Constructing Conjugated Polymer Composite Fluorescent Nanodrug Materials for Treating Abdominal Aortic Aneurysm.

The abdominal aortic aneurysm (AAA) is a dilation of the lower part of the body aorta. AAA has no obvious symptoms in the early stages until the aortic wall ruptures suddenly, resulting in massive blood loss and flow into the abdominal cavity. Currently, there is no effective drug treatment for AAA, and the development of effective drugs is crucial. In this study, a novel approach utilizing chitosan/genipin/zinc oxide (CH/G-ZnO) composite nanoparticles as a drug delivery system is proposed. Compound 1 was loaded onto these nanoparticles to form CH/G-ZnO@1 composite. The composite material exhi

[Mechanism of Notch3 signaling pathway regulating the differentiation of aortic dissection vascular stem cells into smooth muscle cells].

OBJECTIVE: To explore whether the differentiation of vascular stem cells (VSC) into smooth muscle cells (SMC) in aortic dissection (AD) is dysregulated, and to verify the role of Notch3 pathway in this process. METHODS: Aortic tissues were obtained from AD patients undergoing aortic vascular replacement and heart transplant donors at Department of Cardiovascular Surgery, Guangdong Provincial People's Hospital Affiliated to Southern Medical University. VSC were isolated by enzymatic digestion and c-kit immunomagnetic beads. The cells were divided into normal donor-derived VSC group (Ctrl-VSC g

Sodium Nitroprusside Stimulation of Elastic Matrix Regeneration by Aneurysmal Smooth Muscle Cells.

The chronic overexpression of matrix metalloproteases leading to consequent degradation and loss of the elastic matrix with the reduction in tissue elasticity is central to the pathophysiology of proteolytic disorders, such as abdominal aortic aneurysms (AAAs), which are localized rupture-prone aortic expansions. Effecting tissue repair to alleviate this condition is contingent on restoring elastic matrix homeostasis in the aortic wall. This is naturally irreversible due to the poor elastogenicity of adult and diseased vascular cells, and the impaired ability to assemble mature elastic fibers,

Activation of ENS Circuits in Mouse Colon: Coordination in the Mouse Colonic Motor Complex as a Robust, Distributed Control System.

The characteristic motor patterns of the colon are coordinated by the enteric nervous system (ENS) and involve enterochromaffin (EC) cells, enteric glia, smooth muscle fibers, and interstitial cells. While the fundamental control mechanisms of colonic motor patterns are understood, greater complexity in the circuitry underlying motor patterns has been revealed by recent advances in the field. We review these recent advances and new findings from our laboratories that provide insights into how the ENS coordinates motor patterns in the isolated mouse colon. We contextualize these observations by

Stem cell derived extracellular vesicles for vascular elastic matrix regenerative repair.

Abdominal aortic aneurysms (AAA) are localized expansions of the abdominal aorta that develop due to chronic proteolytic disruption of the structural extracellular matrix (ECM) components (elastin and collagen) within the aorta wall. Major limitations in arresting or reversing AAAs lie in naturally poor and aberrant regeneration and repair of elastic matrix structures in the aorta wall. Bone marrow derived mesenchymal stem cells (BM-MSCs) have emerged as a promising regenerative tool and their therapeutic effects are also known to be effected through their paracrine secretions. Extracellular v

NOTCH3 is non-enzymatically fragmented in inherited cerebral small-vessel disease.

The small-vessel disorder cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) arises from mutations in the human gene encoding NOTCH3 and results in vascular smooth muscle cell degeneration, stroke, and dementia. However, the structural changes in NOTCH3 involved in CADASIL etiology are unclear. Here, we discovered site-specific fragmentation of NOTCH3 protein in pathologically affected vessels of human CADASIL-affected brains. EM-based experiments to pinpoint NOTCH3 localization in these brains indicated accumulation of NOTCH3 fragmentation pro

[Treatment of erectile dysfunction caused by cavernous nerve injury: An update].

With the development of prostate surgery and increasing number of patients with pelvic fracture urethral injury, ED caused by cavernous nerve injury (CNI) has attracted more and more medical attention. CNI induces the apoptosis of penile smooth muscle cells and endothelial cells, decreases the density of NOS-positive nerves, and results in the fibrosis of the cavernous smooth muscle. The strategies of nerve regeneration after CNI has been one of the hotspots in the studies of ED. This review focuses on the current treatment strategies to promote the cavernous nerve regeneration and status quo

Direct role for smooth muscle cell mineralocorticoid receptors in vascular remodeling: novel mechanisms and clinical implications.

The mineralocorticoid receptor (MR) is a key regulator of blood pressure. MR antagonist drugs are used to treat hypertension and heart failure, resulting in decreased mortality by mechanisms that are not completely understood. In addition to the kidney, MR is also expressed in the smooth muscle cells (SMCs) of the vasculature, where it is activated by the hormone aldosterone and affects the expression of genes involved in vascular function at the cellular and systemic levels. Following vascular injury due to mechanical or physiological stresses, vessels undergo remodeling resulting in SMC hype

The myogenic mechanism of synchronization and coordination for uterine myocytes contractions during labor.

The article summarized the results of many years studying of synchronization and coordination of uterine myocytes contractile activity during physiological labor contractions. The authors suggest that the physiological basis for these processes is the mechanoreceptor mechanism of feedback for the stretching which is one of the fundamental mechanisms for control contractile activity of uterine myocytes during parturition.

Autoimmunity and extrahepatic manifestations in treatment-naïve children with chronic hepatitis C virus infection.

Hepatitis C virus (HCV) infection has been associated with autoimmunity and extrahepatic manifestations in adults. Few data are available on these topics in children. Nonorgan specific auto-antibodies development is part of the natural course of chronic hepatitis C in children. Smooth muscle autoantibody is the most common autoantibody found, while liver-kidney microsomal type-1 antibody positivity is the most peculiar autoimmune feature of children with HCV infection. The clinical significance of non-organ specific autoantibodies in the course of paediatric chronic hepatitis C is still debate

Genetic variants promoting smooth muscle cell proliferation can result in diffuse and diverse vascular diseases: evidence for a hyperplastic vasculomyopathy.

Genetic predisposition to early onset of occlusive vascular diseases, including coronary artery disease, ischemic stroke, and Moyamoya disease, may represent varying presentations of a common underlying dysregulation of vascular smooth muscle cell proliferation. We discuss mutations in two genes, NF1 and ACTA2, which predispose affected individuals to diffuse and diverse vascular diseases. These patients show evidence of diffuse occlusive disease in multiple arterial beds or even develop seemingly diverse arterial pathologies, ranging from occlusions to arterial aneurysms. We also present the

Dedifferentiation of stromal smooth muscle as a factor in prostate carcinogenesis.

We had earlier established an animal model of prostate carcinogenesis using a combination of testosterone (T) and 17beta-estradiol benzoate (E2) on Noble rats (Wang and Wong, 1998). In the present study we examined the changes in a number of smooth muscle differentiation markers including smooth muscle alpha-actin and myosin, vinculin, desmin, laminin and vimentin as well as changes in fine structure by electron microscopy. Our immunohistochemical (IHC) studies revealed that smooth muscle cells (SMCs) subjacent to dysplastic (precancerous) sites and carcinoma usually exhibited a preferential l

ACTA2 Pathogenic Variants Activating Heat Shock Factor 1 and Increasing Cholesterol Biosynthesis in Smooth Muscle Cells Predispose to Early Onset Atherosclerosis.

BACKGROUND: ACTA2 pathogenic variants predispose to thoracic aortic disease, and a subset of variants lead to early onset atherosclerotic cardiovascular disease (ASCVD). The molecular pathway linking misfolded SMA (α-smooth muscle actin) monomers to augmented atherosclerosis-associated smooth muscle cell phenotypic modulation can be modeled in vitro by stably expressing the ACTA2 p.R149C variant in Acta2-/- smooth muscle cells. METHODS: The Montalcino Aortic Consortium patient registry was used to identify cases with ACTA2 pathogenic/likely pathogenic missense variants. These patients were su

Transplantation of hPSC-Derived Pericyte-Enriched Vascular Assemblies Restores Cerebral Perfusion in the Context of Permanent Brain Ischemia.

Despite advancements in vascular reconstruction techniques, insufficient distal perfusion following ischemic stroke continues to cause irreversible neurological damage, and effective strategies for reconstructing functional microvascular networks remain inadequate. While vascular organoids have shown potential, their clinical translation is limited by the need for invasive surgical resection to accommodate large 3D grafts. This study improves the vascular organoid technology pipeline by developing hPSC-derived Pericyte-Enriched Vascular Assemblies. These assemblies are composed of CD31+ endoth

ER morphological analysis associated with interstitial cells of Cajal and smooth muscle cells in the murine stomach.

In the gastrointestinal tract, several modulators are involved in generating complex motility patterns, including smooth muscle cells (SMCs), interstitial cells of Cajal (ICCs), and PDGFRα+ cells. Notably, ICCs are excitable cells that generate characteristic oscillations in pacemaker activity mediated by the cytoplasmic Ca2+ concentration. Since the primary source of cytoplasmic Ca2+ is the endoplasmic reticulum (ER) or sarcoplasmic reticulum (SR), its distribution and localisation in the cytoplasm may indicate characteristic aspects that help in understanding its physiological functions. Electrophysiological Modeling of Uterine Smooth Muscles Cells During Estrus.

Uterine activity is regulated by sex hormones, notably progesterone and estrogen. Sex hormones play an important regulatory role in preparing the uterus for pregnancy, which includes an influence on contractile function. Computational models have previously been developed to simulate the electrical and contractile activity of pregnant smooth muscle cells. Here, a non-pregnant cell model which includes estrus stages was developed based on an existing pregnant cell model. The estrus cycle variations are modeled by including the effects of sex hormones on the different ionic currents. Root mean s

Multiple cell types including melanocytes contribute to elastogenesis in the developing murine aortic valve.

Elastic fibers are crucial for aortic valve (AoV) function and are generated and maintained by valvular interstitial cells (VICs). VICs exhibit diverse phenotypes, yet the specific subpopulation responsible for producing and regulating elastic fibers remains unclear. This gap in knowledge is significant, given that elastin (Eln) abnormalities lead to congenital AoV defects and initiate AoV diseases. This study characterizes the timing of Eln expression in murine AoV, revealing it peaks during late embryogenesis and early postnatal stages, decreasing in adulthood. Spatial transcriptomics and RT

Single-Cell Analyses Offer Insights into the Different Remodeling Programs of Arteries and Veins.

Arteries and veins develop different types of occlusive diseases and respond differently to injury. The biological reasons for this discrepancy are not well understood, which is a limiting factor for the development of vein-targeted therapies. This study contrasts human peripheral arteries and veins at the single-cell level, with a focus on cell populations with remodeling potential. Upper arm arteries (brachial) and veins (basilic/cephalic) from 30 organ donors were compared using a combination of bulk and single-cell RNA sequencing, proteomics, flow cytometry, and histology. The cellular atl
