Evaluation of acute and subacute toxicity of aqueous extract of Crassocephalum rubens leaves in rats.

ETHNOPHARMACOLOGICAL RELEVANCE: Crassocephalum rubens is found throughout tropical Africa including the Indian Ocean islands. The leaves are commonly eaten in form of soups and sauces in South-Western Nigeria, also in other humid zones of Africa. Traditionally, it is used as an antidote against any form of poisoning; used to treat stomach and liver complaints; and externally to treat burns, sore eyes, earache, leprosy and breast cancer. In this study, acute and subacute toxicity of aqueous extract of C. rubens leaves was evaluated in rats in order to assess its safety profile. MATERIALS AND M

CYP3A4/P-glycoprotein inhibitors related colchicine toxicity mimicking septic shock.

Colchicine toxicity is uncommon when patients receive a therapeutic dose regularly. However, inadvertent drug interactions can result in unpredicted adverse outcomes. The toxicity of colchicine can manifest in various ways, ranging from mild and non-specific symptoms to severe form known as multiple organ dysfunction syndrome. This case highlights (1) the diagnostic challenge that arises when distinguishing between the severe manifestation of colchicine toxicity and septic shock and (2) concomitant prescription of colchicine with potent CYP3A4 and P-glycoprotein inhibitors (ie, clarithromycin)

Construction of low-toxicity cadmium sulfide/nitrogen-doped muti-walled carbon nanotubes for peroxymonosulfate activation: The crucial role of electron transfer.

Cadmium sulfide is widely employed in environmental catalysis due to its excellent catalytic behaviors. However, the inherent toxicity and leaching risk of CdS-based catalyst presents significant challenges for practical applications. This study explored the incorporation of CdS nanowires on the nitrogen-doped multi-wall carbon tubes (N-MWCNTs) substrate to minimize the leaching rate and mitigate the bio-toxicity by regulating the electron transfer process. The low bio-toxicity of CdS/NMWCNT was confirmed by s series of toxicity tests. Additionally, the catalytic performance could be further e…

DPYD genotyping and dihydropyrimidine dehydrogenase (DPD) phenotyping in clinical oncology. A clinically focused minireview.

BACKGROUND: In clinical oncology, systemic 5-fluorouracil (5-FU) and its oral pro-drugs are used to treat a broad group of solid tumours. Patients with dihydropyrimidine dehydrogenase (DPD) enzyme deficiency are at elevated risk of toxicity if treated with standard doses of 5-FU. DPYD genotyping and measurements of plasma uracil concentration (DPD phenotyping) can be applied as tests for DPD deficiency. In April 2020, the European Medicines Agency recommended pre-treatment DPD testing to reduce the risk of 5-FU-related toxicity. OBJECTIVES: The objective of this study is to present the curren

Predictors of Mortality in Adults With Calcium Channel Blocker Toxicity Receiving Extracorporeal Membrane Oxygenation Support: An Extracorporeal Life Support Organization Registry Analysis.

Calcium channel blocker (CCB) toxicity presents a significant mortality risk, highlighting the need for effective treatment strategies such as extracorporeal membrane oxygenation (ECMO). This study analyzes Extracorporeal Life Support Organization (ELSO) registry data to determine the mortality rate in adults treated with ECMO for CCB toxicity and identifies clinical predictors of mortality. A retrospective analysis of the ELSO registry from January 2016 to April 2023 was conducted, focusing on adults diagnosed with CCB toxicity (ICD-10 T46.1) who received ECMO. Data collected included patient

Toxicology of sorbic acid and sorbates.

Sorbic acid and its salts have been subjected to an extensive battery of tests, including acute, short-term and chronic toxicity/carcinogenicity tests, two-generation reproduction and teratogenicity studies. These studies show that sorbic acid and sorbates have a very low level of mammalian toxicity, even in chronic studies at up to 10% of the diet, and are devoid of carcinogenic activity. They are non-mutagenic and non-clastogenic in vitro and in vivo. The low toxicity is explicable by the fact that sorbic acid is metabolized rapidly by similar pathways to other fatty acids. In humans, a few

Hazard Assessment of Ag Nanoparticles in Soil Invertebrates-Strong Impact on the Longer-Term Exposure of Folsomia candida.

Silver nanomaterials (Ag NMs) are widely used, including in consumer products, and they inevitably enter the environment, with the soil compartment acting as a major sink. However, most available toxicity data focus on the reference Ag NM300K and rely on standard tests, even though long(er)-term exposure tests are recognized as particularly important for assessing the risks to soil invertebrates. Hence, the aim of the present study was to investigate the toxicity of commercial Ag NPs (Ag-Sigma, NPs < 150 nm) to the soil ecotoxicology model Folsomia candida (Collembola). Effects were assessed b

Biocompatibility and sub-chronic toxicity studies of phlorotannin/polycaprolactone coated trachea tube for advancing medical device applications.

The phlorotannin-polycaprolactone-coated endotracheal tube (PP tube) has been developed with the aim of preventing tracheal stenosis that can result from endotracheal intubation, a factor that can lead to a serious airway obstruction. Its preventive efficacy has been assessed through both in vitro and in vivo investigations. However, there is a lack of studies concerning its biocompatibility and sub-chronic toxicity in animal models, a crucial factor to ensure the safety of its usage as a functional endotracheal tube. Thus, this study aimed to evaluate the biocompatibility and sub-chronic (13

Alteration of acute toxicity of inorganic and methyl mercury to Daphnia magna by dietary addition.

Acute toxicity of inorganic mercury [Hg(II)] and methylmercury (MeHg) to Daphnia magna was characterized using a 48-h static, non-renewal ac … l triggering mortality varied widely among treatments. The results suggest that standard short-term toxicity tests (water-only) should be supplemented with extra tests with dietary addition to provide a more environmentally relevant estimation of short-term toxicity of chemical compounds.

Ezrin as a complementary marker in ocular toxicity assessment using a three-dimensional reconstructed human corneal-like epithelium model, EpiOcular™.

INTRODUCTION: A variety of in vitro tests to replace the Draize test have been developed; however, there is no available method for assessing the full spectrum of Globally Harmonized System (GHS) categories. Human cornea-like three-dimensional (3D) reconstructed tissue models are the most promising in vitro systems. The objective of this study was to evaluate the ocular toxicity of 11 test substances using the EpiOcular™ model after performing proficiency tests. We further evaluated the effectiveness of ezrin staining as a complementary marker in histological analysis to overcome the limitatio

Hepatic toxicity of adjuvant chemotherapy for carcinoma of the breast.

Four patients developed abnormal liver function tests and focal defects on liver scan while receiving cyclophosphamide, methotrexate and 5-fluorouracil as adjuvant chemotherapy following mastectomy for breast cancer. Liver biopsies showed severe focal inflammation. The biopsy findings and the subsequent clinical course of the patients strongly suggest that these abnormalities were due to hepatic toxicity of the chemotherapy and not metastic breast cancer. A review of serial liver function tests performed on 24 patients in that chemotherapy program revealed that four out of eight patients with

Zebrafish behavioral assessment as a first-tier whole-organism NAM for developmental neurotoxicity: A multi-laboratory evaluation.

The development of New Approach Methodologies (NAMs) for developmental neurotoxicity (DNT) assessment requires demonstrating reproducibility across testing facilities and clearly defining their role in tiered testing strategies. This multi-laboratory study evaluated the zebrafish Light-Dark Transition Test (LDTT) across four independent laboratories testing 33 compounds (28 suspected DNT toxicants and 5 putative DNT-negatives), including organophosphate and pyrethroid insecticides (deltamethrin, parathion), flame retardants (TDCIPP, TMPP, BPDP), and neonicotinoids (nicotine, acetamiprid) using

Species-aware prediction of fish acute toxicity improves hazard screening and prioritization of environmentally detected pharmaceuticals.

Widespread contamination of surface waters by active pharmaceutical ingredients (APIs) leads to ubiquitous exposure in fish, critical sentinels in aquatic food webs. However, ecotoxicity assessment remains constrained by pronounced interspecies differences in sensitivity and by the scarcity of toxicity data for fish species representative of distinct ecoregions. To support hazard screening of environmentally detected APIs, we developed an ensemble model integrating Deep Neural Networks (DNN) with six other machine learning models. Trained on 10,054 acute toxicity data points (4190 groups, 1651

Mapping disease-linked AOPs to inform bioassay selection for chemical hazard assessment.

Modern societies synthesize, distribute, and utilize thousands of chemicals, but their contribution to the global burden of non-communicable diseases remains insufficiently characterized. Improved availability of robust hazard data would enable more systematic assessment, prioritization, and management of chemical-related health risks. New Approach Methodologies (NAM) are promising because they provide high-throughput, mechanistically informative, and animal-free testing that improves current regulatory toxicity testing. However, selecting biologically relevant in-vitro assays capable of predi

Transfer learning for honey bee toxicity prediction: MolFormer versus classical QSAR representations.

Honey bee (Apis mellifera) toxicity assessment is essential for environmental risk evaluation of agrochemicals, yet experimental data are costly and often limited, constraining the development of robust predictive models. This study investigates whether transfer learning via pretrained chemical language models can provide competitive QSAR performance for bee toxicity classification. Using the ApisTox dataset (1,035 compounds; 296 toxic and 739 non-toxic for bees), molecular representations were derived from SMILES in three ways: (i) PaDEL molecular descriptors, (ii) RDKit Morgan fingerprints,

A robust dynamic 3D liver spheroid platform for (sub)chronic toxicity assessment: Metabolic characterization of HepG2 spheroids as a case study.

In vitro assessment of (sub)chronic chemical exposure requires liver models that maintain stable functionality during long-term cultivation and whose baseline behavior is well characterized. HepG2 cells remain widely used in toxicology due to their robustness and reproducibility; however, their hepatoblastoma origin confers persistent tumor‑associated traits. Although three‑dimensional (3D) HepG2 spheroids provide greater physiological relevance than two‑dimensional (2D) cultures, their baseline behavior during prolonged dynamic cultivation remains insufficiently characterized, limiting confid

Comparative multi-generational bioassay on individual and mixture toxicity of silver and zinc oxide nanoparticles: effects on growth and reproduction in Artemia salina.

This research compared multigenerational toxicity of individual and combined silver and zinc oxide nanoparticles (AgNPs and ZnONPs) on Artemia Salina, focusing on mortality, growth (weight and length), and reproductive indices. Three consecutive generations (F0, F1, and F2) of Artemia were exposed to AgNPs (0.6 mg L-1), ZnONPs (0.25 and 0.5 mg L-1), and their combinations (AgNPs|ZnONPs). Higher mortality was recorded for F0 and F1 compared to F2 in individual ZnONPs treatments (F0 > F1 > F2); similarly, the highest mortality (62.5 %) occurred in F0, while the lowest (25 %) in F2 exposed to AgN
