Combined Metabolome and Transcriptome Analysis of Floral Organ Development in Magnolia cavaleriei var. platypetala 'Tanchun'.

Magnolia cavaleriei var. platypetala 'Tanchun' is a newly registered flower variety in China, known for its characteristic floral aroma that intensifies toward full bloom. However, the composition of the volatiles of this aromatic flower remains uncharacterized. Here, we compared the volatile organic compound composition of Tanchun through gas chromatography-mass spectrometry and comparative transcriptome sequencing analyses of the stamen (S), pistil (P), and petals (T) during flower development, i.e., the bud (S1), semi-opened (S2), and bloom (S3) stages. We present a first comprehensive prof

Maternal bis-glycinate bound zinc supplementation alters sow performance and milk metabolomic-lipidomic profiles and mitigates piglet diarrhea.

Dietary zinc supplementation to piglets reduces diarrhea and improves growth, yet the effects of maternal zinc supplementation on milk and subsequent piglet performance remain unclear. This study evaluated the impact of maternal zinc supplementation on sow, piglet, and lactation performances. Thirty-six crossbred sows were randomized to CON (standard diet; n = 18) or TRT (standard diet supplemented with 1 g/sow/day of bis-glycinate bound zinc; n = 18) from day 85 of gestation to day 21 of lactation. Sow and piglet performance were recorded. Blood was collected at farrowing to determine serum m

Integrated Transcriptomics and Metabolomics Reveal Changes in Cell Homeostasis and Energy Metabolism in Trachinotus ovatus in Response to Acute Hypoxic Stress.

Trachinotus ovatus is an economically important mariculture fish, and hypoxia has become a critical threat to this hypoxia-sensitive species. However, the molecular adaptation mechanism of T. ovatus liver to hypoxia remains unclear. In this study, we investigated the effects of acute hypoxic stress (1.5 ± 0.1 mg·L-1 for 6 h) and re-oxygenation (5.8 ± 0.3 mg·L-1 for 12 h) in T. ovatus liver at both the transcriptomic and metabolic levels to elucidate hypoxia adaptation mechanism. Integrated transcriptomics and metabolomics analyses identified 36 genes and seven metabolites as key molecules that

The role of G protein-coupled receptor in neutrophil dysfunction during sepsis-induced acute respiratory distress syndrome.

Sepsis is defined as a life-threatening dysfunction due to a dysregulated host response to infection. It is a common and complex syndrome and is the leading cause of death in intensive care units. The lungs are most vulnerable to the challenge of sepsis, and the incidence of respiratory dysfunction has been reported to be up to 70%, in which neutrophils play a major role. Neutrophils are the first line of defense against infection, and they are regarded as the most responsive cells in sepsis. Normally, neutrophils recognize chemokines including the bacterial product N-formyl-methionyl-leucyl-p

Clozapine Worsens Glucose Intolerance, Nonalcoholic Fatty Liver Disease, Kidney Damage, and Retinal Injury and Increases Renal Reactive Oxygen Species Production and Chromium Loss in Obese Mice.

Clozapine is widely employed in the treatment of schizophrenia. Compared with that of atypical first-generation antipsychotics, atypical second-generation antipsychotics such as clozapine have less severe side effects and may positively affect obesity and blood glucose level. However, no systematic study of clozapine's adverse metabolic effects-such as changes in kidney and liver function, body weight, glucose and triglyceride levels, and retinopathy-was conducted. This research investigated how clozapine affects weight, the bodily distribution of chromium, liver damage, fatty liver scores, gl

Cell autonomous requirement of neurofibromin (Nf1) for postnatal muscle hypertrophic growth and metabolic homeostasis.

BACKGROUND: Neurofibromatosis type 1 (NF1) is a multi-organ disease caused by mutations in neurofibromin 1 (NF1). Amongst other features, NF1 patients frequently show reduced muscle mass and strength, impairing patients' mobility and increasing the risk of fall. The role of Nf1 in muscle and the cause for the NF1-associated myopathy are mostly unknown. METHODS: To dissect the function of Nf1 in muscle, we created muscle-specific knockout mouse models for NF1, inactivating Nf1 in the prenatal myogenic lineage either under the Lbx1 promoter or under the Myf5 promoter. Mice were analysed during

The adaptor protein Gab1 couples the stimulation of vascular endothelial growth factor receptor-2 to the activation of phosphoinositide 3-kinase.

Phosphoinositide 3-kinase (PI3K) mediates essential functions of vascular endothelial growth factor (VEGF), including the stimulation of endothelial cell proliferation and migration. Nevertheless, the mechanisms coupling the receptor VEGFR-2 to PI3K remain obscure. We observed that the Grb2-bound adapter Gab1 is tyrosine-phosphorylated and relocated to membrane fractions upon VEGF stimulation of endothelial cells. We could detect the PI3K regulatory subunit p85 in immunoprecipitates of endogenous Gab1, and vice versa, and measure a Gab1-associated lipid kinase activity upon VEGF stimulation. F

Gastrointestinal and metabolic effects of methylxanthines.

This chapter has covered a wide range of different actions of methylxanthines. They are able to slightly reduce the tone of the lower esophageal sphincter. This is of little concern in the normal individual, but in patients with a reduced initial tone it might lead to heartburn. Coffee intake has been associated with gastritis, but the role of methylxanthines in this effect is obscure. High doses of methylxanthines are also known to be emetic. Practically every function in the intestine can be influenced by high doses of methylxanthines, but the mechanisms involved and the biological significa

Phenotype-Anchored Multi-Omics Reveals Mechanisms Underlying Starvation-Induced Body, Liver, and Gallbladder Discoloration in Rock Carp (Procypris rabaudi).

Visible changes in body and internal organ coloration can serve as practical indicators of nutritional and metabolic stress in aquaculture, but their physiological meaning remains poorly understood. Here, we used starvation as a controlled nutritional stress model to investigate the physiological and molecular changes associated with body color fading and liver and gallbladder discoloration in rock carp (Procypris rabaudi). To assess the effects of food deprivation (0-35 days), fish were examined using growth and biochemical assays, intestinal histology, as well as integrated hepatic transcrip

Metabolic response to anthelmintic treatment in beef cattle with subclinical gastrointestinal nematode infection.

Gastrointestinal nematodes (GINs) represent a recurring challenge for beef cattle production, because infections are predominantly subclinical and current diagnostic tools have limited ability to detect affected animals before significant losses to the production chain occur. With the advent of new technologies, it has become possible to obtain a wide range of information from animal serum. Through metabolomics, it is possible to expand knowledge of the host-parasite relationship, identify biomarkers with diagnostic potential, and prospect new molecules that may aid in treatment. Thus, the pre

Integrated transcriptomic and immune enzymatic analyses uncover coordinated immunometabolic responses in large yellow croaker (Larimichthys crocea) to Metanophrys sp. infection.

Metanophrys sp. (Scuticociliatida) has recently emerged as a significant parasitic threat in large yellow croaker (Larimichthys crocea) aquaculture. To elucidate the host response, we conducted an experimental infection followed by an integrated analysis combining immune enzymatic profiling and transcriptome sequencing. Antioxidant and immune enzyme activities, including SOD, CAT, and MDA in the skin, gill, and liver, and LYZ and Na+/K+-ATPase in the skin and gill were monitored from 0 to 72 hours post-infection (hpi). The results revealed tissue- and time-specific significant changes, indicat

Pathogenesis of Alcoholic Fatty Liver a Narrative Review.

Alcohol effect hepatic lipid metabolism through various mechanisms, leading synergistically to an accumulation of fatty acids (FA) and triglycerides. Obesity, as well as dietary fat (saturated fatty acids (FA) versus poly-unsaturated fatty acids (PUFA)) may modulate the hepatic fat. Alcohol inhibits adenosine monophosphate activated kinase (AMPK). AMPK activates peroxisome proliferator activated receptor a (PPARα) and leads to a decreased activation of sterol regulatory element binding protein 1c (SRABP1c). The inhibition of AMPK, and thus of PPARα, results in an inhibition of FA oxidation. Th

Leukotoxin (LtxA/Leukothera) induces ATP expulsion via pannexin-1 channels and subsequent cell death in malignant lymphocytes.

Leukotoxin (LtxA) (Trade name, Leukothera) is a protein that is secreted from the oral bacterium Aggregatibacter actinomycetemcomitans, which targets and kills activated white blood cells (WBCs) by binding to lymphocyte function associated antigen-1 (LFA-1). Interaction between LtxA and Jurkat T-cells results in cell death and is characterized by increased intracellular Ca2+, activation of caspases, clustering of LtxA and LFA-1 within lipid rafts, and involvement of the Fas death receptor. Here, we show that LtxA can kill malignant lymphocytes via apoptotic and necrotic forms of cell death. We
