The cross-reactivity of 22 arbovirus species (alphaviruses; tick- and mosquito-borne flaviviruses; and selected bunyaviruses) was tested with monovalent immune mouse ascitic fluids by indirect immunofluorescence (IIF) in comparison with classical serological reactions (virus neutralization -- VN; haemagglutination inhibition -- HI; and complement fixation -- CF -- reactions). Known relationships within the virus groups studied were confirmed. As to the differentiation limits, the VN test was followed by IIF. Evaluation of the ratio of heterologous to homologous antibody activities showed that …

KLA 16 virus, recovered from a child with Thailand hemorrhagic fever, produces infant mouse, rat, and hamster disease that is characterized by spontaneous bleeding at multiple sites, notably in the gastrointestinal tract, and by marked abnormalities in hemostatic mechanisms. This virus differs in this respect from Chikungunya virus to which it is immunologically similar and from other Thai hemorrhagic fever viruses.

The SARS-CoV-2 pandemic supercharged global efforts towards cutting-edge vaccination strategies for current threats and emerging viruses. It is imperative to develop and investigate next-generation vaccines such as chimeric Virus-Like Particles (chi-VLPs) vaccines for increased immunogenicity, ease of production, and scalability to supplement the worldwide vaccine supply. This study reports a novel bivalent vaccine design of Chimeric Alphavirus-Coronavirus Virus-Like Particles (ChAC-VLPs), displaying fusion glycoproteins of Chikungunya virus (CHIKV) and a Receptor Binding Domain (RBD) of SARS-…

Chikungunya virus (CHIKV) is a re-emerging mosquito-borne alphavirus for which no specific antiviral therapy is currently available. During the large outbreak in Foshan, Guangdong Province, China, in July 2025, CHIKV rapidly spread to neighboring regions and caused more than 16,000 confirmed cases. In this study, the predominant outbreak strain of CHIKV was selected as the reference sequence to establish a panel of complementary biosafe tools for antiviral compound screening and mechanistic investigation. A virus replicon particle (VRP) system for CHIKV was first constructed and applied to com…

Chikungunya virus, a mosquito-borne alphavirus, causes outbreaks of both acute and chronic musculoskeletal diseases. Despite the recent approval of a live-attenuated and virus-like particle-based vaccine, a stable, safe and efficacious vaccine that can be manufactured at low cost is lacking. To address this need, we engineered Escherichia coli to produce robust biopolymer particles (BPs) densely coated with CHIKV envelope glycoproteins E2 and E1, forming a natively folded heterodimer mimicking the virus surface (E2-BP-E1). Native E2-E1 heterodimer formation was confirmed by monoclonal antibodi…

Cervical cancer, driven by the malignant transformation of keratinocytes via the human papillomavirus (HPV) E7 oncoprotein, remains a significant global health challenge. In this study, we developed an alphavirus-based self-amplifying mRNA (saRNA) vaccine encoding either a genetically engineered HPV16 E7 gene or a fusion of granulocyte-macrophage colony-stimulating factor (GM-CSF) with HPV16 E7, both encapsulated in self-assembling liposome-protamine-RNA (LPR) nanoparticles. SaRNA represents a promising vaccine platform due to its intrinsic ability to amplify and prolong antigen expression, of…

Chikungunya virus (CHIKV), an Alphavirus, emerged in the Americas in 2013 and was first documented in Brazil in September 2014, in the states of Pará and Bahia. Although Alagoas state officially reported its first case in late 2015, this study investigated potential earlier unreported cases by analyzing samples from a 2013-2014 Orthoflavivirus serological survey. We screened sera from patients with acute febrile illness, initially suspected of dengue but negative in molecular tests for the genus Orthoflavivirus, using ELISA (IgM/IgG), viral isolation, PCR, and next-generation sequencing. Two s…

Viruses were identified from male anthropophilic mosquitoes from Mato Grosso (MT) State, Midwest Brazil from February 2017 to January 2018. Mosquitoes tested included Aedes (Stegomyia) aegypti (1139 males; 84 pools), Culex quinquefasciatus (9426 males; 179 pools), Culex sp. (3 males; 3 pools) and Psorophora albigenu (1 male; 1 pool) collected from four cities of MT. Pools were subjected to viral RNA extraction followed by RT-PCRs specific for ten flaviviruses, five alphaviruses and Simbu serogroup of orthobunyaviruses. Positive pools were passaged three times in VERO cells (alphavirus and orth…

Eastern Equine Encephalitis virus (EEEV; family Togaviridae, genus Alphavirus) a highly pathogenic mosquito-borne virus is endemic to eastern North America. The ecology of EEEV in Florida differs from that in other parts of the United States; EEEV in the northeastern United States is historically associated with freshwater wetlands. No formal test of habitat associations of EEEV in Florida has been reported. Geographical Information Sciences (GIS) was used in conjunction with sentinel chicken EEEV seroconversion rate data as a means to examine landscape features associated with EEEV transmissi…

Young chickens were inoculated with 5,000 PFU of eastern equine encephalitis (EEE) virus and bled at intervals thereafter for determinations of hemagglutination-inhibiting (HI), neutralizing (N), immunoglobulin M (IgM), and IgG antibodies. HI, N, and IgM antibodies were first detected 4 days after infection, and IgG was detected 7 days after infection. All four antibodies persisted through day 90 after infection. HI, N, and IgM antibody titers remained elevated and were not cross-reactive with the related alphavirus western equine encephalitis (WEE) virus. IgG antibody titers also remained hig…

Mosquito-borne viruses pose a significant global health challenge, particularly in resource-limited settings where multiple viruses often cause illnesses with similar symptoms that require different treatment. We introduce the first 7-plex reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay in a hand-held device capable of detecting the presence of Chikungunya virus (CHIKV), dengue virus serotypes (DENV 1-4), Mayaro virus (MAYV), and Zika virus (ZIKV) in a single test. If the result is positive from the single-plex device for the 7-plex assay, 3-plex and 4-plex devices

BACKGROUND: Dengue and chikungunya are co-circulating vector-borne diseases with substantial overlap in clinical presentations. It is important to differentiate between them during first presentation as their management, especially for dengue hemorrhagic fever (DHF), is different. This study compares their clinical presentation in Singapore adults to derive predictors to assist doctors in diagnostic decision-making.
METHODS: We compared 117 patients with chikungunya infection diagnosed with reverse transcription-polymerase chain reaction (RT-PCR) with 917 dengue RT-PCR-positive adult patients

Animal models, which mimic human disease, are invaluable tools for understanding the mechanisms of disease pathogenesis and development of treatment strategies. In particular, animal models play important roles in the area of infectious arthritis. Alphaviruses, including Ross River virus (RRV), o'nyong-nyong virus, chikungunya virus (CHIKV), mayaro virus, Semliki Forest virus and sindbis virus, are globally distributed and cause transient illness characterized by fever, rash, myalgia, arthralgia and arthritis in humans. Severe forms of the disease result in chronic incapacitating arthralgia an…

This study was undertaken to evaluate the effectiveness of four complementary and combined strategies to minimize the presence of the invasive mosquito Aedes albopictus, firmly established in Sant Cugat del Vallès, Catalonia, Spain. A quasi-experimental design including six neighbourhoods was performed in 2008-2009. The abundance of mosquitoes was monitored through ovitraps. The multiple intervention strategy consisted of four actions: source reduction; larvicide treatments (Bacillus thuringiensis israelensis and diflubenzuron); adulticide treatments (alfacipermetrin); and cleaning up uncontro

Chikungunya fever is a viral disease transmitted to human beings by Aedes genus mosquitoes. From 1972 to 1986 in Kédougou, Senegal, 178 Chikungunya virus strains were isolated from gallery forest mosquitoes, with most of them isolated from Ae. furcifer-taylori (129 strains), Ae. luteocephalus (27 strains), and Ae. dalzieli (12 strains). The characteristics of the sylvatic transmission cycle are a circulation periodicity with silent intervals that last approximately three years. Few epidemics of this disease have been reported in Senegal. The most recent one occurred in 1996 in Kaffrine where t
