A hybrid multimodal machine learning model for Detecting Alzheimer's disease.

Alzheimer's disease (AD) diagnosis utilizing single modality neuroimaging data has limitations. Multimodal fusion of complementary biomarkers may improve diagnostic performance. This study proposes a multimodal machine learning framework integrating magnetic resonance imaging (MRI), positron emission tomography (PET) and cerebrospinal fluid (CSF) assays for enhanced AD characterization. The model incorporates a hybrid algorithm combining enhanced Harris Hawks Optimization (HHO) algorithm referred to as ILHHO, with Kernel Extreme Learning Machine (KELM) classifier for simultaneous feature selec

Integrated Biomarker-Volumetric Profiling Defines Neurodegenerative Subtypes and Predicts Neuroaxonal Injury in Multiple Sclerosis Based on Bayesian and Machine Learning Analyses.

Background: The clinical-radiological paradox in multiple sclerosis (MS) underscores the need for biomarkers that better reflect neurodegenerative pathology. Serum neurofilament light chain (sNfL) is a dynamic marker of neuroaxonal injury, while brain volumetry provides structural assessment of disease impact. However, the precise link between sNfL and regional atrophy patterns, as well as their combined utility for patient stratification and prediction, remains underexplored. Objective: This study aimed to establish a multimodal biomarker framework by integrating sNfL with comprehensive volum

AI-Based ECG Analysis for Early Detection of Occlusion Myocardial Infarction Using Coronary Angiography Results.

Myocardial Infarction remains a high-mortality cardiovascular disease. While ST-Elevation Myocardial Infarction (STEMI) is the traditional intervention standard, many Occlusion Myocardial Infarction (OMI) cases are misclassified as Non-ST Elevation Myocardial Infarction (NSTEMI), leading to delayed treatment and poor outcomes. This study collected clinical and 12-lead ECG data from AMI patients over five years, categorized into OMI and Non Occlusion Myocardial Infarction (NOMI) groups based on angiography and cardiac enzymes. We developed a ResNet-1D deep learning model to identify OMI from El

Multi-Modal Deep Learning-Based Model to Predict Burkitt Lymphoma Recurrence.

Burkitt Lymphoma (BL) is an aggressive B-cell non-Hodgkin lymphoma. Recurrent BL has extremely poor prognosis, and does not have a standard treatment option. Although the disease pathology is well characterized, there is currently a lack of research specifically on predictive modeling of BL recurrence. We developed a deep learning (DL) model, BLIMP (Burkitt Lymphoma multI-Modal recurrence Predictor) to predict BL recurrence by combining clinical, gene expression, and mutation data, utilizing a cohort of 184 patients from a publicly available dataset. Our approach achieved an AUC of 0.788 on a

Convolutional neural networks can detect orthostatic hypotension in Parkinson's disease using resting-state functional near-infrared spectroscopy data.

Neurological disorders such as Parkinson's disease (PD) often adversely affect the vascular system, leading to alterations in blood flow patterns. Functional near-infrared spectroscopy (fNIRS) is used to monitor hemodynamic changes via signal measurement. This study investigated the potential of using resting-state fNIRS data through a convolutional neural network (CNN) to evaluate PD with orthostatic hypotension. The CNN demonstrated significant efficacy in analyzing fNIRS data, and it outperformed the other machine learning methods. The results indicate that judicious input data selection ca

A Variance-Based Sensitivity Analysis Approach for Identifying Interactive Exposures.

Chemical mixtures can significantly affect human health, but understanding the interactions among various chemical exposures and identifying influential ones in relation to some health outcomes are difficult. Bayesian kernel machine regression (BKMR) is a widely used model for capturing nonlinear dynamics and interactions between multiple exposures and health outcomes. However, tools for quantifying the interactions captured by this flexible model are scarce. Utilizing the inherent connection between BKMR and Gaussian process regressions, we adopt the classic variance-based sensitivity analysi

Infrared micro-spectroscopy for cyto-pathological classification of esophageal cells.

We report results from a study utilizing infrared spectral cytopathology (SCP) to detect abnormalities in exfoliated esophageal cells. SCP has been developed over the past decade as an ancillary tool to classical cytopathology. In SCP, the biochemical composition of individual cells is probed by collecting infrared absorption spectra from each individual, unstained cell, and correlating the observed spectral patterns, and the variations therein, against classical diagnostic methods to obtain an objective, machine-based classification of cells. In the past, SCP has been applied to the analysis

Advancements and challenges in bloodstain pattern analysis: Addressing limitations for enhanced forensic reliability.

Bloodstain Pattern Analysis (BPA) is a technique used in forensic investigations to recreate violent crime scenes by examining the distribution of bloodstain patterns to interpret the dynamics of the occurrence, including the impact directions, movement of the participants, and the type of weapon that may have been used. Its interpretiveness is, however, severely limited due to underlying limitations, such as the ambiguity of theories of causation patterns, large classification error rates (e.g., 23–30% in controlled studies), distortions created by the surface, environmental contaminations, a

Unsupervised machine learning reveals prognostic value of dynamic carbohydrate antigen 125 trajectory in gastric cancer.

BACKGROUND: Carbohydrate Antigen 125 (CA125), a tumor-associated glycoprotein, is an established prognostic marker in gastric cancer. However, the trajectory of CA125 levels incorporating dynamic changes over time have not been investigated. METHODS: CA125 levels were collected during the first year of treatment to form individual trajectories and grouped using the unsupervised K-means algorithm. Cox proportional hazards model assessed the association between trajectory groups and overall survival (primary outcome). RESULTS: A total of 1,015 patients were included, covering stages I-IV. 7,347

Integrated network toxicology and multi-omics profiling reveal cell cycle, DNA damage repair, and metabolic alterations in cyclophosphamide-induced premature ovarian insufficiency.

BACKGROUND: Cyclophosphamide (CTX)-induced chemotherapy injury is a leading iatrogenic cause of premature ovarian insufficiency (POI) and a major barrier to fertility preservation. Yet the systems-level mechanisms underlying its reproductive toxicity remain poorly defined. METHODS: An integrated network toxicology framework incorporating machine learning and Mendelian randomization (MR) was applied to identify candidate targets, evaluate genetic associations, and establish a predictive model. Ovarian transcriptomic and metabolomic profiling, together with in vivo validation, were subsequently

Integrative transcriptomic identification of potential common biomarkers between non-obstructive azoospermia and papillary thyroid cancer via multi-algorithmic machine learning and network biology approaches.

BACKGROUND: Emerging evidence suggests potential mechanistic links between non-obstructive azoospermia (NOA) and papillary thyroid carcinoma (PTC) through shared signaling pathways. However, no integrative transcriptomic study has systematically examined common molecular signatures between these disorders. METHODS: We performed transcriptomic analyses using multiple independent public datasets from GEO and TCGA. Differential expression analysis identified shared mRNAs, lncRNAs, and miRNAs between NOA and PTC. Protein-protein interaction networks were constructed followed by hub gene identific

Identification of Autophagy-Related Biomarker and Molecular Subtypes in Alopecia Areata Based on Bioinformatics Analysis, Machine Learning, and Experimental Validation.

Background: Alopecia areata (AA) is a common autoimmune alopecia disease. Evidence suggests that autophagy-related genes (ARGs) may contribute to its pathophysiology. This study aims to explore and identify potential autophagy-related biomarkers and molecular subtypes in AA. Methods: In this study, autophagy-related differential expression genes (ARDEGs) in AA were identified by comparing the differentially expressed genes (DEGs) in the GSE68801 dataset with the ARGs. Then, we applied three different machine learning methods to identify key hub genes and further verified them on independent da

Photonic platform coupled with machine learning algorithms to detect pyrolysis products of crack cocaine in saliva: A proof-of-concept animal study.

The non-invasive detection of crack/cocaine and other bioactive compounds from its pyrolysis in saliva can provide an alternative for drug analysis in forensic toxicology. Therefore, a highly sensitive, fast, reagent-free, and sustainable approach with a non-invasive specimen is relevant in public health. In this animal model study, we evaluated the effects of exposure to smoke crack cocaine on salivary flow, salivary gland weight, and salivary composition using Attenuated total reflection-Fourier transform infrared (ATR-FTIR) spectroscopy. The exposure to crack cocaine was performed in an acr

BACKGROUND: Fibromyalgia (FM) is a commonly encountered disease featuring chronic generalized pain, sleep disorder, and physical fatigue. Ankylosing spondylitis (AS) causes chronic lumbodorsalgia involving the sacroiliac joint, often clinically complicated with FM. Nevertheless, the pathophysiology of FM secondary to AS is still lacking. METHODS: Gene expression data of the whole blood in FM and AS patients were retrieved from the Gene Expression Omnibus (GEO) database. Differentially expressed genes (DEGs) were evaluated employing the "limma" package. Gene Ontology (GO) and the Kyoto Encyclo

Characterizing spatiotemporal spread of infectious diseases using ellipse-shaped transmission hotspots: application to dengue virus outbreaks.

BACKGROUND: Identifying transmission hotspots associated with micro-clustering patterns at the early stages of epidemics is helpful to characterize spatiotemporal spread of infectious diseases. However, standard methods with statistical validation to establish a dynamic warning system for emerging infectious diseases are lacking. We therefore aimed to integrate a geographic information system-based surveillance system and data-driven methods to identify transmission hotspots, thereby assisting decision-makers to implement appropriate policies at the early stages of epidemics. METHODS: We prop

A Novel Radiomics-based Interpretable Model for Bladder Cancer Grade Prediction Using White-Light Cystoscopy Images.

BACKGROUND AND OBJECTIVE: White-light cystoscopy (WLC) is the standard diagnostic modality for bladder cancer, but preoperative grading remains inaccurate. We developed a multichannel radiomics model to predict tumour grade (low-grade [LG] vs high-grade [HG]) from WLC and to identify imaging biomarkers. METHODS: WLC images were retrospectively collected from 423 patients across two centres. A total of 2624 tumour regions were segmented for training, with 584 and 358 regions for internal and external validation, respectively. Radiomic features were extracted from the greyscale and red-green-bl
