Chikungunya, Influenza, Nipah, and Semliki Forest Chimeric Viruses with Vesicular Stomatitis Virus: Actions in the Brain.

Recombinant vesicular stomatitis virus (VSV)-based chimeric viruses that include genes from other viruses show promise as vaccines and oncolytic viruses. However, the critical safety concern is the neurotropic nature conveyed by the VSV glycoprotein. VSVs that include the VSV glycoprotein (G) gene, even in most recombinant attenuated strains, can still show substantial adverse or lethal actions in the brain. Here, we test 4 chimeric viruses in the brain, including those in which glycoprotein genes from Nipah, chikungunya (CHIKV), and influenza H5N1 viruses were substituted for the VSV glycopro…

Vaccination with Vesicular Stomatitis Virus-Vectored Chimeric Hemagglutinins Protects Mice against Divergent Influenza Virus Challenge Strains.

UNLABELLED: Seasonal influenza virus infections continue to cause significant disease each year, and there is a constant threat of the emerg … ecting against diverse influenza viruses. Here, we describe such a strategy that utilizes vesicular stomatitis virus (VSV) as a vector for chimeric hemagglutinin (cHA) antigens. This vaccination strategy is effective at generating HA stalk-specific, broadly cross-reactive serum antibodies by both intramuscular and intranasal routes of vaccination. We show th …

Vesicular stomatitis virus with the rabies virus glycoprotein directs retrograde transsynaptic transport among neurons in vivo.

Defining the connections among neurons is critical to our understanding of the structure and function of the nervous system. Recombinant viruses engineered to transmit across synapses provide a powerful approach for the dissection of neuronal circuitry in vivo. We recently demonstrated that recombinant vesicular stomatitis virus (VSV) can be endowed with anterograde or retrograde transsynaptic tracing ability by providing the virus with different glycoproteins. Here we extend the characterization of the transmission and gene expression of recombinant VSV (rVSV) with the rabies virus glycoprote…

Histidine-mediated RNA transfer to GDP for unique mRNA capping by vesicular stomatitis virus RNA polymerase.

The RNA-dependent RNA polymerase L protein of vesicular stomatitis virus, a prototype of nonsegmented negative-strand (NNS) RNA viruses, forms a covalent complex with a 5'-phosphorylated viral mRNA-start sequence (L-pRNA), a putative intermediate in the unconventional mRNA capping reaction catalyzed by the RNA:GDP polyribonucleotidyltransferase (PRNTase) activity. Here, we directly demonstrate that the purified L-pRNA complex transfers pRNA to GDP to produce the capped RNA (Gpp-pRNA), indicating that the complex is a bona fide intermediate in the RNA transfer reaction. To locate the active sit…

Potent vesicular stomatitis virus-based avian influenza vaccines provide long-term sterilizing immunity against heterologous challenge.

The emergence in 1997 and continuance today of a highly lethal H5N1 avian influenza virus (AIV) causing human disease has raised concern about an impending pandemic and the need for a vaccine to prepare for such an occurrence. We previously generated an efficacious vesicular stomatitis virus (VSV)-based AIV vaccine expressing H5 hemagglutinin (HA) from the fifth genomic position of VSV (J. A. Schwartz et al., Virology 366:166-173, 2007). Here we have generated and characterized VSV-based vaccines that express the A/Hong Kong/156/1997 (clade 0) H5 HA from the first position of the VSV genome. T…

Effectiveness of vitamin B12 in treating recurrent aphthous stomatitis: a randomized, double-blind, placebo-controlled trial.

BACKGROUND: The frequency of recurrent aphthous stomatitis (RAS), the most common oral mucosa lesions seen in primary care, is up to 25% in the general population. However, there has been no optimal therapeutic approach. Our objective was to confirm our previous clinical observation of the beneficial treatment of RAS with vitamin B(12). METHODS: A randomized, double-blind, placebo-controlled trial was done using primary care patients. A sublingual a dose of 1000 mcg of vitamin B(12) was used in patients in the intervention group for 6 months. RESULTS: In total, 58 patients suffering from RAS…

A recombinant vesicular stomatitis virus replicon vaccine protects chickens from highly pathogenic avian influenza virus (H7N1).

Highly pathogenic avian influenza viruses (HPAIV) of subtypes H5 and H7 cause fatal disease in poultry (fowl plague) but also have zoonotic potential. Currently commercially available vaccines often do not provide sufficient protection and do not allow easy discrimination between vaccinated and infected birds. Therefore, vaccination of domestic poultry against H5 and H7 HPAIV is not allowed in many countries, or is only possible after special permission has been provided. We generated a recombinant marker vaccine based on non-transmissible vesicular stomatitis virus (VSV) expressing the HA ant…

Vesicular stomatitis virus-based vaccines protect nonhuman primates against aerosol challenge with Ebola and Marburg viruses.

Considerable progress has been made over the last decade in developing candidate preventive vaccines that can protect nonhuman primates against Ebola and Marburg viruses. A vaccine based on recombinant vesicular stomatitis virus (VSV) seems to be particularly robust as it can also confer protection when administered as a postexposure treatment. While filoviruses are not thought to be transmitted by aerosol in nature the inhalation route is among the most likely portals of entry in the setting of a bioterrorist event. At present, all candidate filoviral vaccines have been evaluated against pare…

[Prophylactic effect of allopurinol mouthwash against stomatitis induced by the chemotherapy (PMUE regimen) for gastrointestinal malignancies].

For prevention of the chemotherapy-induced stomatitis we administered allopurinol mouthwash to 15 out of 38 patients who underwent PMUE regimen. The severity of stomatitis was graded on five scales according to the criteria of Japan Society For Cancer Therapy. The incidence of stomatitis was defined as patients number with stomatitis above Grade 2/all patients number. Six of 23 patients which were not given allopurinol mouthwash (control group) developed stomatitis of Grade 1 (three patients), Grade 3 (two), or Grade 4 (one). Only one patient given allopurinol mouthwash (AMW group), however, s…

Status spongiousus resulting from intracerebral infection of mice with temperature-sensitive mutants of vesicular stomatitis virus.

Mice infected intracerebrally (i.c.) with wild-type (wt) VSV or temperature-sensitive (ts) mutants, ts 11, ts 22, ts 31 and ts 41, were studied for the development of histopathological lesions in the central nervous system (CNS). Mice infected i.c. with wt VSV exhibited histopathological lesions consisting principally of occasional foci of perivascular mononuclear cell infiltration and rare foci of necrosis. All wt VSV infected mice died within 2 days of i.c. inoculation. In contrast, mice infected i.c. with ts 22 and ts 31 developed spongiform lesions limited to the grey matter of the spinal

Franks & Farquhar (2007) and beyond: The influence of guard and subsidiary cell bio-mechanics on stomatal function.

Stomata are the gatekeepers to leaf gas exchange and as such are essential for CO2 uptake for photosynthesis, water loss for evaporative cooling and ultimately plant growth and productivity. Research has long focused on understanding the mechanisms that govern stomatal aperture adjustments, in particular how changes in turgor pressure in GCs are achieved and how these cause the reversible cell shape distortion that leads to pore opening and closing. Here we discuss the immense impact the paper by Franks and Farquar (2007) has had on this research field, both in the past and more recently, by s

Starch metabolism and stomatal regulation enhance drought resilience in barley: an integrated multi-omics analysis.

Understanding the molecular mechanisms of drought tolerance in barley is crucial for food security. This study employed integrated transcriptomic and metabolomic analyses to investigate drought responses in tolerant (EC_S1) and sensitive (Baudin) barley. We identified key differentially expressed genes (DEGs) and metabolites (DEMs), highlighting genotype-specific strategies in stress adaptation. The drought-tolerant EC_S1 exhibited enhanced activation of pathways related to plant hormone signaling, antioxidant metabolism (e.g., glutathione), and energy mobilization. Functional characterization

Decoupling of stomatal conductance, transpiration and photosynthesis in terrestrial plants under elevated temperature: a meta-analysis.

Elevated temperatures pose challenges to stomatal conductance, which regulates transpiration and photosynthesis. However, the coupling of stomatal conductance, transpiration and photosynthesis may shift with warming. Here, we synthesize evidence from a meta-analysis of 207 studies to assess leaf physiological responses to warming. On average, the responses of stomatal conductance are highly variable, exhibiting no consistent directional trend, whereas transpiration increases and photosynthesis decreases, reflecting a shift towards transpirational cooling. Stomatal conductance declines until te

Dynamic phosphorylation of RopGEF2 by MPK3/6 kinases fine-tunes diurnal stomatal movements in Arabidopsis.

Light triggers diurnal dynamics in stomatal apertures. To balance carbon dioxide uptake for photosynthesis against water loss through transpiration, plants employ mechanisms to prevent excessive stomatal opening. However, the dynamic regulatory mechanism governing stomatal apertures in diurnal stomatal movements remains poorly understood. Here, we report that ROP (RHO OF PLANTS) GUANINE NUCLEOTIDE EXCHANGE FACTOR 2 (RopGEF2), a negative regulator of light-induced stomatal opening, exhibits dynamic phosphorylation in Arabidopsis (Arabidopsis thaliana) guard cells along with changes in stomatal

Concurrent Temperature and Light Intensity Fluctuations Promote Stomatal Opening and Non-Steady State Photosynthesis.

In nature, fluctuations in light intensity (FL) are tightly coupled to rapid changes in leaf temperature (Tleaf), yet the short-term physiological effects of these concurrent drivers remain largely unresolved. Here, we combined rapid infrared induced Tleaf fluctuations with controlled step changes in light intensity for leaf gas exchange measurements to disentangle how each factor, and their interaction, affects stomatal conductance (gs) and net CO2 assimilation (A) in cucumber plants grown under constant light or FL. Modest but rapid increases in Tleaf (~3°C) alone triggered a pronounced wron

PSKR: An LRR-RLK, enhances oxidative stress tolerance by modulating ROS-driven stomatal closure.

Stomatal closure is a key adaptive response that plants employ to mitigate oxidative stress, a process tightly regulated by hydrogen peroxide (H₂O₂) signaling in guard cells. Despite its importance, the molecular mechanisms controlling H₂O₂ homeostasis during stomatal regulation remain poorly understood. In this study, we identify the leucine-rich repeat receptor-like kinase (LRR-RLK) Phytosulfokine Receptor (PSKR) as a positive regulator of stomatal closure under oxidative stress. Overexpression of AtPSKR1 and OsPSKR10 in Arabidopsis conferred enhanced oxidative stress tolerance, evidenced by
