Increased body weight in C57BL/6 female mice after exposure to ionizing radiation or 60Hz magnetic fields.

PURPOSE: The purpose of this investigation was to determine whether early treatment with ionizing radiation and/or chronic magnetic field (MF) exposure affected body weight in female mice. MATERIALS AND METHODS: Weanling C57BL/6 female mice were irradiated with four equal weekly cobalt-60 exposures (total cumulative doses: 3.0, 4.0, 5.1Gy) and/or received chronic lifetime exposure to 1.4 mT 60 Hz circularly polarized MF or ambient MF. The body weights of 2280 mice were recorded at 35 age intervals, and analysis of variance was used to compare the mean differences from baseline weights between

Evaluation of chromosomal aberrations, micronuclei, and sister chromatid exchanges in hospital workers chronically exposed to ionizing radiation.

Cytogenetic analysis was performed in peripheral blood lymphocytes from hospital workers chronically exposed to ionizing radiation in comparison to matched non-exposed individuals. The accumulated absorbed doses calculated for the radiation workers ranged from 9.5 to 209.4 mSv. The endpoints used were chromosomal aberrations (CA), micronuclei (MN), and sister chromatid exchanges (SCE). The frequencies of CA/100 cells observed for the exposed group were significantly (P=0.018) higher than in the control group: 3.2 and 2.6, respectively. Similarly, the mean numbers of SCE per cell were statistic

Spontaneous and ionizing radiation induced mutations involve large events when selecting for loss of an autosomal locus.

The mouse P19H22 embryonal carcinoma cell line contains two distinct chromosome 8 homologs, one derived from Mus musculus domesticus (M. domesticus) and the other derived from Mus musculus musculus (M. musculus). It also contains a deletion for the M. musculus aprt allele, which is located on chromosome 8. In this study, cells with spontaneous or induced aprt deficiencies were isolated from P19H22 and examined to determine the nature of the mutational events that had occurred. Ultraviolet radiation (UV), ethyl methanesulfonate (EMS), and two forms of ionizing radiation, 137Cs and 252Cf, were u

Childhood cancer and paternal exposure to ionizing radiation: preliminary findings from the Oxford Survey of Childhood Cancers.

Paternal occupational data already collected as part of the Oxford Survey of Childhood Cancers have been reviewed. Information on occupations during or before the relevant pregnancy was sought for 15,279 children dying from cancer in England, Wales, and Scotland in the period 1953-81, and for an equal number of matched controls. Estimates were made for paternal exposure to human-made external ionizing radiation in the six months before conception of the survey child--as judged from job histories and dates of birth. Assessments were also made for potential exposure to unsealed sources of radion

The ionizing radiation-induced replication protein A phosphorylation response differs between ataxia telangiectasia and normal human cells.

Replication protein A (RPA), the trimeric single-stranded DNA-binding protein complex of eukaryotic cells, is important to DNA replication and repair. Phosphorylation of the p34 subunit of RPA is modulated by the cell cycle, occurring during S and G2 but not during G1. The function of phosphorylated p34 remains unknown. We show that RPA p34 phosphorylation is significantly induced by ionizing radiation. The phosphorylated form, p36, is similar if not identical to the phosphorylated S/G2 form. gamma-Irradiation-induced phosphorylation occurs without new protein synthesis and in cells in G1. Mut

Resistance of the stromal cell in murine long-term bone marrow cultures to damage by ionizing radiation.

The ability of bone marrow stromal cells to survive and function after exposure to ionizing radiation remains controversial. Therefore, we used the murine long-term bone marrow culture system to analyze the effects of single doses of ionizing radiation (9-500 Gy) on the function of a preexisting, nearly confluent stroma that was supportive of hematopoiesis. Hematopoiesis ceased promptly in all the irradiated cultures and did not recover unless fresh marrow cells were inoculated. Radiation doses less than or equal to 100 Gy caused no obvious morphologic change in the cells. Total RNA, total pro

Toxicity and mutagenicity of low dose rates of ionizing radiation from tritiated water in human lymphoblastoid cells.

In order to study the toxic and mutagenic effects of low-dose-rate exposure to ionizing radiation, human lymphoblast cells were grown continuously in tritiated water (3H2O) for up to 8 days. Dose rates ranged from 0.0054 to 0.064 rad/min. Mutation to trifluorothymidine resistance (TK locus) and 6-thioguanine resistance (HGPRT locus) was measured; comparable results were observed at both loci. The mutant fraction as a function of total absorbed dose was independent of dose rate over the range studied. At the lower doses, the dose-response curve was linear, with no indication of a threshold. Ove

Abnormal levels of UV-induced unscheduled DNA synthesis in ataxia telangiectasia cells after exposure to ionizing radiation.

In cultured cells from normal individuals and from patients having ataxia telangiectasia (AT) the rate of unscheduled DNA synthesis (UDS) induced by UV light was investigated by autoradiography. The number of grains in 6 different AT cell strains was similar to that observed in normal cells. Exposure of normal cells to doses of X-rays up to 20 krad had no influence on the rate of UV-induced UDS. In contrast, the UV-induced UDS was significantly modified in AT cells by treatment with X-rays. In AT cell strains that were reported to have reduced levels of gamma-ray-induced repair DNA synthesis (

Effect of ionizing radiation on the human brain: white matter and gray matter T1 in pediatric brain tumor patients treated with conformal radiation therapy.

OBJECTIVE: To test a hypothesis that fractionated radiation therapy (RT) to less than 60 Gy is associated with a dose-related change in the spin-lattice relaxation time (T1) of normal brain tissue, and that such changes are detectable by quantitative MRI (qMRI). METHODS: Each of 21 patients received a qMRI examination before treatment, and at several time points during and after RT. A map of brain T1 was calculated and segmented into white matter and gray matter at each time point. The RT isodose contours were then superimposed upon the T1 map, and changes in brain tissue T1 were analyzed as

Shielding the skin: do African medicinal plants possess the ability to mitigate ionizing radiation-induced skin alterations?

BACKGROUND AND PURPOSE: Therapeutic exposure to ionizing radiation (IR) is fundamental to radiotherapy; however, radiation-induced skin toxicity remains a common and dose-limiting adverse effect. Although synthetic radioprotectors such as amifostine reduce radiation-mediated damage through free radical scavenging and DNA repair enhancement, their clinical application is constrained by significant side effects. Natural products, particularly antioxidant-rich medicinal plants, represent a promising complementary strategy for protecting skin tissue against radiation-induced oxidative and genotoxi

Perceptions and concerns regarding medical exposure to ionizing radiation in women of reproductive age: A systematic review.

INTRODUCTION: Ionizing radiation plays a fundamental role in clinical practice for diagnosis and therapy. However, patients with reproductive potential, including women of reproductive age who may be pregnant or become pregnant in the future, often report limited knowledge and increased anxiety regarding these procedures. Although medical exposures involve inherent risks, when examinations are appropriately justified and optimized, clinical benefits generally outweigh potential radiation-related risks. Nevertheless, misconceptions and risk overestimation may lead to refusal or delay of necessa

Ensembling Unets for Rare Chromosomal Aberration Detection in Metaphase Images, Uncertainty Quantification, and Ionizing Radiation Dose Estimation.

In biological dosimetry a radiation dose is estimated using the average number of chromosomal aberrations per peripheral blood lymphocytes. This analysis is still manually performed on 2D metaphase images depicting the 23 pairs of chromosomes because the false discovery rate of current automated detection systems is too high and variable because of sensitivity to small variations in image quality (chromosome spread, illumination variations …). Therefore, the current systems are only used to assist human experts. Designing more performant automatic and reliable chromosomal aberration detection

Ionizing radiation promotes lung injury by inducing ferroptosis-driven senescence in epithelial cells via NCOA4-mediated ferritinophagy.

Radiation-induced lung injury (RILI) is a dose-limiting factor in thoracic radiotherapy, and effective treatments are currently lacking. Ionizing radiation (IR) induced senescence of lung epithelial cells is considered a central process in the development and progression of RILI. Ferroptosis, a form of regulated cell death characterized by iron dependency, has been implicated in various IR-induced injuries, including RILI. However, whether ferroptosis participates in the pathological process of cellular senescence remains unreported. In this study, we investigated the potential association bet

Effects of low and moderate doses of ionizing radiation on colon carcinogenesis: Experimental models and current evidence.

Millions of people are exposed annually to low and moderate doses of ionizing radiation (LMDR), mostly as a result of medical diagnostic procedures. These individuals may be at increased risk of developing colon cancer (CC), as predicted by the linear no-threshold model used for radioprotection. However, risk estimates derived from epidemiological studies remain subject to considerable uncertainty, necessitating a better mechanistic understanding of how LMDR may contribute to CC development. This review examines available experimental models for CC research and current evidence describing the

Usp11 maintained the survival of marginal zone B cells under ionizing radiation by deubiquitinating DLL1 and JAG2.

Efficacy of radiation therapy is compromised by hematopoietic and immune impairments, with elusive underlying causes. This study aimed to elucidate Usp11's role in radiation-induced injuries and uncover related mechanisms. Utilized ARS mouse model to observe survival rates of Usp11-/- (KO) mice post-TBI (Total Body Irradiation). Assessed lymphocyte and MZ B (Marginal Zone B) cell rates using histological analysis, single-cell sequencing, immunofluorescence (IF), immunohistochemistry (IHC), and flow cytometry (FCM). Conducted Co-IP and ubiquitination experiments for mechanism elucidation. Quant

A human 3D culture-organ-on-chip platform for investigating the tumor microenvironment response to ionizing radiation.

Studying complex human responses, such as radiation-induced effects in the tumor microenvironment (TME), requires advanced in vitro systems. Here, we present the Apparatus to Simulate Tumor and Reproduce Organs in an Interactive and Dynamic System (ASTEROIDS), which integrates three-dimensional cell culture with organ-on-chip technology. To characterize the ASTEROIDS, a lung TME was reproduced by co-culturing cancer spheroids with endothelial and fibroblast cells mimicking vascular and stromal compartments. The ASTEROIDS maintained cell viability, endothelial barrier formation, and spheroid zo
