Tissue invasion by tumor cells induces a host inflammatory response that variably impacts tumorigenesis. This has been well documented for tumor-associated macrophages (TAMs) that could play a pro/M2- or an anti/M1-tumoral function. TAMs frequently infiltrate diffuse large B-cell lymphoma (DLBCL), an aggressive neoplasm arising from germinal center-experienced B cells. However, the pathway leading to the presence of TAMs in DLBCL remains unknown, and their impact is unclear. Here, we show that some DLBCL tumor cells expressed the chemokine CCL5, enabling the differential recruitment of blood m

Identification of neoepitopes as tumor-specific targets remains challenging, especially for cancers with low mutational burden, such as ovarian cancer. To identify mutated human leukocyte antigen (HLA) ligands as potential targets for immunotherapy in ovarian cancer, we combined mass spectrometry analysis of the major histocompatibility complex (MHC) class I peptidomes of ovarian cancer cells with parallel sequencing of whole exome and RNA in a patient with high-grade serous ovarian cancer. Four of six predicted mutated epitopes capable of binding to HLA-A*02:01 induced peptide-specific T cell

The efficacy of cancer vaccines has been limited by the immunosuppressive tumor microenvironment, which can be alleviated by immune checkpoint inhibitor (ICI) therapy. Here, we tested if oncolytic viruses (OVs), similar to ICI, can also synergize with cancer vaccines by modulating the tumor microenvironment. VSV-GP, a chimeric vesicular stomatitis virus (VSV) pseudotyped with the glycoprotein (GP) of the lymphocytic choriomeningitis virus, is a promising new OV candidate. Here, we show that in mouse B16-OVA melanoma, combination treatment of VSV-GP with an ovalbumin (OVA) peptide-loaded dendri…

INTRODUCTION: Canine mast cell tumors (MCTs) are common neoplasms with heterogeneous biological behavior, and histological grading alone may not fully reflect their aggressiveness. The tumor microenvironment, particularly tumor-associated tissue eosinophilia (TATE), may provide complementary information on MCT biology.
METHODS: This study evaluated whether intratumoral eosinophil infiltration, expressed as the eosinophil-to-mast cell ratio (EMR), was associated with histological grade in canine MCTs. A secondary objective was to assess the relationship between peripheral blood inflammatory ma…

The acidic tumor microenvironment is a common feature of tumors, and boric acid-functionalized quantum dots (BA-QDs) exhibit pH-sensitive boron affinity effects and fluorescence emission characteristics. In this study, CdTe QDs were prepared using the water phase synthesis method. Additionally, BA-QDs were prepared by modifying QDs with 4-mercaptophenylboric acid. Hesperetin, baicalein, quercetin, and other model drugs were used, with QDs and BA-QDs serving as carriers, to create a drug-loaded system of QDs with tumor microenvironment-responsive drug release performance. The physical and chemi…

Carotid body tumor (CBT) is a rare neck tumor located at the adventitia of the common carotid artery bifurcation. The prominent pathological features of CBT are high vascularization and abnormal proliferation. However, single-cell transcriptome analysis of the microenvironment composition and molecular complexity in CBT has yet to be performed. In this study, we performed single-cell RNA sequencing (scRNA-seq) analysis on human CBT to define the cells that contribute to hypervascularization and chronic hyperplasia. Unbiased clustering analysis of transcriptional profiles identified 16 distinct

Adipocyte-rich omentum offers "good soil" for disseminating ovarian cancer (OvCa), contributing to therapeutic difficulty. However, little is understood about the association between adipocytes and tumor growth at peritoneal dissemination site. Herein, we report the induction of adipocyte dedifferentiation by OvCa cells and pro-tumorigenic effects of resulted adipocyte-derived fibroblasts. We confirmed that malignant ascites promoted the dedifferentiation of the primary human adipocytes obtained from surgical omental specimen into omental adipocyte-derived fibroblast (O-ADF) that possess both

This study aimed to characterize the tumor microenvironment (TME) in patient-derived xenograft (PDX) models of oral squamous cell carcinoma (OSCC) and compare histological findings with primary tumors of origin (PTT). OSCC samples from five donor patients were implanted into NOD/SCID mice (PDX0) and subsequently re-implanted into new animals (PDX1), yielding three groups for analysis: PTT, PDX0, and PDX1 (n = 5 each). Histological slides with sections were stained with hematoxylin and eosin for grade analysis and subjected to immunohistochemical reactions with antibodies against SMA, CD4, CD8,…

The intricate tumor microenvironment presents formidable obstacles to the efficacy of adoptive T cell therapy in the management of solid tumors by limiting the infiltration and inducing exhaustion of the transferred T cells. Here, we developed a bacterial-based adjuvant approach that augments the efficacy of adoptive T-cell therapy for solid tumor treatment. Our study reveals that intratumor injection of E. coli MG1655 normalizes tumor vasculatures and reprograms tumor-associated macrophages into M1 phenotype that produce abundant CCL5, together facilitating tumor infiltration of adoptively tr…

With the success of immunotherapy in cancer, understanding the tumor immune microenvironment (TIME) has become increasingly important; however in pediatric brain tumors this remains poorly characterized. Accordingly, we developed a clinical immune-oncology gene expression assay and used it to profile a diverse range of 1382 samples with detailed clinical and molecular annotation. In low-grade gliomas we identify distinct patterns of immune activation with prognostic significance in BRAF V600E-mutant tumors. In high-grade gliomas, we observe immune activation and T-cell infiltrates in tumors th

Tumor-associated macrophages (TAMs) are major infiltrating immune cells in liver cancer. They are polarized to anti-tumor M1 type or tumor-supporting M2 type in a dynamic changing state. Tramadol, a synthetic opioid, exhibits tumor-suppressing effect in several cancers, but whether it plays a role in TAMs polarization is uncertain. In the present study, the potential influence of tramadol on TAMs polarization was explored in liver cancer. An orthotopic murine Hepa 1-6 liver cancer model was constructed. The potential function of tramadol was evaluated by cell viability assay, EdU incorporation

Vulvo-vaginal epithelial tumors are uncommon in mares, and data on the epithelial-to-mesenchymal transition (EMT) and the tumor-immune microenvironment (TIME) are still lacking. This is a study investigating the equus caballus papillomavirus type 2 (EcPV2) infection state as well as the EMT process and the tumor microenvironment in vulvo-vaginal preneoplastic/ benign (8/22) or malignant (14/22) epithelial lesions in mares. To do this, histopathological, immunohistochemical, transcriptomic, in situ hybridization, and correlation analyses were carried out. Immunohistochemistry quantification sho…

Pancreatic ductal adenocarcinoma (PDAC) exhibits the highest incidence of perineural invasion among all solid tumors. The intricate interplay between tumors and the nervous system plays an important role in PDAC tumorigenesis, progression, recurrence, and metastasis. Various clinical symptoms of PDAC, including anorexia and cancer pain, have been linked to aberrant neural activity, while the presence of perineural invasion is a significant prognostic indicator. The use of conventional neuroactive drugs and neurosurgical interventions for PDAC patients is on the rise. An in-depth exploration of

INTRODUCTION: Despite recent advances, triple-negative breast cancer (TNBC) patients remain at high risk for recurrence and metastasis, which creates the need for innovative therapeutic approaches to improve patient outcomes. Cryoablation is a promising, less invasive alternative to surgical resection, capable of inducing tumor necrosis via freeze/thaw cycles. Necrotic cell death results in increased inflammatory signals and release of preserved tumor antigens, which have the potential to boost the local and systemic anti-tumor immune response. Thus, compared to surgery, cryoablation enhances

Tumor-associated macrophages (TAMs) are central orchestrators of immune evasion, therapeutic resistance, and clinical outcome across diverse solid tumors. However, the classical M1/M2 paradigm fails to capture the lineage diversity, spatial heterogeneity, and functional plasticity of TAMs within the tumor microenvironment. Unlike previous reviews that mainly summarize macrophage polarization or TAM subtype classification, this review focuses on how emerging single-cell and spatial multi-omics technologies redefine TAM ontogeny, phenotypic diversity, spatial organization, and therapeutic vulner…

Colorectal cancer frequently spreads to the liver, and current treatments often damage healthy liver tissue, while targeting tumors. Diffusing Alpha-emitters Radiation Therapy (Alpha DaRT) disperses alpha-emitting atoms from an intra-tumoral implanted source to a range of a few millimeters in the tumor. Alpha radiation is highly efficient in killing cancer cells, compared to other radiation types, and its short range in the tissue, potentially enables sparing healthy tissue adjacent to the tumor. Alpha DaRT's efficacy was previously demonstrated in subcutaneous tumors from various histotypes.

Background: Oral squamous cell carcinoma (OSCC) remains a clinically challenging malignancy characterized by aggressive behavior and limited therapeutic options. Bitter taste receptors (TAS2Rs), expressed across multiple tissues and cancer types, have recently emerged as regulators of tumor biology and immune responses; however, their functional significance in OSCC remains poorly understood. Methods: Immunohistochemical analysis was performed using surgically resected human tongue OSCC specimens and a tissue microarray (TMA) cohort. In parallel, four TAS2R agonists were evaluated in SCC7 cell
