'-cyclic AMP or its dibutyryl derivative. The effects of these substances on behaviour, electrocortical activity and body temperature were studied.2. Behavioural and electrocortical sleep with fall in body temperature were obtained with intraventricular noradrenaline, alpha-methylnoradrenaline and isoprenaline; dopamine was ineffective. The doses required to elicit sleep were smaller

The fate of newly synthesized proteins in nerve and glial cells following nerve injury as shown by electron microscope radioautography.

Following peripheral crush of hypoglossal nerve the fate of newly synthesizing proteins in corresponding nerve and glial cells was traced out using light and high resolution radioautography after intraventricular injection of 3H-leucine. Radioautographic analysis was applied on the normal and chromatolytic neurons and their glial surroundings 45 min after injection of labelled precursor. Both chromatolytic neurons and surrounding glia showed higher incorporation of labelled proteins than those of control nerve and glial cells. Relative specific radioactivity (RSR) of ergastoplasm and lysosomes
