Di-2-ethylhexyl Phthalate Reprograms Epithelial-Mesenchymal Transition and Lenvatinib Responsiveness in Hepatoma Cells via SPAG4-Dependent MAPK/ERK Signaling.

Environmental chemical exposure has emerged as an important modulator of cancer progression and therapeutic outcomes. Di-2-ethylhexyl phthalate (DEHP), a ubiquitous environmental plasticizer, has been widely recognized for its endocrine-disrupting effects; however, its impact on intracellular signaling reprogramming and cancer drug responsiveness remains incompletely understood. In this investigation, we aimed to elucidate the toxicological mechanisms by which DEHP alters cellular phenotypes and therapeutic sensitivity in hepatocellular carcinoma (HCC) cells. We demonstrate that DEHP exposure

mTOR-driven integrin β4-enriched extracellular vesicles from lenvatinib-resistant hepatocellular carcinoma fuel lung metastasis via fibroblast-niche formation.

Tumor-derived extracellular vesicles (EVs) play crucial roles in facilitating the colonization and growth of metastatic cancer cells in distant organs. Nevertheless, the precise mechanisms by which EVs contribute to therapy-resistant cancer dissemination remain poorly understood. In the present study, we aim to investigate how EVs derived from lenvatinib-resistant (LR) hepatocellular carcinoma (HCC) drive lung metastasis and identify potential therapeutic targets for both inhibiting metastasis and overcoming lenvatinib resistance. Using LR HCC models and omics analysis, we demonstrated that LR

Lenvatinib enhances the response of biliary tract cancer to anti-HER2 therapy.

Human epidermal growth factor receptor 2 (HER2) is frequently overexpressed or amplified in biliary tract cancer (BTC). Although NCCN clinical practice guidelines recommend HER2-targeted agents as subsequent-line therapy, drug resistance often restricts the clinical benefits of existing regimens. Here, we demonstrate that HER2 inhibitors have heterogeneous effects on the proliferation of BTC cells. Further bioinformatic analysis and functional experimental validation revealed that HER2 inhibitors significantly activated fibroblast growth factor receptor (FGFR) signalling pathway in HER2high BT

Evaluation of the combination lenvatinib and pembrolizumab in endometrial cancer; a real world multi-institutional review of practice patterns, efficacy and tolerability.

OBJECTIVE: KEYNOTE-775 defined lenvatinib/pembrolizumab as the new standard-of-care for patients with proficient mismatch repair (pMMR) recurrent EC. However, the regimen required dose reductions in 66.5 % of participants and the generalizability of these results was uncertain. We conducted an observational study to determine the prescribing patterns, outcomes and side effects in a real-world setting. METHODS: A national multidisciplinary consortium was utilized to study treatment patterns of patients with advanced/recurrent EC treated with lenvatinib/pembrolizumab from 2019 through 2022. Tre

Synthesis and biological evaluation of asiatic acid derivatives as c-Kit inhibitors.

Totally fifteen inhibitors of c-Kit based on asiatic acid (AA) derivatives were designed and synthesized with modification at C-2, C-3, C-23 and C-28 of AA. Their structures were confirmed by HRMS,1H NMR and 13C NMR. In vitro activity assay showed that these compounds can inhibit the proliferation of A549, SGC-7901 and MCF-7 cells, and especially compound II2 showed better inhibitory activity on these tumor cells, similar to the positive control drug Sorafenib. Mechanistically, selected compound may inhibit the proliferation of A549 cells by inhibiting c-Kit protein expression, which may be a

Pharmacokinetic exposure and treatment outcomes of lenvatinib in patients with renal cell carcinoma and differentiated thyroid carcinoma.

PURPOSE: After initial approval of lenvatinib for radioiodine-refractory differentiated thyroid cancer (DTC), it has also shown promising outcomes in among others metastatic renal cell carcinoma (mRCC). Given that trial populations typically do not represent routine clinical care populations, questions arise about how applicable trial outcomes are in clinical practice. This study aims to compare the pharmacokinetics (PK), toxicity patterns, and survival data of lenvatinib in a real-world cohort with DTC and mRCC to those observed in pivotal clinical trials. A positive allosteric modulator of α7 nicotinic receptor reduces levodopa-induced dyskinesias in hemi-parkinsonian mice.

Parkinson's disease is a prevalent neurodegenerative disorder characterized by motor impairments including rigidity, bradykinesia and tremor. L-3,4-dihydroxyphenylalanine (L-DOPA) keeps being the standard treatment for Parkinson's disease. But long-term treatment often leads to L-DOPA induced dyskinesias (LIDs): abnormal involuntary movements (AIMs) that significantly impact patients' quality of life. Drugs acting on nicotinic acetylcholine receptors (nAChRs) have emerged as a potential treatment for managing LIDs, since nicotine, shows promise in alleviating LIDs in animal models. Positive al

Obesity paradox role in the immunosuppressive treatment of hepatocellular carcinoma.

The "obesity paradox" in hepatocellular carcinoma (HCC) suggests patients with obesity may experience better treatment outcomes compared to patients without obesity. Wang et al highlighted this paradox in HCC immunotherapy, demonstrating superior progression-free survival and overall survival in patients with overweight and obesity treated with lenvatinib and camrelizumab, focusing on hepatitis B virus-related HCC. Mechanisms such as better nutritional reserves, leptin-mediated immune modulation, and reduced protein breakdown may explain these outcomes. Obesity's role in anti-programmed cell d

Molecular biology of cholangiocarcinoma and its implications for targeted therapy in patient management.

Cholangiocarcinoma (CCA) remains a devastating malignancy and a significant challenge to treat. The majority of CCA patients are diagnosed at an advanced stage, making the disease incurable in most cases. The advent of high-throughput genetic sequencing has significantly improved our understanding of the molecular biology underpinning cancer. The identification of 'druggable' genetic aberrations and the development of novel targeted therapies against them is opening up new treatment strategies. Currently, 3 targeted therapies are approved for use in CCA; Ivosidenib in patients with IDH1 mutati

Albumin-Binding Lutetium-177-Labeled LLP2A Derivatives as Theranostics for Melanoma.

Very late antigen-4 (VLA-4) is a transmembrane integrin protein that is highly expressed in aggressive forms of metastatic melanoma. A small-molecule peptidomimetic, LLP2A, was found to have a low pM affinity binding to VLA-4. Because LLP2A itself does not inhibit cancer cell proliferation and survival, it is an ideal candidate for the imaging and delivery of therapeutic payloads. An analog of [177Lu]Lu-labeled-LLP2A was previously investigated as a therapeutic agent in melanoma tumor-bearing mice, resulting in only a modest improvement in tumor growth inhibition, likely due to rapid clearance

Efficacy and toxicity of immune checkpoint inhibitors combination therapy for advanced renal cell carcinoma: a systematic review and network meta-analysis.

BACKGROUND: Although immune checkpoint inhibitors (ICIs) show a significant overall survival advantage over standard advanced renal cell carcinoma (aRCC) therapies, tumor response to these agents remains poor. Some studies have shown that combination therapy including an ICI appears to be the best treatment; however, the overall benefit in terms of efficacy and toxicity still needs to be assessed. Thus, we performed a network meta-analysis to evaluate the differences in the efficacy of several combinations that include an ICI to provide a basis for clinical treatment selection. METHODS: We co
