Bisphenol-A bis(diphenyl phosphate) (BDP) has been detected in biological organisms, but its toxicological effects remain unclear. In this study of 4376 participants, the characteristic BDP metabolite diphenyl phosphate was associated with gastrointestinal diseases via a nonlinear dose-response relationship, and was identified as a risk factor (OR = 1.02; 95% CI: 1.01-1.04; P = 0.013). Pregnant rats received 50 mg/kg/day BDP during gestation (GD 14-20) or gestation plus lactation (GD 14-PND 35). Both regimens triggered colonic inflammation and gut microbiome dysbiosis in offspring. The combine

Processing-bodies (PBs) are cytoplasmic membraneless condensates essential for RNA regulation. They share structural and functional similarities with germ granules and are critical for primordial folliculogenesis, a developmental process highly susceptible to exogenous insults. Aristolochic acid I (AAⅠ), a pervasive environmental toxin from Aristolochiaceae plants, is known to cross the placental barrier and induce fetal ovarian oxidative stress. Here, using a mouse model of maternal AAⅠ exposure (16.5 to 18.5 days post-coitus), we demonstrate that this prenatal insult causes profound defects

Beyond being one of the most widely prescribed medications for type 2 diabetes, metformin has been increasingly used during pregnancy for gestational diabetes and polyendocrine metabolic ovarian syndrome. While it is considered safe for pregnancy and lactation, metformin is known to induce gut microbial changes in non-pregnant individuals with type 2 diabetes, and maternal and offspring microbial alterations are known to influence offspring development. Therefore, understanding whether metformin drives microbiota changes beyond the confounding effects of maternal hyperglycaemia holds significa

BACKGROUND AND OBJECTIVES: Concerns about modafinil and armodafinil use in pregnancy emerged from animal reproductive toxicity and limited human data, including a US registry suggesting increased risk of major congenital malformations (MCMs). Subsequent studies have reported conflicting findings. This study aimed to evaluate the risk of MCMs and other adverse pregnancy outcomes following prenatal modafinil exposure in the French population.
METHODS: This retrospective cohort study identified pregnancies during 2012-2019 using the French national health database (SNDS). Pregnancies exposed and

Prenatal polycyclic aromatic hydrocarbon (PAH) exposure may contribute to neural tube defects (NTD), but combined effects and underlying mechanisms remain unclear. We conducted a matched case-control study (128 pairs) in six high-risk counties in Shanxi Province, China (2004-2016). At delivery or pregnancy termination, placenta samples were collected. Sixteen placental PAHs and 11 hydroxylated metabolites were quantified by GC-MS/MS as biomarkers of fetal intrauterine exposure, alongside 16 DNA adducts measured by UPLC-MS/MS. Complementary ICR mouse experiments with benzo[a]pyrene (BaP, 100 mg

Zika virus (ZIKV) is an arbovirus commonly transmitted to humans through the bite of an infected Aedes mosquito. Clinically, ZIKV is known to cause a wide range of symptoms, from febrile rash to severe neurological conditions, with congenital microcephaly being one of the most notable. Herein, we present a case of concurrent ZIKV infection in dizygotic twins, where only one twin developed microcephaly and severe encephalic malformations, while the other exhibited normal encephalic architecture. Our findings shed light on the complex interplay between ZIKV infection and the manifestation of neu

The early-life microbiome plays a pivotal role in host development and lifelong health. Maternal factors are increasingly recognized as crucial in shaping offspring microbiome. However, how maternal preconception perturbations affects offspring health remain unclear. Thus, we combined animal and clinical data to elucidate whether preconception microbial perturbations disrupt microbial succession and increase offspring susceptibility to colitis. In animals, preconception antibiotic exposure induced long-lasting disruptions in offspring microbial ecology, through enhanced maternal-offspring micr

Aristolochic acid I (AAI), the predominant compound in Aristolochiaceae plants and Asarum species, is a widespread environmental contaminant capable of accumulating in soil, contaminating water and crops, ultimately entering the human body. Its nephrotoxic, carcinogenic, and reproductive toxic effects pose significant health concerns. This study investigates the impact of maternal AAI exposure on meiotic prophase I (MPI) during early fetal oogenesis. Pregnant mice were orally administered AAI at doses of 0.03125, 0.125, and 1 mg/kg from 14.5 to 16.5 dpc, with fetal ovaries collected at 17.5 dp

Infant body size at birth has been suggested as a good marker of fetal development. However, in Vietnam, impacts of maternal TCDD exposure originating from Agent Orane on body size indices at birth have not been investigated in infants from birth cohorts around former US airbases. We investigated the effects of maternal TCDD exposure and TEQ levels of dioxin congeners other than TCDD (non-TCDD) on body size indices at birth among 504 infants from three Vietnamese birth cohorts in two contaminated areas and one unexposed area. The effects of two exposure factors, TEQ-PCDD and TEQ-PCDF in breast

Opioids are a class of pain-relieving drugs known to cross the placental and blood brain barriers, exposing the fetus in utero. Rates of opioid use disorder amongst pregnant individuals in the United States are on the rise, and intravenous routes of opioid administration are highly associated with hepatitis C (HCV) infection. Newborns with prenatal opioid exposure (POE) are more likely to be small for gestational age and have increased rates of neurodevelopmental delay. Microglia are brain-resident macrophages that originate from yolk-sac precursors that play critical role in neurodevelopment.

Hypobaric hypoxia encountered at high altitudes impairs reproductive health and fertility across species. Previous findings have demonstrated that maternal hypoxia exposure disrupts granulosa cell (GC) viability and oocyte maturation in female offspring; however, its transgenerational impact on male reproductive outcomes remains poorly elucidated. In this study, pregnant mice (F0) were subjected to hypoxic conditions, and male progeny across four generations (F1‒F4) were evaluated. Results revealed that maternal hypoxia induced mild alterations in sperm DNA methylation in F1 males but caused p

L-mimosine, a compound found in legumes of the Leucaena genus, is known for its ability to disrupt the cell cycle and its toxic properties. Its effects during critical periods such as gestation and lactation remain poorly understood. This study aimed to evaluate the potential toxic effects of L-mimosine in rats exposed during these periods, as well as its transfer through biological fluids, such as milk, to offspring. Oral doses of 140, 240, or 340 mg/kg of maternal body weight were administered, and a control group (CO), which received only the vehicle, was included. During gestation, dams re

Liver is an important target organ of ambient fine particulate matter (PM2.5). Numerous studies have shown that PM2.5 exposure can cause liver lipid metabolism disorders and other liver damage in mammals. However, the impact of PM2.5 on liver health during pregnancy, a sensitive life stage, remains understudied, and the underlying mechanisms are also unknown. Given the critical role of adenosine 5'-monophosphate activated protein kinase (AMPK) in regulating lipid metabolism and inflammation, we hypothesize that AMPK activation may mitigate maternal hepatic lipid metabolism disorders, reduce in

BACKGROUND: Maternal stress during pregnancy may impact offspring development via changes in the intrauterine environment. However, genetic and environmental factors shared between mothers and children might skew our understanding of this pathway. This study assesses whether prenatal maternal stress has causal links to offspring outcomes: birthweight, gestational age, or emotional and behavioral difficulties, triangulating across methods that account for various measured and unmeasured confounders.
METHODS: We used data from the Norwegian Mother, Father, and Child Cohort Study (MoBa), includi

With the growing crisis of plastic pollution, the severe environmental problem threatens human and ecosystem health. And nanoplastics can carry other environmental contaminants, thereby causing severe toxic effects. Cadmium (Cd), as a metal, has been widely concerned because of its long biological half-life, high toxicity and low excretion rate. Additionally, Cd as an endocrine disrupting chemical also has reproductive toxicity and genotoxicity. Studies have found that co-exposed to Cd and PS-NPLs complexes may lead to more severe adverse effects in aquatic and mammals. And the utilization of

The composition of particulate matter (PM) in poultry farms differs significantly from that of atmospheric PM as there is a higher concentration of microbes on farms. To assess the health effects of PM from poultry farms on pregnant animals, we collected PM from duck houses using a particulate sampler, processed it via centrifugation and vacuum concentration, and subsequently exposed the mice to airborne PM at 0.48 mg/m3 (i.e., low concentration group) and 1.92 mg/m3 (i.e., high concentration group) on the fifth day of pregnancy. After exposure until the twentieth day of pregnancy or spontaneo
