Accelerated long-term forgetting in children with neurofibromatosis type 1.

AIM: To investigate the presence of accelerated long-term forgetting after a 7-day delay in children with neurofibromatosis type 1 (NF1), using an adapted verbal memory recall task, Experimental Word Recall Task (EWRT), and to examine its relationship with other cognitive abilities. METHOD: This cross-sectional design involved 60 children with NF1 and 88 typically developing children who completed the EWRT and were aged between 6 years and 15 years 11 months. Statistical analyses were carried out in individuals who met the EWRT learning criteria (i.e., were able to successfully learn the list

Metabolic Disorder Therapeutics and their Effects on Memory.

Diabetes is one of the major metabolic disorders of this era. It not only impacts a person's lifestyle but also has a long-term impact on the brain. It has a detrimental effect on a person's health when combined with hypertension and hyperlipidaemia. Several studies have suggested that the incidence of dementia is higher in people with metabolic syndrome. Investigations are underway to determine whether antidiabetic, hypolipidemic, hypercholesteraemic, anti-hypertensive, and other combination medicines can minimize the incidence of cognitive impairment. Some studies have suggested that anti-di

Progress and Challenges Toward Generation and Maintenance of Long-Lived Memory T Lymphocyte Responses During COVID-19.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causing the coronavirus disease 2019 (COVID-19) pandemic is a serious global threat until we identify the effective preventive and therapeutic strategies. SARS-CoV-2 infection is characterized by various immunopathological consequences including lymphocyte activation and dysfunction, lymphopenia, cytokine storm, increased level of neutrophils, and depletion and exhaustion of lymphocytes. Considering the low level of antibody-mediated protection during coronavirus infection, understanding the role of T cell for long-term protection is

Patients with or without a clonal mast cell disease undergoing venom immunotherapy differ in their CD8 T memory cell response.

Hymenoptera venom immunotherapy (VIT) is usually effective in inducing long-term tolerance to the allergen. In patients with a clonal mast cell disease (CMD) and Hymenoptera venom-induced anaphylaxis, however, life-long VIT is recommended, as tolerance to the venom is often lost after VIT termination. We compared blood samples from patients with CMD and non-CMD with severe Hymenoptera venom-induced anaphylaxis before and more than 3 years after VIT to identify differences that might explain the lack of long-term tolerance in patients with CMD. Serum tryptase, IgE, IgG4, KIT D816V variation, as

Gene driven analytical learning model for accurate breast cancer diagnosis.

Patients diagnosed with breast cancer exhibit a diverse range of prognostic outcomes due to the varied nature of the disease across different patient groups. To address this complexity and enhance prognostic predictions based on gene expression data from breast cancer samples, this study has developed an integrated deep learning method that combines Convolutional Neural Networks (CNN) with Bidirectional Long Short-Term Memory (BiLSTM) networks. This automated pipeline conducts a correlation analysis using Pearson correlation to derive a reliable 236-gene set, ensuring no data contamination fro

Meprin β Modulates Brevican Proteolysis Impairing Neural Plasticity and Memory Formation.

The metalloprotease meprin β is known for its multifunctional involvement in various physiological processes throughout the body including the brain. However, its broader functions within the brain besides amyloid β generation remain largely unexplored. To investigate this, we utilized a mouse model overexpressing meprin β in neurons within the cortex and hippocampus, regions crucial for learning and memory. Behavioral assessments, employing the Morris' Water Maze paradigm test, revealed impaired cognitive functions in animals overexpressing meprin β. Furthermore, electrophysiological recordin

SFRP1 upregulation causes hippocampal synaptic dysfunction and memory impairment.

Impaired neuronal and synaptic function are hallmarks of early Alzheimer's disease (AD), preceding other neuropathological traits and cognitive decline. We previously showed that SFRP1, a glial-derived protein elevated in AD brains from preclinical stages, contributes to disease progression, implicating glial factors in early pathogenesis. Here, we generate and analyze transgenic mice overexpressing astrocytic SFRP1. SFRP1 accumulation causes early dendritic and synaptic defects in adult mice, followed by impaired synaptic long-term potentiation and cognitive decline, evident only when the ani

Ventral Tegmental Area Dopamine Neural Activity Switches Simultaneously with Rule Representations in the Medial Prefrontal Cortex and Hippocampus.

Multiple brain regions need to coordinate activity to support cognitive flexibility and behavioral adaptation. Neural activity in both the hippocampus (HPC) and medial prefrontal cortex (mPFC) is known to represent the spatial context and is sensitive to reward and rule alterations. Midbrain dopamine (DA) activity is key to reward-seeking behavior and learning. There is abundant evidence that midbrain DA modulates HPC and PFC activity. However, it remains underexplored how these networks engage dynamically and coordinate temporally when animals adjust their behavior according to changing rewar

Early sensory cortex is activated in the absence of explicit input during crossmodal item retrieval: evidence from MEG.

Crossmodal associations form a fundamental aspect of our daily lives. In this study we investigated the neural correlates of crossmodal association in early sensory cortices using magnetoencephalography (MEG). We used a paired associate recognition paradigm in which subjects were tested after multiple training sessions over a span of four weeks. Subjects had to learn 12 abstract, nonlinguistic, pairs of auditory and visual objects that consisted of crossmodal (visual-auditory, VA; auditory-visual, AV) and unimodal (visual-visual, VV; auditory-auditory, AA) paired items. Visual objects included

Imbalanced pattern completion vs. separation in cognitive disease: network simulations of synaptic pathologies predict a personalized therapeutics strategy.

BACKGROUND: Diverse Mouse genetic models of neurodevelopmental, neuropsychiatric, and neurodegenerative causes of impaired cognition exhibit at least four convergent points of synaptic malfunction: 1) Strength of long-term potentiation (LTP), 2) Strength of long-term depression (LTD), 3) Relative inhibition levels (Inhibition), and 4) Excitatory connectivity levels (Connectivity). RESULTS: To test the hypothesis that pathological increases or decreases in these synaptic properties could underlie imbalances at the level of basic neural network function, we explored each type of malfunction in

Melatonin, consciousness, and traumatic stress.

Descartes intuitively anticipated the so-called 'binding problem' of consciousness and thought that the pineal gland enables spatio-temporal integration in cognitive processing. Recent findings indicate that a major role in the process of temporal integration and binding involve neurons in suprachiasmatic nuclei, specifically targeting the pineal gland and other structures, and control the neuroendocrine rhythms. Melatonin is an endocrine output signal of the clock and provides circadian information as an endogenous synchronizer which stabilizes and reinforces circadian rhythms. This integrati

Preexposure to drug administration context blocks the development of tolerance to sedative effects of diazepam.

The development of tolerance to benzodiazepine (BDZ) and other psychoactive agents such as morphine, alcohol, and barbiturates is thought to be a contingent or learning phenomenon. In a previous report, we demonstrated a positive correlation between the development of tolerance to the sedative effects of diazepam (DZ) and hippocampal synaptic plasticity. The results of the present work show that the development of tolerance to the hypolocomotor action of DZ (5 mg/kg) for 4 days and the associated increase in synaptic plasticity are context specific. Because animal preexposure to the drug admin

Ecstasy (MDMA) and memory function: a meta-analytic update.

A meta-analysis was conducted to examine the impact of recreational ecstasy use on short-term memory (STM), long-term memory (LTM), verbal and visual memory. We located 26 studies containing memory data for ecstasy and non-ecstasy users from which effect sizes could be derived. The analyses provided measures of STM and LTM in 610 and 439 ecstasy users and revealed moderate-to-large effect sizes (Cohen's d) of d = -0.63 and d = -0.87, respectively. The difference between STM versus LTM was non-significant. The effect size for verbal memory was large (d = -1.00) and significantly larger than the

Toxic encephalopathy due to 1,1,1-trichloroethane exposure.

Twenty-eight workers with long-term repetitive high exposures to 1,1,1-trichloroethane were evaluated for complaints of short-term memory loss, disequilibrium, moodiness, irritability, and decreased ability to concentrate. As a group, they had significant deficits in memory, intermediate memory, rhythm, and speed as measured by the Luria-Nebraska Neuropsychological Battery. Platform posturography demonstrated deficits in vestibular, somatosensory, and ocular components of balance. The encephalopathic picture in these patients is similar to what has been described after exposure to other solven

Cathepsin B controls the persistence of memory CD8+ T lymphocytes.

The persistence of memory T lymphocytes confers lifelong protection from pathogens. Memory T cells survive and undergo homeostatic proliferation (HSP) in the absence of Ag, although the cell-intrinsic mechanisms by which cytokines drive the HSP of memory T cells are not well understood. In this study we report that lysosome stability limits the long-term maintenance of memory CD8(+) T cell populations. Serine protease inhibitor (Spi) 2A, an anti-apoptotic cytosolic cathepsin inhibitor, is induced by both IL-15 and IL-7. Mice deficient in Spi2A developed fewer memory phenotype CD44(hi)CD8(+) T

Antidepressant-like and memory-enhancing effects of the N-type calcium channel blocker ziconotide in rats.

The lack of oral or injectable formulations of ziconotide (ω-conotoxin peptide), a novel analgesic agent, limits research on potential neurobehavioral protective properties of this substance, including antidepressant-like effects. Here we expose rats to a stress paradigm that induces depression and memory impairment to assess the effects of ziconotide treatment. Ziconotide was administered intracerebroventricular (i.c.v.) to rats undergoing stereotaxic surgery at a single dose (1 μg/rat) or in repeated long-term applications (dosage groups: 0.1, 0.3, and 1 μg/rat). The antidepressant activity

Enhanced Mesenchymal Stromal Cells or Erythropoietin Provide Long-Term Functional Benefit After Neonatal Stroke.

BACKGROUND AND PURPOSE: Perinatal stroke is a common cause of life-long neurobehavioral compromise. Mesenchymal stromal cells (MSCs) and EPO (erythropoietin) have each demonstrated short-term benefit with delayed administration after stroke, and combination therapy may provide the most benefit. The purpose of this study is to determine the long-term histological and functional efficacy of enhanced, intranasal stem cell therapy (MSC preexposed to EPO) compared with standard MSC or multidose systemic EPO. METHODS: Transient middle cerebral artery occlusion or sham surgery was performed in postn

Consequences of value-directed remembering in working memory on memory strength and memory interference.

Critical details are important to remember, whereas trivial information is not. The present research examined whether there were differences in relative susceptibility to proactive interference (PI) for high-value, low-value, zero-value, or negative-value information within working memory (WM) in younger adults. PI occurs when previously learned information disrupts current performance. Across four experiments, participants (N = 255) completed a recent probes WM task, where verbal memoranda were arbitrarily assigned point values after encoding and memory was tested after a 3-s delay. A surpris
