Insecticidal dipping fluid emulsions, mixed in vitro in dam water containing suspended clay particles and 1% w/v zinc sulphate, were analysed to determine rates of settling of diazinon, cyhalothrin and cypermethrin. Fifteen minutes after mixing, the concentration of the insecticides 5 cm below the surface had declined by 72.5%, 72.8% and 89.4%, respectively. On remixing, the concentration of insecticide in suspension was close to or greater than the initial concentration. In 2 trials, lice were eradicated from sheep showered with dip wash mixed in cloudy dam water to which 1% w/v of zinc sulph

One hundred and one subjects with head louse infestation were entered into two separate studies, in which a phenothrin aqueous/alcoholic lotion was compared to a carbaryl lotion and a malathion lotion. Fifty subjects were treated with a single application of the phenothrin lotion, 28 with the carbaryl lotion and 23 with the malathion lotion. In the comparative study of the phenothrin and malathion lotions an inspection on the day following treatment showed no live lice remained, but that six of the subjects treated with malathion lotion still had evidence of viable eggs (p less than 0.05). Lambda-cyhalothrin (LCT) is a widely used pyrethroid insecticide frequently detected in environmental and food-associated matrices, yet its effects on host bile acid metabolism remain unclear. Male C57BL/6 mice were orally exposed to LCT for 28 days and analyzed using integrated bile acid metabolomics, hepatic and ileal gene-expression profiling, 16S rRNA sequencing, and shotgun metagenomics. LCT reduced hepatic total bile acids but increased plasma and fecal bile acids, indicating compartment-specific bile acid redistribution. This response was accompanied by hepatic Cyp7a1/Cyp27a1 downregula

"Wasping" is an emerging trend where pyrethroid-based insecticides are intentionally inhaled. Michigan's pesticide surveillance program identified events in Michigan wherein correctional officers and incarcerated persons required medical evaluation at local hospitals after "wasping" exposures. Michigan's pesticide surveillance program reviewed case reports involving acute pesticide exposures. Demographic characteristics, signs and symptoms, and clinical outcomes were evaluated and described for each exposed correctional officer and incarcerated person. Reports identified 11 separate events at

Voltage-gated sodium channels (VGSCs) are primary targets of pyrethroid insecticides, yet aphids possess a unique architecture with gene fission and a noncanonical DENS selectivity filter. Here, the VGSC of Myzus persicae was characterized using heterologous expression, electrophysiology, structural modeling, and molecular docking. Individual channel fragments failed to produce sodium currents, whereas linker concatenation partially restored activity with reduced amplitude, indicating impaired interdomain coupling. Despite the DENS motif, sodium selectivity was retained, indicating that ion di

Deciphering the molecular drivers of insecticide resistance is paramount to extending the effectiveness of malaria vector control tools. Here, we demonstrate that a P450 haplotype spanning a Glu205→Asp (E205D) amino acid point mutation in the CYP6P3 gene drives pyrethroid insecticide resistance in the mosquito malaria vector Anopheles gambiae. Pooled whole-genome DNA sequencing data from A. gambiae mosquitoes detected a major P450-linked locus (CYP6 haplotype) on chromosome 2R adjacent to the locus encoding a sodium channel. In vitro metabolism assays with recombinantly expressed CYP6P3 protei

BACKGROUND: The extent to which insecticide resistance is affecting malaria vector control in community and home is not fully understood. This study assessed the implications of insecticide resistance for entomological efficacy of long-lasting insecticidal nets (LLINs) against wild free-flying field Anopheles gambiae s.l. and Anopheles funestus s.l. in experimental hut trials (EHT) in northeastern Tanzania before and after the evolution of pyrethroid-resistance.
METHODS: Evaluations of LLIN efficacy were conducted according to World Health Organization (WHO) guidelines in ten EHT commissioned

BACKGROUND: Insecticide-treated nets (ITNs) remain the primary vector control method across sub-Saharan Africa to prevent malaria transmission, with 2.2 billion pyrethroid (PY) ITNs distributed since 2004. Widespread PY resistance has contributed to stalling progress in recent years; however, new ITNs containing the pyrrole, chlorfenapyr, or the synergist piperonyl butoxide can provide improved efficacy. With more vector control options but limited resources, malaria programs face complex decisions influenced by local transmission intensity, mosquito species, and dynamic insecticide resistance

In the Americas, visceral leishmaniasis (VL) is caused by infection with Leishmania infantum, which has the domestic dog as its main reservoir in urban areas and is transmitted through the bite of infected female Lutzomyia longipalpis. VL has been expanding geographically in Brazil, and prevention and control actions are essential to reduce the impact on populations affected by these neglected diseases. Since 2021, Brazil has implemented the use of deltamethrin-impregnated collars (DM) for the prevention and control of VL in priority municipalities, based on evidence of the effectiveness of th

Insecticides are valuable and widely used tools for the control of insect pests. However, we know very little about the genes and processes involved in the key steps of the poisoning sequence. Besides, the excessive and indiscriminate use of insecticides is generating mechanisms of resistance in various insect pests. To improve our understanding of insecticide toxicity, we need a powerful model organism to help describe the processes underlying insecticide poisoning and the use of genomic tools to identify and analyze the genetic basis of the toxicological response of the insecticides. In this

Health concerns over mixed pesticide residues highlight the need for rapid detection methods. This study developed a multifunctional point-of-use (POU) sensor that integrates separation, enrichment, and surface-enhanced Raman spectroscopy (SERS) using high-index Au trisoctahedrons (TOHs) and metal-organic frameworks (MOFs) on polyamide 6 (PA-6) film. The Au TOHs with sharp tips and high index facets offer excellent SERS performance, while MOFs facilitate analyte enrichment and separation. The flexible PA-6 film was used as an absorbent for the initial adsorption. Using the Au TOHs@ZIF-67/PA-6,

BACKGROUND: Insecticide-treated nets (ITNs) are washed 20 times as part of experimental hut trials to simulate the loss of active ingredient (AI) occurring over their intended 3-year lifespan and estimate insecticidal durability. The ability of the 20-wash method to predict the end-of-life performance of ITNs has not been empirically validated.
METHODS: We performed an experimental hut trial to compare the efficacy of new ITNs unwashed and washed 20 times to field-aged ITNs withdrawn from households 3 years post-distribution against a pyrethroid-resistant vector population in Covè, Benin. Fou

Insecticide-based vector control, which comprises the use of insecticide-treated bed nets (ITNs) and indoor residual spraying (IRS), is the key method of malaria control in Ethiopia. However, its effectiveness is threatened as malaria vectors become resistant to insecticides. Thus, the aim of this study was to monitor the insecticide susceptibility status of malaria vectors. WHO susceptibility tests were used to detect knock-down and mortality rate in the wild malaria vectors collected in Gondar zuria woreda, Northwest Ethiopia. The WHO diagnostic doses of 0.75% permethrin, 0.05% deltamethrin,

BACKGROUND: India plans to eliminate malaria by 2030, targeting zero local cases by 2027. However, malaria remains prevalent in endemic regions. As a means of addressing this issue, insecticide-treated nets (ITNs) are crucial for prevention. In areas with pyrethroid-resistant vectors, studies indicate that ITNs containing pyrethroids and piperonyl butoxide (PBO) are more effective. However, their physical and chemical durability and bioefficacy in this setting require further evaluation. This study assessed the durability of two pyrethroid-PBO-treated nets and one pyrethroid-only-treated net t

INTRODUCTION: In southern Zambia, malaria transmission is low, with outdoor biting Anopheles mosquitoes playing a significant role in malaria transmission. Locals cook in outdoor, open-walled kitchen shelters, exposing them to these outdoor biting vectors and malaria. Volatile pyrethroid spatial emanators (SE) operate through a mode of action which could provide local protection around these kitchens. In this study, SE devices containing the pyrethroid transfluthrin were deployed to local kitchens, where human landing collection (HLC) was utilized to determine differences in host-landing rates

Aedes aegypti is the primary vector of arboviruses in Senegal, yet this species is not typically targeted by routine vector control programs. Through entomological surveillance, we investigated over a one-year (2022-2023) the risk of arbovirus transmission in Dakar, Senegal, the spatial distribution of insecticide resistance and the underlying resistance mechanisms. Weekly ovitraps were deployed in 15 localities (10 per locality), and monthly adult mosquito collections were conducted in six localities. Arboviruses were detected in adult Ae. aegypti using Real-time quantitative reverse transcri

BACKGROUND: Insecticide resistance among malaria vector species now occurs in 84 malaria-endemic countries and territories worldwide. Novel vector-control interventions, including long-lasting insecticidal nets (LLINs) that incorporate new active ingredients with distinct modes of action, are urgently needed to delay the evolution and spread of resistance and to alleviate reversals in malaria-control gains. We aimed to assess the longitudinal effect of two dual-active-ingredient LLINs on insecticide resistance during a cluster-randomised, controlled trial in Benin.
METHODS: This 3-year, three

BACKGROUND: The emergence of pyrethroid resistance has threatened the elimination of Triatoma infestans from the Gran Chaco ecoregion. We investigated the status and spatial distribution of house infestation with T. infestans and its main determinants in Castelli, a municipality of the Argentine Chaco with record levels of triatomine pyrethroid resistance, persistent infestation over 2005-2014, and limited or no control actions over 2015-2020.
METHODS: We conducted a 2-year longitudinal survey to assess triatomine infestation by timed manual searches in a well-defined rural section of Castell
