Effect of coffee consumption on the risk of gastric cancer: a systematic review and meta-analysis of prospective cohort studies.

BACKGROUND AND OBJECTIVES: Results from observational epidemiologic studies on the relationship between coffee consumption and gastric cancer are inconsistent and inconclusive. To assess the association between coffee consumption and the risk of gastric cancer, we summarized evidence from prospective cohort studies. METHODS: Relevant studies were retrieved through computer searches (PubMed, EmBase and the Cochrane Library) and a review of references up to December 2014. The quality of the included studies was evaluated by Newcastle-Ottawa quality assessment scale. We used a meta-analytic appr

Association between class III obesity (BMI of 40-59 kg/m2) and mortality: a pooled analysis of 20 prospective studies.

BACKGROUND: The prevalence of class III obesity (body mass index [BMI]≥40 kg/m2) has increased dramatically in several countries and currently affects 6% of adults in the US, with uncertain impact on the risks of illness and death. Using data from a large pooled study, we evaluated the risk of death, overall and due to a wide range of causes, and years of life expectancy lost associated with class III obesity. METHODS AND FINDINGS: In a pooled analysis of 20 prospective studies from the United States, Sweden, and Australia, we estimated sex- and age-adjusted total and cause-specific mortality…

The disappointing quality of published studies on operative techniques for rectovaginal fistulas: a blueprint for a prospective multi-institutional study.

BACKGROUND: Treatment of rectovaginal fistulas is difficult, and many surgical interventions have been developed. The best surgical interven … ble. LIMITATIONS: The major limitation of this review was the limited availability of high-quality prospective studies, making it impossible to perform a meta-analysis. CONCLUSIONS: No conclusion about the best surgical intervention for rectovaginal fistulas could be formulated. More large studies of high quality are needed to find the best treatment for rectovagin …

Linked clinical trials--the development of new clinical learning studies in Parkinson's disease using screening of multiple prospective new treatments.

Finding new therapies for Parkinson's disease (PD) is a slow process. We assembled an international committee of experts to examine drugs potentially suitable for repurposing to modify PD progression. This committee evaluated multiple drugs currently used, or being developed, in other therapeutic areas, as well as considering several natural, non-pharmaceutical compounds. The committee prioritized which of these putative treatments were most suited to move immediately into pilot clinical trials. Aspects considered included known modes of action, safety, blood-brain-barrier penetration, preclin

Body mass index in midlife and late-life as a risk factor for dementia: a meta-analysis of prospective studies.

The relationship between body mass index (BMI) (in midlife and late-life) and dementia was investigated in meta-analyses of 16 articles reporting on 15 prospective studies. Follow-ups ranged from 3.2 to 36.0 years. Meta-analyses were conducted on samples including 25 624 participants evaluated for Alzheimer's disease (AD), 15 435 participants evaluated for vascular dementia (VaD) and 30 470 followed for any type of dementia (Any Dementia). Low BMI in midlife was associated with 1.96 [95% confidence interval (CI): 1.32, 2.92] times the risk of developing AD. The pooled relative risks for AD, Va…

Methods for prospective studies of adverse effects as applied to prostate cancer patients treated with surgery or radiotherapy without hormones.

BACKGROUND: Recently two new methods for prospective studies of adverse effects after treatment have been developed: Proportions of patients regaining 90% of baseline function score (PBS-90) and Generalized Estimating Equation (GEE). We compared these methods to examine changes of sexual, urinary, and bowel functions after robot-assisted prostatectomy (RALP) and conformal external beam radiotherapy (EBRT) in patients without androgen deprivation therapy (ADT). METHODS: The post-treatment functional course was studied prospectively in 254 patients (N = 150 RALP and N = 104 EBRT) with PBS-90 an…

The independent predictive value of peritraumatic dissociation for PTSD symptomatology after type I trauma: a systematic review of prospective studies.

We conducted a systematic review of prospective studies examining the independent predictive value of peritraumatic dissociation (PD) for posttraumatic stress disorder (PTSD) symptomatology following single traumatic events. Insight into the independent predictive value may help to identify victims at risk for PTSD symptomatology. For this purpose a literature search was carried out using the online databases PsycINFO, Medline/Pubmed, and PILOTS. Studies were included if they were published in peer-reviewed journals (before 2007), focused on more or less single traumatic events (Type I trauma)…

Importance of faecal sample collection in prospective cohort studies to evaluate the effect of the gut microbiome on cancer development.

The gut microbiome has been linked to cancer in many cross-sectional studies. These studies compared individuals with and without cancer, but they do not establish causation. Prospective cohort studies, with faecal samples collected before a cancer diagnosis, will be instrumental in assessing the role of the gut microbiome in human cancer risk. We identified 18 prospective cohort studies, each of which collected faecal samples from at least 5000 individuals with expected future cancer linkages, and calculated the projected numbers of colorectal, stomach, pancreatic, and female breast cancers.

The circulating proteome and cancer risk: a systematic literature review and meta-analysis of 26 prospective studies with genetic validation.

BACKGROUND: Proteins have an integral role in cancer aetiology and inform cancer detection, treatment, and prognosis. While published data from prospective proteomic studies identifying early detection cancer risk markers have rapidly increased in the past five years, the landscape of evidence remains unclear. We aim to synthesise the evidence on circulating proteins and cancer risk. METHODS: We conducted a systematic review and meta-analysis. We searched Embase and Medline up to December 2023 with reference-list screening and hand-searching up to August 2025. Prospective cohort studies were

Cognitive Impairment and Incident Atrial Fibrillation in Individuals without Stroke: Evidence from Two Prospective Cohort Studies and Mendelian Randomization Analysis.

AIMS: Emerging evidence suggests that neurocognitive dysfunction may contribute to the pathogenesis of atrial fibrillation (AF). We aimed to investigate whether cognitive impairment precedes incident AF independent of stroke, and to explore the potential causal relationships within a brain-heart axis framework. METHODS AND RESULTS: Included were 18,392 participants from the Kailuan study who completed the Mini-Mental State Examination (MMSE) and 377,248 participants from the UK Biobank who completed domain-specific cognitive tests. All participants were aged ≥ 45 years and were free of stroke

Comparative analysis of Antigliotic Guiding Regenerative Gel (AGRG - LDP-916) for peripheral nerve reconstruction: Evidence from prospective randomized controlled studies.

INTRODUCTION: Peripheral nerve injuries (PNIs) with segmental defects remain a challenge. Autologous nerve grafts (ANG) are the gold standard but are limited by donor-site morbidity and variable outcomes in long-gap repairs. Processed allografts and synthetic conduits provide alternatives but lack biological cues for optimal regeneration. RESEARCH QUESTION: This article presents a structured, author-driven narrative synthesis of published prospective randomized controlled preclinical studies evaluating the Antigliotic Guiding Regenerative Gel (AGRG), also known as LDP-916, a bioactive hydroge

Assessment of the relationship between living alone and suicidal behaviors based on prospective studies: a systematic review and meta-analysis.

Suicide, a global health concern, can be influenced by living arrangements. Hence, the objective of this systematic review was to assess the extent of the association between living alone and suicidal behaviors. We conducted a comprehensive search across eight databases for prospective studies. Hazard ratios (HR) and 95% confidence intervals were pooled using Stata software. Nine large-scale prospective studies with 3,663,205 participants proved eligible. The results of the meta-analysis showed that the pooled prevalence of living alone was 32%. Living alone was associated with suicide attempt…
