A commercial formulation of glyphosate inhibits proliferation and differentiation to adipocytes and induces apoptosis in 3T3-L1 fibroblasts.

Glyphosate-based herbicides are extensively used for weed control all over the world. Therefore, it is important to investigate the putative … on. On the other hand, GF treatment severely inhibited the differentiation of 3T3-L1 fibroblasts to adipocytes. According to our results, a glyphosate-based herbicide inhibits proliferation and differentiation in this mammalian cell line and induces apoptosis suggesting GF-mediated cellular damage. Thus, GF is a potential risk factor for human health and the …

Tumor necrosis factor alpha is a negative regulator of resistin gene expression and secretion in 3T3-L1 adipocytes.

Resistin has recently been implicated as an adipocytokine leading to insulin resistance and, therefore, potentially linking obesity and diabetes. To further characterize the regulation of this fat-secreted protein by insulin sensitivity-modulating hormones, 3T3-L1 adipocytes were treated with tumor necrosis factor (TNF) alpha, angiotensin (AT) 2, as well as growth hormone (GH), and resistin gene expression and protein secretion were determined by quantitative real-time reverse transcription-polymerase chain reaction and Western blotting. Interestingly, both, resistin mRNA expression and protei…

Activation of the sodium pump blocks the growth hormone-induced increase in cytosolic free calcium in rat adipocytes.

GH promptly increases cytosolic free calcium ([Ca2+]i) in freshly isolated rat adipocytes. Adipocytes deprived of GH for 3 h or longer are incapable of increasing [Ca2+]i in response to GH, but instead respond in an insulin-like manner. Insulin blocks the GH-induced increase in [Ca2+]i in GH-replete cells and stimulates the sodium pump (i.e. Na+/K+-ATPase), thereby hyperpolarizing the cell membrane. Blockade of the Na+/K+-ATPase with 100 microM ouabain reversed these effects of insulin and enabled GH to increase [Ca2+]i in GH-deprived adipocytes. Both insulin and GH activated the sodium pump i…

Wortmannin and LY294002 inhibit myo-inositol accumulation by cultured bovine aorta endothelial cells and murine 3T3-L1 adipocytes.

We have previously reported that myo-inositol uptake and metabolism is reduced in human fibroblasts derived from patients with ataxia telangiectasia (AT). Treating normal fibroblasts with 10-100 microM wortmannin duplicates some of the phenotypic properties of AT fibroblasts including the decrease in myo-inositol accumulation. In the present study we examined whether treatment of other types of mammalian cells with wortmannin or LY294002 altered myo-inositol uptake. Cultured bovine aorta endothelial cells or 3T3-L1 adipocytes were incubated with either wortmannin or LY294002, and afterwards, m…

Effects of riboflavin deficiency upon age-related changes in beta-adrenergic and adenosine receptor binding in rat adipocytes.

The binding of (-)(3H)dihydroalprenolol (DHA) and (-)-N6-(3H)phenyl-isopropyladenosine (PIA) to beta-adrenergic and adenosine receptors, respectively, was determined in membranes of adipocytes isolated from riboflavin-deficient rats and age-matched, pair-fed controls for feeding periods of three to forty weeks. In normal chow-fed animals, the binding of both ligands to receptors markedly decreased in older compared to that in younger animals. In young, riboflavin-deficient animals, DHA binding diminished compared to that in age-matched controls. No further decrease in binding of DHA occurred i…

Structure of covalent insulin-receptor complexes (I-S-S-R) in isolated rat adipocytes and human placental membranes.

The structure of naturally-formed covalent disulphide-linked complexes between insulin and its receptor was examined by sodium dodecyl sulphate/polyacrylamide-gel electrophoresis. To prevent destabilization of disulphide bonds at alkaline pH the standard discontinuous electrophoresis conditions were changed to a continuous buffer system at pH 7.0. 125I-insulin was first bound to either rat adipocytes or human placental membranes for 10 min at 37 degrees C. After washing, non-dissociable radioactivity was extracted from cells or membranes in Triton X-100 and immunoprecipitated with an antiserum…

The context-dependent roles of PPAR-γ in adipocyte differentiation and obesity: a master regulator with dual functions.

Obesity has become a global public health crisis. It is characterized by pathological proliferation of adipose tissue resulting from an energy metabolism imbalance. Adipocyte differentiation is the core process driving this proliferation and is tightly regulated by a complex molecular network. Peroxisome proliferator-activated receptor gamma (PPAR-γ) is the primary regulator of adipocyte differentiation. It also plays a key regulatory role in lipid and glucose metabolism as well as inflammatory responses. Its dysregulation is closely associated with the onset and progression of obesity. This r

Discovery of circadian clock activators with anti-obesity efficacy via suppression of adipocyte development and hypertrophy.

Circadian clock-driven orchestration of metabolic pathways in adipocytes is required to maintain nutrient homeostasis, and clock disruption predisposes to the development of obesity and insulin resistance. However, pharmacological means to promote beneficial clock actions for metabolic disease interventions remain to be explored. Here, we report the identification and characterization of clock-activating molecules with developmental stage-dependent inhibition of adipocyte development and hypertrophy, leading to robust anti-obesity efficacy in vivo. Both chlorhexidine and a new derivative CM002…

ALKBH5-mediated microglia-neuron crosstalk inhibits sympathetic innervation and adipocyte browning in male mice.

Interactions between nervous system and adipose tissue are involved in the regulation of adipocyte browning. However, whether microglial activation participates in the regulation of cold exposure-induced sympathetic innervation and adipocyte browning is unclear. Here, we demonstrate that RNA m6A demethylase ALKBH5 in the microglia nucleus of hypothalamus (PVH) region inhibits cold exposure-induced adipocyte browning in the subcutaneous white adipose tissue (sWAT) of male mice. The single nuclear-RNA sequencing, brain slice electrophysiological assay and pathological analyses show that microgli

Osteocytes secrete adiponectin and display adipocyte-like phenotype under control of PPARG nuclear receptor.

Osteocytes and adipocytes represent cells with disparate functions. Osteocytes regulate bone metabolism (remodeling) and bone homeostasis, while adipocytes regulate energy metabolism and energy storage. Here, we demonstrate that osteocyte phenotype consists of adipocytic features which are under control of peroxisome proliferator-activated receptor gamma (PPARG), a master regulator of adipocyte differentiation and function. Using a mouse model with osteocyte-specific deletion of PPARG (OTγKO) and osteocyte cellular model of MLO-Y4 cells edited with CRISPR/Cas9 for PPARG deficiency, we are demo…

Calpain inhibition by calpeptin modulates adipocyte lipid metabolism and secretome-mediated inflammatory crosstalk with hepatocytes.

OBJECTIVE: Calpeptin, a calpain inhibitor with potential for treating inflammatory diseases, reduces extracellular vesicle (EV) secretion. While adipose tissue is a recognized target of calpeptin, its effects on lipid metabolism remain unknown. We investigated calpeptin's impact on fatty acid (FA) profiles and metabolic pathways in human Simpson-Golabi-Behmel Syndrome adipocytes and their EVs. METHODS: Adipocytes were treated with 25 or 50 µM calpeptin, and EVs were isolated from conditioned media (CM) by ultracentrifugation. Immortalized human hepatocytes (IHHs) were pre-treated with 0 or 40…

Adipocyte-Derived KIF13B Aggravates Obesity-Associated Metabolic Dysfunction via LRP1/PPARγ Signaling.

Obesity is driven by maladaptive adipose expansion, yet the intracellular machinery linking motor protein-associated intracellular regulation to adipogenic transcription remains incompletely understood. Here, we identify adipocyte-derived kinesin family member 13B (KIF13B) as a regulator of obesity-associated metabolic dysfunction through lipoprotein receptor-related protein 1 (LRP1)/peroxisome proliferator-activated receptor γ (PPARγ) signaling. KIF13B expression is markedly upregulated in adipose tissue from humans and mice with obesity. Adipocyte-specific Kif13b deletion protected mice from

All-Trans Retinoic Acid Attenuates Inflammation and Insulin Resistance Induced by Adipocyte-Macrophage Coculture.

Obesity is characterized by chronic low-grade inflammation, largely driven by macrophage infiltration into adipose tissue, which contributes to the development of insulin resistance. All-trans retinoic acid (ATRA), a biologically active metabolite of vitamin A, has demonstrated anti-inflammatory properties. This study examined the effects of ATRA on inflammation and insulin resistance using a coculture model comprising hypertrophied 3T3-L1 adipocytes and RAW264.7 macrophages. Coculture markedly elevated the production of pro-inflammatory mediators-including nitric oxide, monocyte chemoattracta…

Adipocyte-specific Mlkl knockout mitigates obesity-induced metabolic dysfunction by enhancing mitochondrial functions.

Obesity is a global epidemic characterized by chronic low-grade inflammation and metabolic dysfunction, with adipose tissue playing a pivotal role in these processes. The mixed lineage kinase domain-like pseudokinase (MLKL) is a critical mediator of necroptosis but also exhibits noncanonical roles in metabolic regulation. This study aimed to investigate the adipocyte-specific functions of MLKL in obesity. Using adipocyte-specific Mlkl knockout (MlklAdi-KO) mice, we observed reduced susceptibility to high-fat diet (HFD)-induced obesity, enhanced glucose tolerance, and improved insulin sensitivi

Atgl-Dependent Adipocyte Lipolysis Promotes Lipodystrophy and Restrains Fibrogenic Responses during Skin Fibrosis.

During skin fibrosis, extracellular matrix proteins are overproduced, and resident lipid-filled mature dermal adipocytes are depleted in both human disease and mouse models. However, the mechanisms underlying this reduction in lipid-filled adipocytes during fibrosis are poorly understood. In this study, we found that adipocyte lipolysis through the rate-limiting enzyme Atgl is required for loss of adipose tissue during skin fibrosis in mice. We found that in 2 fibrotic mouse models, adipocyte lipolysis occurred early during skin fibrosis development, and lipid storage was re-established during…

ADH1B, the adipocyte-enriched alcohol dehydrogenase, plays an essential, cell-autonomous role in human adipogenesis.

Alcohol dehydrogenase 1B (ADH1B) is a primate-specific enzyme which, uniquely among the ADH class 1 family, is highly expressed both in adipose tissue and liver. Its expression in adipose tissue is reduced in obesity and increased by insulin stimulation. Interference with ADH1B expression has also been reported to impair adipocyte function. To better understand the role of ADH1B in adipocytes, we used CRISPR/Cas9 to delete ADH1B in human adipose stem cells (ASC). Cells lacking ADH1B failed to differentiate into mature adipocytes manifested by minimal triglyceride accumulation and a marked redu…

Tirzepatide modulates the regulation of adipocyte nutrient metabolism through long-acting activation of the GIP receptor.

Tirzepatide, a glucose-dependent insulinotropic polypeptide/glucagon-like peptide 1 receptor (GIPR/GLP-1R) agonist, has, in clinical trials, demonstrated greater reductions in glucose, body weight, and triglyceride levels compared with selective GLP-1R agonists in people with type 2 diabetes (T2D). However, cellular mechanisms by which GIPR agonism may contribute to these improved efficacy outcomes have not been fully defined. Using human adipocyte and mouse models, we investigated how long-acting GIPR agonists regulate fasted and fed adipocyte functions. In functional assays, GIPR agonism enh
