Multifaceted nature of milk: potential source of parasites and a natural approach to parasite infection control.

Milk, a vital dietary staple across cultures, exhibits a complex interplay of properties that extend beyond its nutritional value. This review explores the multifaceted nature of milk, highlighting its role as a potential carrier of parasitic pathogens, matrix for detecting parasitic infections, and natural methods for controlling parasitic infections. The potential presence of parasites in milk may represent a public health concern depending on factors such as animal species, regional parasite prevalence, hygienic practices, and consumption of raw or unpasteurized milk. Concurrently, the uniq…

High parasite virulence necessary for the maintenance of host outcrossing via parasite-mediated selection.

Biparental sex is widespread in nature, yet costly relative to uniparental reproduction. It is generally unclear why self-fertilizing or asexual lineages do not readily invade outcrossing populations. The Red Queen hypothesis predicts that coevolving parasites can prevent self-fertilizing or asexual lineages from invading outcrossing host populations. However, only highly virulent parasites are predicted to maintain outcrossing, which may limit the general applicability of the Red Queen hypothesis. Here, we tested whether the ability of coevolving parasites to prevent invasion of self-fertiliz…

Host defense or parasite cue: Skin secretions mediate interactions between amphibians and their parasites.

Amphibian skin secretions (substances produced by the amphibian plus microbiota) plausibly act as a first line of defense against parasite/pathogen attack, but may also provide chemical cues for pathogens. To clarify the role of skin secretions in host-parasite interactions, we conducted experiments using cane toads (Rhinella marina) and their lungworms (Rhabdias pseudosphaerocephala) from the range-core and invasion-front of the introduced anurans' range in Australia. Depending on the geographical area, toad skin secretions can reduce the longevity and infection success of parasite larvae, or

Host life history and host-parasite syntopy predict behavioural resistance and tolerance of parasites.

There is growing interest in the role that life-history traits of hosts, such as their 'pace-of-life', play in the evolution of resistance and tolerance to parasites. Theory suggests that, relative to host species that have high syntopy (local spatial and temporal overlap) with parasites, host species with low syntopy should have lower selection pressures for more constitutive (always present) and costly defences, such as tolerance, and greater reliance on more inducible and cheaper defences, such as behaviour. Consequently, we postulated that the degree of host-parasite syntopy, which is nega…

Parasite control practices and public perception of parasitic diseases: A survey of dog and cat owners.

Drugs used in the control of internal and external parasites in companion animals play a crucial role in Animal and Public Health. To ensure continuing protection, these drugs should be administered regularly and in intervals, as suggested by the manufacturers. To assess parasite control practices and other related factors, including the degree of public awareness on the topic, 312 dog and cat owners were surveyed while attending the Small Animal Hospital, Faculty of Veterinary Medicine, Lisbon University. Results showed that 89.7% of the dogs were currently being treated with endoparasitic dr…

Visceral leishmaniasis in eastern Sudan: parasite identification in humans and dogs; host-parasite relationships.

In 1996, an epidemic outbreak of visceral leishmaniasis (VL) started in Barbar el Fugara, a village in Gedarif State (eastern Sudan). From 1997 to 2000, regular epidemiological studies were carried out in the human population, as well as in mammals and sand flies. In symptomatic patients, 46/69 lymph node, 6/20 post kala-azar dermal leishmaniasis (PKDL) and 1/4 cutaneous cultures in NNN medium were positive. In 69 dogs, 23/79 lymph node cultures were positive. In other mammals (47 rodents, five donkeys, one mongoose and one monkey) spleen and/or blood cultures were negative. Characterization o

Anthelmintic Resistance in Companion Animal Parasites: Diagnostic and Treatment Challenges.

Anthelmintics are central to parasite control in companion animals, but drug targets in helminths remain limited. Anthelmintic resistance is a clinically significant threat, with emerging multiple anthelmintic drug-resistant Ancylostoma caninum, macrocyclic lactone-resistant Dirofilaria immitis and praziquantel-resistant Dipylidium caninum. Each treatment is a selection event which eliminates susceptible parasites and enriches resistant survivors. Drug-resistant helminths often go undetected due to limited diagnostics and sole reliance on treatment. While some molecular tools can detect gene p…

Validation of Random Transgene Integration and Expression in Eimeria Parasites.

Validation of transgenic Eimeria parasites is essential for confirming stable genomic integration and expression of exogenous genes. This protocol describes a comprehensive workflow for verifying transgene insertion and protein expression in Eimeria tenella. The procedure begins with genomic DNA extraction from sporulated oocysts, followed by PCR amplification to preliminarily verify the presence of the target fragment. To further confirm genomic integration, whole-genome resequencing is performed to identify insertion sites and evaluate the stability of the integrated construct in the parasit…

Targeting Infected Host Cell Heme Metabolism to Kill Malaria Parasites.

Background/Objectives: Malaria remains a major global health burden, increasingly complicated by resistance to artemisinin-based therapies. Because artemisinin activation depends on heme and porphyrin chemistry, we sought to exploit host red blood cell (RBC) heme metabolism as a therapeutic vulnerability. This study aims to develop and evaluate a host-directed "bait-and-kill" strategy that selectively sensitizes malaria-infected RBCs to artemisinin. Methods: We integrated quantitative proteomics, erythropoiesis transcriptomic analyses, flow cytometry, and in vitro malaria culture assays to cha

Mechanistic insights into coordinated var transcriptional switching in malaria parasites.

The exceptional virulence of the human malaria parasite, Plasmodium falciparum, is attributed to the adhesive properties of infected red blood cells and the parasite's ability to avoid antibody recognition through antigenic variation. Both properties are derived from the hypervariable surface protein PfEMP1, which is encoded by members of the multi-copy var gene family. Waves of parasitemia during an infection are thought to correspond to var transcriptional switching, enabling parasites to avoid elimination by antibodies targeting previously expressed forms of PfEMP1. The mechanisms underlyin…

Parasites trigger epithelial cell crosstalk to drive gut-brain signalling.

Parasitic infections modulate both immune and sensory responses, but how these systems collaborate to elicit protective behaviours remains incompletely understood. The gut epithelium contains specialized sensory cells that detect pathogens and irritants. These include cholinergic tuft cells, which sense parasites and initiate type 2 immune responses1-3, as well as serotonergic enterochromaffin (EC) cells, which detect irritants and communicate with afferent nerve fibres to transmit nociceptive signals4-6. Here we show that paracrine signalling between these cells constitutes a mechanism for ne…

New experimental insights into the biology of avian malaria parasites.

BACKGROUND: Despite much research on avian malaria, the details of development of Plasmodium species in birds remain insufficiently investigated. This study aimed to gain new experimental information on avian malaria parasites regarding their host specificity, molecular characterization, persistence, and association with Matryoshka RNA viruses (MaRNAV), which are linked to haemosporidian infections. METHODS: Three species of widespread avian Plasmodium-P. circumflexum pTURDUS1, P. vaughani pSYAT05, and P. relictum pCOLL7-were found in naturally infected wild birds of the Muscicapidae, Turdida…

Interactions between bacterial microbiota and protozoan parasites: an intimate relationship.

Interactions between parasites and the gut microbiota play a key role in immune responses and susceptibility to zoonotic diseases; however, many aspects of how protozoan infections alter microbial diversity and how these changes influence parasite pathogenicity and host defense remain poorly understood. This review integrates recenst studies using murine models of the Muroidea superfamily, highlighting patterns in microbial diversity, functional interactions, and potential immunometabolic mechanisms underlying these relationships. Both consistent findings and discrepancies among studies are id…

Hybrid Molecules as Efficient Drugs against Multidrug-Resistant Malaria Parasites.

This review is focused on hybrid molecules defined as chemical entities with two or more structural domains, as antimalarial drug-candidates, over the past 25 years. Due to their different pharmacophores, such hybrids can interact with a single biological target by different and complementary mechanisms; they can also act simultaneously on several targets having complementary biological functions (dual mode of action), and can theoretically reduce the selection of parasite drug-resistance. This review is not an exhaustive report of all hybrid drugs tested on malaria parasites but a selection o…
