OCAM: A new member of the neural cell adhesion molecule family related to zone-to-zone projection of olfactory and vomeronasal axons.

Zone-to-zone projection of olfactory and vomeronasal sensory axons underlies the topographic and functional mapping of chemoreceptor expression zones of the sensory epithelia onto zonally arranged glomeruli in the main and accessory olfactory bulbs. Here we identified OCAM (R4B12 antigen), an axonal surface glycoprotein expressed by subsets of both olfactory and vomeronasal axons in a zone-specific manner. OCAM is a novel homophilic adhesion molecule belonging to the immunoglobulin superfamily with striking structural homology to neural cell adhesion molecule. In both the main and accessory ol…

Leigh Syndrome Pathomechanism Involves Region-Specific Innate Immune Activation in Ndufs4 Knockout Mice.

Although recent evidence suggests that the immune system contributes to the pathogenesis of paediatric Leigh syndrome, detailed mechanistic insights are still lacking. Here, we investigated the involvement of immune system activation and inflammation in brain tissue in Leigh syndrome, using hypothesis generating methods. We compared the transcriptomes of olfactory bulb and cerebellum from male Ndufs4−/− (knockout) mice (n = 5–6), a well-established model of paediatric Leigh syndrome. Relative to wildtype animals, knockout mice displayed enrichment of innate immune system pathways in the olfact…

Three consecutive cases of rhinocerebral mucormycosis in calves caused by Lichtheimia ramosa infection on a beef cattle farm.

Three Japanese Black calves died within a two-month period on a beef cattle farm. Necropsy consistently revealed liquefactive necrosis of the ventral brain (extending from the olfactory bulb to the midbrain) and thymic atrophy. Histopathologically, necrosuppurative encephalitis characterized by abundant fungal hyphae was observed in all three cases. Fungal lesions also involved the olfactory mucosa, olfactory nerves, and olfactory bulb. Fungi isolated from the cerebrum of all three calves were identified as Lichtheimia ramosa. These cases were diagnosed as rhinocerebral mucormycosis, originati…

Parallel processing of afferent olfactory sensory information.

KEY POINTS: The functional synaptic connectivity between olfactory receptor neurons and principal cells within the olfactory bulb is not well understood. One view suggests that mitral cells, the primary output neuron of the olfactory bulb, are solely activated by feedforward excitation. Using focal, single glomerular stimulation, we demonstrate that mitral cells receive direct, monosynaptic input from olfactory receptor neurons. Compared to external tufted cells, mitral cells have a prolonged afferent-evoked EPSC, which serves to amplify the synaptic input. The properties of presynaptic glutam…

Neurotrophins are not required for normal embryonic development of olfactory neurons.

Neurons of the vertebrate olfactory epithelium (OE) regenerate continuously throughout life. The capacity of these neurons to regenerate and make new and precise synaptic connections in the olfactory bulb provides a useful model to study factors that may control or mediate neuronal regeneration. Expression and in vitro studies have suggested potential roles for the neurotrophins in the olfactory system. To directly examine whether neurotrophins are required for olfactory neuron development, we characterized in vivo the role of the neurotrophins in the primary olfactory system. For this, we gen…

Glial fibrillary acidic protein and vimentin expression in the frog olfactory system during metamorphosis.

In the present study, we investigated glial cell organization in the olfactory system of adult and tadpole Xenopus laevis using glial fibrillary acidic protein and vimentin antibodies. Our results showed for the first time that glial fibrillary acidic protein was strongly expressed at the level of the olfactory nerve from tadpole to adult and was likely to be expressed by ensheathing glia. In the olfactory bulb, the nerve layer was stained, and no staining was observed in glomeruli. By contrast, vimentin decorated radial glia in the bulb but faintly stained the olfactory nerve. Interestingly, …

[Comparison of the effects of anosmia induced by either peripheral lesion or bulbectomy upon the feeding pattern of the rat (author's transl)].

In order to support the contention that the feeding pattern seen after olfactory bulb removal is due to a sensory loss, the feeding pattern of rats was studied after a peripheral chemical lesion of the olfactory mucosa. A conditioned smell aversion procedure was used to assess the occurence and duration of anosmia after the topical application of zinc sulfate to the olfactory mucosa. It was found that the sensory deficit induced by the peripheral lesion lasted from four to six days. The occurrence of the disrupted feeding pattern in the peripherally lesioned rats coincided in time with the sho…

A novel method using intranasal delivery of EdU demonstrates that accessory olfactory ensheathing cells respond to injury by proliferation.

Olfactory ensheathing cells (OECs) play an important role in the continuous regeneration of the primary olfactory nervous system throughout … n, to visualise OECs, and main/accessory olfactory neurons, respectively. We surgically removed one olfactory bulb including the accessory olfactory bulb to induce degeneration, and found that accessory OECs in the nerve bundles that terminate in the accessory olfactory bulb responded by increased proliferation with a peak occurring 2 days after the injury. Wnt/Frizzled family members mediate olfactory sensory neuron axon extension.

A comprehensive model has yet to emerge, but it seems likely that numerous mechanisms contribute to the specificity of olfactory sensory neuron (OSN) axon innervation of the olfactory bulb. Elsewhere in the nervous system the Wnt/Fz family has been implicated in patterning of anterior-posterior axes, cell type specification, cell proliferation, and axon guidance. Because of our work describing cadherin-catenin family member expression in the primary olfactory pathway, and because mechanisms of Wnt-Fz interactions can depend in part on catenins, we were encouraged to explore Wnt-Fz expression a…

Projections from the posterolateral olfactory amygdala to the ventral striatum: neural basis for reinforcing properties of chemical stimuli.

BACKGROUND: Vertebrates sense chemical stimuli through the olfactory receptor neurons whose axons project to the main olfactory bulb. The main projections of the olfactory bulb are directed to the olfactory cortex and olfactory amygdala (the anterior and posterolateral cortical amygdalae). The posterolateral cortical amygdaloid nucleus mainly projects to other amygdaloid nuclei; other seemingly minor outputs are directed to the ventral striatum, in particular to the olfactory tubercle and the islands of Calleja. RESULTS: Although the olfactory projections have been previously described in the…

Altered odor perception in Dlgap2 mutant mice, a mouse model of autism spectrum disorder.

Olfactory dysfunction has been observed in patients with Autism Spectrum Disorder (ASD). A microdeletion at the 8p23 terminal regions of chromosome 8p23 was identified in a Taiwanese patient with ASD, suggesting a potential association with mutations in the DLGAP2 gene. DLGAP2 is expressed in the olfactory bulb in rodents. The current study investigated olfactory phenotypes of Dlgap2 mutant mice. The results indicated that odor detection capabilities were comparable between wild-type (WT) and Dlgap2 mutant mice. However, homozygous mutant (Homo) mice showed less interest in sniffing odors of b…

Molecular and cellular properties of mammalian primary olfactory axons.

How are the axonal projections of olfactory and vomeronasal receptor neurons to the olfactory bulb formed during development? How are the primary olfactory axonal connections functionally organized? With progress in molecular biological techniques and histochemical methods, it became possible to study cellular strategies and molecular mechanisms which guide the primary olfactory axons of the main and accessory olfactory systems to the target glomeruli in the bulb. In addition, new methodologies have begun to elucidate various subsets of the primary olfactory axons with distinctive central conn…

Odor Enrichment Increases Hippocampal Neuron Numbers in Mouse.

The hippocampus and olfactory bulb incorporate new neurons migrating from neurogenic regions in the brain. Hippocampal atrophy is evident in numerous neurodegenerative disorders, and altered hippocampal neurogenesis is an early pathological event in Alzheimer's disease. We hypothesized that hippocampal neurogenesis is affected by olfactory stimuli through the neural pathway of olfaction-related memory. In this study, we exposed mice to novel pleasant odors for three weeks and then assessed the number of neurons, non-neuronal cells (mainly glia) and proliferating cells in the hippocampus and ol…

Lhx2-dependent specification of olfactory sensory neurons is required for successful integration of olfactory, vomeronasal, and GnRH neurons.

Inactivation of the LIM-homeodomain 2 gene (Lhx2) results in a severe defect in specification of olfactory sensory neurons (OSNs). However, the ramifications of lack of Lhx2-dependent OSN specification for formation of the primary olfactory pathway have not been addressed, since mutant mice die in utero. We have analyzed prenatal and postnatal consequences of conditionally inactivating Lhx2 selectively in OSNs. A cell-autonomous effect is that OSN axons cannot innervate their target, the olfactory bulb. Moreover, the lack of Lhx2 in OSNs causes unpredicted, non-cell-autonomous phenotypes. Firs…
