Advancing Drug Discovery for Neurological Disorders Using iPSC-Derived Neural Organoids.

In the last decade, different research groups in the academic setting have developed induced pluripotent stem cell-based protocols to generate three-dimensional, multicellular, neural organoids. Their use to model brain biology, early neural development, and human diseases has provided new insights into the pathophysiology of neuropsychiatric and neurological disorders, including microcephaly, autism, Parkinson's disease, and Alzheimer's disease. However, the adoption of organoid technology for large-scale drug screening in the industry has been hampered by challenges with reproducibility, sca

Acrophobia Quantified by EEG Based on CNN Incorporating Granger Causality.

The aim of this study is to quantify acrophobia and provide safety advices for high-altitude workers. Considering that acrophobia is a fuzzy quantity that cannot be accurately evaluated by conventional detection methods, we propose a comprehensive solution to quantify acrophobia. Specifically, this study simulates a virtual reality environment called High-altitude Plank Walking Challenge, which provides a safe and controlled experimental environment for subjects. Besides, a method named Granger Causality Convolutional Neural Network (GCCNN) combining convolutional neural network and Granger ca

The extrinsic risk and its association with neural alterations in spasmodic dysphonia.

INTRODUCTION: Spasmodic dysphonia (SD) is an isolated focal dystonia characterized by laryngeal spasms during voluntary voice production. Environmental factors have been assumed to play a role in SD pathophysiology; however, the exact extrinsic risk factors and their association with neural alterations remain unknown. METHODS: A total of 186 SD patients and 85 healthy controls completed a structured 177-question survey, consisting of questions on general biographical information, medical history, symptomatology of dystonia. Data were imputed in a stepwise regression model to identify extrinsi

Morphological and genetical changes of endothelial progenitor cells after in-vitro conversion into photoreceptors.

BACKGROUND: Retinal degeneration is a condition ensued by various ocular disorders such as artery occlusion, diabetic retinopathy, retrolental fibroplasia and retinitis pigmentosa which cause abnormal loss of photoreceptor cells and lead to eventual vision impairment. No efficient treatment has yet been found, however, the use of stem cell therapy such as bone marrow and embryonic stem cells has opened a new treatment modality for retinal degenerative diseases. The major goal of this study is to analyze the potential of endothelial progenitor cells derived from bone marrow to differentiate int

Malformations in a chornobyl-impacted region.

OBJECTIVE: One of the populations most exposed to chronic low-dose radiation from Chornobyl (Chernobyl in Russian) lives in Polissia, the region representing the northern half of Rivne Province (Oblast) in Ukraine. Here the patterns and population rates of malformations are reported and possible etiologic factors and regional contrasts are explored. PATIENTS AND METHODS: Malformations, as defined by international standards, noted among all 96 438 births in Rivne between 2000 and 2006, were analyzed statistically. Contrasts of rates in Polissia compared with the rest of Rivne also were investi

Coping with emotions past: the neural bases of regulating affect associated with negative autobiographical memories.

BACKGROUND: Although the ability to adaptively reflect on negative autobiographical experiences without ruminating is critical to mental health, to our knowledge no research has directly examined the neural systems underlying this process. METHODS: Sixteen participants were scanned using functional magnetic resonance imaging (fMRI) as they focused on negative autobiographical memories using cognitive strategies designed to facilitate (feel strategy) versus undermine (analyze and accept strategies) rumination. RESULTS: Two key findings were obtained. First, consistent with prior emotion regul

A novel method for the topographic analysis of neural activity reveals formation and dissolution of 'Dynamic Cell Assemblies'.

The study of synchronous oscillations in neural systems is a very active area of research. However, cognitive function may depend more crucially upon a dynamic alternation between synchronous and desynchronous activity rather than synchronous behaviour per se. The principle aim of this study is to develop and validate a novel method of quantifying this complex process. The method permits a direct mapping of phase synchronous dynamics and desynchronizing bursts in the spatial and temporal domains. Two data sets are analyzed: Numeric data from a model of a sparsely coupled neural cell assembly a

Human brain parenchymal microglia express CD14 and CD45 and are productively infected by HIV-1 in HIV-1 encephalitis.

Microglia are endogenous brain macrophages that show distinct phenotypes such as expression of myeloid antigens, ramified morphology, and presence within the neural parenchyma. They play significant roles in a number of human CNS diseases including AIDS dementia. Together with monocyte-derived (perivascular) macrophages, microglia represent a major target of HIV-1 infection. However, a recent report challenged this notion based on findings in SIV encephalitis. This study concluded that perivascular macrophages can be distinguished from parenchymal microglial cells by their expression of CD14 a

Neural elements in the pineal complex of the frog, Rana esculenta, I: Centrally projecting neurons.

The pineal complex of anuran amphibians is a directly photosensory organ, encompassing both an extracranial portion, the frontal organ, and an intracranial portion, the pineal organ proper. The projection neurons of the frontal organ respond differentially according to the wavelengths of the light stimuli. The pineal organ, on the other hand, functions mainly as a luminosity meter. Most of its centrally projecting neurons respond to all increases in ambient illumination with decreases in spontaneous firing of action potentials, although some neural units in the pineal organ may respond accordi

Amniotic fluid alpha-fetoprotein and acetylcholinesterase in early genetic amniocentesis.

Early identification of fetal abnormalities is possible as a result of improved ultrasound resolution, chorionic villus sampling, and early genetic amniocentesis. A potential advantage of early genetic amniocentesis over chorionic villus sampling is its ability to detect neural tube defects. We obtained 476 amniotic fluid samples between ten and 15 weeks' gestation and analyzed them for karyotype and alpha-fetoprotein (AFP); 142 were also tested for acetylcholinesterase. Amniotic fluid AFP levels rose to a peak at 12-13 weeks' gestation and then gradually declined, closely approximating the pa

Interruption of the aortic arch and coarctation of the aorta: pathogenetic relations.

The pathologic anatomic features and associated cardiac anomalies of 63 cases of interruption of the aortic arch (IAA) type A (54 reported and 9 observed) and 145 cases of type B (124 reported and 21 observed) were analyzed and compared with those seen in 57 autopsy cases of infant coarctation of the aorta (CA). There was no significant sex predominance within the 3 groups and the prognosis without surgery was uniformly poor. Ventricular septal defect was much more common in IAA type B than in type A or CA. Anomalous origin of the subclavian artery and DiGeorge syndrome were commonly associate

Classifying Alzheimer's Disease Using a Finite Basis Physics Neural Network.

The disease amyloid plaques, neurofibrillary tangles, synaptic dysfunction, and neuronal death gradually accumulate throughout Alzheimer's disease (AD), resulting in cognitive decline and functional disability. The challenges of dataset quality, interpretability, ethical integration, population variety, and picture standardization must be addressed using deep learning for the functional magnetic resonance imaging (MRI) classification of AD in order to guarantee a trustworthy and practical therapeutic application. In this manuscript Classifying AD using a finite basis physics neural network (CA

Selectively predicting the onset of ADHD, oppositional defiant disorder, and conduct disorder in early adolescence with high accuracy.

INTRODUCTION: The externalizing disorders of attention deficit hyperactivity disorder (ADHD), oppositional defiant disorder (ODD), and conduct disorder (CD) are common in adolescence and are strong predictors of adult psychopathology. While treatable, substantial diagnostic overlap complicates intervention planning. Understanding which factors predict the onset of each disorder and disambiguating their different predictors is of substantial translational interest. MATERIALS AND METHODS: We analyzed 5,777 multimodal candidate predictors from children aged 9-10 years and their parents in the AB

Neuro-tumor-immune crosstalk in pancreatic cancer: Mechanisms and therapeutic insights.

The nervous system functions as a central regulatory network that maintains physiological homeostasis through the coordinated actions of the central and peripheral nervous systems. The emergence of cancer neuroscience has unveiled novel mechanisms underlying bidirectional regulation among the nervous system, tumor cells, and the immune system. This review focuses on pancreatic cancer, often referred to as the "king of cancers," a malignancy characterized by aggressive behavior, early perineural invasion, and dismal prognosis. We systematically analyze the interactions within the neuro-tumor-im

Reduction in calcium responses to whisker stimulation in the primary somatosensory and motor cortices of the model mouse with trigeminal neuropathic pain.

OBJECTIVE: Chronic constriction injury (CCI) of the infraorbital nerve induces neuropathic pain, such as allodynia and hyperalgesia, in the orofacial area. However, the changes in the local circuits of the central nervous system following CCI remain unclear. This study aimed to identify the changes following CCI in Thy1-GCaMP6s transgenic mice. METHODS: Neural activity in the primary somatosensory cortex (S1) and motor cortex (M1) following whisker stimulation was assessed using in vivo Ca2+ imaging. CCI-induced changes in responses were analyzed. RESULTS: Before CCI, whisker stimulation ind

Mesenchymal Stem Cell-Derived Mitochondria Enhance Extracellular Matrix-Derived Grafts for the Repair of Nerve Defect.

Peripheral nerve injuries (PNI) can lead to mitochondrial dysfunction and energy depletion within the affected microenvironment. The objective is to investigate the potential of transplanting mitochondria to reshape the neural regeneration microenvironment. High-purity functional mitochondria with an intact structure are extracted from human umbilical cord-derived mesenchymal stem cells (hUCMSCs) using the Dounce homogenization combined with ultracentrifugation. Results show that when hUCMSC-derived mitochondria (hUCMSC-Mitos) are cocultured with Schwann cells (SCs), they promote the prolifera

Artificial Neural Networks Coupled with MALDI-TOF MS Serum Fingerprinting To Classify and Diagnose Pathological Pain Subtypes in Preclinical Models.

Pathological pain subtypes can be classified as either neuropathic pain, caused by a somatosensory nervous system lesion or disease, or nociplastic pain, which develops without evidence of somatosensory system damage. Since there is no gold standard for the diagnosis of pathological pain subtypes, the proper classification of individual patients is currently an unmet challenge for clinicians. While the determination of specific biomarkers for each condition by current biochemical techniques is a complex task, the use of multimolecular techniques, such as matrix-assisted laser desorption/ioniza
